        인제한에 따른 Scenedesmus quadricauda의 광합성 특성 및 질소, 이 함량 변화

        Photosynthetic parameters of Scendesmus quadricauda, such as the maximum photosynthetic rate ($P_{max}$), photosynthetic efficiency (α) and the initial saturation intensity of irradiance for photosynthesis ($I_K$) were obtained using photosynthesis-irradiance (P-I) curve in a phosphorus-limited chemostat. S. quadricauda exhibitied no photoinhibition until at 200 μmol·$m^{-2}$ . $P_{max}$ (r=0.963, P=0.002) and $I_K$(r=0.904, P=0.013) showed linear relationships with growth rate. Chlorophyll-α concentration and cell dry weight decreased at higher growth rates, ut chlorophyll-α content per cell dry weight increased. The increase in photosynthetic rates at higher growth rates was due to the increase of $P_{max}$ and $I_K$ which was caused mainly by the increase in the absolute amount of chlorophyll-α rather than the increased photosynthetic efficiency of individual chlorphyll-α. The α did not show a significant relationship with growth rate (r=0.714, P=0.111). The cell quota of carbon (r=0.554, P=0.254) was not correlated with growth rate, but cell quota of nitrogen (r=0.818, P=0.047) and phosphorus (r=0.855, P=0.030) exhibited linear correlations with growth rate.

        TiO2-SnO2 나노입자로 부터 고굴절 하드코팅 도막의 제조

        TiO2 나노입자의 광촉매 반응을 억제하기 위해 평균 직경 3~5 nm의 TiO2-SnO2 나노입자가 titanium tetraisopropoxide(TTIP)와 tin chloride의 가수분해 반응에 의해 합성되었다. 생성된 TiO2-SnO2 나노입자를 졸-겔법에 의해 3-glycidoxypropyl trimethoxysilane(GPTMS)과 반응시킴에 의해 유-무기 혼성 코팅 용액이 제조되었다. 그 후 코팅 용액을 기재인 polycarbonate(PC) 시트 위에 스핀 코팅시키고, 120 oC에서 열경화 시켜 고굴절률 하드코팅 도막이 제조되었다. TiO2-SnO2 나노입자로부터의 코팅 도막은 TiO2 나노입자로부터 얻어진 코팅 도막의 2H에 비해 증가된 3H의 연필경도를 보였다. 또한 TiO2-SnO2 나노입자로부터의 코팅 도막의 굴절률은 Sn/Ti 몰 비가 0에서 0.5로 증가함에 따라 633 nm 파장에서 1.543으로부터 1.623으로 향상되었다. TiO2-SnO2 nanoparticles with an average diameter of 3~5 nm were synthesized by hydrolysis of titanium tetraisopropoxide (TTIP) and tin chloride to depress the photocatalytic activity of TiO2 nanoparticles. Organic-inorganic hybrid coating solutions were prepared by reacting the TiO2-SnO2 nanoparticles with 3-glycidoxypropyl trimethoxysilane (GPTMS) by the sol-gel method. The hard coating films with high refractive index were obtained by curing thermally at 120 oC after spin-coating the coating solutions on the polycarbonate (PC) sheets. The coating films from TiO2- SnO2 nanoparticles showed an improved pencil hardness of 3H compared to 2H of the coating films from TiO2 nanoparticles. Besides, the refractive index of the coating films from TiO2-SnO2 nanoparticles enhanced from 1.543 to 1.623 at 633 nm as the Sn/Ti molar ratio increased from 0 to 0.5.

        N: P ratio 조절에 의한 미세조류 생장과 경쟁 제어

        Microalgae can grow autotrophically with the supply of light, carbon dioxide and inorganic nutrients in water through photosynthesis. Generally, microalgal growth is limited by the concentrations and relative ratio of nitrogen (N) and phosphorus (P) among the nutrients in the aquatic environment. Each microalga has its specific optimum N : P ratio resulting in dominance in a particular water having similar nutrient composition. Algal bloom is an immense growth of certain microalga commonly cyanobacterium and can be sequestrated by reducing the limiting nutrient, generally P in the freshwater. Moreover, dominance of a less toxic blooming strain can be established by manipulating N : P ratio in the water. On the other hand, microalgal biomass of a certain species can be enhanced by increasing limiting nutrient and adjusting the N : P ratio to the target species. The above-mentioned eco-physiological features of microalgae can be more completely interpreted in connection with their genomic informations. Consequently, microalgal growth regulation which can be achieved on the basis of its eco-physiological and further genomic insights would be helpful not only in the control of algal bloom, but also for an increased yield of algal biomass.

