        나군대 잎의 약리 효과에 관한 연구

        Crinum asiaticum var. japonicum is a wild plant growing only in Jeju-island, Korea, and in Japan. The whole part of this plant has been known to have the pharmacological actions such as analgesic, anti-inflammatory, platelet-aggregation inhibitory, antitussive, and expectorant. With these assumed actions, the leaves (Crinum folium) of this plant has been used in the folk remedies for arthritis and arthralgia. There is, however, no scientific evidences for the pharmacological actions of Crinum asiaticum var. japonicum. In the present study, the analgesic, anti-inflammatory, and platelet-aggregation inhibitory actions of Crinium folium were evaluated using writhing test, tail-flick test, carrageenin antiedema test, in vitro thromboxane B₂ quanti tation assay and in vitro platelet aggregation test. In order to obtain the partially purified fraction whose pharmacological action is excellent, the methanol extract of Crinium folium was fractionated consecutively into four biological fractions such as ether, ethyl acetate, butanol, and water fractions and their pharmacological actions of the fractions were investigated. Putting our results together, Crinum folium, especially ethyl acetate fraction was proven to have significant analgesic, anti-inflammatory and platelet-aggregation inhibitory actions by inhibition of prostanoids biosynthesis as one of its mechanism of action.

        가미사물탕 수침 엑기스가 쥐의 생식에 미치는 효과

        This study was carried out to investigate the effect of Kamisamool-Tang water extract on the reproduction in female rats. Three groups, treated for 14 days before mating, were administered with 1.56, 3.12 and 4.68g/㎏/day of water extract respectively. The fourth group was administered with 3. 12g/㎏/day and treated for 10 days of early pregnancy. Autopsy was performed on day 16 of gestation to note the number of pregnant animals, the number of implantation sites, the number of live fetuses, and the number of corpora lutea. The results obtained are as follows: 1) In pregestation study, no significant change of the number of implantation sites and live fetuses were obtained. But a significant decrease (p<0.05) of the number of implantation sites and live fetuses was occurred in early gestation. 2) Considering above results, it is suggested that Kami-Samool-Tang has anti-implantation activities in early gestation, but no ovulatory-suppression activities in pregestation.

        도파민 수송체의 기능적 특성 및 발현에 관한 연구

        Brain dopamine systems play a central role in the control of movement, hormone release, and many complex behavior. The action of dopamine at its synapse is terminated predominately by high affinity reuptake into presynaptic terminals by dopamine transporter(DAT). The dopamine transporter(DAT) is membrane protein localized to dopamine-containing nerve terminals and closely related with cocaine abuse, Parkinsonism, and schizophrenia. In present study, the recombinant plasmid pRc/CMV-DAT, constructed by subcloning of a cDNA encoding a bovine DAT into eukaryotic expression vector pRc/CMV, was stably transfected into CV-1 cells(monkey kidney cell line). The DAT activities in the cell lines selected by Geneticin(R) were determined by measuring the uptake of [3H]-dopamine. The transfected cell lines showed 30-50 fold higher activities than untransfected CV-1 cell line, and this result implies that DAT is well expressed and localized in transfected cells. The transfected cells accumulated [3H]-dopamine in a dose-dependent manner with a Km of 991.6nM. Even though high doses of norepinephrine, epinephrine, serotonin, and choline neurotransmitters inhibited the uptake of [3H]-dopamine, DAT in transfected cell line was proven to be much more specific to dopamine. The psychotropic drugs such as GBR12909, CFT, normifensine, clomipramine, desipramine, and imipramine inhibited significantly the dopamine uptake in tissue culture cells stably transfected with DAT cDNA. Radioactive in situ hybridization was done to map the cellular localization of DAT mRNA-containing cells in the adult rat central nervous system. The strong hybridization signals were detected only in the substantia nigra pars compacta and ventral tegmental area. The restricted anatomical localization of DAT mRNA-containing cells confirms the DAT as a presynaptic marker of dopamine-containing cells in the rat brain

        작약감초탕이 미성숙 흰쥐에서 난포성숙 및 Estrogen생성에 미치는 효과

        The traditional herbal medicine, Jackyakgamcho-tang(JGT), was reported to decrease serum testosterone levels and make pregnancy possibel in anovulatory woman and rat. JGT contains Paeoniae Radix(PR) and Glycyrrhizae Radix(GR) in equal amount. This study was designed to investigate the effect of JGT and its components(PR, GR, paeoniflorin and glycyrrhizin) on uterine and ovarian responses, follicular development, and estrogen secretion in the immature rat. The samples(water extracts of JGT, PR, GR; pure compound of paeoniflorin and glycyrrhizin) were administered orally to rats from the 21th day of age to the 28th or 30th days of age for 7 or 9 days. JGT(400mg/kg) and PR(100mg/kg, 200mg/kg) treatments significantly increased serum estradiol above levels in control rats, but both GR and glycyrrhizin had no effect on this parameter. Gross observation and histological analysis revealed that an increased number of growing follicules was observed in the ovaries of JGT and PR treated rat. However the lutenized follicles and ova present in the oviducts were not observed in all rats except one treated with estrogen as a positive control. These results indicate that JGT stimulates the estrogen production and follicular maturation in the immature rat and PR is the main component to induce such reaction.

