Campbell Early 잎 추출물이 Isoprenaline 유발 심근병증 랫트에서 심장-신장 개선에 미치는 영향

결론적으로, ISO 주입으로 심근병증을 유발하여 심장과 신장의 구조 변화 및 기능 손상(심신증후군)을 일으켰으며, Campbell early 잎이 이를 개선시키는 것으로 확인했다. 비록 용량 의존적인 효과를 보이진 않았지만 대체로 고농도의 Campbell early 잎 추출물이 더욱 심장 및 신장의 기능 이상을 개선하는 효과가 있다는 것을 알 수 있었다. 따라서 추후 심장-신장 상호 기전에 대한 후속 연구에 유용한 결과로 사용될 수 있을 것으로 보이며, 포도는 열매뿐만 아니라 잎 또한 심혈관계 질환 개선 소재로 활용이 가능할 것이라 사료된다. Many studies have shown that grapes have antioxidant, nerve protection, liver protection, and anti-cancer effects, but their effectiveness on leaves is not known. The study was conducted to identify the effectiveness of Campbell early leaves (CEL) in heart and kidney dysfunction. Cardiomyopathy was induced by a treatment with isoprenaline (ISO) in rats to confirm the improved effect of cardiac-kidney dysfunction (cardio-renal syndrome). ISO (10 ㎎/㎏/d) was administered intraperitoneally for two weeks and was divided into five groups: CONT, control; ISO, isoprenaline 10 ㎎/㎏/d; PRO, ISO+propranolol 10 ㎎/㎏; CEL100, ISO+CEL 100 ㎎/㎏/d; CEL200, ISO+CEL 200 ㎎/㎏/d. The experiment was conducted for four weeks. As a result, the CEL groups showed an improvement in the value of the left ventricle mass index after exposure to ISO compared to the control group. Furthermore, the echocardiogram measurements showed that the CEL groups could recover the decrease in ejection fraction, fractional shortening, and stroke volume. To identify the heart-kidney functionality, albumin, osmotic, and blood nitrogen was measured in each group, which showed a significant increase in the ISO group, decreasing in the CEL group. The fibrosis of the kidneys also recovered in the CEL group. In conclusion, in cardiomyopathy caused by ISO-induced rats, CEL improves the heart and kidney function. Therefore, grape leaves and fruit are considered useful for improving the heart-kidney functional disorders in grape leaves.

Hypertension has been approved to cause disharmony between the heart and kidney such as cardiac hypertrophy and kidney dysfunction. In traditional oriental medicine Paeo-tang (PET) has been shown to have effects on blood circulation improvement. However, the beneficial effect of PET on hypertension remains unknown. In this study, we investigated that PET attenuates blood pressure and improves cardiovascular and renal function in NG-nitro-L-arginine methylester (L-NAME) rat model. Hypertensive rat models were induced by the administration of L-NAME (40 mg/kg/day) and then PET (50 or 100 mg/kg/day) or Olmetec was treated for 2 weeks. PET treatment significantly suppressed the systolic blood pressure and decreased intima-media thickness in the thoracic aorta. PET ameliorated endothelium-dependent and independent vascular relaxation in the L-NAME-induced vascular dysfunction. PET ameliorated the functional decline in the kidney such as albumin and blood urea nitrogen in plasma. These results demonstrated that PET possesses protective effects against L-NAME-induced hypertension.
TNF-α로 유도된 혈관내피세포의 혈관염증에 미치는 오적산(五積散)의 억제 효과

Objectives : Ojeoksan, originally recorded in an ancient Korean medicinal book named “Donguibogam” and has been used for the treatment of circulation disorder of blood which was called blood accumulation (血積) in Korean medicine. Therefore, this study was carried out to investigate the beneficial effect of OJS on vascular inflammation in HUVECs. Methods : We evaluated the effect of OJS on the expression of cell adhesion molecules and protective role in HUVEC stimulated by TNF-α by using Western blot. Results : Pretreatment with OJS decreased the adhesion of HL-60 cells to TNF-α-induced HUVEC. OJS suppressed TNF-α-induced expression level of cell adhesion molecules such as intracellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1(VCAM-1), and endothelial cell selectin (E-selectin). Moreover, OJS significantly decreased TNF-α-induced production of intracellular reactive oxygen species (ROS); and inhibited the phosphorylation of IκB-α in the cytoplasm compared to the experimental group. Pretreatment with OJS inhibited the trans-location of NF-κB p65 to the nucleus. OJS also inhibited phosphorylation of MAPKs compared to the experimental group. OJS significantly increased the protein expression of Nrf2 and HO-1. Conclusions : Ojeoksan has a protective effect on vascular inflammation, and might be a potential therapeutic agent for early atherosclerosis.

Objectives : It is well known that Sibjotang (Shizaotang), traditional herbal medicine formula, regulates the body fluid blood pressure homeostasis. This study is to investigate whether Sibjotang improves diabetic renal dysfunction in type II diabetes mellitus animal model, db/db mice. Methods : The animals model were divided into three groups at the age of 8 weeks; control group (C57BLKS/J-db/m mice), diabetic group [(C57BLKS/J+Lepr)-db/db mice], and Sibjotang group [(C57BLKS/J+Lepr)-db/db mice + Sibjotang 100 ㎎/㎏/day]. During 8 weeks of treatment, blood glucose and urinary albumin excretion were checked in metabolic chamber at 8, 12, and 16 weeks of age, respectively. Results : Body weight and food intake of diabetic group were significantly higher than control group after 8 weeks administration. However, there were not significant different between the diabetic group and Sibjotang group. Urinary albumin excretion was significantly decreased in the Sibjotang group than the diabetic group. In addition, supplementation with Sibjotang significantly lowered levels of blood glucose, insulin, and homeostatic model assessmentinsulin resistance (HOMA-IR), suggesting reduced insulin resistance. The ratio of mesangial matrix/glomerular area was markedly larger in diabetic group than control group, whereas Sibjotang significantly reduced this expansion. Moreover, immunohistological study revealed that Sibjotang attenuated the increase of transforming growth factor(TGF)-β expression in kidney. Conclusion : Sibjotang ameliorates diabetes-associated renal injury through the improvement of the blood glucose and insulin sensitivity, and inhibiting the TGF-β1 expression. Therefore, Sibjotang may be a new therapeutic formula for the treatment of diabetic-associated renal dysfunction.
