        공사현장의 품질관리를 위한 방수공사의 하자분석

        Waterproof work is closely related to the performance of the building structure and a falling-off in durability. This study focuses on finding out the defect characteristics and problems of waterproof construction classified by work types and construction methods, through waterproof work companies. The defections of construction method are classified into defect, design, construction, material, surface for working, and site control, in order to find out defect types and defect characteristics which will be used to construct devised the improvement plan for quality management of waterproof construction. The result of this study can be summarized as follows: there are many defections due to not observing the specific regulations, and after construction, defections caused by desquamation are shown in various places- Therefore, the improvement plan for quality management of waterproof construction is that adequate construction followed by technical specifications and construction workers' improvement in technical skill are necessary.

        흰쥐 재생간의 5´-Nucleotidase 및 Gamma-Glutamyl Transpeptidase의 활성치

        A study was made on the changes in activities of the followings during 6 post-operative days : Plasma membrane, microsomal, ,nuclear and cytosolic 5'nucleotidase and gamma-glutamy1 transpeptidase of regenerating rat livers, ant rat serum 5'nucleotidase and gamma-glutamy1 transpeptidase after 70% (median and left lateral lobes) partial hepatectomy. The activities of alanine aminotransferase in both serum and cytosol of regenerating rat liver were also measured. The activities of 5'-nucleotidase in the normal rats hepatic subcellular fractions were " 59.29±11.35(n mol Pi/mg protein/min)in the plasma membrane ; 52.86±3.88 in the microsome ; 10.56±2.12 in the mitochondria ;11.48±3.62 in the nuclei, and 3.55±0.54 in the cytosol. Those of gamma-glutamy1 transpeptidase were: 1.96±0.46 (n mol p-nitroaniline/mg protein/min) in the plasma membrane ; 3.38±0.62 in the microsome ; 0.88±0.12 in the mitochondria ; 1.40±0.13 in the nuclei, and 0.76±0.12 in the cytosol. Normal activity of alanine aminotransferase was 330±56 Karmen unit/mg protien in the rat hepatic cytosol. After partial hepatectomy in the rats, activities of plasma membrane bound gamma-glutamy1 transpeptidase in regenerating livers significantly increased in the span between the second and the third days but plasma membrane bound 5'-nucleotidase showed no change. The activity of microsomal 5'- nucleotiadase in the regenerating liver tremendously from the second to the sixth days after partial hepatectomy. And the activity of hepatic microsomal gamma-glutamy1 transpeptidase also elevated from the second and the third days after partial hepatectomy. Tomy activity of mitochondrial 5'-nucleotidase in the regenerating liver decreased between the third and the sixth days after partial hepatectomy. And the activity of hepatic microsomal gamma-glutamy1 trsnspeptidase also elevated from the second and the third days after partial hepatectomy. The activity of mitochondrial 5'-nucreotidase in the regenerating liver decreased between the third and the sixth days of partial hepatectomy. The hepatic nuclear and cytosolic 5'-nucleotidase decreased in the regenerating stage. The decreased values were seem after two days in cytosolic 5'-nucleotidase and after three days in nuclear 5'-nucleotidase following the operations. However, no significant changes in hepatic mitochondrial, nuclear and cytosolic gramma-glutamy1 were ntoed through out the experiments. The activity of serum alanine aminotransferase markedly elevated from 12 hours to three days after partial hepatectomy, but serum 5'-nucleotidase and gamma-glutamy1 transpeptidase activities remained normal.

