<P><B>Abstract</B></P><P>Scapular winging (SW) is defined as increased prominence of the whole medial border of the scapula. Many researchers recently recommended knee push-up plus (KPP) for enhancing serratus anterior (SA) activation. However, during push-up plus, thoracic kyphosis (TK) may usually occur as a compensatory movement. Thus, the purpose of this study was to investigate the effect of Thera-Band application during KPP on rectus abdominis (RA) activity, TK angle (TKA), SA activity, and amount of SW in subjects with SW.</P><P>Fifteen subjects performed KPP with Thera-Band applied to different posterior body parts (no Thera-Band, in the occiput, and in the thoracic region). Electromyography was used to record the RA and SA activities. Image J software was used to calculate the compensatory TKA during KPP, and a scapulometer was used to measure SW in the quadruped position. One-way repeated-measures analysis of variance was used to test for significance.</P><P>KPP with Thera-Band in the occiput showed significantly lower RA activity (<I>P</I> = .001) and TKA (<I>P</I> < .001) than KPP with no Thera-Band. SA activity (<I>P</I> = .020, <I>P</I> = .047) and SW (<I>P</I> < .001, <I>P</I> < .001) were significantly lower with Thera-Band applied to the occiput and thoracic regions than in KPP with no Thera-Band.</P><P>Thera-Band applied to the occiput and thorax can be beneficial as it decreases RA and SA muscle activity and reduces TKA and SW during KPP in subjects with SW.</P>

<P>Asian ginseng (<I>Panax ginseng</I> C.A. Meyer) is widely used as an Oriental medicine in the East Asian regions, particularly Korea and China. In the study, the strontium isotope ratios (87Sr/86Sr), multielements, and metabolite profiles of 35 ginseng samples collected from Korea and China were examined in an attempt to develop a method to distinguish the origin of ginsengs from the two countries. A multivariate statistical approach was performed to analyze the multielements and the 1H nuclear magnetic resonance (NMR) data. Results of a <I>t</I>-test for Mg, Fe, Al, and Sc showed significant variation between Korean and Chinese ginsengs, indicating potential tracers for discriminating them. Discriminating between the ginsengs from the two countries was generally successful when both the 87Sr/86Sr ratios and rare earth element (REE) contents were used together. Moreover, principal component analysis (PCA) derived from the 1H NMR data revealed a significant separation between the ginsengs originating from the two countries. The major metabolites responsible for differentiation were sugars such as glucose, xylose, and sucrose. The results suggest that this multiplatform approach offers a comprehensive method to distinguish the origin of ginsengs.</P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/jf201696j'>ACS Electronic Supporting Info</A></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/jf201696j'>ACS Electronic Supporting Info</A></P>

<P>Plant phytochromes are photoreceptors that mediate a variety of photomorphogenic responses. There are two spectral photoisomers, the red light-absorbing Pr and far-red light-absorbing Pfr forms, and the photoreversible transformation between the two forms is important for the functioning of phytochromes. In this study, we isolated a Tyr-268-to-Val mutant of Avena sativa phytochrome A (AsYVA) that displayed little photoconversion. Interestingly, transgenic plants of AsYVA showed light-independent phytochrome signaling with a constitutive photomorphogenic (cop) phenotype that is characterized by shortened hypocotyls and open cotyledons in the dark. In addition, the corresponding Tyr-303-to-Val mutant of Arabidopsis (Arabidopsis thaliana) phytochrome B (AtYVB) exhibited nuclear localization and interaction with phytochrome-interacting factor 3 (PIF3) independently of light, conferring a constitutive photomorphogenic development to its transgenic plants, which is comparable to the first constitutively active version of phytochrome B (YHB; Tyr-276-to-His mutant). We also found that chromophore ligation was required for the light-independent interaction of AtYVB with PIF3. Moreover, we demonstrated that AtYVB did not exhibit phytochrome B activity when it was localized in the cytosol by fusion with the nuclear export signal and that AsYVA exhibited the full activity of phytochrome A when localized in the nucleus by fusion with the nuclear localization signal. Furthermore, the corresponding Tyr-269-to-Val mutant of Arabidopsis phytochrome A (AtYVA) exhibited similar cop phenotypes in transgenic plants to AsYVA. Collectively, these results suggest that the conserved Tyr residues in the chromophore-binding pocket play an important role during the Pr-to-Pfr photoconversion of phytochromes, providing new constitutively active alleles of phytochromes by the Tyr-to-Val mutation.</P>

