N/A Microsomal △6 desaturase is a key enzyme in the metabolism of polyunsaturated fatty acids, the activity of which is regulated by insulin. To determine if changes in d △6 desaturase activity in experimental diabetes correspond to changes in fatty acid compoistion of microsome and RBC membrane, 18 Sprague-Dawley rats were studied for 6 weeks. The animals were divided into 3 groups with 6 rats in each group. The first group was the contrals, the 2nd 3rd groups were streptozotocin diabetic rats but insulin was injected to the 3rd group. The △ 6 desaturase activities of diabetic rats were 68pg of those of controls (p<0.05) and increased to 119% of controls (p<0.05) with insulin treatment. Linolic (LA), palmitic (PA) and docosahexaenoic acids (DHA) were increased and arachidonic (AA) & oleic acids (OA) decreased in liver microsome of diabetic rats, which were reversed with insulin treatment, respectively. In erythrocyte membrane LA & stearic acids (SA) were increased, and PA, AA, and palmitoleic acids were decreased which were not reversed with insulin treatment, however. To summarize, insulin controls the △ 6 desaturase activities, which in turn affect fatty acid compositions of microsome and RBC membranes. Theee findings suggest that adequate insulin therapy is required for the correction of the polyunsaturated fatty acid metabolism as well as blood glucose control in the insulin dependent diabetics.

제주재래종 감귤류 10종에 대해서 수확시기별 총 폴리페놀 함량과 활성산소종 소거활성 변화를 조사하였다. 총 폴리페놀 함량은 지각과 홍귤이 미숙과인 8월 하순과 9월 하순에 200 ㎎% 내외로 가장 높았고 대부분의 감귤종이 수확시기가 늦어질수록 감소하는 경향을 보였다. 수확시기별 감귤과피의 superoxide anion radical 소거활성은 홍귤과 지각에서 60% 이상으로 가장 높았고 사두감이 가장 낮은 활성을 보였으며 홍귤과 지각을 제외한 감귤류의 수확시기별 차이는 크지 않았다. Hydrogen peroxide 소거활성은 8월 하순에 사두감이 73.8%로 가장 높은 소거활성을 나타내었고 그 다음에 지각이 거의 70%의 소거활성을 보였으며, 대부분의 감귤종이 수확시기가 늦어질수록 감소하는 경향을 보였다. Hydroxy radical 소거활성은 대부분의 재래감귤종의 과피에서 비교적 높은 활성을 보였으며 지각과 당유자가 8월 하순에 각각 75.1%와 74.6%로 가장 높은 소거활성을 보였고, 그 외 감귤종도 60% 이상의 소거능을 보였으며 수확시기별 차이는 거의 없었다. 감귤이 성숙됨에 따라 hydrogen peroxide 소거능, superoxide anion 라디칼 소거능은 감소하는 경향을 보이나 nitric oxide 라디칼 소거활성은 증가하는 추세를 보였고, 병귤이 58.4%로 가장 활성이 높았으며 다른 활성산소종에서 활성이 높았던 사두감, 지각, 당유자가 활성이 가장 낮았다. 본 실험결과 제주재래종 감귤 중 특히 지각이 대부분의 활성산소종에 높은 소거활성을 보였으며, 이에 대한 구체적인 연구가 진행될 필요성이 있을 것으로 판단된다. This study was conducted to investigate total polyphenolic contents and reactive oxygen species (ROS) scavenging effects of extracts from peels of ten Jeju native citrus fruits according to the harvest from August 2006 to February 2007. Total polyphenolic contents from methanol extracts of citrus peel were the highest in Jigak (Citrus aurantium) and Hongkyool (C. tachibana) by over 200 ㎎% in the unmatured period, from the late August to the late September, and all the citrus peels mostly decreased while ripening. Scavenging effect of superoxide anion radical showed good correlation with total polyphenolic contents. The unmatured periods of Hongkyool and Jigak were the highest with more than 60%. Hydrogen peroxide scavenging activity was the highest in Sadoogam (C. pseudogulgul) at 73.8% in late August and the second highest activity was observed in Jigak at near 70%, and all the citrus peels decreased during ripening. Hydroxy radical scavenging activity were the highest among all the ROS scavenging activities, especially in the Jigak and Dangyooja (C. grandis) at 75.1% and 74.6%, respectively, and not much affected by increased maturity of the fruits. Nitric oxide radical scavenging activity was the highest in Bungkyool (C. platymama) at 58.4% in late February, and increased with fruit ripening. In this study, Jigak was generally the highest in the polyphenolic contents and ROS scavenging activities, so the further studies are needed for industrial applications.
