      • 습사료에 첨가한 유용미생물 및 한약재 혼합제(한방천ㆍ어력천) 특성과 혼합 첨가제가 넙치간의 활성에 미치는 효과

        유용 미생물과 한약재가 포함된 새로운 사료첨가제의 다양한 농도(0.3, 0.6 및 0.9%)에 따른 넙치의 간기능의 활성에 미치는 영향 및 사료첨가제의 특성에 대해서도 조사하였다. 첨가제에 포함된 유용미생물(유산균, 바실러스균, 효모 및 광합성균)의 총수는 한방천이 5.6${\times}$$10^{8}$ CFU/g이었으며, 어력천은 3.0${\times}$$10^{8}$ CFU/g 이었다. 앙식에 일반적으로 사용되는 습사료의 병원미생물(Edwardsiella tarda, Vibrio anguillarum and Streptorwccus sp.)은 모든 첨가군에 있어서 첨가제의 농도의존적으로 유의한 감소를 나타내었다. 간중량 지수는 0.3%의 첨가군에서 유의하게 증가하였다. 혈중의 단백질량은 모든 첨가군이 유의한 증가를 나타내었으나. 간장내의 단백질량은 대조군이 높은 수치를 나타내었다. 생체방어기작을 하는 catalase 및 superoxide dismutase는 0.3% 및 0.6% 첨가군에서 각각 높은 활성을 나타내었다. 이상의 결과로 한약재와 유용미생물 혼합 사료첨가제의 사용으로 넙치의 항산화효소의 활성을 증대시켜 어류스트레스의 면역증대를 기대할 수 있을 것으로 여겨진다. The effects of different concentrations (0, 0.3, 0.6 and 0.9%) of fish feed additives (Hanbangchun and Olyukchun) utilizing effective microorganisms and herb medicine on activity of liver function were examined in olive flounder, Paralichthys olivaceus, Moreover, we investigated the characteristics of the additives. Total number of microorganisms (Lactic acid bacteria, Bacillus subtilis, Saccharomyces cerevisiae, Photosynthetic bacteria and Azotobactor) in the additives was 5.6${\times}$10$^{8}$ CFU/g in the Hanbangchun and 3.0${\times}$10$^{8}$ CFU/g in the Olyukchun. Levels of three typical pathological microorgamisms (Edwardsiella tarda, Vibrio anguillarum and Streptococcus sp.) in moist pellets (MP) were significantly decreased by the additives in a concentration-dependent way. Hepatosomatic index of fish in the 0.3% group was significantly increased. Total serum protein was increased in all the groups containing additives, but the protein content in liver was higher in the control group. Higher activities of catalase and superoxide dismutase which are involved in physiological defense mechanisms were found in the dietary groups containing 0.3% and 0.6%, respectively. These results suggest that the additives, Hanbangchun and Olyukchun, can increase tolerance of olive flounder against stress and hypoxic conditions by increasing activities of body antioxidant enzymes.

        넙치 Paralichthys olivaceus 초대 배양 간세포의 난황 전구물질 합성에 미치는 estradiol-$17{\beta}$와 2,4-D의 영향

