The anti-inflammatory effects of 4'‑O‑methylalpinumisoflavone (OMAI) has been reported in recent years. To develop effective and safe skin whitening agents, we investigated the anti-oxidative and melanogenic effects of OMAI isolated from fruit of Maclura tricuspidata Carrière (Cudrania tricuspidata) in macrophage and melanoma cell lines. In our results, OMAI showed effective superoxide scavenging activity and suppressed production of lipopolysaccharide (LPS)-induced intracellular reactive oxygen species (ROS) in RAW264.7 cells. In addition, α-melanocyte stimulation hormone (MSH)-induced production of melanin was also reduced by OMAI in B16F10 cells. Finally, OMAI significantly inhibited tyrosinase activity in B16F10 cells. These results suggest that OMAI suppressed melanin production via scavenging reactive oxygen species and inhibition of tyrosinase activity.

In previous report, Seungmagalgeun-tang (SGT) could exert its anti-inflammatory actions in the BV-2 microglial cells. However, study on the anti-inflammatory effect of SGT in mast cells has not been identified. Therefore, we examined on the anti-inflammatory effect of SGT on the phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187-induced rat basophilic leukemia (RBL-2H3) cells. SGT inhibited the release of β-hexosaminidase and secretion and expression of pro-inflammatory cytokines such as tumor necrosis factor (TNF)-α and interleukin (IL)-4 on RBL-2H3 cells, without affecting cell viability. The protein expression level of nuclear factor (NF)-κB (p65) was decreased in the nucleus by SGT. In addition, SGT suppressed the degradation of inhibitory protein IκB-α protein, the activation of p38 mitogen-activated protein kinase (MAPK), and the expressions of cyclooxygenase (COX)-2 mRNA and protein level in RBL-2H3 cells. These results suggest that SGT could be involved anti-allergic effect by control of NF-κB (p65) translocation into the nucleus through inhibition of IκB-α degradation and suppression of COX-2 expression.

Sophora flavescens, as a traditional herbal medicine, has been used to treat with a variety of disesases. In previous reports, S. flavescens and sophoraflavanone G (a prenylated flavonoid from S. flavescens) inhibited cytokines productions in LPS-induced Raw 264.7 macrophages cells and BV2 microglial cells. We examined on the anti-allergic effect of S. flavescens on the PMA plus A23187-induced rat leukemia (RBL-2H3) cells. S. flavescens inhibited the release of β-hexosaminidase and productions and expressions of tumor necrosis factor (TNF)-α, interleukin (IL)-4 and cyclooxygenase (COX)-2 in a dose-dependent manner on stimulated RBL-2H3 cells, however, S. flavescens not affect cell viability. The protein expression level of nuclear factor (NF)-κB (p65) was decreased in the nucleus and suppressed the degradation of inhibitory protein IκB-α protein, the activation of extracellular signal-regulated kinases (ERK) mitogen-activated protein kinase (MAPK) by S. flavescens. These results suggest that S. flavescens could be involved anti-allergic effect by control of NF-κB (p65) translocation into the nucleus through inhibition of IκB-α degradation and suppression of pro-inflammatory cytokines expression.

Objective: Previously, the methanol extracts of the semen of Xanthium strumsrium could involved anti-inflammatory effects in lipopolysaccharide (LPS)-stimulated Raw 264,7 cells, We evaluated the anti-allergic effects of X. strumarium on rat basophilic leukemia (RBL-2H3) cells, Methodes : To investigate the effect of X. strumarium on the phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187-induced RBL-2H3 cells. The effects of X. strumarium on the degranulation and the pro-inflammatory cytokines secretion and expression from RBL-2H3 cells were evaluated with $\beta$-hexosaminidase assay, ELISA, and RT-PCR analysis, In addition, we examined the effects of X. strumarium on nuclear factor (NF)-${\kappa}B$ activation and $I{\kappa}B-\alpha$ degradation using Western blot analysis. Results : X. strumarium inhibited degranulation and secretions and expressions of pro-inflammatory cytokines, such as tumor necrosis factor-alpha ($TNF-\alpha$), interleukin (IL)-4 and cyclooxygenase (COX)-2, on stimulated RBL-2H3 cells, however, X. strumarium not affect cell viability. In stimulated RBL-2H3 cells, the protein expression level of nuclear factor-kappa B (NF-${\kappa}B$) was decreased in the nucleus by X. strumarium. In addition, X. strumarium suppressed the degradation of inhibitory protein $I{\kappa}B-{\alpha}$ protein in RBL-2H3 cells. Conclusion : These results suggest that X. strumarium inhibits the degranulation and secretion of pro-inflammatory cytokines through blockade of NF-${\kappa}B$ activation and I $I{\kappa}B-{\alpha}$ degradation.

