<P>In this study, a silica xerogel-chitosan hybrid is utilized as a coating material to incorporate bone morphogenic protein-2 (BMP-2) on a porous hydroxyapatite (HA) scaffold for bone tissue engineering. BMP-2 is known as a therapeutic agent for improving bone regeneration and repair. Silica xerogel-chitosan hybrids have been used for the delivery of a growth factor as well as osteoconductive coatings. The biological properties of the hybrid coating incorporated with BMP-2 were evaluated in terms of the BMP-2 release behavior, osteoblastic cellular responses and in vivo performance. BMP-2 was continuously released from the hybrid coating layer on the porous HA scaffold for up to 6 weeks. The hybrid coating containing BMP-2 showed significantly enhanced osteoblastic cell responses in comparison with the hybrid coating and HA substrate. Consequently, new bone formation was significantly increased within the hybrid coating containing BMP-2. These results reveal that the hybrid coating containing BMP-2 has the potential to be used as a bone implant, whose osteogenic properties are promoted by the release of BMP-2 in a controlled manner for a prolonged period of time.</P>

In chondrocytes, fibroblast growth factors (FGFs) inhibitchondrocytes proliferation by upregulation of the cell cycleinhibitor p21cip/waf. In this report, we first investigated theroles of fibronectin (FN)-mediated cell adhesion in the modulationof FGF-1's antiproliferative function in chondrocytes. In thisstudy, we found that FN-mediated signaling could rescue cell cyclearrest induced by FGF-1 in primary human chondrocytes. Thisprevention of cell cycle arrest induced by FGF-1 was due to thesuppression of the cell cycle inhibitor p21cip/waf expression onadhesion to FN and its downstream activation of signalingpathways. Finally, we showed that this rescue induced byFN-mediated adhesion is dependent on the extracellular regulatedkinase (ERK) signaling pathway. Taken together, these studiessupport that, despite FGF??FGF receptor's growth-inhibitoryfunction, the FN-mediated signaling can collaborate to compensatefor its negative effect on chondrocytes proliferation, providingevidence for cross talk between signals emerging from these cellsurface molecules in chondrocyte.

The newly invented ‘Hyperloop’ derived from the ideas of Elon Musk, the current CEO of Tesla Electric vehicle. This idea attracted a lot of attention from around the world, including from Obama, the previous President of the U.S., and even various nations of Europe. There has been high hopes of creating Hyperloop-related products, which require high technology, and many companies have been investing in the idea. So far, they have completed a detailed test run. These technical skills are being exported to Southeast Asia and is dominating the ‘Hyperloop’ market. Korail has been developed with continuous advances ever since its launching, and it arose as a national railroad powerhouse that is now recognized everywhere. We expect it will make a good match with ‘Hyperloop’. Therefore, we will introduce the tube vehicle development and its status within and outside of Korea and which direction this study will lead to in the future.

Analysis of metabolomic patterns in thoroughbreds before and after exercise

Objective: Evaluation of exercise effects in racehorses is important in horseracing industry and animal health care. In this study, we compared metabolic patterns between before and after exercise to screen metabolic biomarkers for exercise effects in thoroughbreds. Methods: The concentration of metabolites in muscle, plasma, and urine was measured by $^1H$ nuclear magnetic resonance (NMR) spectroscopy analysis and the relative metabolite levels in the three samples were compared between before and after exercise. Subsequently, multivariate data analysis based on the metabolic profiles was performed using orthogonal partial least square discriminant analysis (OPLS-DA) and variable important plots and t-test was used for basic statistical analysis. Results: From $^1H$ NMR spectroscopy analysis, 35, 25, and 34 metabolites were detected in the muscle, plasma, and urine. Aspartate, betaine, choline, cysteine, ethanol, and threonine were increased over 2-fold in the muscle; propionate and trimethylamine were increased over 2-fold in the plasma; and alanine, glycerol, inosine, lactate, and pyruvate were increased over 2-fold whereas acetoacetate, arginine, citrulline, creatine, glutamine, glutarate, hippurate, lysine, methionine, phenylacetylglycine, taurine, trigonelline, trimethylamine, and trimethylamine N-oxide were decreased below 0.5-fold in the urine. The OPLS-DA showed clear separation of the metabolic patterns before and after exercise in the muscle, plasma, and urine. Statistical analysis showed that after exercise, acetoacetate, arginine, glutamine, hippurate, phenylacetylglycine trimethylamine, trimethylamine N-oxide, and trigonelline were significantly decreased and alanine, glycerol, inosine, lactate, and pyruvate were significantly increased in the urine (p<0.05). Conclusion: In conclusion, we analyzed integrated metabolic patterns in the muscle, plasma, and urine before and after exercise in racehorses. We found changed patterns of metabolites in the muscle, plasma, and urine of racehorses before and after exercise.
