        Phylogenetics, Safety and In Vitro Functional Properties of Bacillus Species Isolated from Iru, a Nigerian Fermented Condiment

        Bacillus species were isolated from iru, a traditional fermented condiment in Nigeria. Polyphasic approach was used to evaluate the phylogenetic relationship and strain sub-type of the isolated species. Additionally, the phylogenetic profiles of the species isolated from iru were compared with those of bacilli isolated from different continents. The phylogenetic diversity analysis was performed using the combination of 16S rRNA gene sequencing, ITS-PCR, ITS-PCR-RFLP, and M13 RAPD-PCR. The analysis revealed that Bacillus subtilis U170B and B. subtilis U146A isolated from iru were the closest relatives of strains belonging to the phylogeny of B. subtilis sensu stricto and were related to other bacilli isolated from different continents that had functional benefits. The two isolated species exhibited resistance to acidic pH (pH 2.0). The survival rates of B. subtilis U170B, B. subtilis U146A, and B. clausii UBBC-07 (commercial probiotic strain) cultured at pH 2.0 for 3 h were 33.45, 12.44, and 9.53%, respectively. The strains were highly tolerant to bile salts [0.3% (w/v)]. B. subtilis U170B exhibited the highest cell viability (43.45%) when cultured for 3 h in the presence of bile salts, followed by B. subtilis U146A (25%) and B. clausii UBBC-07 (18.94%). B. subtilis U170B and B. subtilis U146A did not exhibit haemolytic activity and were susceptible to different antibiotics. Additionally, these two strains exhibited weak antagonistic activity against B. cereus. The diverse wild strains of B. subtilis can be used as a safe multifunctional starter culture for the industrial production of condiments with health benefits.

        Association between exercise-induced asthma and parental socio-economic status among school-aged adolescents in a semiurban community in Nigeria

        This study investigated the prevalence and association between exer-cise-induced asthma (EIA) and parental socio-economic status (PSES) among school-aged adolescents in a semiurban community in Nigeria. Three hundred and eighty-five adolescents (185 male and 200 female adolescents) whose ages ranged between 10 and 19 years participated in this cross-sectional study. Participants were recruited from four gov-ernment approved secondary schools in Ido-Ekiti using a multistage sampling technique. Peak expiratory flow rate (PEFR) was assessed at baseline and after 6-min run test (6-MRT) on a level playing ground us-ing a standard peak flow meter. PEFR measurements were repeated at 5th, 10th, 15th, and 20th min post 6-MRT. Participants who had >15.0% PEFR fall were considered to have EIA. PSES was assessed using a validated socio-economic status questionnaire. Descriptive and infer-ential statistics were used to analyze data. Alpha level was set at P<0.05. The mean ages of males and females were 13.9±2.0 and 13.8±1.6 years, respectively. More than half of the participants, 58.2% had EIA (>15.0% PEFR scores) while 53.5% belonged to middle PSES class. There was no significant difference between PEFR scores of male and female participants at baseline and 5th min post 6-MRT. How-ever, male participants had significant higher PEFR than the female counterparts at 10th (t=2.090, P=0.037), 15th (t=2.162, P=0.031), and 20th min (t=2.978, P=0.003). There was significant association between EIA and PSES (χ2=152.4; P=0.001). The prevalence of EIA is very high among school-aged adolescents in Nigeria and was significantly asso-ciated with PSES.

        Effects of polyphenols of Cocos nucifera husk fibre on selected indices of cardiovascular diseases in mice

        Cocos nucifera (C. nucifera) oil is indigenously used to treat cardiovascular diseases. However, coconut husk fibre (which is rich in polyphenols) has not been screened for this property. Based on the ethnomedicinal use of polyphenols in treating cardiovascular diseases, this study was carried out to evaluate the effects of polyphenols of C. nucifera husk fibre on selected cardiovascular disease indices in mice. Fifty adult male Swiss albino mice were assigned randomly into five groups (A-E). Mice in groups B, C, D and E were administered 31.25, 62.5, 125, and 250 mg/kg body weight polyphenols of ethyl acetate extract of C. nucifera husk fibre respectively while the control group (A) mice received 5% DMSO for seven days. The mice were sacrificed twenty four hours after the last administration of polyphenols. Heart and plasma lactate dehydrogenase (LDH), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) activities and plasma lipid profile were determined. Results revealed significant reduction (*p < 0.05) in plasma levels of total cholesterol and LDL-cholesterol with no significant change (*p > 0.05) in HDL-cholesterol, triglyceride and VLDL levels in the plasma at all doses of polyphenols administered compared to controls. There was significant reduction (*p < 0.05) in the activities of heart AST and LDH while plasma ALT, AST, and ALP activities were not significantly altered (*p > 0.05) at all doses of polyphenols administered compared to controls. These results suggest that the polyphenols of C. nucifera husk fibre possess cardio-protective properties and also indicate their possible use in the treatment of cardiovascular diseases.

