        전기영동에 의한 황어의 계통발생 단계 및 성체 조질들에서 동위효소의 유형

        Developing embryos of dace, Tribolodon hakonensis, were analyzed with regard to both gross morphological changes and specific enzymatic changes from the unfertilized egg stage until some 3 week posthatching. The patterns of gene expression among the different tissues in adults were also investigated. The tissue and developmental expression of 13 loci encoding various enzymes and isozymes were visualized by histochemical staining after polyacrylamide gel electrophoresis. Distribution of enzymes and isozymes in the adult tissues usually show tissue specific patterns. The expression of LDH-C and G6-PD-1 loci was the more restricted than the other loci, and these loci were expressed only predominantly in the liver. Many of the enzyme systems investigated exhibited rich patterns (all loci except for MDH-1A locus) of ontogenetic change, while a few (MDH-1A) remained relatively unchanged throughout the period studied. The appearance of several rather tissue - specific isozymes (LDH-A, C and MDH-1B) was closely correleated with the morphological and functional differentiation of the muscle, liver and gill tissue.

      • 마우스의 악하선 세포의 분리 및 배양조건 확립

        마우스의 악하선 세포를 배양하기 위하여 악하선 조직으로부터 세포를 분리하는 조건과 분리된 세포의 배양조건을 조사하였다. 세포분리에는 0.25% trypsin을 사용하였으며 배양액은 여러 농도의 fetal bovine serum (FBS) 또는 low protein serum replacement(LPSR)가 첨가된 Dulbecco's modified Eagle's medium(DME) 이었다. 배양된 세포의 대부분 상피형 세포로 확인 되었으며, 배양시 5-10%의 FBS를 첨가하였을 경우에 가장 높은 DNA합성능을 보였으나 이보다 높은 농도의 FBS 첨가시에는 오히려 DNA 합성능이 저하되었다. 혈청 대체물인 LPSR 첨가에 의한 악하선 세포를 배양 했을 때 population doubling time은 42.5 시간이었고 세포의 포화밀도는 1.2 $\times$10 5 cells / $cm^2$이었다. Dihydrotestosterone (DHT)은 악하선 배양세포의 DNA 합성에 관여하지 않거나, 관여한다면 DNA 합성을 억제하는 것으로 보였다. 이에 반해 Thyroxine (T4)은 악하선 배양세포의 DNA 합성을 현저히 증가시켰다. T4와 DHT 모두다 배양세포의 단백질 합성능을 증가시켰다. 또한 이 호르몬들이 악하선 배양세포의 epidermal growth factor(EGF) 분비를 증가시킨 결과는 DHT와 T4는 악하선 세포의 EGF 생산 뿐만 아니라 EGF 분비의 조절에도 관련되어 있음을 시사한다. 본 실험에서 정해진 악하선 세포의 배양조건은 악하선 세포의 증식과 기능의 변조를 탐구하는데 유용하게 응용될 수 있을 것으로 생각된다. The purpose of this research was to establish the culture condition for dissociated submandibu -lar gland (SG) cells. After trypsin digestion of SG from 3-4 weeks old mice, dissociated cells were cultured in 1OO/o fetal bovine serum-Dulbecco's modified Eagle's medium (FBS-DME) or 0.5-2% low protein serum replacement-DME (LPSR-DME) on plastic surface to form monolayer. The effects of FBS, LPSR and hormones on the growth and function of cultured SG cells were examined. SG cells dissociated by enzyme were successfully cultured and were characterized as epithelial-like cells by light and electron microscope. The maximal DNA synthesis of cultured SG cells was achieved by DME containing 5-10% FBS. The same results were obtained when the effects of LPSR on cell proliferation were examined up to a LPSR concentration of 2%. SG cells cultured in 20/o LPSR-DME expressed a population doubling time of 42.5 hrs and a saturation density of 1.2 $\times$10 5cell/cm$^2$. Dihydrotestosterone (DHT) in medium did not influence on the DNA synthesis of the cultured SG cells, but stimulated protein synthesis of the SG cells. Thyroxine (T4) stimulated protein synthesis of the SCI cells markedly in a dose-dependent fashion. EGF secretion by the cultured SG cells increased significandy by DHT and or T4 trearment. This finding indicated that secretion of EGF by the SG cells was under the control of the hormones such as androgen and thyroid hormones. It seems to be that the culture condition described here can be used as a useful tool for further research on the SCI cells.

