        엽록체 DNA를 이용한 마약성 양귀비 판별법 개발

        Among a variety of molecular markers, insertions and deletions (InDel) markers are proposed for the identification of narcotic opium poppies in this study. For the design of InDels markers, the chloroplast genome sequences of 3 papaver species (P. sominiferum, P. orientale, and P. rhoeas) were completed using Illumina Hiseq 2000 and the specific genetic differences among species were found. On the basis of the InDel size differences, three InDel markers were developed with the advantages of easy to set up and high accuracy to distinguish narcotic and non-narcotic poppies. As a result of applying the 3 InDel markers to the drug case samples, these markers correctly differentiated the samples contained the Thebaine, Codeine, and/or Morphine. In addition, it was confirmed that narcotic poppy could be identified in seeds without narcotic substances, in damaged or decomposed samples, and in samples containing a mixture of various plants. Therefore, the newly developed 3 InDel markers for the identification of narcotic poppies would be highly useful in drug investigations by complementing gas chromatography-mass spectrometry (GC-MS

        마약성과 비마약성 대마 품종의 식별을 위한 카나비놀 생합성 유전자 분석법

        Cannabis sativa is an important industrial plant utilized to produce fiber, oil, and medicinal ingredients. A chemotype of cannabis is divided into “Drug type” with predominance of tetrahydrocannabinolic acid (THCA) and “Fiber type” with cannabidiolic acid (CBDA). To develop molecular markers for the discrimination of these two types, nucleotide sequences of THCA synthase and CBDA synthase as well as their pseudogenes were retrieved from the recently published cannabis genome in chromosome scale. Gene-specific SNPs were discovered by multiple alignment of these sequences, and 2 dominant marker sets from each gene were designed for selective amplification. Our markers successfully identified “Drug type” and “Fiber type” cannabis plants as well as forensic samples including processed materials. Our molecular markers will provide a fast and efficient system for molecular-based identification of the cannabis plant.

        현장감정물 시료의 체액흔 확인을 위한 상용화키트의 유효성 평가

        사건현장에서 얻어진 증거물에 대해 DNA 감정과 더불어 중요한 증거로 자리매김 하고 있는 것은범죄현장의 흔적이 어떤 체액에서 유래된 것인지를 확인하는 것이다. 특히, 성범죄의 경우 남성의 정액 유무는 범죄 행위를 입증하는데 큰 영향을 미치기 때문에 정액흔 판단 검사를 기본으로 수행하고 있고, 신속하고 정확한 확인이 가능한 상용화키트를 주로 사용한다. 현재, 상용화 키트로 확인할 수 있는 체액의 종류는 정액, 혈액, 타액, 소변 등이 있으며 여기에서는 사건 감정에서 확인 빈도가 높은 정액 및 혈액을 대상으로 기존에 검증되어 사용하고 있는 RSID와 ABAcard 키트 외에 의료용 또는 법과학용으로 국내·외에서 판매되고 있는 3개의 키트를 선정하여 유효성 평가를 실시하였다. 정액과 혈액에 대한 민감도와 특이도 실험을 수행하였고, 실험결과 정액은 103 희석비율까지 양성 결과를 보였으며, 혈액은 키트 종류에 따라 102 또는 103 희석비율에서 양성 결과를 확인하였다. 특이도 실험결과 혈액 키트는 모두 음성으로 확인되었으나, 정액키트의 경우 남성 소변에서 RSID 키트를 제외한 나머지 4개 키트에서 양성 또는 양성(약함)의 결과를 보였다. 본 연구에서 확인한 각 키트의 특성 및 실험결과를 바탕으로 현장증거물에 대한 체액흔 해석지침을 만들고 감정사례에 따라 적합한 키트를 선정할 수 있도록 기준을 마련하고자 한다. Determination of the origin of the body fluids recovered at crime scenes has become essential evidence along with DNA in the forensic science field. Particularly, in the case of sexual crimes, the presence or absence of semen crucially determine the type of crimes or sentence, thus a semen test is performed mainly and commercial kits are used for more accurate and efficient results. Currently, the body fluids that can be identified using commercial kits include semen, blood, saliva, and urine. In this study, a comparative experiment was conducted with commercial kits against semen and blood, which are frequently used body fluids in forensic cases. Five commercial kits were selected to perform sensitivity and specificity tests on semen and blood. For the sensitivity test, the semen kits showed positive results up to 103 dilution, and the blood was identified as positive at 102 or 103 dilution depending on the kit. As a result of the specificity test, all blood kits were observed as negative for other body fluids and all semen kits except the RSID kit that appeared positive or weak positive for male urine. Based on the features and results of the experiments, we intend to establish guidelines for analysis of body fluids from trace evidence and standards to select suitable kits for cases.

