Antiinflammatory activities of the solvent fractionates of several herbal medicines were investigated and SKI306X was prepared from the active principles of three herbal medicines, Prunella vulgaris, Trichosanthes kirilowii and Clematis mandshurica. SKI306X was shown to have strong inhibitory effects on acetic acid-induced pain, carrageenan-induced paw edema and adjuvant-induced arthritis. LD50 of SKI306X was more than 5g/kg in rat, so generally nontoxic. Chemical analysis revealed that oleanolic acid and rutin, which are known to have various antiinflammatory activities, were contained in it. These results suggest SKI306X may become a useful drug for the treatment of inflammatory diseases such as rheumatoid arthritis.

The acetone precipitates of the hot water extract of Torilis fructus showed strong hemostatic activity which was not inhibited by aspirin. This activity was not through platelet activation but possibly through activating some coagulation factors in plasma. The hemostatic action of the precipitates was not active by oral adminitration and no behavioral toxicity appeared in treated mice. However, mice treated with the acetone precipitates through tail vein showed serious tremor and then were killed probably by the thrombus produced in the body. The hemostatic activity was still remained after treatment with beta-glucosidase, beta-galactosidase, alpha-amylase, subtilisin BPN'', or trypsin but completely lost by acid hydrolysis. The active components seemed to be a complex of unidentified macromolecules to which some phenolic compounds were strongly bound.

SKI306X is a herbal extract prepared from three herbs Clematis mandshurica, Trichosanthes kirilowii and Prunella vulgaris. It showed strong antiinflammalory actions on carrageenan-induced edema, acetic acid-induced pain, adjuvant-induced arthritis, and oxygen radical-generated reactions. In this study, the acute toxicity of SKI306X was evaluated in rats by a single oral administration. Thirty male and thirty female rats were divided into 6 groups according to the dose levels, respectively. After oral administration of SKI306X with several doses (5.0 g/㎏, 3.3 g/㎏, 2.2 g/㎏, 1.5 g/㎏, 1.0 g/㎏), mortality, clinical signs, body weight, and gross findings in organs were examined. No toxic effect was shown in terms of mortality, clinical signs, body weight changes and gross findings. It is suggested the LD_(50) of SKI306X would be more than 5.0 g/㎏ in rats.

For developing plant anti-inflammatory agents, extracts from Lonicera japonica were obtained and evaluated for their anti-inflammatory and analgesic activities using acute/chronic inflammatory models and writhing tests. It was found that the extracts with a modified extraction method showed higher anti-inflammatory and analgesic activities than those of the extracts based on the ancient literatures.

Anti-inflammatory and analgesic activities and skin irritation of piroxicam patch were investigated. Piroxicam patch increased the pain threshold in rat hind paw inflamed by carrageenan and inhibited writhing induced by acetic acid in mice. Piroxicam patch also inhibited the carrageenan-induced edema in rat hind paw as well as the increased vascular permeability induced by histamine in rats. In adjuvant arthritis of rats, piroxicam patch showed anti-inflammatory effects. Skin irritation of piroxicam patch was tested in Newzealand White rabbits and evaluated by Primary Irritation Index of Draize. The results from skin irritation test showed that piroxicam patch seemed practically non-irritating. The result from the present study indicates that piroxicam may be useful without serious side effects as anti-inflammatory analgesics in this patch form.

Type 2 diabetes mellitus (T2DM) is characterized by insulin resistance (IR). T2DM is correlated with obesity and most T2DM medications have been developed for enhancing insulin sensitivity. Silk protein fibroin (SPF) from spiders has been suggested as an attractive biomaterial for medical purposes. We generated transgenic rice (TR) expressing SPF and fed it to diabetic BKS.Cg-m+/+Leprdb mice to monitor the changes in blood glucose levels and adipose tissue proteins associated with energy metabolism and insulin signaling. In the present study, the adipocyte size in abdominal fat in TR-SPF-fed mice was remarkably smaller than that of the control. Whereas the adenosine monophosphate-activated protein kinase (AMPK)-activated protein kinase and insulin receptor substrate 1 (IRS1) protein levels were increased in abdominal adipose tissues after TR-SPF feeding, levels of six-transmembrane protein of prostate 2 (STAMP2) proteins decreased. Phosphorylation of AMPK at threonine 172 and IRS1 at serine 307 and tyrosine 632 were both increased in adipose tissues from TR-SPFfed mice. Increased expression and phosphorylation of IRS1 at both serine 307 and tyrosine 632 in adipose tissues indicated that adipocytes obtained from abdominal fat in TR-SPF-fed mice were more susceptible to insulin signaling than that of the control. STAMP2 protein levels decreased in adipose tissues from TR-SPF-fed mice, indicating that STAMP2 proteins were reducing adipocytes that were undergoing lipolysis. Taken together, this study showed that TR-SPF was effective in reducing blood glucose levels in diabetic mice and that concurrent lipolysis in abdominal adipocytes was associated with alterations of AMPK, IRS1, and STAMP2. Increased IRS1 expression and its phosphorylation by TR-SFP were considered to be particularly important in the induction of lipolysis in adipocytes, as well as in reducing blood glucose levels in this animal model.
