        대장균에서 과잉생산된 Bacillus licheniformis B1의 ${\beta}$-1,4-Glucanase 특성

        The ${\beta}$-1,4-glucanase gene of Bacillus licheniformis B1 was expressed in Esherichia coli BL21, and a protein with a mass of 50 kDa that was soluble was overproduced. A protein with a mass of 37 kDa was secreted from B. licheniformis. It seems that the ${\beta}$-1,4-glucanase produced in E. coli contained the leader peptide and unprocessed carboxy-terminal region, but its processing occurred in the carboxyterminal in Bacillus. The optimal temperature of ${\beta}$-1,4-glucanase was $40^{\circ}C$. The enzyme still had 76% maximal activity at $60^{\circ}C$. The optimal pH of the enzyme was 7. The enzyme retained considerable activities over the weak-acidic, neutral, and weak-basic pH range. Acidic fungal cellulases are used in food, detergent, pulp, paper, textile industries. However, studies about neutral and alkaline cellulase are not enough. The cellulase developed in this study may be useful for industrial applications in the fields of biofuel development. Bacillus licheniformis B1의 ${\beta}$-1,4-glucanase 유전자는 Escherichia coli BL21에서 발현되어, 이로부터 수용성의 50 kDa 단백질이 과량생산되었다. 반면에 B. licheniformis에서는 37 kDa의 형태가 분비되었다. E. coli에서 발현된 ${\beta}$-1,4-glucanase는 leader peptide가 제거되지 않고 포함되어 있고 Bacillus에서는 효소의 carboxy 말단에서 processing이 일어난 것으로 보인다. E. coli에서 생산된 ${\beta}$-1,4-glucanase의 최적온도는 $40^{\circ}C$이었지만, $60^{\circ}C$에서도 최대치의 76% 활성을 유지하였다. 효소의 최적 pH는 7이었고, 효소의 활성은 전체적으로 보면 약산성, 중성 및 약알칼리 영역에 널리 걸쳐 있었다. Cellulase는 식품, 세제, 펄프, 제지, 섬유산업 등 다양한 분야에서 주로 산성의 곰팡이계 효소가 이용되고 있으나, 중성 및 알칼리성 cellulase 연구 및 개발은 미흡한 편이다. 본 연구에서 개발된 중성 cellulase가 바이오 연료 개발 등의 분야에서 활용되기를 기대해 본다.

        남성의 헤어서비스 이용동기에 따른 이용만족도

        The aim of this study is to find out the motivation of visits and satisfaction on hair service among the men clients dividing into groups with similar types. The method is that finding out the motivation to visit, then dividing into groups by the reason of visit, and doing analysis of variance to analyse the satisfaction level. First of all, men in their 20 were the majority of subjects, not-married were the highest portion. Second, after dividing groups by factor of visit motive, groups were re-divided by image oriented, indifferences, and trend adapters. Third, the result of level of satisfaction by groups, was drawn by these six factors-personal service, facilities and environment, trustworthiness, skills and technique, kindness of workers and entertainment and beverage service. Image oriented group's satisfaction level was dependant on personal service and facilities and environment and every factor of satisfaction level was high in trend adopter group.

        학령전기 아동의 내러티브 과제에서 나타나는 어휘사용 특성 : AAC 적용을 위한 기초연구

        배경 및 목적: 보완대체 의사소통 영역에서 어휘 관련 연구가 꾸준히 이루어져왔음에도 불구하고, 어린 아동 대상의 어휘 연구는 찾아보기 어렵다. 본 연구에서는 학령전기 아동을 대상으로 내러티브 발화를 수집하여 낱말 단위로 분석한 후 어휘 사용 양상을 살펴보고자 하였다. 방법: 연구대상은 4-6세 일반아동 53명(남 26명, 여 27명)으로, 이들은 3가지 내러티브 산출과제(이야기 다시 말하기, 개인적 경험 말하기, 이야기 구성하기)를 수행하였다. 산출된 발화는 과제유형과 연령집단에 따른 고빈도 어휘, 전체어휘에서 핵심어휘가 차지하는 비율, 핵심어휘에서 나타나는 구조어와 내용어의 비율, 전체 과제에서 나타나는 부수어휘 및 비율 등으로 분석하였다. 결과: 고빈도 어휘는 과제 및 연령의 영향을 받는 것으로 나타났으며, 핵심어휘는 전체낱말의 72%를 차지하여 소수의 핵심어휘가 전체어휘의 많은 부분을 대표하는 것으로 나타났다. 또한 구조어와 내용어의 비율은 19:81로 내용어가 차지하는 비율이 월등히 높은 반면, 부수어휘가 전체낱말에서 차지하는 비율은 7.45%로 낮게 나타났다. 논의 및 결론: 어린 아동의 어휘 사용은 과제의 영향을 크게 받음을 확인하였다. 대화체가 아닌 긴 이야기를 수집하여 학령전기 아동의 핵심어휘 목록을 제시하였다는 데 의의가 있다. Objectives: Despite the continuous body of research on vocabulary in augmentative and alternative communication, it is difficult to find related studies on vocabulary use in young children. The purpose of this study was to investigate vocabulary use patterns in preschool children by collecting narrative data, and analyzing it by word units. Methods: The subjects were 53 normal children between the ages of 4 and 6 (26 boys and 27 girls). Subjects were asked to carry out three narrative production tasks (retelling a story, personal experience, and script). Produced utterances were analyzed for high frequency words according to task type and age group, percentage of core vocabulary in the total vocabulary, percentage of structure words and content words in the core vocabulary, and use and percentage of fringe vocabulary in total tasks. Results: The appearance of high frequency words differed according to task type and age group. Core vocabulary consisted of 72% of the total word production, representing a large proportion of the total vocabulary. The ratio of structure words to content words was 19:81, showing that content words were more dominant, while the percentage of fringe vocabulary of the total vocabulary was much lower at 7.45%. Con-clusion: Vocabulary use in children is affected by the type of task. This study was significant in that it collected narrative utterances to analyze the core vocabulary in preschool children.