수확시기별 제주재래종 감귤착즙액의 Flavonoids 분포 및 항산화 활성

제주재래종 감귤류 14종의 착즙액에 대해서 수확시기별 flavonoids 13종의 함량변화를 분석하고 항산화성을 검토하고자 하였다. 감귤착즙액의 flavonoids 함량은 수확시기가 늦어질 수록 큰 폭으로 감소하다가 4~5차 채취기 이후 서서히 증가하는 경향을 보였다. Quercatagetin, narirutin, hesperidin, neohesperidin은 각각 지각 573.73 ㎎/100 ㎖, 사두감 393.99 ㎎/100 ㎖, 소유자 29.63 ㎎/100 ㎖, 지각 201. 23 ㎎/100 ㎖으로 미숙과인 8월 하순에 가장 높았다. Polymethoxyflavone류 중 nobiletin, sinensetin, tangeretin은 홍귤에서 8월 하순에 각각 7.39 ㎎/100 ㎖, 2.24 ㎎/100 ㎖, 0.63 ㎎/100 ㎖으로 가장 높았고, 다음으로 진귤이 많이 함유되어 있었다. 3,5,6,7,8,3',4'-Heptamethoxyflavone은 편귤에서 0.27 ㎎/100 ㎖으로 가장 높았고, 3',4',7,8-tetramethoxyflavone, 3',4'-dimethoxyflavone, 4'-methoxyflavone, 5,6,7,3',4',5'-hexamethoxyflavone, scutellarein tetramethylether는 모든 품종에서 ND~0.08 ㎎/100 ㎖로 함량이 낮았다. 총 폴리페놀 함량은 대부분의 시료가 미숙과인 8월 하순에 가장 높았다가 11월까지 감소하다가 12월 하순부터 증가하는 경향을 보였다. 감귤착즙액의 전자공여능은 대체적으로 높은 것으로 나타났으나 flavonoids와 총폴리페놀 함량과의 상관관계는 높지 않은 경향을 보였고 당유자가 가장 높은 항산화활성을 나타내었다. 재래 감귤류 중 지각, 당유자는 quercetagetin이 다른 감귤류에 비해 월등히 높았고, polymethoxyflavone류인 nobiletin과 tangeretin은 미숙과인 홍귤과 진귤에서 함량이 높아 소재 개발로서 경쟁력이 있을 것으로 사료된다. This study aims to evaluate the changes of flavonoid contents and antioxidants activity of Jeju native citrus fruits juice according to the harvest date. Flavonoids such as quercatagetin, narirutin, hesperidin and neohesperidin were contained most plentifully in the juice of Jigak (Citrus aur- antium) by 573.73 ㎎/100 ㎖, Sadoogam (C. pseudogulgul) by 393.99 ㎎/100 ㎖, Soyooja by 29.63 ㎎/100 ㎖ and Jigak (C. aurantium) by 201.23 ㎎/100 ㎖ in the late August, respectively. The highest contents of nob-iletin, sinensetin and tangeretin among polymethoxyflavones were found in the juice of Hongkyool (C. tachibana) by 7.39 ㎎/100 ㎖, 2.24 ㎎/100 ㎖, 0.63 ㎎/100 ㎖ in the late August, respectively. 3,5,6,7,8,3’,4’- Heptamet- hoxyflavone recorded the highest amount in Punkyool (C. tangerina) by 0.27 ㎎/100 ㎖ in the late August, but the other polymethoxyflavones including 3’,4’,7,8-tetramethoxyflavone, 3’,4’-dimethoxyflavone, 4’-methoxyflavone, 5,6,7,3’,4’,5’-hexamethoxyflavone, scutellarein tetramethylether were observed only trace amount in all the citrus fruits. Flavonoid contents in the citrus fruit juices were the highest during early maturation and decreased rapidly while ripening. Total polyphenol contents were the highest in the late August and decreased with ripening. However from the late December, the contents were increased again. Antioxidant activities of the fruits were evaluated as electron donating ability and were the lowest in the late September and increased with the fruit ripening. These results suggest that quercetagetin among all the flavonoids was most plentiful in Jigak and Dangyooja (C. grandis), so that the fruits could be used for industrial material of flavonoids and antioxidant agents.

