SKI306X is a herbal extract prepared from three herbs Clematis mandshurica, Trichosanthes kirilowii and Prunella vulgaris. It showed strong antiinflammalory actions on carrageenan-induced edema, acetic acid-induced pain, adjuvant-induced arthritis, and oxygen radical-generated reactions. In this study, the acute toxicity of SKI306X was evaluated in rats by a single oral administration. Thirty male and thirty female rats were divided into 6 groups according to the dose levels, respectively. After oral administration of SKI306X with several doses (5.0 g/㎏, 3.3 g/㎏, 2.2 g/㎏, 1.5 g/㎏, 1.0 g/㎏), mortality, clinical signs, body weight, and gross findings in organs were examined. No toxic effect was shown in terms of mortality, clinical signs, body weight changes and gross findings. It is suggested the LD_(50) of SKI306X would be more than 5.0 g/㎏ in rats.

Objectives. This study surveyed medical students attending clerkship to assess their attitude toward and knowledge about breastfeeding, and self-confidence to manage common breastfeeding problems. Methods. A self-report questionnaire was administered to the 323 medical students at four medical colleges in Taegu in May 1997. The response rate was 92.9%, but the respondents used in the final data analysis were 245 (75.8%) due to missing variables. Results. Overall, respondents showed equivocal attitudes toward breastfeeding (mean score 2.9 on a 6-point scale). Knowledge about breastfeeding was substantially low with the median % correct 39.2%. Of nine knowledge areas, weak areas were especially 'contraindications and barriers to breastfeeding', 'use of breastfeeding aids', 'expression and storage of breast milk'. Those reported to be confident to manage common breastfeeding problems were only 25.7%. Correlations between knowledge and self-confidence were not statistically significant except in college A (r=0.35, p<0.05). Correlations between knowledge and attitude were not significant. Conclusions. These results suggest that medical students attending clerkship in Taegu show equivocal attitudes toward breastfeeding, low self-confidence to manage common breastfeeding problems and have substantially limited level of knowledge. There should be more concerted efforts to improve this situation on the part of those involved in breastfeeding education.

This study was performed to determine the subacute toxicities of SKI306X, an antiinflammatory herbal extract, in rats. SKI306X was administered orally to rats once a day for 4 weeks at doses of 0.3, 1.0, and 3.0 g/㎏/ day. Each group consisted of 20 male and 20 female rats, including 5 male and 5 female rats per group for an interim study at the end of 2-week administration and for a 2-week recovery study, respectively. Throughout the study, all rats survived and no adverse clinical signs were observed. Although male rats treated with high dose (3.0 g/㎏/day) of SKI306X showed slight loss of body weight (approximately 5%) in comparison with control animals during the administration period, their body weight loss was normally restored during the recovery period. No significant change was found in all hematological parameters of SKI306X-treated groups except for the decreased number of red blood cells in all female groups at the interim study. Statistically significant changes were observed in several blood enzyme levels of SKI306X-treated groups; however, most of these significant changes were within normal range and statistically significant values did not show dose-related responses. In SKI306X-treated groups, the absolute and relative weights of liver, heart, and stomach were statistically different from those of control group, but these differences disappeared at the end of recovery period and also drug-related gross and histopathological findings in these organs were not found. No other drug-related gross and histopathological findings were observed. It is concluded from the results of this study that non-toxic dose of SKI306X was estimated to be between 0.3 and 1.0 g/㎏/day and the maximum tolerated dose of SKI306X was assumed to be higher than 3.0 g/㎏/day.

Anti-inflammatory and analgesic activities and skin irritation of piroxicam patch were investigated. Piroxicam patch increased the pain threshold in rat hind paw inflamed by carrageenan and inhibited writhing induced by acetic acid in mice. Piroxicam patch also inhibited the carrageenan-induced edema in rat hind paw as well as the increased vascular permeability induced by histamine in rats. In adjuvant arthritis of rats, piroxicam patch showed anti-inflammatory effects. Skin irritation of piroxicam patch was tested in Newzealand White rabbits and evaluated by Primary Irritation Index of Draize. The results from skin irritation test showed that piroxicam patch seemed practically non-irritating. The result from the present study indicates that piroxicam may be useful without serious side effects as anti-inflammatory analgesics in this patch form.
Elimination of Lead by TTFD and TPD from Central Nervous System of Postnatally Lead - exposed Rats

Amount of lead burden in a tissue reflects poisoning of lead in that tissue, so is the removal of lead directly connected to curement of lead poisoning. The purpose of present study was to investigate the relative effects of penicillamine and thiamine tetrahydrofurfuryl disulfide (TTFD) or thiamine propyl disulfide (TPD) in the removal of lead from rat brain tissue treated with excessive lead. Wistar rat pups of both sexes were used in this experiment. Within 1 day of parturition, experimental mothers nursing their pups as well as rat pups were given drinking water containing 0.2% lead acetate, TTFD 20 mg/1.2 L (2 mg/kg/day), TPD 20 mg/1.2 L (2 mg/kg/day), penicillamine 40 mg/1.2 L (40 mg/kg/day), 0.2% lead acetate+TTFD 20 mg/1.2 L (2 mg/kg/day), 0.2% lead acetate+TPD 20 mg/1.2 L (2 mg/kg/day) or 0.2% lead acetate+penicillamine 40 mg/1.2 L (40 mg/kg/day) ad libitum, throughout the entire period of experiment. Rat pups in the control group received normal tap water. The animals were sacrificed by decapitation on the day when they become 2 or 8 weeks of age. Brains were dissected into five regions: telencephalon, diencephalon, midbrain, pons/medulla and cerebellum. The dissected brain tissues were lyophillized and then solubilized by acid mixture (nitric acid+sulfuric acid). Lead levels in the solubilized brain tissues were measured by the inductively coupled plasma. In lead-exposed rats, lead levels were significantly higher than those of control group in all brain regions, lead levels in brain regions of TTFD or TPD group were generally lower than those of control group. The simultaneous administration of lead with TTFD or TPD to animals caused significant decrement of lead from all brain regions. In the elimination of lead from brain regions, effectiveness of TTFD or TPD was equivalant to penicillamine.
