        재배지역에 따른 검정콩 부위별 추출물의 항산화 활성

        With the aim of developing region specialized crops, this study was conducted to clarify effects of variant and cultivation region on antioxidative activities in various black soybean (Glycine max L.) seed tissues. Three black soybean varieties (SCEL-1, Wonheug, and Cheongja 3) were each cultivated in 4 different regions (Jeonju, Pyeongchang, Paju, and Cheonan). Harvested seeds were used to assess DPPH and ABTS radical scavenging activity, and total polyphenol, flavonoid and anthocyanin content. SCEL-1 soybean hull contained higher DPPH and ABTS radical scavenging activity (61% and 85% respectively) compared to Wonheug (40% and 50% respectively). SCEL-1 cultivated in Pyeongchang displayed the highest total polyphenol and flavonoid content (1,189 mg GAE/100g sample and 951 mg CTE/ 100g sample, respectively). Total anthocyanin content was ranked in the following order: SCEL-1>Wonheug>Cheongja 3. All black soybeans showed much higher antioxidant activity in the soybean hull than in the dehulled soybean. The antioxidant activity of black soybeans cultivated at high latitudes was high. These results suggest that the best black soybean variant for high beneficial biological activities is the SCEL-1 variant. For a complete understanding of the potential of black soybean as functional foods, we plan to further analyze their antioxidant activities in future studies.

        F344 랫드를 이용한 이황화메틸의 아급성 흡입독성연구

        이황화메틸의 반복 흡입노출에 의한 아급성 독성 잠재력을 평가하기 위해 암수 랫드에게 0, 5, 25 및 125 ppm용량으로 21일간 반복 흡입노출하고, 일반증상과 체중, 사료섭취 량, 혈액치, 혈청생화학치 및 부검소견을 관찰하였다. 시험결과, 랫드에게 이황화메틸을 3주간 반복 흡입노출하면 125 ppm의 농도에서 체중증가의 억제와 사료섭취 량의 감소를 유발하나, 혈액 및 혈청생화학치에는 어떠한 이상도 유발하지 않는 것으로 나타났다. 본 시험 조건 하에서 이황화메틸의 표적 장기는 관찰되지 않았으며, 무해용량은 암수 모두 25 ppm으로 사료된다. The purpose of this study was to investigate the potential subacute toxicity of dimethyl disulfide by 3 weeks inhalation in F344 rats. The test article, dimethyl disulfide was exposed by inhalation to male and female rats at dose levels of 0, 5, 25, or 125 ppm/6 hrs/ day for 3 weeks. Five rats/ sex/ group were sacrificed on day 4 after the initiation of treatment, while 5 rats/ sex/ group were sacrificed at the end of treatment period. During the test period, clinical signs, mortality, body weights, food consumption, hematology, serum biochemistry, and gross findings were examined. Slight decreases in body weight gain were noted in both sexes of the highest dose group in a dose dependent manner but were only statistically different from the control animals in males of the group. A slight non-significant reduction in food consumption were also noted in the both sexes of the highest dose group. There were no adverse effects on mortality, clinical signs, hematology, serum biochemistry, and necropsy findings at any dose tested. Based on these results, it was concluded that the 3 weeks repeated dose of dimethyl disulfide by inhalation resulted in suppressed body weight gain and decreased food consumption at 125 ppm of both sexes. In the present experimental conditions, the target organ was not determined in rats. The no­observed-adverse-effect level (NOAEL) was considered to be 25 ppm/6 hrs/day for both sexes.

        Effects of α - Tocopherol and β - Carotene Supplementation on Oxidative Damage by Lipid Oxidation in Rat Liver