      • 복합생약제 가미육군자탕이 3T3-L1 세포의 증식과 분화에 미치는 영향

        The purpose of this research was to investigate the effect of Kami Yukkunja Tang(KYT) on the proliferation and differentiation of preadipocytes, 3T3-L1, and to establsh the in vitro screeninig method for the effects of combined preparation of crude drugs. The extract of KYT inhibited the proliferation of 3T3-L1 at concentrations of 10^-4 to 10^-3g/ml, but enhanced at 10^-7g/ml. The adipose conversion of 3T3-L1 was decreased by treatment of 10^-4g/ml of KYT extract in 2 days of initial-culture and by 10^-7. 10^-6g/ml of KYT in 5 days of final-culture. In spontaneous differentiation, KYT extract increased the adipose conversion at the concentrations of 10^-7 to 10^-4g/ml in 2 days of initial culture and in 7 days of whole term of culture, and in 5 days of final-culture, KYT extract increased the differentiation at higher concentrations (10^-5 and 10^-4g/ml). The proliferation and differentiation of 3T3-L1 were inhibited significantly by the 5% and 10% serum(KTY-serum) from KYT-treated mice. The protein synthesis of 3T3-L1 cells at confluent stage was decreased by KYT-serum significantly but not by KYT extract. These findings suggest that KYT and KYT-serum have potencies in interfering with the process of adipocyte differentiation and the assay system described in this paper can be used as a useful tool for further research on the adipose conversion by several other combined preparations of crude drugs.

      • Brain microsomal ATPase activity and effects of HgCl_2 on it during chick embryo development

        본 연구에서 발생중이 계배(Chick embryo)로부터 분리된 뇌 소포체막에 붙어있는 ATPase의 일반적인 특성과 그에 미치는 Hg ion의 영향에 대하여 알아보았고 CBM ATPase activity에 미치는 Hg ion의 영향과 그 기작을 밝혀보고자 Dixon, Lineweaver-Burk plot 등을 작성하였다. 본 연구의 결과는 다음과 같이 요약된다. 1. 발생중인 여러 시기의 ATPase activity는 서서히 증가하였으며, 0.9∼1.1μM, Pi/㎎ Protein/10min의 범위에 있었다. 2. CBM ATPase는 Ca^2+와 Mg^2+에 의하여 활성화 되고 4mM MgCl^2의 존재하에서 Ca^2+의 최적농도는 100μM이었다. 3. 효소의 activity에 미치는 Hg^2+의 저해효과는 Hg^2+의 농도가 증가함에 비례하였다. 4. Hg에 의한 50% 저해는 Hg^2+의 농도가 10∼25μM일 때 일어났으며, 이 값은 Dixon plot에서 얻은 Ki값과 거의 일치하는 것이다. 5. Lineweaver-Burk plot에서 얻은 ATP의 Km값은 1.2mM이었다.

      • 子官癌細胞(HeLa)에 대한 血府逐瘀湯과 數種 抗癌劑의 倂用投與 效果

        The studies were conducted to investigate the combined effects of Hyulbu Chuko Tang (HCT) and several anti-cancer drugs. The effects of HCT and several anti-cancer drugs on the proliferation of HeLa Colls, human cervical cell line, was estimated by MTT colorimetric assays. The 50% inhibitory concentrations of mitomycin C(MMC), cisplatin(CPT), mercaptopurine (MCP) and 5-fluorouracil(5-FU) on HeLa cell were 0.313, 0.565, 22.745 and 28.325/㎖, repectively. HCT extract inhibited the proliferation of HeLa cell at 10^-6-10^-3g/㎖. The inhibitory action of the MCP treated group on the proliferation of HeLa cell was increased by HCT and MCP. When the mice were treated by MMC, the number of leukocyte was decreased significantly at the 1st and 3rd day, but recovered at the 7th day. In the groups of MMC treated with HCT, the number of leukocytes was increased significantly than the group of MMC treated only at the 1st and 3rd day. The HCT extract increased the number of plaque forming cells(PFC), but the MMC treated group decreased the number of PFC. The combined treatment of MMC and HCT increased the number of PFC significantly than the MMC treated group. The HCT extract increased the proliferation of T cells, but the MMC treated group decreased the proliferation of T cells. The combined treatment of MMC and HCT increased the T cell proliferation significantly than MMC treated group.