        익모초 물추출물이 흰쥐 대퇴동맥의 혈관경련에 미치는 영향

        N/A Objective : In order to study the effect of water extract of Leonuri herba (ELS) on the subarachnoid hemorrhage animal model, the autologous whole blood was applied to the vicinity of the sprague dawley rat right femoral artery. Methods : Following periarterial application of autologous whole blood (PAB) and intraperitoneal injection of ELS (10, 100 and 300 mghg, P.o.), the vasomotor responses to ELS were identified and the histological changes, neovascularized blood vessel were observed. Results : Contractile response to ELS (10, 100 and 300 mg/kg, p.0.) was significantly decreased in the rat femoral arteries of PAB 5 and 7 days. In PAB 5 and 7 days, significantly increased vessel wall thickness and narrowed lumen were observed in comparison with the control group. In the arteries exposed to autologous whole blood for 5 and 7 days, a lot of newly generated capillaries in association with markedly curved blood vessel wall thickness, elastic lamina and irregularly oriented endothelial cells were observed in the periphery of the vessel wall. The number of neovascularized blood vessels were significantly decreased in the periphery of PAB with ELS (10, 100 and 300 mg/kg, p.0.) 5 and 7 days. In VEGF expression, Blood plus ELS (10, 100 and 300 mg/kg, p.0.)-exposed vessel were more weak expression than blood-exposed vessel to vein autologous whole blood for 5 and 7 days. Conclusions : From these results, we concluded that ELS inhibited posthemorrhagic chronic vasospasm and neovascularization in rat femoral artery model.

        새로운 프로톤 펌프 억제제 , IY-81233 의 항위염과 항궤양작용

        This study was designed to determine the effect of newly synthesized antiulcer agent, 5-pyrrolyl-6-halo-2-(pyridyl-2-methylthio)benzimidazole derivatives (IY-81233), on various experimental ulcers and on the secretion of prostaglandin E² (PGE²) into the gastric lumen of rat. IY-81233 was previously reported to have a strong inhibitory effect on H^+/K^+-ATPase and on gastric acid secretion in rats. Oral administration of IY-81233 at concentrations of 0.2, 2.0, and 20 mg/kg inhibited gastric lesions and duodenal ulcer induced by indomethacin, HCl-ethanol, water-immersion stress, cysteamine, and acetic acid in a dose dependent manner. Their IC_(50) values were 3.4, 1.4, 0.8, 1.3, and 1.2 mg/kg, respectively. These results indicate that IY-81233 is a potent antiulcer agent although it is slightly less potent than omeprazole in healing of gastritis and ulcers. The secretion of PGE² into gastric lumen was also investigated in relation to the cytoprotective effect by IY-81233 in rats. The PGE² level was not changed significantly by an oral administration of IY-81233, suggesting that IY-81233 has little effect on the gastric protection. Therefore, it can be concluded that IY-81233 exerts prominent antiulcer activity by suppressing gastric acid secretion via an inhibition of a proton pump and not by protecting the gastrointestinal mucosa against various ulcerative stimuli.

      • 생약 및 생약제제의 규격에 관한 연구(ⅩⅤ) : 모려가루의 품질평가 및 칼슘함량 분석 The evaluation on powdered oyster shell by the monograph in K.P. and the assay of calcium