        지방자치단체의 공공디자인진흥계획 수립에 관한 연구 : 경상남도의 권역별 특성에 따른 범주화 방안

        우리나라 공공디자인은 20여 년이 넘는 시행착오의 과정에서 체계와 법률적 바탕이 정립되어 왔지만, 공공디자인의 기존계획이 기본적인 방향이나 본질적인 문제를 개선하기보다는 가시적인 효과를 얻을 수 있는 방안만을 주목하고, 공공디자인의 개념 및 범위가 각 기관마다 내용이 서로 다름으로 공공디자인 계획의 전반적인 변화가 필요하게 되었다. 과거 하드웨어적인 접근에서 소프트웨어적인 접근으로 디자인에 대한 사회적인 변화와 산업적인 변화가 확장됨에 따라 여러 분야에서 문제해결을 위하여 시대에 맞는 디자인 도입이 필요하다. 따라서 지자체 공공디자인 진흥계획에 대한 면밀한 이해는 필수적이라 판단되며, 이에 대한 사례 분석과 현장설문 및 인터뷰를 통해 공공디자인 정책의 기본목표와 추진 방향을 연구하였고, 역사·문화, 지역경제, 생활 등을 고려한 공공디자인 정책의 방향성을 제시했으며, 공공디자인의 향상을 위한 전략 및 세부방안을 제안하고 공공디자인 진흥에 관한 제도 개선사항 및 주요사업들을 발굴하여 시·군과 도의 공공디자인 진흥계획의 역할 및 연계실행 방안을 제시하였다. Public design in Korea has been established in the process of trial and error for more than 20 years, but the existing plan of public design is a way to obtain visible effects rather than approached basic directions or essential problems for improvement. As the content of the concept and scope of public design by institution is different, it is necessary to change the public design plan. Because of social and industrial changes in design froms the previous hardware approach to software approach expand, it is necessary to introduce designs for problem to solve in various fields. Therefore, a deep understanding of the public design promotion plan for local governments is considered essential, and entire case analysis, field surveys, and interviews, the basic goals and directions of public design policies were studied. And history, culture, life, and local economy were studied. The direction of public design policy in consideration, proposed strategies, plans, and detailed plans for public design improvement, and identified system improvements and major projects related to public design promotion were suggested. In addition the role and connection of the city/county public design promotion plan were The implementation plan was also presented.

        주정 중독 흰쥐에서 총담관 결찰이 간의 Mitochondria 및 Microsome의 총 Monoamine oxidase와 혈청의 Benzylamine oxidase활성에 미치는 영향

        The activities of the hepatic mitochondrial and microsomal total monoamine oxidase(MAO), and serum benzylamine oxidase were studied for cholestasis induced by common bile duct ligation and acute ethanol intoxication developed, or cholestasis after chronic ethanol intoxication for manifestation of the biochemical background of alcohol intoxication in hepatobiliary disease. There were marked increases in the mitochondrial and microsomal total MAO activities of the liver in the rats treated with chronic ethanol intoxication, and showed a significant increase in the mitochondrial total MAO after treatment with acute ethanol intoxication. When common bile duct was ligated in the rats, mitochondrial total MAO activity in the liver significantly decreased. When common bile duct was ligated after chronic ethanol intoxication and acute ethanol intoxication was done after common bile duct ligation, the activity of the mitochondrial total MAO in the liver decreased more significantly than the group only with ethanol intoxication was performed. However, the activity showed a higher degree on the same time points than the groups only with the common bile duct ligation. The serum benzylamine oxidase ectivity in the group with only common bile duct ligation or the ethanol intoxication with cholestasis were measured between the 7th and 14th day, with no difference between two groups. According to the above results, the hepatic MAO seems to be an enzyme in which its activity increases in acute and chronic ethanol intoxicaion. And total MAO in the liver is the enzyme with increased activity in ethanol intoxication with cholestasis more than in cholestasis. The cause of the increase is the development of biosynthesis. Accordingly, these results will be the data supporting that alcoholic drink is enzymologically harmful in hepatobillary disases.

      • Testosterone 처리에 의한 넙치, Paralichthys olivaceus 난소에서 doublesex-and mab-3-related transcription factor-1 (DMRT-1) mRNA의 발현 유도

        본 연구에서는 넙치(Paralichthys olivaceus) 정소로부터 DMRT-1 partial cDNA를 분리하였다. 넙치 DMRT-1은 317개의 염기로 구성되어 있으며, Atlantic halibut, 감성돔 및 무지개 송어와 각각 94%, 85%, 82%의 높은 상동성을 보였다. RT-PCR을 이용하여 DMRT-1 mRNA 발현은 난소보다 정소에서 높게 나타난 것을 관찰할 수 있었다. 또한, 암컷 넙치에 웅성호르몬인 testosterone 처리시 처리 농도에 따라 난소에서 DMRT-1 mRNA 발현이 증가함을 볼 수 있었다. 따라서, DMRT-1은 수컷에서 특이적으로 발현되는 유전자임을 알 수 있으며, 본 연구의 결과는 넙치의 성 전환 및 유도에 대한 기초자료를 제공할 것이다. We isolated a 317 bp of partial cDNA for doublesex-and mab-3-related transcription factor-1 (DMRT-1) from the testis of olive flounder, Paralichthys olivaceus using RT-PCR. Based on the multiple sequence alignment, olive flounder DMRT-1 shared relatively high sequence homology (82 to 94%) with orthologues from other teleost species such as Atlantic halibut, Hippoglossus hippoglossus, black porgy, Acanthopagrus schlegeli and rainbow trout, Oncorhynchus mykiss. DMRT-1 mRNA was predominantly expressed in the testis of olive flounder. In our investigation for the effect of testosterone treatment in vivo on induced expression of ovarian DMRT-1 transcript, mRNA levels of DMRT-1 in ovary were significantly up-regulated by testosterone treatments (0.3 or $3.0{\mu}g$ testosterone/g body weight for 12 to 36 hours) as judged by RT-PCR analysis. In overall, transcriptional stimulation of DMRT-1 during treatments was more affected by doses of testosterone than treatment durations. This result strongly suggests that the regulation of DMRT-1 be tissue- and gender-specific in olive flounder, and also provides useful baseline knowledge on the testosterone-mediated regulation in the reproductive physiology of this species.