        Screening of Bioactive Materials from Freshwater Microalgae

        One hundred and fifty four micro algal strains, newly isolated from nationwide freshwaters in Korea, were screened for their anticancer, ant diabetic, and antibiotic activities. The micro algal strains were cultured with different nutritional conditions that were divided into 4 groups as follows; a normal Allen medium, nitrogen (N)-limited medium, phosphorus (P)-limited medium, and N and P-limited medium. Algal biomass was extracted with a mixture of acetone:H₂O (1:1, v:v) and the extracts were used for the screening of bioactive materials. Anticancer, ant diabetic, and antibiotic materials were screened by the methods of vaccinia Hl-related protein tyrosine phosphates (VHR DS-PTPase) inhibition, protein tyrosine phosphates 1B (PTP1B) inhibition, and paper disk. The inhibition activity of VHR DS-PTPase was observed in 18 strains, having a maximum 79% inhibition from Anabaena affinis and the inhibition activity of PTP1B was observed in 9 strains, having a maximum 97% from Sphaerocystis schroeteri. Microcystis aeruginosa incubated in an N and P-limited medium showed antibiotic activity in 8 species out of 13 pathogenic bacteria. As a whole, it seemed that the stressed condition such as N and/or P limitation increased the production of bioactive materials in micro algae.

        cpcBA-Intergenic Spacer Region을 이용한 Cyanobacteria의 다양성 분석

        The cyanobacterial diversity was analyzed by restriction fragment length polymorphism (RFLP) of PCR-amplified rpcBA-Intergenic Spacer (IGS) genes and cpcBA-IGS gene sequencing with a sample collected at Chuso-ri in Daechung Reservoir on March 15, 2005, The Shannon-Weiner diversity index was 0.65, indicating that the cyanobacterial community structure was simple. PCR-RFLP profiles obtained were Phormidium spp. (58 clones), Anabaena spp. (14 clones), Microcystis spp. (4 clones), Spirulina sp. (1 clone) and uncultured cyanobacteria (2 clones). The PCR-RFLP of cpcBA-IGS revealed that Phormidium spp. and Anabaena spp. dominated in the invested sample. As a consequence, it seems that the analysis of functional genes such as cpcBA-IGS can be used for the species identification and community analysis of cyanobacteria. 대청호에서 수화 발생이 빈번한 추소리 수역에서 2005년 3월 15일에 채취한 시료를 대상으로 유전자 분식에 의한 cyanobacteria의 다양성을 조사하였다. rpcBA-Intergenic Spacer (IGS)는 cyanobacteria에 특징적 색소인 phycocyanin을 합성하는 유전자와 유전자 사이의 부분으로, 환경시료에서 cyanobacteria의 다양성을 조사하기에 매우 유용한 기능 유전자이다. cpcBA-IGS를 이용하여 restriction fragment length polymorphism (RELP)으로 cyanobacteria의 다양성을 분석한 결과 Phomidium 속은 58 clones, Anabaena 속은 14 clones, Microcyxtis 속은 4 clones, Spirulina 속은 1 clone 그리고 uncultured cyanobacteria 2 clones가 존재하였다. 전반적으로 Phormidium 속이 우점하였으며, 여름철에 수화를 일으키는 Anabaena 속과 Microcystis 속도 많이 분포하였다. 따라서 cyanobacteria는 cpcBA-IGS와 같은 기능 유전자에 의한 종 동정 및 군집분석이 가능함을 보였다.

