蔘蘇飮과 蔘蘇飮加味方의 抗알레르기효과에 관한 實驗的 硏究

Background : Although Samsoeum has been used frequently on allergic rhinitis, but there isn't any experimental research for that. Objective : This study was performed to investigate the anti-allergic effects of Samsoeum and Samsoeumgamibang. Materials and Methods : Katayama's method was used to observe the vascular permeability response induced by serotonin and histamine. Muller's method was used to observe the contact dermatitis response induced by picryl chloride. Miller's method was used to observe the delayed type hypersensitivity response to SRC. Results : 1. In the vascular permeability response to intradermal injection of serotonin, 2,600mg/kg, p.o. group of Samsoeum(蔘蘇飮), 1,300 and 2,600mg/kg, p.o. group of Samsoeumgamibang(蔘蘇飮加味方) showed significant inhibitory effects on the leakage of Evan's blue solution. 2. In the vascular permeability response to intradermal injection of histamine, 1,300 and 2,600mg/kg, p.o. group of Samsoeum, also 1,300 and 2,600mg/kg, p.o. group Samsoeumgamibang showed significant inhibitory effects on the leakage of Evan's blue solution. 3. In the contact dermatitis response induced by picryl chloride, 1,300 and 2,600mg/kg, p.o. group of Samsoeum, 1,300 and 2,600mg/kg, p.o. group of Samsoeumgamibang showed significant inhibitory effects on ear swelling formation. 4. In the delayed type hypersensitivity response to SRC. 2,600mg/kg, p.o. group of Samsoeum and 2,600mg/kg, p.o. group of Samsoeumgamibang showed significant inhibitory effects on foot swelling. Conclusion : This study shows that Samsoeum and Samsoeumgamibang may have anti-allergic effects. So Samsoeum and Samsoeurngamibang can be helpful to treat allergic rhinitis.
산풍고삼환(散風苦蔘丸)이 알레르기성 접촉피부염에 미치는 영향

Objectives: To study the effectiveness of Sanpunggosamhwan(SGH) against Allergic Contact Dermatitis(ACD), the contact hypersensitivity assay, change of cutaneous shape, anti-allergic effect, anti-inflammatory effect, and the effect on skin barrier were observed. Methods: The 200g rats were divided into three groups of 15 rats. The first group is the Normal group which was applied Acetone olive oil only. The second group is the ACD group which has intentionally activated Allergic Contact Dermatitis by DNCB. The third group is the SGH group which was given medication of Sanpunggosamhwan extract after the induction of Allergic Contact Dermatitis by DNCB. Each group of rats was observed after 24, 48 and 72 hours. Results: I. With the result of contact hypersensitivity assay, at 24hours SGH group showed appreciably less ear swelling than ACD group. 2. Regarding general change of skin, SGH group showed less hyperplasia of epidermis, less damage of epidermis than ACD group. 3. Regarding the number of WBC, ACD group showed significantly less than normal and SGH group at 72 hours. 4. Regarding the number of RBC in blood, ACD and SGH group showed significantly more RBC than normal group at 24, 48, 72 hours. 5. Regarding the ratio of neutrophil in WBC, ACD and SGH group showed significantly high percentage than normal group at 24, 48 hours. 6. Regarding the ratio of lymphocyte in WBC, ACD and SGH group showed significantly high percentage than normal group at 48 hours. 7. Regarding the erythema, SGH group showed significantly more erythema than normal and ACD group at 48 hours. 8. Regarding the melanin, SGH group showed significantly less melanin than normal group at 24 hours.9. Regarding the skin hydration, SGH group showed significantly high value than and ACD group at 72 hours. 10. Regarding the skin pH, ACD group showed significantly high value than normal and SGH group af 24 hours. 11. Regarding the number of Total IgE, ACD and SGH group showed more Total IgE than normal. g개up at 24 hours. 12. At Electro microscope-morphologic changes of skin, the damage of epithelium was decreased and regeneration power of skin was increased in the SGH group. Conclusions: The Sanpunggosamhwan extract administration was effective on the mitigation of skin damage in rats with allergic contact dermatitis.