        Estradiol-$17{\beta}(E_2)$와 2,4-dichlorophenoxy acetic acid (2, 4- D)가 난황 전구물질의 합성에 미치는 영향을 넙치 Paralichthys olivaceus 간세포의 초대 배양을 통하여 조사하였다. 배양간세포의 생존율은 배양온도 $27^{\circ}C$에 서 가장 높게 나타났으며, $15^{\circ}C$에서는 생존율이 급격히 감소하여 약 50%의 생존율을 나타내었다. $E_2$에 의한 VTG 의 합성은 $10^{-6}M$에서 최대치를 나타내었다. 2, 4-D $10^{-7}\sim10^{-5}M$의 첨가에 의해 VTG의 합성은 이루어지지 않았다. 그러나, 저농도인 $10^{-8}M$에서는 VTG의 합성이 증가하였다. $E_2$ 및 2, 4-D에 의해 합성된 VTG는 $10^{-6}M$ tamoxifen의 첨가에 의해 유의하게 억제되었다 (P<0.01). 본 연구 결과에서 $E_2$와 2, 4-D의 동시 첨가는 VTG의 합성을 억제하지 않았다. 이러한 결과는 2, 4-D의 작용이 $E_2$와 유사한 작용을 가지지만, VTG의 합성에 있어서 $E_2$ 수용체에의 작용 양식은 서로 다른 것으로 추정된다. Effects of Estradiol-$17\beta(E_2)$ and 2, 4-dichlorophenoxy acetic acid (2, 4-D) on vitellogenin(VTG) production were investigated in primary hepatocyte culture of olive flounder, Paralichthys olivaceus. Highest survival rate of hepatocyte were observed at $27^{\circ}C$, which markedly declined equal to 50% of those of $15^{\circ}C$. Vitellogenin production peaked at the concentration of $10^{-6}M\;E_2$. No effect was observed on VTG production at various concentrations of 2, 4-D. However, a low concentration of 2, 4-D (ie, $10^{-8}M$) only appeared increased VTG production. $E_2$ or $10^{-8}M$ 2, 4-D-primed VTG production was markedly inhibited by the addition of $10^{-6}M$ tamoxifen to the culture medium(P<0.01). Inhibition was not affected by combinational treatment with $10^{-6}M$ $E_2$ and $10^{-6}M$ 2, 4-D. These results from the current investigation suggest that 2, 4-D mimics $E_2$, but the mechanism of reaction in inducing the $E_2$ receptor are different in VTG production in oliver flounder hepatocytes.

        For numerical simulation of the deformation and residual stress development in metals during laser peening, the constitutive equations describing material behavior at various strain rates and plastic properties at high strain rate impact conditions are prerequisite. However, these parameters are often unknown for various engineering materials. In this study, the parameters of Johnson-Cook constitutive equation of duplex stainless steel were determined by comparing the residual stress profile predicted by numerical simulation using trial values and the measured residual stress profile with x-ray diffraction.

        환경수 내의 다양한 Ca²+ 농도에 따른 시클리드(Maylandia lombardoi )의 생리적 변화

        We investigated physiological changes in the freshwater cichlid, fish Maylandia lombardoi in response to different calcium concentrations in rearing water. Four different calcium concentrations (0, 2, 4 and 8 mM) were prepared in rearing water by the addition of ionized calcium (CaO) to examine the effect of various calcium concentrations in rearing water on physiological changes in the cichlid fish, M. lombardoi. Total calcium concentrations in plasma and body increased in a calcium concentration-dependent manner and reached the maximum at 8-mM calcium concentra-tion. Stress-related cortisol was significantly decreased in the 8-mM group compared to the control group (0 mM). Lysozyme activity also significantly decreased in the 8-mM group. These results suggest that CaO in rearing water increases calcium uptake in fish and affects the body by decreasing stress and improving immunity in fish.