        α-토코페롤과 β-카로틴을 강화한 식이가 흰쥐 간의 산화적인 손상을 줄이는 데 미치는 영향을 살펴보기 위하여 국립보건 연구원으로 부터 분양 받은 Sprague-Dawley계 숫쥐 45마리를 9군으로 나누어 식이지방과 항산화제 첨가 유무를 달리한 식이로 12주 동안 사육하였다. 식이지방원으로는 정어리유, 대두유, 그리고 라드를 이용하여 AIN-76 사료조제법에 따라 탄수화물이 총에너지의 65%, 단백질이 15%, 그리고 지방이 20%되게 하였으며 여기에 α-토코페롤과 β-카로틴을 AIN-76에 사용한 양의 각 200%, 그리고 150% 만큼 첨가였다. 간의 MDA와 protein carrbonyl값은 정어리유군에서 가장 높아 고도의 불포화지방을 다량 함유한 식이군의 지방산화가 가장 현저히 일어남을 알 수 있었으며 이에 항산화제를 강화한 식이를 섭취시킨 경우 지방의 산화를 억제하는 효과도 정어리유군에서 가장 현저히 볼 수 있었다. α-토코페롤 강화한 정어리유군에서 MDA의 생성이 대조군에 비해 35%, β-카로틴 첨가군에서는 15%, 그리고 protein carbonyl의 생성은 각각 44%, 32% 감소되었음을 볼 수 있었고 그 감소의 정도는 모두 유의적이었다. 항산화제를 강화한 군의 지방산화 억제효과는 protein carbonyl값의 측정시 더욱 현저하게 볼 수 있어 지방의 산화정도를 측정하는 데 있어서 이 방법이 더 민감함을 알 수 있었다. Cu, Zn-SOD와 catalase의 활성을 측정하여 지방산화에 따른 효소의 손상정도를 살펴보았을 때, 역시 정어리유군에서 효소의 활성이 대두유군이나 라드군에 비해 현저하게 감소되었음을 볼 수 있었고, 여기에 항산화제를 강화시켰을 때 α-토코페롤 첨가군에서는 효소 활성이 높아지는 경향을 볼 수 있었으나 β-카로틴을 첨가한 군에서는 효소의 활성이 더욱 감소하는 반대의 결과가 나타났다. 이러한 현상은 식이에 첨가한 β-카로틴이 소장에서 Vt A로 전환되어 간에 축적되어 나타난 Vt A 독성 때문으로 추측된다. 쥐의 경우 식이로 섭취된 카로틴은 소장 점막에서 비타민 A로 거의 모두 전환된다는 보고 (4)가 있다. The effect of α-tocopherol and β-carotene supplementation on reducing the oxidative damage in the liver of rats were studied. Forty-five male Sprague Dawley aged 4 weeks were randomly assigned to 9 groups of five for the 12 weeks of the study. Nine groups, sardine oil, sardine oil+Vt E, sardine oil+β-carotene, soybean oil, soybean oil+Vt E, soybean oil+β-carotene, lard, lard+Vt E, lard+β-carotene group, were prepared. Sardine oil, soybean oil, or lard was used for dietary fat and 200% of α-tocopherol or 150% of β-carotene was supplemented to each diet. Each diet supplied 65% of total energy as carbohydrate, 15% as protein, and 20% as lipid. The MDA value and protein carbonyl contents of sardine oil group were significantly different (p<0.05) to those of other fat groups indicating that the most severe lipid oxidation occurred in the group fed diet containing highly polyunsaturated fatty acid. When α-tocopherol or β-carotene was supplemented to the sardine oil diet, MDA value (-35%, -15%, respectively) and protein carbonyl content (-44%, -32%, respectively) decreased significantly (p<0.05). Cu, Zn-superoxide dismutase (SOD) and catalase activities of three different sardine oil groups with or without antioxidants were lower than those of soybean oil or lard group. The reducing effect of α-tocopherol on oxidative damage in sardine oil group supplemented with α-tocopherol was noticeable (p<0.05). However the adverse effect of β-carotene was observed. SOD and catalase activities of β-carotene supplemented groups were the lowest among the same fat groups, but the differences were not statistically significant. The possible cause of decreased enzyme activity seemed to be related to the vitamin A(Vt A) toxicity in the liver where retinol converted from dietary β-carotene in the intestinal mucosa was stored.

        고착협 내탈립 기계수확 적응 장류⋅두부용 콩 품종 ‘새금’

        The soybean cultivar, ‘Saegeum’, has been developed for preparing soy-paste and tofu. The soybean cultivars ‘Daepung’ and ‘SS98207-3SSD-168’ were crossed in 2003 to obtain ‘Saegeum’. Single seed descent method was used to advance the generation from F3 to F5, and the plant lines with promising traits were selected from F6 to F7 by pedigree method. The preliminary yield (PYT) and advanced yield trials (AYT) were conducted from 2009 to 2010, and the regional yield trial (RYT) was conducted in 12 regions between 2011 and 2013. The morphological characteristics of ‘Saegeum’ were as follows: determinate plant type, white flower, tawny pubescence color, and brown pod color. Flowering and maturity dates were August 2, XXXX and October 17, XXXX, respectively. Plant height, first pod height, number of nodes, number of branches, and number of pods were 79 cm, 18 cm, 16, 2.3, and 44, respectively. The seed characteristics of ‘Saegeum’ were as follows: yellow spherical shape, yellow hilum, and the 100-seed weight was 25.4 g. ‘Saegeum’ was resistant to bacterial pustule and SMV in the field test, and its lodging resistance was mildly strong, whereas its shattering resistance was excellent. The ability of this cultivar to be processed into tofu, soybean malt, and other fermented products was comparable with that of ‘Daewonkong’. The yield of ‘Saegeum’ in the adaptable regions was 3.02 ton ha-1. Thus, ‘Saegeum’ is adaptable to mechanized harvesting because of its high first pod height, as well as lodging and shattering resistance. (Registration number: 5929)