        한국산 Gobiid fish 16 종의 핵형

        A karyological analysis was carried out on different Korean species of the family Gobiidae (10 genera, 3 subfamies). The preparations were made according to the flame drying method, and the karyotypes were analyzed by conventional Giemsa staining. Diploid chromosome number ranged between 40 and 50 (Acantho-gobius hasta, 44; A. sp., 42; A, lactipes, 40; A. flavimanus, 44; Acentro-gobius pflaumi, 50; Rhinogobius brunneus, 44; Chasmichthys dolichognathus, 44; C. gulosus, 44; Chaenogobius annularis, 44; C. castaneus, 42; C. mororanus, 42; Mugilogobius abei, 46; Favonigobius gymnauchen, 48; Odontobutis platycephala, 44; Tridentiger obscures, 44; T, sp., 44), and arm number was in the range of 40-96. Some detected chromosomal characteristics permitted establishing some karyotypic relationships among the different species investigated.

        Effect of glycyrrhizin on nicotine-induced behavioral disturbance of planarian, Dugesia japonica

        Nicotine is a component of tobacco products and is one of the most commonly abused substances that leads to addiction. Therefore, the present study was performed to investigate the behavioral pattern and toxicity by nicotine exposure in planarians. Basically, planarians were exposed to different concentration of nicotine for 5 min. To investigate detoxification effect, planarians were exposed to nicotine for 5 min, and treated glycyrrhizin for 5 min, then motility and seizure-like behavior were observed for 5 min. As a result, the motility of nicotine-exposed planarians decreased approximately more than 50% compared to freshwater control. However, the motility of glycyrrhizin-exposed planarians recovered than nicotine- exposed planarians. In the assessment of seizure-like behavioral pattern, planarians exposed to nicotine showed head-bop or c-like type rather than screw-like or snake-like patterns. However, planarians exposed to glycyrrhizin showed no seizure-like behavior. To examine the oxidative stress response, planarians were cultured in fresh water containing 1 mM nicotine for 1 day. Planarians were homogenized and extracted to assay the contents of reactive oxygen species (ROS), lipid hydroperoxides (LH), glutathione (GSH), catalase (CAT), and superoxide dismutase (SOD). The result showed that a significantly higher level of ROS, LH indicated in planarians exposed to nicotine, on the other hand, glycyrrhizin-exposed planarians were significantly decreased ROS, LH levels. In conclusion, the motility decreased when planarians were exposed to nicotine, in a dose-dependent manner, whereas seizure- like behavior increased. Nicotine induced behavioral disturbances and cell toxicity in planarians were recovered by glycyrrhizin, suggesting a candidate substance for nicotine addiction treatment.

        항암제 Mitomycin C의 부작용에 대한 加味十全大補湯의 영향

        The purpose of this research was to investigate the effect of Kamisipjeondaebo-tang water extract (KSDT) on an adverse reaction of mitomycin C (MMC), antitumor drug. MMC decreased the cell viability and the production of cytokines, such as interferon-γ, interleukin-2 and interleukin-4 in murine thymocytes, but KSDT enhanced the cell viability and the production of interferon-γ. In addition, MMC increased the production of tumor necrosis factor-α and nitric oxide secreted from peritoneal macrophages, but KSDT decreased its effects. Also, KSDT increased the phagocytic activity decreased by MMC in peritoneal macrophages. KSDT did not recover the number of leucocytes decreased by MMC. These results suggest that KSOT may be reduce the adverse reaction of MMC, such as immune-suppressive action, via increase of cell viability and interferon-γ production in thymocytes and phagocytic activity in peritoneal macrophages.