        성스테로이드 호르몬과 고수온 처리가 조피볼락 , Sebastes schlegeli 의 성분화에 미치는 영향

        조피볼락, S. schlegili 치어에 estradiol-17β (E₂)와 17α-methyltestosterone (MT) 그리고 고수온을 처리하여 성분화에 미치는 영향을 조사하였다. 실험어는 출산 후 56일부터 72일까지 E₂20, 40, 60㎍/g diet 농도로 경구투여 하였고, MT는 20, 50㎍/g diet 농도로 경구투여 하였다. 고수온 처리는 대조구 그리고 호르몬 처리구와 비교하여 약 10℃ 이상인 27.0±0.5℃로 사육하였다. 출산 후 56일까지 성분화 과정에서, 생식소는 대부분 생식원세포로 구성되어 있어 성적으로 미분화 단계였다. 그러나 출산후 128일째에, 난소는 난소강과 난소박판으로 구성되어 있고, 난원세포와 주변인기난모세포는 난소박판에 분포하고 있다. 그리고 정소는 많은 정세관으로 이루어 졌고, 이들 세관내에는 정원세포들이 무리를 지어 분포하고 있고 또한 색소포들이 정소의 수질층과 기부에 드문드문 분포하고 있다. 성비에 있어, 대조구와 비교하였을 때 모든 E₂처리구는 더 많은 암컷이 관찰되었고, 반면에, 모든 MT 처리구와 고수온 처리구에서는 더 많은 수컷이 관찰되었다. 이러한 결과들에서, 성스테로이드 호르몬의 농도와 종류 그리고 고수온 사육이 조피볼락의 성분화 과정에 있어서 성결정에 영향을 주고 있다고 볼 수 있다. This study investigated the effects of estradiol-17β (E₂), 17α-methyltestosterone (MT) and high temperature (WT) on gonadal sex differentiation in black rockfish, Sebastes schlegeli. Fish were reared to oral adminstration of E₂ at nominal concentrations of 20, 40 and 60㎍/g diet, and MT at nominal concentration of 20 and 50㎍/g diet from 56 days to 77 days after parturition. In the treatment of WT, water temperature of the breeding tanks ranged 27.0±0.5℃ more than 10℃ approximately, in comparison to the control and other experimental group. In the process of sex differentiation until 56 days after parturition, gonads were composed of mostly gonia cells, sexually undifferentiated. In contrast, 128 days after parturition, the ovaries were composed of ovarian cavity and lamellae, and oogonia and perinucleolus oocytes were distributed in the lamellae of ovary, and the testes were composed of a number of seminiferous tubule, and spermatogonia were distributed in the seminiferous tubule, and also melanophore scattered in the matrix layer of testis. In the sex ratio, more females than male were observed from E₂ treatment groups when compared to the control, but more males than females were observed from MT and WT treatment groups when compared to the control. In the results of the present study, the concentration and kinds of the sex steroid hormones, and also the rearing high temperature caused to the factor of sex determination in the process of sex differentiation of black rockfish.

        쏨뱅이(Sebastiscus marmoratus)의 Vitellogenin 분석을 위한 효소면역측정법(ELISA) 및 면역크로마토그래피분석법(ICG) 개발

        We tested biomarker systems [enzyme-linked immunosorbent assay (ELISA) and immunochromatography assay (ICG) kits] for the screening of endocrine-disrupting chemicals in contaminated environments using antibodies resulting from 17β-Estradiol-induced vitellogenin (Vtg) in the wild scorpion fish Sebastiscus marmoratus. Monoclonal antibodies of two clones (S28 and S15) were used as capture and tracer antibodies for ELISA and ICG assays. ELISA detected Vtg at levels greater than 0.1 μg/mL, while ICG detected Vtg at levels greater than 1 μg/mL. However, the ICG system was able to detect antibodies from 17β-Estradiol-induced Vtg serum that had been diluted 1,000 times. Our results suggest that previously developed biomarker assays can be used as detection systems to detect known endocrine-disrupting chemicals in contaminated environments, and to measure their activity