        Purpose: This study classified normal adults according to age, and made them carry out nonword repetition task, and investigated an effect of scoring methods and syllable length on performance. Besides, variation of response was examined. Methods: 4 groups, i.e., young adults (20~29), prime-aged adults (30~49), middle-aged adults (50~64) and older adults (65 or above) heard nonword, and carried out 4-repetition task. Item-, syllable- and phoneme-based scoring was applied to item of the first response among 4 responses. And with regard to variation measurement, it was scored, applying all items of 4 responses. Results: Repeated measures analysis of variances with mixed design was carried out so as to examine a difference in variation of articulatory production and accuracy according to syllable length between groups in each scoring method. With regard to item-, syllable- and phoneme-based accuracy, a between-group main effect was significant. With regard to item-, syllable- and phoneme-based accuracy according to syllable length, the within-group difference was significant. A significant interaction effect according to group and syllable length was observed in syllable- and phoneme-based scoring except item-based one. Between-group main effect was not significant in variation of production. Within-group difference was significant in variation of production according to syllable length. Significant interaction effect according to group and syllable length was not found. Conclusions: It was shown that syllable- and phoneme-based scoring method was more sensitive in distinguishing between age groups than item-based one. Within-group difference according to syllable length was shown in all of item-, syllable- and phoneme-based methods. Particularly, it was verified that between-group discrimination increased in 5 and 6 syllables. It was impossible to divide age groups according to variation of articulatory production.

        The Sea dike tidal closure works has traditionally ended in the failure. Therefore, the success of the tidal closure in Hwaong in the sea near Inchon where has the greatest tidal gap in our country deserves to consider its meaning. While its process was going on, we had the difficulty like bad weather, unexpected ground settlement, flow velocity beyond the imagine, etc. Moreover, environment group's rush was so fierce that the works could have been delayed. This works showed that not only technical aspects but also social problems(environment group's rush and public discontents) were important factors in accomplishing the works. Finally, all of the difficulties were overcome and the tidal closure works were successful. So, This success is the best example in a kind of the another works in itself.

        귀리 첨가 호상 요구르트의 저장 중 항산화 활성 및 품질 특성

        Recently, consumers' awareness of the importance of the intestinal action of lactic acid bacteria and intestinal microbes is increasing, as well as interest in yogurt. In this study, yogurt was prepared with three mixed strains (lactic acid bacteria combination, Lactobacillus acidophilus, Lactobacillus delbrueckii subsp., and Bulgaricus, Streptococcus thermophilussei, 1:1:1) by adding oats flour, and the quality characteristics of yogurt were investigated, while stored at a storage temperature of 4℃ for 12 days. According to the storage period, the control as wel as the oat yogurt showed slight decrease in pH, and no significant change in acidity. Sugar content slightly increased. and brightness decreased, in the control and the oat yogurt. Visible cell numbers increased during storage, and decreased on the 12th day. Viscosity in the oat yogurt was 7,580 cP, which was approximately eight times higher than that of the control group, and decreased gradually according to the storage period. Antioxidant activity (DPPH) was approximately two times higher in the oat-added yogurt, and slightly increased with the storage period, decreased on the 12th day of storage, and β-glucan was detected only in oat-added yogurt.

        It has been reported that bacterial diseasees of domestic soybean are bacterial pustule (Xanthomonas axonopodis pv. glycines), wildfire ( Pseudomonas syringae pv. tabaci), bacterial blight (Pseydomonas savastanoi pv. glcines) and bacterial brown spot (Pseudomonas syringae pv. syringae) in Korea. Bacterial pustule had been the most issue in soybean diseases. In recently wildfire also occurrence in soybean and Bacterial blight and bacterial brown spot have been reported to less incidence in Korea. Each bacterial diseases were seed borne and then they caused diseases as orumary casual agent. in this study, direct OCR assay was applied to detect casual agent of bacterial diseases and survey on seed contamination on 44 soybean cultivars and 14 genctic resources in Korea, The pathogens were detected frin the seed samples incubated for 12h with 160rpm shaking. The result of surbey on seed contamination showed that bacterial pustule, wildfire and bacterial blight were detected from some cultivars and genetic resources. We also conducted resistance screening in other to select resistance cultivars of Baterial pustule and Wildfire. we inoculated on 128 cultivars with Baterial pustule and Wildfire respectively. These cultivars were classified through pathoggenecity from 1 to 9. In addition, 35 and 50 resistant cultivars were confirmed against Baterial pustule and Wildfire respectively

        랫드에 투여된 규회석이 호흡기계에 미치는 영향 연구

        Occupational exposure to wollastonite can occur during its mining, processing and using. Wollastonite was analyzed for its physicochemical properties by transmission electron microscope and scanning electron microscope equipped with energy dispersive X-ray spectrometer. Wollastonite fibers were 3.3㎛ in diameter(av.) and were 30.5㎛ in length(av.). The atomic composition of wollastonite was Si 56.41% and Ca 43.59%. Respiratory toxicity of wollastonite has been studied in term sequential in Sprague-Dawely rats. UICC chrysotile(average diameter 0.03㎛, average length 2.93㎛) was applied as the positive control. The effects of 2㎎ wollastonite on respiratory system and pathological changes were evaluated after 1, 4, 8, 12 weeks instilled into rat lungs. Inflammation response broke out from 1 week after instilled with wollastonite and the pathological examination further showed increased legions of granulomatous inflammation after 4 weeks, but decreased granulomatous inflammation after 8 weeks, whereas chrysotile induced progressive interstitial granulomatous inflammation and fibrosis as a function of time.