The purpose of this thesis is to investigate the way how the image of Korean costume could be introduced into the world fashion market, and appreciated by foreigners. For this purpose. I focus on analyzing the works of Lee. Yong-Hee presented in Pre^t-a`-Porter Paris Collection and interpreting the image of Korean costume in her works. I propose some imperative factors essential to promote the participating of Korean costume into the world fashion market. Firstly, the Korean costume should be reevaluated and moderated in a degree, to be acceptable or accessible to foreigners. Secondly, the commerciality is to be stressed in the light of fashion industry dealing with the practical function of the costume. Thirdly, Our open-mindness to other culture would also make our costume to be more prevalent in world fashion market paradoxically. The Seoul Collection in 2000 fall was opened by Korean government on the recognition that the fashion industry is highly productive and helpful not only in cultural basis but in economics.

RBL-2H3 cell line을 이용하여 백선피의 항알레르기 효과를 알아보기 위하여 PMA와 A23187로 자극한 후 RBL-2H3 내의 과립에 Histamine와 같이 존재하는 ${\beta}-hexosaminidase$의 유출량의 억제 정도 및 합성된 후 분비되는 $TNF-{\alpha}$, IL-4의 합성조절 정도를 살펴본 결과, 백선피는 세포생존율에는 영향을 미치지 않고 백선피의 농도가 증가함에 따라 ${\beta}-hexosaminidase$의 세포 위로의 유출을 억제하였으며, 또한 알레르기 반응과 밀접한 관련이 있는 사이토 카인 중에 하나인 $TNF-{\alpha}$, IL-4의 mRNA 수준에서의 발현의 조절을 통한 합성도 감소하였다. 이 결과로 보아 백선피는 알레르기와 관련된 질환에 응용이 가능할 것으로 사료된다.

황기건중탕은 보비익기(補裨益氣) 및 거습열(祛濕熱)하는 효능(效能)으로 신체(身體)의 허약(虛弱)을 개선하며 염증(炎症)을 유발할 수 있는 외사(外邪)의 침입을 방어하여 인체의 면역기능(免疫機能)을 강화하는 처방으로 면역관련 세포중 하나인 microglial cell를 대상으로 면역세포에 황기건중탕이 어떠한 효능을 발휘하는 지를 살펴보았다. 물로 전탕(煎湯)한 황기건중탕을 여과한 후 실험한 결과 BV2 microglial cells에서 LPS로 자극하여 염증을 유발하는 주요 물질인 NO의 분비를 세포독성 없이 억제하였다. 또한NO를 생성하는 효소인 Cox-2의 발현도 감소시켰다. 그리고 기타 염증전구 물질들이 $TNF-{\alpha}$, $IL-1{\beta}$, IL-12도 황기건중탕의 용량이 증가함에 따라 의존적으로 감소함을 알 수 있었다. 이러한 결과를 보아 황기건중탕은 면역관련세포인 microglial cell에서 염증 관련 인자들의 분비 및 생성을 억제를 통하여 면역관련 상태를 개선시키는 것으로 사료된다.
Inhibitory Effect of Chan-Su on the Secretion of PGE2 and NO in LPS-stimulated BV2 Microglial Cells

Chan-Su (Venenum bufonis) has long been for a variety of other purposes including treatment of inflammation in the folk medicine recipe. Since nitric oxide (NO) is one of the major inflammatory parameters, we first studied the effects of Chan-Su on NO production in lipopolysaccharide (LPS)-stimulated BV2 microglial cells. Chan-Su inhibited the secretion of NO in BV2 microglial cells, without affecting cell viability. The protein level of inducible nitric oxide synthase (iNOS) was decreased by Chan-Su. And Chan-Su also inhibited production of prostaglandin E2 (PGE2) and expression of cyclooxygenase (COX)-2. Proinflammatory cytokines, such as tumor necrosis factor (TNF)-α, interleukin (IL)-1β and IL-12, were inhibited by Chan-Su in a dose-dependent manner. And Chan-Su inhibited the degradation of IκB-α, which was considered to be inhibitor of nuclear factor (NF)-κB, one of a potential transcription factor for the expression of iNOS, COX-2 and proinflammatory cytokines. These results suggest that Chan-Su could exert its anti-inflammatory actions by suppressing the synthesis of NO through inhibition of IκB-α degradation.
MC903으로 유도하는 아토피피부염 생쥐 동물 모델 최적화: 유발 기간 단축 및 부작용 감소를 중심으로

This study was designed to establish an atopic dermatitis (AD) model using MC903 and was conducted to find out the optimal challenge concentration that can cause dermatitis symptoms enough to be experimentally verified while reducing adverse effects such as weight loss. MC903 was treated at concentrations of 2, 3, and 4 nmol/day, and evaluation of skin surface symptoms, water contents, histopathological changes, body weight changes, and spleen/body weight ratio was conducted. In addition, the expression level of thymic stromal lymphopoietin (TSLP) was also observed. In our results, MC903 induced AD skin lesions such as erythema, scab and fissure and lowered skin moisture level significantly. In addition, water holding capacities in the 3 or 4 nmol groups were diminished significantly compared to that in the control group. On the other hand, 4 nmol treatment induced a weight loss of up to 20% compared to control group. Finally, a higher level of TSLP expression was observed in the 3 nmol group than in the 2 nmol group or the 4 nmol group. Taken together, we propose a total 14-day protocol consisting of 3 days of sensitization (2 nmol/day) and 6 days of challenge (3 nmol/day).