The inhibitory effect of polyphenol extracts (SeapolynolTM, SPN) of the marine brown algae Ecklonia cava and dieckol, a major component of SPN, on hyperlipidemia was investigated in ICR mice fed a high-fat diet (HFD) for five weeks. For analysis of the anti-hyperlipidemic effects of SPN and dieckol, these two agents were given orally on a daily basis to HFD-fed mice for four weeks, starting one week after the beginning of HFD feeding. Groups administered with SPN as well as dieckol showed lower body weight gains than the HFD only group. Administration of SPN and dieckol also resulted in a significant reduction of the level of total cholesterol (TCHO), triglyceride (TG), and low-density lipoprotein (LDL) cholesterol in the serum of HFD-fed mice. In Oil Red O staining using 3T3-L1 preadipocytes, it was shown that both SPN and dieckol markedly inhibited lipid accumulation of 3T3-L1 cells. Furthermore, SPN and dieckol (50 ㎍/mL) significantly inhibited 3-hydroxyl-methyl glutaryl coenzyme A (HMGCoA) reductase activity in vitro. Taken together, these results suggest that polyphenols of Ecklonia cava (SPN) and dieckol reduce body weight gain and fat accumulation in HFD-induced obese mice, and that their hypolipidemic effect is related to the inhibition of adipogenesis of adipocytes and HMGCoA reductase activity.

ABSTRACT: Serratus anterior muscle weakness causes scapular winging, characterized by scapular downward rotation and scapular anterior tipping in the resting position. Knee push-up plus (KPP), a modified push-up plus that is generally used in early rehabilitation programs, is performed to elicit SA activation. However, excessive thoracic kyphosis can easily occur as a compensatory movement during KPP. Therefore, the aim of this study was to examine the effect of tactile cues during KPP on activities of the thoracic erector spinae, rectus abdominis, lower trapezius, serratus anterior, and upper trapezius; scapular winging; and thoracic kyphosis in subjects with scapular winging.Thirteen men with scapular winging performed KPP exercises under 3 different tactile cue conditions (no cue, to the thoracic spinous process [TSP], and to the interscapular region [ISR]). Electromyography was used to collect the muscle activity; in addition, a scapulometer and ImageJ software were used to measure the amount of scapular winging and thoracic kyphosis, respectively. One-way repeated-measures analysis of variance and Bonferroni correction were used to assess for statistical significance. The activity of the thoracic erector spinae, rectus abdominis, serratus anterior, and upper trapezius had significant difference among three conditions (P < .05). Lower trapezius activity was no significantly different among 3 conditions. Degrees of scapular winging and thoracic kyphosis had also significant difference among 3 conditions (P < .05).These results suggest that the tactile cue to the TSP and ISR in KPP exercise can be an effective method for decreasing scapular winging as well as correcting compensatory thoracic kyphosis during the KPP in subjects with scapular winging.

The CRISPR-Cas9 system has significantly advanced regenerativemedicine research by enabling genome editing in stemcells. Due to their desirable properties, mesenchymal stem cells(MSCs) have recently emerged as highly promising therapeuticagents, which properties include differentiation ability and cytokineproduction. While CRISPR-Cas9 technology is applied todevelop MSC-based therapeutics, MSCs exhibit inefficient genomeediting, and susceptibility to plasmid DNA. In this study,we compared and optimized plasmid DNA and RNP approachesfor efficient genome engineering in MSCs. The RNP-mediatedapproach enabled genome editing with high indel frequencyand low cytotoxicity in MSCs. By utilizing Cas9 RNPs, we successfullygenerated B2M-knockout MSCs, which reduced T-celldifferentiation, and improved MSC survival. Furthermore, thisapproach enhanced the immunomodulatory effect of IFN-r priming. These findings indicate that the RNP-mediated engineeringof MSC genomes can achieve high efficiency, and engineeredMSCs offer potential as a promising therapeutic strategy.

Background: Round shoulder posture, results from excessive flexed posture of the thorax, is defined as a position of scapular protraction, anterior tipping, and downward rotation. However, previous studies have focused on only passive position of the thorax during scapular posterior tilting (SPT) and have not reported on SPT combined with correction of flexed posture. Objects: The aim of this study was to compare effects of SPT and SPT with prone trunk extension (SPT + PTE) on activities of the lower trapezius, serratus anterior, and thoracic erector spinae and degree of posture in subjects with round shoulder and flexed posture. Methods: Fifteen subjects with round shoulder and flexed posture were recruited. The caliper was used to measure the degree of round shoulder and flexed posture. Electromyography was performed to collect data of muscle activities. Paired t-test was used to compare two exercise (α=.05). Results: When SPT + PTE was applied, the degree of round shoulder posture (p=.001) and flexed posture (p=.039) significantly decreased compared with that when SPT was applied. The lower trapezius activity significantly increased in the SPT + PTE condition compared with that in the SPT condition (p=.026). There were no significant differences in serratus anterior activity between SPT + PTE and SPT. The thoracic erector spinae activity significantly increased in the SPT + PTE condition compared with that in the SPT condition (p=.014). Conclusion: SPT + PTE might be one of the effective methods to enhance activities of lower trapezius and thoracic erector spinae, and to reduce round shoulder posture and flexed posture in subjects with round shoulder and flexed posture.