        주정 중독 흰쥐에서 총담관 결찰이 혈청 및 간의 Lactate Dehydrogenase활성에 미치는 영향

        The activities of the liver and serum lactate dehydrogenase were studied when cholestasis was induced by common bile duct(CBD) ligation in rats and chronic ethanol intoxication developed, or cholestasis following acute ethanol intoxication for manifestation of the biochemical background of alcohol intoxication in hepatobiliary disease. There was no significant change in the liver and serum lactate dehydrogenase activities of rats treated with both acute or chronic ethanol intoxition. The liver lactate dehygrogenase activity showed a significant decrease at the 2nd , 3rd, 7th and 14th days follwing the ligation in both the group with the ligation follwing the chronical ethanol intoxication and the groups only with CBD ligation. The acute intoxication with ethanol which was done after 14 days of the CBD ligation, the rats liver lactate dehydrogenase activities decreased considerably and the same seen in the group with the 14th day following the CBD ligation. The groups that received CBD ligation after being chronically intoxicated with ethanol showed a marked increade at the 1st, 2nd, 3rd, 7th and 14th days floolwing the ligaion in the sera lactate dehydrogenase activities. However, the activities showed a far higher degree than groups of the CBD ligation. For the groups of acute intoxication with ethanol done after 14 days of the CBD ligation, the sera lactate dehydrogenase activities increased markadly, but the activities showed a higher degree than the group with the 14th day following the CBD ligation. According to the above results, when the acute and chronic ethanol intoxication with cholestasis occurred, the sera lactate dehydrogenase activities are higher than in cholestasis because of increaded liver cell damage. which causes the enzyme to leak into the blood in great quantity. According, these results will be the data supprorting alcoholic drink is enzymologically harmful in hepatobiliary disease.

        주정 중독 흰쥐에서 총담관결찰이 간 및 혈청의 Malate Dehydrogenase 활성에 미치는 영향

        The activities of the liver and serum malate dehydrogenase(MDH) were studied when cholestasis was induced by common bile duct ligation and chronic ethonic intoxication developed, or cholestasis following acute ethanol intoxiaction in hepatobiliary disease. The rats liver cytosolic MDH activities showed a significant increase in both chronically ethanol intoxicated groups and acutely ethanol intoxicated groups. But the rats liver mitochondrial MDH activities showed no significant changes in both chronically ethanol intoxiacted groups and acutely ethanol intoxicated groups. The groups that received common bile duct(CBD) ligation after being chronically intoxicated with ethanol showed a considerable decrease at the 2nd, 3rd, 7th and 14th days following the ligation in the liver cytosolic MDH activities. However, the activities showed a higher degree than groups of the CBD ligation. On the other hand, the liver mitochondrial MDH activities showed a significant decrease at the 3rd, 7th and 14th days following the ligation, but the activities showed a higher degree on the same time points than the groups only with the CBD ligation. At the 1.5th and 24th hours following the acute intoxication with ethanol done after 14days of the CBD ligation, the rats showed a slight decrease in the liver cytosolic and mitochondrial MDH activities than the goup only with the acute ethanol intoxication. However, the activities showed more remarkable increase than the goup sacrificed on the 14th day following the CBD ligation. The groups that receved CBD ligation after being chronically intoxicated with ethanol showed a dramatic increase at the 1st, 2nd, 3rd, 7th and 14th days following the ligation in the sera MDH activities. However, the activities showed a far higher degree than groups of the CBD ligation. For the goups of acute intoxication with ethanol done after 14 days of the CBD ligation, the sera MDH activities increased markedly, but the activities showed a higher degree than the goup with the 14th day following the CBD ligation. According to the above results, the liver cytosolic and mitochondrial MDH seems to be an enzyme which increases activities in both acute and chronic ethanol intoxication with cholestasis more than in cholestasis; the cause of the increase seems to be the development of biosynthesis. Especially, when the acuted and chronic ethanol intoxication with cholestsis occurred, the sera MDH are higher than in chlestasis because of increased liver cell damage, which causes the enzyme to leak into the blood in great quantity. Accordingly, these results will be the data supporting that alcoholic drink is enzymologically harmful in hepatobiliary disease.
