        형질전환 토마토에서 Cytosine Methylation에 의한 유전자발현 억제

        토마토 형질전환체에서 NPTII 및 TPSP 유전자의 발현양상을 조사하였다. 4개 line 토마토 형질전환체가 선발배지에서 kanamycin에 저항성을 보이며 생산되었다. 그러나, 1번 line과 11번 line의 후대에서 NPTII 유전자의 발현이 억제되었으며, Kanamycin 저항성을 보이지 못했다. Southern 분석 결과, 1번 11번 line은 여러 copy의 외래 유전자를 가지고 있었으며, 2번, 10번 line은 1-2 copy의 외래유전자를 가지고 있었다. 형질전환 line 11번은 methylation sensitive 제한효소처리 후 DNA methylation 분석 결과, TPSP coding부위 및 도입유전자의 발현을 조절하는 CaMV35S 프로모터 주위에서 CpG methylation이 축적되었음이 확인되었다. 따라서 여러 copy의 외래유전자를 가진 토마토 형질전환 line 11번에서 NPTII 유전자 및 TPSP유전자의 발현이 억제된 것은 transcriptional gene silencing 기작에 의한 것으로 추정되었다. Transgene expression was analyzed in tomato plants. Four lines of neomycin phosphotransferase II gene (NPTII) and the trehalose biosynthetic fusion gene (TPSP) transformed $T_0$ plants showed kanamycin resistance on selection medium. However, the analysis of phenotype (kanamycin resistance) and mRNA expression in $T_1$ plants indicated that the expression of the NPTII and TPSP transgenes was down-regulated to an undetectable level in two independent lines 1 and 11. Southern analysis demonstrated that the lines 1 and 11 had multicopies of the transgenes, whereas the typical transgenic lines 2 and 10 had 1 or 2 copies. DNA methylation analysis using methylation sensitive enzyme detected accumulated CpG DNA methylation on TPSP coding region and CaMV35S promoter region in the line 11, but not the typical transgenic line 2. These results suggest that multicopy transgene in plants is attributed to down-regulation of the transgene expression via transcriptional gene silencing.

        어린이집 보육교사의 감성지능, 의사소통능력, 셀프리더십이 교사역량에 미치는 영향 연구

        This study aims to find out the relationship between emotional intelligence, self-leadership, and teacher competency of daycare center teachers, and the influence of emotional intelligence and self-leadership on teacher competency. The collected data verified statistical significance through descriptive statistics, independent sample t-verification and one-way variance analysis, Pearson's titration correlation analysis, and multiple regression analysis using the SPSS 20.0 statistical program. Looking at the differences according to background variables, self-leadership showed significant differences in age and career, and teacher competency showed significant differences in institutional type, educational background, certificate, career, and age in charge. Second, it was found that there was a significant positive correlation between the emotional intelligence, self-leadership, and teacher competency of daycare center teachers. Third, the emotional utilization of children's emotional intelligence of children's emotional intelligence, but emotional utilization was not significantly affected by teachers' history. Based on these results, the paper is meaningful in that it laid the foundation for the development of teacher education programs using emotional intelligence and self-leadership factors as a strategy to improve teacher competency, which is the most basic quality as an infant teacher.

      • 생물전환 공법을 이용한 천연 유래 화장품 소재 개발

        최근의 기술적 진보를 통해 화학적 공정 기반이 아닌 천연유래의 화장품 소재 개발이 각광받고 있으나 국내 화장품 산업에서 천연소재 원료의 80% 이상은 수입에 의존하고 있다. 천연소재 화장품은 기존의 화학적 합성제품에 비해 부가가치가 높아 고급화장품 또는 특정 제품에 제한적으로 사용되어지며 점차 그 활용도가 높아지고 있다. 본 연구는 스테롤과 다양한 지방산간의 에스테르화 반응을 통해 새로운 형태의 친환경적 유화제를 개발하고자 진행되었으며 다양한 기능성 친환경 소재의 국산화를 이루기 위해 진행되었다. 생물전환공법을 사용한 천연 유화제를 합성하기 위해 스테롤과 지방산의 에스테르화에 관여하는 효소의 screening을 진행하였다. 총 19개의 효소에 대한 생촉매로써의 기능을 검토하였고 최종 적합효소를 선정하였다. 선정된 효소를 사용한 생성물의 정성·정량 분석을 통해 반응 수율을 결정하였고 최적의 반응 조건을 설정하기 위한 실험을 진행하였다.

        국내 간호사의 환자안전관리활동에 대한 메타경로분석

        Purpose: This study aimed to test a hypothetical model of Korean nurses’ patient safety management activities using meta-analytic path analysis. Methods: A systematic review, meta-analysis, and meta-analytic path analysis were conducted following the PRISMA and MOOSE guidelines. Seventy-four studies for the meta-analysis and 92 for the meta-analytic path analysis were included. The R software program (Version 3.6.3) was used for data analysis. Results: Four variables out of 49 relevant variables were selected in the meta-analysis. These four variables showed large effect sizes (ESr = .54) or median effect sizes (ESr = .33∼.40) with the highest k (number of studies) in the individual, job, and organizational categories. The hypothetical model for the meta-analytic path analysis was established using these variables and patient safety management activities. Twelve hypothetical paths were set and tested. Finally, the perception of the importance of patient safety management and patient safety competency directly affected patient safety management activities. In addition, self-efficacy, the perception of the importance of patient safety management, patient safety competency, and patient safety culture, indirectly affected patient safety management activities. Conclusion: Self-efficacy, the perception of the importance of patient safety management, patient safety competency, and the organization’s patient safety culture should be enhanced to improve nurses’ patient safety management activities.