        상아모세포의 조건배지를 이용한 백악모세포의 분화와 석회화 조절

        For the regeneration of periodontal tissues, the microenvironment for new attachment of connective tissue fibers should be provided, At this point of view, cementum formation in root surface plays a key role for this new attachment. This study was performed to figure out which factor promotes differentiation of cementoblast Considering anatomical structure of tooth, we selected the cells which may affect the differentiation of cementoblast - Ameloblast, OD11&MDPC23 for odontoblasts, NIH3T3 for fibroblsts and MG63 for osteoblasts. And OCCM30 was selected for cementoblast cell line. Then, the cell lines were cultured respectively and transferred the conditioned media to OCCM30. To evaluate the result, Alizarin red S stain was proceeded for evaluation of mineralization. The subjected mRNA genes are bone sialoprotein(BSP), alkaline phosphate(ALP) , osteocalcin(OC), type I collagen(Col I), osteonectin(SPARC ; secreted protein acidic and rich in cysteine). Expression of the gene were analysed by RT-PCR, The results were as follows: 1. For alizarin red S staining, control OCCM30 didn't show any mineralized red nodules until 14 days. But red nodules started to appear from about 4 days in MDPC-OCCM30 & OD11-OCCM30. 2. For results of RT-PCR, ESP mRNAs of control-OCCM30 and others were expressed from 14 days, but in MDPC23-OCCM30 & OD11-OCCM30 from 4 days. Like this, the gene expression of MDPC23-OCCM30 & OD11-OCCM30 were detected much earlier than others. 3. For confirmation of odontoblast effect on cementoblast, conditioned media of osteoblasts(MG63) which is mineralized by producing matrix vesicles didn't affect on the mineralized nodule formation of cementoblasts(OCCM30). This suggest the possibility that cementoblast mineralization is regulated by specific factor in dentin matrix protein rather than matrix vesicles. Therefore, we proved that the dentin/odontoblast promotes differentiation/mineralization of cementoblasts. This new approach might hole promise as diverse possibilities for the regeneration of tissues after periodontal disease.

        헤어종사자의 외모 이미지와 직업특성이 외모 관리에 미치는 영향

        This study is to find out the effect of the appearance of the hairdressers and the character of the occupation on their appearance management, concluding the factors, how the appearances of hairdressers affect the character of the work, and appearance management. First, Women, in between the ages of 21 and 30 were the most prevalent, six months to one-year work period made up the highest portion, and when deciding a work place, the most important factor was answered as facilities. Second, the appearance groups were drawn in ``casual-formal``, ``conservative-innovative``, and ``calm-outstanding``. The appearance management was divided into two factors:external confidence and external adapted. Third, in the case of the effect of the appearance on the character of the occupation, an idealappearance management and ideal work ability were definitively related to those three factors. In the case of the effect of the appearanceon appearance management, the more relevant they were to the three appearance factors, the more they had external confidence andexternal adaption. The two factors of the character of the occupation and external confidence/external adaption were directly proportional to each other.

        뇌수모세포종 및 가족성 선종성 용종증으로 발현한 Turcot 증후군 1예

        저자들은 수모세포종이 있는 24세 여자 환자에서 가족성 선종성 용종증이 동반된 Tucot 증후군 1예를 경험하였으며 문헌고찰과 함께 보고하는 바이다. Turcot syndrome is characterized by the concurrence of a primary neuroepithelial brain tumor and multiple colorectal polyposis. We report a case of a 24-year-old woman diagnosed with Turcot syndrome. At first, the patient was diagnosed as having a medulloblastoma after a tumorectomy of the 4th ventricle mass. The patient underwent radiotherapy and chemotherapy. After high-dose chemotherapy, neutropenic fever and severe mucositis developed. For an evaluation of the persistent hematochezia and diarrhea, a colonoscopy was performed. It revealed pseudomembranous colitis and multiple polyps in the entire colon. According to the family history, her father had undergone a total colectomy due to colon cancer and polyposis of the entire colon. Her brother also was found to have multiple polyps in the colon by a colonoscopy. The patient was diagnosed with Turcot syndrome.