        On the Performance of Cuckoo Search and Bat Algorithms Based Instance Selection Techniques for SVM Speed Optimization with Application to e-Fraud Detection

        Support Vector Machine (SVM) is a well-known machine learning classification algorithm, which has been widely applied to many data mining problems, with good accuracy. However, SVM classification speed decreases with increase in dataset size. Some applications, like video surveillance and intrusion detection, requires a classifier to be trained very quickly, and on large datasets. Hence, this paper introduces two filter-based instance selection techniques for optimizing SVM training speed. Fast classification is often achieved at the expense of classification accuracy, and some applications, such as phishing and spam email classifiers, are very sensitive to slight drop in classification accuracy. Hence, this paper also introduces two wrapper-based instance selection techniques for improving SVM predictive accuracy and training speed. The wrapper and filter based techniques are inspired by Cuckoo Search Algorithm and Bat Algorithm. The proposed techniques are validated on three popular e-fraud types: credit card fraud, spam email and phishing email. In addition, the proposed techniques are validated on 20 other datasets provided by UCI data repository. Moreover, statistical analysis is performed and experimental results reveals that the filter-based and wrapper-based techniques significantly improved SVM classification speed. Also, results reveal that the wrapper-based techniques improved SVM predictive accuracy in most cases.

        EFFECTS OF POLYPHENOLS OF Cocos Nucifera HUSK FIBRE ON SELECTED KIDNEY FUNCTION INDICES IN MICE

        Decoction of Cocos nucifera husk fibre is used indigenously in Nigeria for malaria treatment. Polyphenols have been identified as the phytochemicals responsible for the antimalarial activity of Cocos nucifera husk fibre, though their toxicity has not been evaluated. The polyphenols of Cocos nucifera husk fibre were therefore evaluated for their effects on selected kidney function indices in mice. Fifty mice were randomly divided into five groups (A-E) of ten mice each. Mice in group A were orally administered 5% DMSO solution while those in groups B, C, D and E were orally administered 31.25, 62.5, 125 and 250 mg/Kg body weight of the polyphenols respectively for seven days. Serum urea, creatinine and uric acid concentrations were determined. Serum levels of sodium, potassium, chloride and calcium ions and kidney alkaline phosphatase (ALP), glutamate dehydrogenase (GDH) and gamma-glutamyltransferase (GGT) activities were also determined. The results showed that the polyphenols significantly reduced (p<0.05) urea concentration at 250 mg/Kg body weight and creatinine concentration at all doses compared to controls. The polyphenols caused no significant alteration (p>0.05) in serum uric acid concentration and kidney ALP, GGT and GDH activities compared to controls. There was significant increase (p<0.05) in serum sodium ion concentration at 31.25, 125 and 250 mg/Kg body weight of polyphenols whereas significant increase (p<0.05) in serum potassium and chloride ions was observed at 62.5 and 250 mg/Kg body weight compared to controls. Thus, polyphenols of Cocos nucifera husk fibre may adversely affect some osmoregulatory functions of the kidney, especially at higher concentrations.

        Antidiabetic potential of Chromolaena Odorata leave extract and its effect on Nrf2/keap1 antioxidant pathway in the liver of diabetic-induced Wistar Rats

        The incidence of liver dysfunction increases in diabetes due to insulin resistance and oxidative stress. Chromolaena odorata is a dominant medicinal plant used in traditional medicine as a remedy for many diseases, particularly diabetes mellitus but the underlying mechanism of the plants chemicals is yet to be documented. This study investigated possible mechanisms behind the folkloric use of C. odorata in the management of diabetes. The effect of this plant on genes linked with glucose metabolism and oxidative stress was investigated in the liver of diabetic rats. The rats were divided into diabetes control, normal control, C. odorata extract (200 mg/kg) and diabetic + C. odorata extract (200 gm/kg). Diabetes was induced in the rats by intraperitoneal injection of a buffered solution of STZ (65 mg/kg). Ethanolic extract of C. odorata leaves (200 mg/kg) was orally administered to the treatment groups for a period of three weeks. The mRNA expression of Glut2, glucokinase, Nrf2 and keap1 were quantified using RT-PCR. Molecular docking studies were further explored to denote compounds from C. odorata that might be responsible for its liver protective effect in diabetes. In the diabetic group, liver mRNA expressions of Glut2, glucokinase and Nrf2 were down-regulated (p < 0.05) while the expression of keap1 was upregulated (p < 0.05), relative to the control group. The treatment of diabetic rats with ethanolic extract of C. odorata leaves (200 mg/kg), however, upregulated the mRNA expression of Glut2, glucokinase and Nrf2 while repressing keap1 mRNA expression. Docking results showed that 5,7-dihydroxy-6–4-dimethoxyflavanone and luteolin as possible compounds from C. odorata potentiating this protective role. These findings suggest that C. odorata could mitigate liver dysfunction induced by diabetes.