        모려71루는 Osfreafigas말Itunberg의 껄질로 대한약전, 일본약국방 및 신농본초경의 상품으로 수재되어 았으며,진경안신 (뜰론촌혀), 평갈잠얄(꾸쁠탄), 수렴고삽(찬찮르ie), 연견산결 (3k몰곰) 등의 약리작용을 나타내며, 쬐근에는 칼슘보급제로 널리 사용피고 있다. 따라서 모려가루의 칼슘함쟌을 0.051w-EDTA로 적정하여 측정한 결과 칼슘으로써 11.I~34.6%였고, 개별 중금속 둥의 오염을 파악하고자 As, Cd, Cu, Mg, Mn, Pb, Sb, Sn, Zn둥 금속함량을 ICPS로 분석한 결자 As, Cd, Cu, Pb, Sb, Sn, Zn 은 검출되지 않았고, Mg은 0.es~e.707ppm을, Mn은 0.OO3~0.296ppm을 함유하고 있는 겄으로 나타났다. 또한 모려가루의 품질을 평가하고자 대한약전에 따라 화인시칩 , 순도신험, 건조감량 등을 실시하였으며, 그 결과 순도시험항의 산불용물의 경우 기준치에서 상당히 벗어났으며, 칼슘함량이 갇소할수록 산불용물은 증가하는 경향을 보였으므로 원료의 전처리과정에 대한 규제가 필요하며 , 표준 모려가루의 확보가 시급하며 표준 모려가루애 따라 전반적으로 규격기준이 재설정되어야 할 것으로 사료된다. According to the monograph of powdered Oyster shell in the Korea Pharmacopoeia, we carried out the quality evaluation in 13 samples. All of them were passed on identification except for color change. The impurity test on acid insoluble residues was difficult to determine because of the vague test method. So we established new test method which decreases the sample size and modifies the procedure. Contents( % ) of Calcium in powdered Oyster shell were from 11.1 to 34.6% with 0.05M-EDTA titration. And the levels of trace metals were determinated using inductively coupled plasma spectrometer. The metals of As, Cd, Cu, Pb, Sb, Sn and Zn were not detected but the levels of Mg were from 0.05 to 0.707ppm and Mn were from O.003 to 0.296ppm.

      • 의약품 품질평가에 관한 연구(Ⅰ) : 효소제 의약품의 품질평가(염화리소짐 제제) Evaluation of Enzymatic Drugs(Lysozyme chloride preparations)

        떵화리소잗은 분자량 약 a4.5 kDa의 뮤코다당류 분해효쑈로서 점액분해,출혈억제 및 용글작용이 있어 종합감기약,치질치료제 및 항생제둥과 복합처방되거나 단일제제로서 만성부비동염, 객담 ·객출의 곤란, 수술중 또는 수술후위 출혈에 사용되는 소염효소제이다. 염화리소짐의 정량법으로는 효소-기질반응에 의하여 기질의 투과도변화를 측정하는 시험법으로서 이러한 효소학적 절량법은 숙련된 실험자를 됐요로 하며t 특히 시험자간의 오차가 크고, 시험의 재현성이 떨어지는 문제점이 제기되어 왔다. 따라서 본 연구에서는 효소제 의약풍의 정화한 품질평가에 활용하기 위하여 모세관전기영동분석기(CE)를 이용하껴 염화리소짐 및 그 함유제제에 대한 재현성과 정확성이 높은 분석법을 개발하고자 하였다. 염화리소짐 제제를 가지고 모세관, 전채질액, 검출기 , 전하둥 CE 분석조건과 표준액 및 김액 전처리법, 내부표준물질등을 검토하여 CE의 분석조건을 확립하였고, 검량션작성, 재현성시험, 시판품의 정량, 효소학적 시험볍과 의 상관성 분석등을 실시하였다. 염화리소짐 제제의 CE 분석조건으로 모세관; Silica(fused, Sormx40cm),검출기; 자외부흡왔광도계』측정파장 220nm), Run buffer', 50mD7 인산염 용액(pH 2.5), Run voltage :15kV를 사용하였다. 염화리소짐에 대한 CE분석법은 재현성이 우수하필 복잡한 효소학적 시험결과와 상관성이 높아 염화리소짐제제의 품질평가를 위한 시험법쁘로 꽐응될 수 있겠다. Lrsozyrne chloride is the mucolytic enzyme with anti-inflammatory properties that is used for the treatments of chronic rhinitis and formulated with many antipyretic or antitussive com-ponents in drugs.7he old enzymological identification or determination mrthod of Iysozyme chloridehas been required skillfulness, low reproducibitity and cost consuming in quality controls. In order tosuitable and reproducible analysis of Iysozyme chloride in pharrnaceuticals as well as biological crudecell extracts, a capillary electrophoresis(CE) inetho,4 has been developed. The influences of the electro-lyte pH and composition, instrumental temperature and voltage have been iuvestigated. Therefore Iyso-gyme rhloride was anaayzea by 50mM phosphate buffer (pH B.S) and UV detection (220nm) within lessthan 25 min with good reproducibilitr giving a relative standard deviation of less than 3%. The preci-sion of the method was evaluated from inter and intra-day replicate injection of Iysozyme chloride standard solutions.