        DGGE를 이용한 대청호 수화 발생시기의 세균군집 분석

        대청호에서 수화 발생시기인 2003년 7월에서 10월까지 분자생태학적 방법의 하나인 DGGE (denaturing gradient gel electrophoresis)를 이용하여 시간에 따른 세균군집구조의 변화를 연구하였다. 조사기간 동안 출현한 식물플랑크톤을 형태학적으로 분류한 결과 cyanobacteria, 규조류 및 녹조류가 발견되었고, 이 중 Microcystis, Chroococcus, Oscillatoria, Phormidium 속이 크게 우점하였다. 16S rDNA의 DGGE pronto 분석에 의하여 Microcystis flos-aquae와 Oscillatoria spp.가 우점하는 것으로 확인되었으며, Aphanizomenon flos-aquae는 8월 중순에 우점하는 것으로 확인되었다. DGGE profile을 토대로 cluster analysis를 적용하여 다양한 미생물 군집의 유사성을 비교한 결과, 9월 2일의 미생물 군집이 다른 시기의 시료와 확연히 다른 그룹으로 구분되었다. 결과적으로 분자 생물학적 방법은 형태적 분석방법과 유사한 결과를 보였으며, 일부 세균의 분포 및 변화, 미생물 군집의 유사성 등에 대한 추가적 정보를 제공하였다. The change of bacterial communities during cyanobacterial bloom was analyzed by DGGE in Daechung Reservoir from July to October in 2003. The traditional morphological analysis showed that the genera of Microcystis, Chroococcus, Oscillatoria, and Phormidium were dominated. The most frequent band in the DGGE profile by 16S rDNA sequence analysis was identified as Microcystis flos-aquae and the cyanobacterial bloom was peaked on September 2. Oscillatoria spp. were also identified and Aphanizomenon flos-aquae dominated in the middle of August. Judging from the analysis of the digitalized DGGE profiles using the cluster analysis technique, the microbial community on September 2 was considerably different from others. Consequently, it seems that the gene fingerprinting method can give not only the similar results to the traditional morphological method but also additional information on the bacterial species and similarity among the examined microbial communities.

        Perchloroethylene과 Trichloroethylene의 혐기적 탈염소화 및 미생물 군집 분석

        울산, 여수 등 공단지역의 토양, 하천의 저니, 해양의 준설토 둥을 이용하여 난분해성 염소화합물인 PCE (perchloroethylene)및 TCE (trichloroethylene)의 혐기성 탈염소화에 관련하는 미생물을 탐색하고 이들의 탈염소화 효율을 조사하였다. 혐기성 상호대사에 의한 탈염소화 효율을 조사하기 위해 전자공여체로 acetate를 사용하여 혐기성 회분식 실험을 실시하였으며, 이와 병행하여 분자생물학적인 기법인 16S rDNA의 PCR-Double Gradient DGGE (DG-DGGE)를 이용하여 미생물의 군집을 분석하였다. 그 결과 울산 태화강 및 여수 하남천의 저니를 접종한 경우 PCE는 $70\%,\;65\%$, TCE는 $50\%,\;45\%$의 높은 탈염소화 효율을 나타내었다. 또한 16S rDNA의 PCR을 이용한 DG-DGGE로 미생물 군집을 분석한 결과, 탈염소화 효율이 높은 지역의 저니에는 Desulfovibrio sup.의 미생물이 주로 존재함을 확인하였다. In this study, the anaerobic enrichment cultivation was performed with the sediments and the dredged soils from the cities of Ulsan, Masan, Yeosu, Gwangyang, Ansan and Seongnam. Acetate as an electron donor and PCE (perchloroethylene) or TCE (trichloroethylene) as an electron acceptor were injected into the serum bottle with an anaerobic medium. After the incubation of 12 weeks, the removal efficiency of PCE was highest at $70\%$ in the treatment with the sediment of Ulsan. Also, the bacterial community structure was analyzed by D-DGGE (double denatured gradient gel electrophoresis) through PCR-based 16S rDNA approaches. The dominant species id the anaerobic enrichment were found to belong to the genus of Desulfovibrio.