We developed nephelometric assays on the Beckman Array 360 Nephelometry analyzer that allow a quantification of IgG subclass concentrations in 100 samples quickly and reproducibly. We have established new reference ranges the human IgG subclass in sera of adults. These reference ranges are the central 95th percentile of results from a study of 100 apparently healthy individuals(44 females, 56 males). The reference ranges of IgG subclasses in sera are IgG1 447-1260 mg/dL, IgG2 246-900 mg/dL, IgG3 15-147.6 mg/dL, and IgG4 12-104.4 mg/dL respectively.
Glucomannan이 노인성 제2형 당뇨병 환자의 당질 및 지질 대사에 미치는 영향

연구배경: 본 연구자들은 소장에서 당분과 지방질의 흡수를 저해하는 효과가 있는 것으로 알려져 있는 glucomannan을 노인성 제2형 당뇨병 환자에게 투여하여 혈당, 지질 및 인슐린저항성에 어떠한 영향이 있는지 조사하고자 하였다. 방법: 연세대학교 의과대학 내분비내과에 내원한 혈청 총콜레스테롤 농도가 높은 당뇨병 환자 중 60세 이상의 76명을 대상으로 실험군 39명, 대조군 37명으로 나누어 실험군에게는 하루 glucomannan을 대조군에게는 placebo 약제를 총 8주간 투여하였다. 공복혈당과 식후 2시간 혈당은 치료 0, 4, 8주에 측정하였으며, 치료 0, 8주에는 당화혈색소 및 총콜레스테롤, 중성지방, 고밀도 지단백 콜레스테롤, 저밀도 지단백 콜레스테롤을 측정하였으며 혈청 인슐린, C-peptide를 측정하고 인슐린저항성을 파악하기 위해 HOMA score를 산출하였다. 결과: 본 연구를 시행한 결과 다음과 같은 결과를 얻었다. 1) 공복혈당은 glucomannan을 8주간 투여한 후에 유의한 감소를 나타내었으며 식후 2시간 혈당은 glucomannan을 투여한 후 4주부터 감소하였으며 8주째에도 유의한 감소를 보였다. 2) 혈청 총콜레스테롤, 중성지방, 저밀도 지단백 콜레스테롤은 glucomannan 투여 후에 유의한 감소를 보였으나 고밀도 지단백 콜레스테롤은 유의한 변화가 없었다. 3) glucomannan을 8주간 투여한 후 HbA₁c는 투여 전에 비하여 유의하게 감소하였다. 4) HOMA-IR 값은 glucomannan 투여 후에 감소하였으며, HOMA-β 값은 투여 8주에 유의한 증가를 나타내었다. 5) glucomannan 투여가 간기능, 신장기능 및 전해질에는 특별한 영향을 미치지 않았다. 결론: 이상의 연구 결과를 통하여, 노인성 제2형 당뇨병 환자에게서 glucomannan은 혈당 및 지질대사 개선에 효과가 있는 것으로 확인되었다. glucomannan의 작용기전은 식후 고혈당 개선을 통한 인슐린저항성 감소 및 인슐린분비능의 개선으로 생각된다. Background: Glucomannan is known to be effective in reducing glucose and lipid absorption from small intestine. In this study, we tried to investigate the relationship between glucomannan and glucose, lipid profile, and insulin resistance in the elderly type 2 diabetes patients. Method: We have taken 76 type 2 diabetes patinets with hypercholesterolemia, aged over 60 years old, who have visited our endocrine clinic of Yonsei University of Medical College and categorized them into two groups. The experimental group, which consisted of 39 subjects, received daily glucomannan, while the control group, which consisted of 37 patients, received daily placebo for 8 weeks. We have measured fasting glucose, 2hr post-prandial glucose at 0, 4, 8 weeks and HbA1C, total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, insulin, c-peptide levels at 0, 8 weeks, and calculated HOMA score in order to determine insulin resistance. Results: The following results were obtained. 1) 2hr postprandial glucose level was significantly reduced after taking glucomannan for 4 and 8 weeks, and fasting glucose level declined after taking glucomannan for 8 weeks. 2) Serum total cholesterol, triglyceride, LDL-cholesterol levels were significantly lowered after treatment. Serum HDL-cholesterol level had a tendency to increase in the experimental group, but was not statistically significant. 3) HbA₁c was significantly decreased after taking glucomannan for 8 weeks. 4) HOMA-IR was reduced at 8 weeks, and HOMA-β showed a significant increase at 8 weeks. 5) Glucomannan did not have adverse effects on CBC, liver function, renal function, and electrolyte levels. Conclusion: The above study results demonstrate that glucomannan is effective in improving glucose and lipid metabolism in elderly type 2 diabetes patients. Glucomannan enhances insulin resistance by lowering postprandial gluocse level and stimulating insulin secretion.
