We have developed a wavelength-swept fiber laser by using semiconductor optical amplifiers with different center wavelengths. The wavelength-swept laser can be utilized as a broadband source for a frequency-domain Optical Coherence Tomography (OCT) system. The wavelength-swept fiber laser was constructed with a dual cavity. Each laser having an identical cavity structure was governed by different lasing conditions, which resulted in a reduced insertion loss for the laser cavity. The two semiconductor optical amplifiers had the same gain bandwidth of 40 nm. The dual-cavity fiber laser had a 0.1-nm instantaneous spectral linewidth and a 1.2-mW average output power at wavelength of 1320 nm. The time-encoded Fourier domain OCT system with a dual cavity was used to image a rat eye and showed potential for application to biological imaging. 본 연구에서는 두 개의 서로 다른 중심파장을 가진 반도체 광증폭기를 이용하여 보다 넓은 대역폭을 가지는 광결맞음 단층 영상 시스템용 파장 가변형 광섬유 레이저를 개발 제작 하였다. 두 반도체 증폭기 간의 간섭을 최소화하고, 삽입 손실을 최소화하기 위하여 공진 구조를 단순화하여 최적화된 이중 공진 구조형 파장 가변 광섬유 레이저를 제작하였다. 두 반도체 광증폭기의 각각의 이득 대역폭은 약 40 nm 이며, 이중 공진 구조를 구성하였을 때 1320 nm 의 중심 파장 대역에서 최종 이득 대역폭은 약 60 nm 이다. 발진 레이저의 선폭은 약 0.1 nm 였으며, 평균 출력 세기는 약 1.2 mW 였다. 이를 바탕으로 광결맞음 단층 영상 시스템을 구성하였으며, 적출된 실험용 쥐의 안구에 대한 단층 영상을 획득하여 제안된 이중 공진 구조 파장 가변형 광섬유 레이저의 성능을 확인 하였다.
펨토초 레이저 기반의 광대역 파장 가변광원을 이용한 주파수영역의 광단층이미징

본 연구에서는 펨토초 펄스 레이저와 외부 가변필터를 이용한 광대역 파장 가변 광원을 제안하고 이를 이용하여 구현된 주파수 영역 OCT (optical coherence tomography)의 성능을 제시하고자 한다. 단일 모드 광섬유에 입사된 빛은 0.5 nm의 순간 선폭과 1 mW의 광세기를 나타내었으며 1 kHz의 반복률로 740 nm에서 850 nm의 파장대역을 가변 할 수 있었다. 6 $\mu$m의 고분능이 가능한 본 시스템은 두 개의 글래스 사이의 공기층을 구분할 정도의 성능을 가짐을 보였으며 펄스 레이저를 주파수 영역의 OCT에 활용하는데 있어서 매우 높은 가능성을 보여주었다. A novel wavelength-swept broadband source based on a femtosecond pulsed laser and an external tunable filter is proposed for the application of frequency-domain optical coherence tomography (FD-OCT). A laser beam coupled into a single-mode fiber, which provided a 0.5-nm instantaneous spectral linewidth with a 1-mW average output power, was tuned from 740 nm to 850 nm at a 1-kHz repetition rate. The system, with an axial resolution of 6 $\mu$m, was used to perform OCT imaging of the air gap between glass plates, revealing a promising potential for application of the pulsed laser source to FD-OCT.

To develop a biomarker predicting tumor treatment efficacy is helpful to reduce time, medical expenditure, and efforts in oncology therapy. In clinics, microvessel density using immunohistochemistry has been proposed as an indicator that correlates with both tumor size and metastasis of cancer. In the preclinical study, we hypothesized that vascular morphometrics using optical coherence tomography angiography (OCTA) could be potential indicators to estimate the treatment efficacy of breast cancer. To verify this hypothesis, a 13762-MAT-B-III rat breast tumor was grown in a dorsal skinfold window chamber which was applied to a nude mouse, and the change in vascular morphology was longitudinally monitored during tumor growth and metronomic cyclophosphamide treatment. Based on the daily OCTA maximum intensity projection map, multiple vessel parameters (vessel skeleton density, vessel diameter index, fractal dimension, and lacunarity) were compared with the tumor size in no tumor, treated tumor, and untreated tumor cases. Although each case has only one animal, we found that the vessel skeleton density (VSD), vessel diameter index and fractal dimension (FD) tended to be positively correlated with tumor size while lacunarity showed a partially negative correlation. Moreover, we observed that the changes in the VSD and FD are prior to the morphological change of the tumor. This feasibility study would be helpful in evaluating the tumor vascular response to treatment in preclinical settings.