        생장 및 부상성이 향상된 남세균 돌연변이 균주 Arthrospira platensis M20CJK3

        A photosynthetic cyanobacterium Arthrospira platensis, well known for health food supplement, was studied as a target species for atmospheric $CO_2$ removal as well as biomass production. Although the biomass of A. platensis was massively produced in many countries, the recovery cost of its biomass is still high. The purpose of this study was to develop the A. platensis mutant strains which have enhanced growth rate and floatation activity to reduce the recovery cost. A. platensis KCTC AG20590 was treated with 0.24% ethyl methanesulfonate (EMS) for 20 min at room temperature. The mutant strain A. platensis M20CJK3 was finally selected by its morphological and physiological features. The morphology of the mutant A. platensis M20CJK3 was changed from loose-coiled form to tight-coiled form showing short pitch. The growth and $CO_2$ uptake rate of A. platensis M20CJK3 were improved about 15% and 17% compared with A. platensis KCTC AG20590, respectively. The floatation activity of A. platensis M20CJK3 was enhanced in 2-fold compared with that of A. platensis KCTC AG20590. Soluble proteins extracted from two strains were analyzed by two dimensional electrophoresis (2-DE) and MALDI-TOF MS/MS. Among 15 protein spots induced in 2-DE analysis, two spots were the proteins related to photosynthesis and electron transfer system of the other cyanobacteria. As a consequence, it seems that the tight-coiled mutant A. platensis M20CJK3 has an advantage of high growth rate and floatation activity which are beneficial for the mass cultivation and recovery. 건강보조식품 등으로 이용되는 Arthrospira platensis는 세계적으로 대량생산되고 있으나 생산공정 중 수확단계에서 많은 비용이 소요된다. 본 연구에서는 부상을 이용한 효과적인 수확을 위해 균주의 개량을 시도하였으며, 개량균주의 생리적 물리적 특성을 파악하고자 하였다. Ethyl methanesulfonate (EMS)를 모균주 A. platensis KCTC AG20590에 0.24%의 농도로 10, 20, 30분씩 처리하여, 형태 및 부상성이 우수한 균주 A. platensis M20CJK3 균주를 분리하였다. A. platensis M20CJK3은 느슨한 형태에서 촘촘한 형태로 세포사(trichome)의 길이 및 코일간 간격이 감소하였으며, 생장 및 $CO_2$ 고정능이 각각 15%, 17% 향상되었다. 또한, 개량균주의 부상성은 모균주에 비해 2배 이상 향상되었다. 이차원 전기영동 분석을 통해 모균주와 개량균주의 단백질 발현양상을 비교분석한 결과 광합성 관련 색소의 구조와 광전자전달계에 관련된 단백질의 발현 양상이 차이를 보였다. 본 연구에서 개발된 A. platensis M20CJK3은 고밀도 대량배양 및 수확에 유리하며, A. platensis 유전자 연구의 유용한 모델 균주로 활용될 수 있을 것으로 사료된다.

        대청호에서 강우와 식물플랑크톤 군집의 관계

        2001, 2003년의 7월부터 10월까지 대청호에 위치한 댐 지역에서 식물플랑크톤 군집, 환경요인 및 강우량을 조사하였다. 조사 기간동안의 월평균 강우량은 2001, 2003년 각각 91.3, 265.3 mm로 조사되었다 최대 Chl. α의 농도는 2001, 2003년 각각 45.4, 131.5 μg L^(-1)로 조사되었다. 남조류 세포수는 2003년 최대 49,000ce11s mL^(-1)로 조사되었고, 2001년은 최대 200,000cells mL^(-1) 이상으로 조사되었다. 그리고 식물플랑크톤 군집에서 남조류의 상대 풍부도는 2001년 97%, 2003년 74%였다. 2001년은 Microcystis spp.가 조사기간동안 우점종으로 조사되었고, 2003년은 Oscillatoria spp., Phormidium spp., Chroococcus spp. 그리고 Microcystis spp. 순으로 우점종의 변화를 보여줬다. 결론적으로 강우량은 식물플랑크톤의 종 다양성 및 수화현상을 일으키는 남조류 세포 밀도에 영향을 주었다. The phytoplankton community, environmental factors, and rainfall were investigated from July to October in 2001 and 2003 on Dam site in Daechung Reservoir. The monthly average rainfall in the investigated period were 91.3 and 265.3 mm in 2001 and 2003, respectively. The maximum chlorophyll a concentration was observed higher at 131.5 μg L^(-1) in 2003 than at 45.41 μg L^(-1) in 2001. The cyanobacterial number in 2001 was counted up to over 200,000 cells mL^(-1), which was much higher than the maximum number of 49,000 cells mL^(-1) in 2003. The relative abundance of cyanobacteria in the phytoplankton community was about 97% in 2001 and 74% in 2003. Microcystis spp. were absolutely dominant species in 2001, while a couple of cyanobacteria such as Oscillatoria spp., Phormidium spp. Chroococcus spp. and Microcystis spp. were dominant species in 2003. Consequently, it seemed that rainfall affected the diversity of phytoplankton species and decreased the density of bloom-forming cyanobacteria.