        감귤박 및 감태추출물의 사료첨가제 급여에 따른 둥근전복 (Haliotis discus discus)의 성장 및 생리적 변화

        Here, we report the physiological changes and growth in disk abalone, Haliotis discus discus, in relation to dietary supplementation with citrus pomace (CP) 6%, Ecklonia cava residue (ECR) 6%, and CP + ECR (3% + 3%). The composition and nutrient content, survival rate and growth rate were measured 0, 4, 8 and 12 weeks after feeding the supplemented diets of CP and/or ECR. Moreover, the experiment of low salinity stress (25psu) for environmental resistance was examined for a period of 48 hours after feeding the supplemented diets for 12 weeks. The activities of superoxide dismutase (SOD), catalase (CAT), lysozymes, respiratory burst, and phenoloxidase were measured. The moisture content and crude protein condition of the body were increased with the addition of ECR only (P<0.05). We observed higher levels of survival in the experimental group compared with the control group. Moreover, the growth disk abalone that were fed a diet containing ECR was higher compared with the control group. However, the growth of abalone fed a diet containing CP was similar to the control group. With a rearing condition of low salinity stress, survival rate and lysozyme activity were increased in the ECR group compared with the control group. Dietary ECR reduced the level of CAT activity to approximately 30% of the control, however the level of CAT activity in the ECR group was similar to the start level of the previous stress. These results suggest that dietary ECR gives rise to an enhanced immunity in disk abalone, as a result of the decrease in CAT and lysozyme activity in particular. Accordingly, the growth and survival rate were increased by feeding an ECR-supplemented diet in the rearing of disk abalone, Haliotis discus discus.

        저선량 60Co감마선 조사 후 생산된 둥근전복( Haliotis discus discus)치패의 초기성장 변화에 관한 연구

        We observed that the growth and physiological change in Haliotis discus discus by low-dose irradiation with gamma ray.Irradiation with gamma ray was undertaken by using the low-level irradiation facility (60CO) in cooperation with the Institute of Nuclear Science and Technology at Jeju National University. The parent abalones were attached one by one and then fixed by using the rubber band to the front side of the fabricated case for irradiation with gamma ray. The experimental plots of irradiation with gamma ray were set as 10, 15, 20 and 25 Gy and the 25 female abalones and 10 male abalones were utilized for each experimental plot. The sperms and eggs were fertilized by setting an interval for each dose to prevent mixing with other experimental plots when fertilizing the sperms and eggs for each dose of irradiation with gamma ray. As for the fertility, it was confirmed to be 85% the control and 10 Gy groups, whereas it was found to be 80%, 65% and 50% in the 15 Gy, 20 Gy and 25 Gy groups, respectively. As a conclusion, the hatching rate and attachment rate were higher at 10 and 15 Gy than the other experimental plots, and the growth rate was higher at 20 Gy than the other experimental plots. Also the changes in lysozyme activity in accordance with the stress of water temperature were found to have a significant increase in the other experimental plots as compared with the control plot at the end of 0 h. The changes in lysozyme activity have remained constant in all the experimental plots at the end of 12 h. These results allowed us to confirm that lysozyme was undertaking the biodefense action by reacting sensitively to the stress of water temperature in the control experimental plot. As for the other experimental plots, they are believed to avoid the biodefense mechanism due to the high degree of anti-parasite mechanism and anti-viral mechanism. Thus, it is believed that it would be imperative to conduct development and research on breeds that were potent for environmental tolerance by applying the method of irradiation with gamma ray to other marine animals and plants.

        사육수의 pH변화가 숭어(Mugil cephalus)에 미치는 생리적 영향

        We examined changes in the physiological responses of gray mullet Mugil cephalus exposed to acidic seawater (pH 6.0, 6.5, 7.0) and normal seawater (pH 8.0, control) for 15 days. As pH decreased, survival rate and body weight also decreased. Levels of aminotransferase, total protein and triglycerides also differed significantly with changes in pH, presumably due to stress caused by exposure to acidic water. The level of osmotic pressure was significantly higher in the pH 6.0 group than in other groups. Superoxide dismutase was significantly higher in the pH 6.5 and 7.0 groups than in the pH 8.0 group, and glutathione level was lowest in the pH 6.0 group. We conclude that decreasing the pH level of seawater induces a stress response in fish, damaging their ability to control their hematological and osmotic pressure. Antioxidant enzymes are generally sensitive to osmotic stress; in this study, antioxidant activity significantly changed with pH level. These results indicate that physiological stress induced by exposure to acidification reduces survival rates and inhibits growth in M. cephalus.