      • 한국인 집단에서 새로운 Quadruplex STR 시스템의 개발과 분석

        본 연구에서는 한국인 집단에서 높은 식별력(discrimination power)을 가지는 4가지 STR 유전자좌(D2S1371, D8S1477, D12S391, D20S470)를 한 번의 PCR 반응으로 분석할 수 있는 Quadruplex PCR 시스템을 개발하였다. 또한 nucleotide sequencing을 통하여 각 유전자와 대립유전자들의 정확한 반복회수를 결정하고 대립유전자 명명에 사용하였다. 한국인 집단에 대한 각 대립유전자의 빈도를 산출하였다. 이렇게 작성된 대립유전자 빈도는 카이제곱 검정, 정화검정법을 통하여 4가지 유전자좌가 모두 Hardy-Weinberg Equilibrium을 이루고 있는 것으로 평가되었다. 본 연구에서 완성된 Quadruplex 시스템은 범인 식별, 친자확인 등의 범과학(Forensic Science) 분야에 유용하게 쓰일 수 있을 것으로 생각된다.

        DNA 바코드 및 유전자은행 정보 분석을 통한 환제(丸劑)의 구성약재 감별

        The herbal pill is usually made into several kinds of herbal medicines and supplementary excipients, such as starch, wheat flour, and honey. However, the verification of correct constituents of herbal pill is very difficult because it is composed of powders mixed with diverse herbal medicines. To determine the material composition of an unknown herbal pill, we analyzed DNA barcode sequences and NCBI GenBank nucleotide database on trial. Among the several DNA barcodes, nuclear ribosomal RNA gene internal transcribed spacer 2 region (ITS2) amplified sufficient PCR products to analyze the nucleotide sequences and provided valuable informations to determine the constituents of herbal pill on GenBank blast search. These valuable information is enough to estimate approximately the main herbal medicines as well as excipients. Therefore, we suggest DNA barcode sequence analysis depending on the GenBank database as a tool of quality control method with chemical analysis for herbal pills.

        Construction of an integrated barcode database for the molecular identification of species

        Species identification based on molecular markers is a major concern in the field of phylogenetic systematics because it has a wide range of applications such as molecular forensics and biodiversity research. Therefore, a well-organized and curated database is essential to meet this need. Barcode of Life Database (BOLD) is the main database system that stores a large variety of species identification markers. However, BOLD does not contain all of the barcode information available in other databases such as GenBank and the Korea Barcode Of Life database (KBOL). As a result, users must navigate all of these databases to obtain integrated information on barcodes and species. To solve this issue, we have constructed a molecular marker database for the integration of all available barcode information. BOLD, the National Center for Biological Information database, and KBOL were used as external sources for database construction. We carefully annotated all data from each database for the exclusion of duplicate information and unreliable data. As a result, the database contains 622,913 molecular markers that identify 104,487 species. Among these, 5,320 makers can be used for the identification of 2,326 Korean species. This database is freely available on the internet (http://203.234.220.113/).

        한국인 집단에서 새로운 Quadruplex STR 시스템의 개발과 분석

        A quadruplex PCR system for polymorphic 4 STR loci that shows high discrimination power (D2S1371, D8S1477, D12S391, and D20S470) has been developed. Nucleotide sequence of alleles from each locus has been determined to verify the exact repeat motif and allele size. The developed quadruplex system has amplification product size range of 96-321 bp, which can be easily separated by conventional denaturing PAGE. The allele and genotype distributions satisfied Hardy-Weinberg equilibrium. This should be useful for the construction of National DNA Intelligence Database as well as forensic analyses.
