This study was undertaken to examine the protein content of GCF and serum from normal population in order to standardise the sample loading on SDS/PAGE gels. The results were as follows : 1. The protein concentration of serum was not different between normal group and diseased group. 2. In GCF, the bands of lower molecular weight protein than albumin were heavily stained, but in serum, higher molecular weight protein bands are did. 3. The profile of protein in normal GCF was characterized by heavily staining bands at 77, 77, 55, 26 KDa corresponding to the positions of transferrin, albumin, heavy and light chains of Ig G. Also 47, 37 KDa nonplasma proteins are found.

Glutathione(GSH),a tripeptide thiol, found in virtually all cells, functions in metabolism tranasport and cellular protection. It protects cells against the destructive effects of reactive oxygen intermediates and free radicals. Also ${\gamma}-Glutamyl$ transpeptidase(${\gamma}-GTT$), an enzyme of major importance in GSH metabolism, initiates GSH degradation. In order to explore the $GSH-{\gamma}-GTT$ system as periodontal disease activity indicator, we observed the ${\gamma}-GTT$ and arachidonic acid metabolits according to clinical groups(Control, Adult periodontitis, Rapidly progressive Periodontitis). From the experiments, the following results were obtained. 1. When compared with normal, ${\gamma}-GTT$ of A. P. and R. P. P. were increased, and only the change of ${\gamma}-GTT$ of R. P. P. was statistically significant(P<0. 05). 2. The amounts of arachidonic acid metabolites were not different with statistical significance among the clinical groups. 3. ${\gamma}-GTT$ may by useful adjuncts as new cytoprotective indicator and periodontal disease activity indicator in accordance with positive corelation pocket depth, attachment level and ${\gamma}-GTT$.

The neuropeptide substance P(SP) has been recognized to modulate immune systems, with close proximity between peptidergic sensory nerve endings and immune cells. These include the macrophage and neutrophil activation, IL-2 production in T cell, augmentation of Ig synthesis, mast cell degranulation, $PGE_2$ and collagenase secretion in synoviocytes. In this study I examined SP-induced various biological activities such as antimicrobial action, cytokine production, and mast cell degranulation in the presence or absence of other inflammatory cell activators. Antimicrobial studies showed that undifferentiated HL-60 cells were not affected by SP. However, SP significantly enhanced antimicrobial action of TPA-treated or dbcAMP-treated HL-60 cells which had been differentiated into PMN or macrophage/monocyte. I could not find synergistic relationship between SP and LPS in parallel experiments of the above. SP did not induce IL-l production from murine macrophage cell line RAW264.7 whether costimulated with LPS or not. Mast cell degranulation was occured only when stimulated with high dose ($10^{-5}M$) of SP and the degree of this activation was slightly reduced by simultaneous application of $MIP-1{\alpha}$. In addition, CGRP which is known to be a common coexisting neuropeptide with SP within specific fibers did not augment the function of SP on mast cell degranulation. These results suggest that immunoregulatory activities of SP could be mediated through direct upregulation of various functions of immune cells and also upregulation of responsiveness of immune cells to other immune activators.

An animal-dedicated coil was manufactured to improve the image quality in developing a coil for mice used in animal experiments. Various parameters (e.g. the pulse sequence) that can evaluate a range of clinical diseases were changed to determine the optimal high-resolution MRI technique for mouse brains. Balb/C nude mice were used in this experiment. A syringe was used to inject an anesthetic solution (300 ml/kg) into the abdominal cavity of the mice. A 3.0 T system (MAGNETOM Trio, A Tim System, Germany) was used. The manufactured receive coil was a mouse body coil, which was a receive-only coil with four channels. The coil had a diameter of 40 mm, length of 100 mm and an overlapped distance between the channels of 4 mm. Image evaluation was conducted based on the signal to noise ratio (SNR) and image uniformity for each pulse sequence. The Q value measured in the manufactured coil was 87.7 for mice, which showed a very sharp resonance curve. The measured difference was 39 dB or higher in the case of isolation between the phased array channels. When the SNR and image uniformity were evaluated, the dual echo steady state 3 dimensions (DESS 3D, 65.07 ± 19.42, 77.86 ± 16.74) showed the highest value, whereas the turbo inversion recovery magnetism 2 dimensions (TIRM 2D, 05.49 ± 01.54, 10.19 ± 03.82) showed the lowest value. In conclusion, the 4-channel phased array coil allowed imaging of a mouse brain with a small diameter. The parameters were changed depending on the selected pulse sequence to determine the optimal conditions for images that can quickly (within an average of 5 minutes and 30 seconds) acquire high-resolution MR images with a resolution of ≤1 mm.
A Study on Development of Phase Array Coil for MRI of Mouse in the 3.0 T High Magnetic Field

<P>In this study, we manufactured an animal-dedicated radio frequency (RF) coil with a small diameter and multiple channels that could improve both image uniformity and signal-to-noise ratio (SNR). Quality of images from the manufactured coil was assessed compared to the loop coil used primarily for animal experiments and that had the same diameter. For our experiments, we used Balb/C nude mice that are most commonly used, and 3.0 T systems. The mouse body coil was a receive-only (Rx-only) coil with four channels. The coil had a diameter of 40 mm, length of 100 mm, and an overlapped distance between channels of 4 mm. A dual echo steady state (DESS) 2D pulse sequence, the type most frequently used for animal experiments, was applied. To evaluate the SNR of the coil cylinder that had a different number of channels, we used the loop coil typically designated for human use and the manufactured coil specific for animal use to obtain and compare phantom and mouse images. According to the results the image analysis, image uniformity and SNR were high for the image obtained from the animal-specific coil that had many channels. In conclusion, our experiment demonstrated that a 3.0 T phase array coil with multiple channels designed for animal use only produced images with excellent uniformity and SNR.</P>