N/A Objectives: Thyroid nodule is a common clinical problem but there is a few cases of malignant lesions requiring surgery. So it is a critical issue to distinguish benign from malignant nodules and to select a patient for surgery. Although there are some variations in the accuracy of the diagnosis, fine needle aspiration biopsy of thyroid nodule has been accepted as a accurate diagnostic procedure in the management of nodular thyroid disease. So it was desirable to verify the usefulness of the fine needle aspiration (FNA) cytology in differentiating from malignant lesions and to clarify the problematic issues concerning the cytopathologic features of indeterminate cytology, which is equivocal to describe benign or malignant lesion, Methods: We examined the medical records retrospectively of all patients who wore engaged in fine needle aspiration cytology or thyroidectomy due to thyroid nodules from 1988 to 1992. Results: 1) Eighty hundreds fifty nine cases of FNA cytology was done in 1992 and 659 cases(75.9%) were adequate for cytopathologic diagnosis. And the diagnoses showed 558 cases of benign, 37 cases of malignant and 57 cases of indeterminate features. 2) The sensitivity of benign nodules were 91.2% and that of malignant lesion were 85.7%, respectivety. The overall accuracy of fine needle aspiration cytology of the thyroid nodule was 88.4%. 3) From 1988 to 1992, the medical records of 140 patients showing indeterminate cytology of thyroid nodules by FNA were reviewed. Among them, 67 patients were operated for the thyroid nodule till June 1993 and pathologic diagnoses were confirmed, The pathologic diagnoses were consisted of 27 cases(40.3 %) of benign lesions and 40 cases(59.7%)of malignant lesions. Conclusion: FNA cytology of the thyroid nodule was useful in distinguishing benign from malignant nodules and surgical excision of the cases showing in- determinate cytopathologic features seems reasonable.
본태성 고혈압 환자의 적혈구 Na+-Li+ Countertransport 와 적혈구막 지방산 구성에 대한 연구

Since the changes of Na^+-Li^+ countertransport (SLC) Vmax in essential hypertension have been reported. it has been known that hyperlipidemia could affect SLC independently of blood pressure. To reveal the relation between SLC Vmax and RBC membrane fatty acid conposition in the hypertensive patients, we divided patients into two groups according to their levels of SLC Vmax of erythrocytes(elevated SLC Vmax: n=4, normal SLC Vamx: n=6). The results were as follows: 1) There was no difference in body mate index, waist-hip ratio, blood pressure, serum cholesterol level between two groups. Triglyceride were higher in elevated group (278.0±180.0 ㎎/㎗) than normal group (124.3±60.7 ㎎/㎗) but there was no statistical significance. HDL-cholesterol levels were significantly lower in elevated group (32.0±3.6 vs. 44.8±8.9 ㎎/㎗) and free fatty acid concentrations, intracellular (RBC) Na^+ concentrations were similar between two groups. 2) The results of oral glucose tolerance test showed no differences in the glucose and insulin area, however the incremental insulin areas of elevated group (108.8±31.3, μU. min/㎖×10²) were significantly higher compared with those of normal group (56.3±20.7 μU. min/㎖×10²). 3) In the elevated group, palmitic acid proportions of erythrocytes (22.1±0.7%) were higher than those of normal group (20.1±0.7%) and SLC Vmax showed significant correlation (r=0.81, p$lt;0.01) with palmitic acid proportion. These results suggest that the abnormal lipid metabolism may change the RBC Na^+ transport system through the alterations of RBC membrane fatty acid composition.