        환자안전사건과 관련된 임상간호사의 이차피해경험이 간호실무변화에 미치는 영향: 대처의 매개효과

        Purpose: This study was investigated the mediating effect of coping behaviors in the relationship between the second victim experiences after patient safety incidents and the nursing practice changes. Methods: A cross-sectional survey was performed using structured questionnaires. Participants were 218 clinical nurses in general tertiary hospitals in South Korea. Data were collected through an online survey and snowball sampling from August 11 to September 6 2020. Data were analyzed using SPSS 23.0 program. A mediation analysis was performed using multiple regression and a simple mediation model applying the PROCESS macro with 95% bias-corrected bootstrap confidence interval. Results: The mean scores of second victim experiences was 3.41/5. Approach coping (β = .55, p < .001) and the avoidant coping (β = - .23, p = .001) showed mediation effects in the relationship between second victim experiences and constructive change in nursing practice. Avoidant coping (β = .29, p < .001) showed a mediation effect in the relationship between second victim experiences and defensive change in nursing practice. Conclusion: Coping behaviors has a mediating effect on the relationship between second victim experiences and nursing practice changes. To ensure that nurses do not experience second victim, medical institutions should have a culture of patient safety that employs a systematic approach rather than blame individuals. They also need to develop strategies that enhance approach coping and reducing avoidant coping to induce nurses’ constructive practice changes in clinical nurses in experiencing second victims due to patient safety incidents.

        생육상과 온실에서 게발선인장 ‘핑크듀’의 엽상경별 CO₂ 흡수율 비교

        CAM 식물은 야간에 기공을 열어 CO₂를 흡수하는 식물로 환기하지 않는 야간에 시설 내 발생한 잉여 CO₂를 광합성에 이용할 수 있다. 게발선인장은 다화성으로, 엽상경 수가 많을수록 상품성이 높아진다. 본 연구는 게발선인장의 야간 CO₂ 시비 적용 가능성을 파악하기 위하여 생육 적정 환경인 생육 상과 실제 농가의 환경에 따른 온실에서 엽상경별 광합성 특성을 조사하였다. 생육상 내 게발선인장 상위 엽상경은 주간에만 CO₂ 흡수를 하는 C₃ 특성을 보였으며, 2차 엽상경은 주/야간 모두 CO₂ 흡수를 하는 C₃–CAM 특성을 보였다. 온실에서 상위 엽상경은 CO₂ 흡수를 하지 않았으나, 2차 엽상경은 야간에 CO₂ 흡수를 하는 CAM 특성을 보였다. 생육상 내 게발선인장의 기공전도도와 수분이용효율은 온실 내 식물에 비하여 모든 엽상경에서 높은 값을 나타냈다. 게발선인장의 수분이용효율은 생육상과 온실에서 모두 CAM 특성이 나타나는 2차 엽상경에서 상위 엽상경에 비하여 높았다. 일일 총 CO₂ 흡수율은 온실 내 식물에 비하여 생육상 내 식물에서 더 높게 나타났으며, 생육상 내 게발선인장의 2차 엽상경에서 값이 155mmol·m<SUP>–2</SUP>·d<SUP>–1</SUP>로 가장 높았다. 게발선인장 ‘핑크듀’는 성숙한 엽상경에서 야간에 CO₂ 흡수가 활발히 일어나는 CAM 특성을 나타낸다. 다화성으로 시장성이 높은 게발선인장 ‘핑크듀’는 적정 환경과 성숙한 엽상경에서 효율적인 야간 탄소 흡수가 가능한 식물이다. Crassulacean acid metabolism (CAM) plants use surplus CO₂ generated by cooling and heating at night when ventilation is not needed in a greenhouse. Schlumbergera truncata ‘Pink Dew’ is a multi–flowering cactus that needs more phylloclades for high–quality production. This study examined photosynthetic characteristics by the phylloclade levels of S. truncata in a growth chamber and a greenhouse for use of night CO₂ enrichment. The CO₂ uptake rate of the S. truncata’s top phylloclade in a growth chamber exhibited a C₃ pattern, and the second phylloclade exhibited a C₃–CAM pattern. The CO₂ uptake rate of the top phylloclade in a greenhouse showed a negative value both day and night, but those of the second phylloclade exhibited a CAM pattern. The stomatal conductance and water–use efficiency (WUE) of S. truncata at both the top and second phylloclades were higher in a growth chamber than in a greenhouse. The WUE of S. truncata in a growth chamber and a greenhouse was higher at the second phylloclade, which is a CAM pattern compared with those of the top phylloclade. The daily total net CO₂ uptake of S. truncata was higher in a growth chamber than in a greenhouse. The daily total net CO₂ uptake of S. truncata at the second phylloclade had the highest value of 155 mmol·m<SUP>–2</SUP>·d<SUP>–1</SUP> in a growth chamber. The night total CO₂ uptake of S. truncata at the second phylloclade was 3–fold higher in a growth chamber than in a greenhouse. S. truncata’s second phylloclade exhibited a CAM pattern that uptake CO₂ at night, and the second phylloclade, was more mature than the top phylloclade. A multi–flowering cactus S. truncata ‘Pink Dew’ efficiently uptake night surplus CO₂ in the proper environmental condition with matured phylloclade.
