        선박평형 수 내 유해 와편모조류(Dinophyceae)의 분자생물학적 검출

        선박평형 수는 유독 와편모조류 및 다양한 미세조류의 국제적인 이동경로로 알려져 있다. 본 연구에서는 선박평형 수에 있는 와편모조류의 다양성을 조사하기 위하여 와편모조류 특이적인 PCR primer와 종 특이적인 real-time PCR 유전자 탐침자를 이용하였다. 선박평형 수 시료에 대한 광학현미경 조사에서는 와편모조류가 매우 낮은 농도로 관찰되었지만, SSU rDNA의 cloning 및 염기서열 분석 결과에서는 기생 와편모조류, 초미세플랑크톤, 어패류 폐사 원인종 등 다양한 종류가 확인되었다. 본 연구 결과는 종 톡이적 PCR primer와 같은 분자생물학적 방법이 선박 평형 수에 외래 유입종의 신속 정확한 진단에 유용함을 보여주고 있다. Ballast water has been known as a major vector for global dispersal of toxic dinoflagellates and other microalgae. In this study, biodiversity in ships’ ballast water was examined using a dinoflagellate-oriented PCR primer set and species-specific real-time PCR. While motile dinoflagellates could be observe at very low cell densities by light microscopy,a wide range of dinoflagellate taxa including parasitic and phototrophic pico-dinoflagellates as well as harmful species to marine fish/shellfish was detected when techniques for cloning/sequencing of SSU rDNA of sample cells were used. Present result suggests that molecular methods including species-specific PCR primers may offer rapid and accurate detection of invasive species in ballast water.

        소형펀치 크리프 시험중 측정하는 변수에 대한 연구

        An effect is made in this study to deepen understanding of small punch(SP) creep testing which has been a round for about 10 years as a substitute for the conventional uniaxial creep testing. Even though considerable numbers of SP creep test program have been performed, most of the tests were aimed at measuring creep rupture lives only. Very flew studies showed interest on the meaning of what we were really measuring during the SP creep tests. In this paper meanings of the parameters measured during the SP creep testing, such as punch load and punch displacement rate are investigated using finite element analysis. It was shown that the measured parameters must represent the stress and strain rates of the material at the annular region located at about 0.65 mm from the center of the SP specimen. The material in this location would go through constant maximum stress and strain rate during the testing. Experimental verification is also discussed.

      • p$^+$ Si 외팔보 구조를 이용한 광학 소자용 마이크로 구동기의 제작

        The paper represents the design and fabrication of an electrostatic micro actuator with $p^+$, Si cantilevers. The micro actuator consists of a plate suspended by four $p^+$, silicon cantilevers and an electrode on a glass substrate. The $p^+$, Si structure is fabricated by the boron diffusion process and the anisotropic wet etch process. The cantilevers of the micro actuator curl down because of the residual stress gradient in $p^+$, silicon. When the electrostatic forec is applied to the $p^+$, cantilevers, the vertical displacement of the plate can be achieved. The deflection of the cantilever due to the residual stress gradient and the vertical displacement by electrostatic force were calculated. The displacement of the plate was measured with a laser displacement meter for various input voltages and frequencies. The feasibility of the proposed micro actuator for the applications to optical pickup devices or optical communication devices was confirmed by the experiments.

        예비전류를 이용한 자동차용 프리실드 강판의 저항 점 용접성 향상에 관한 연구

        In the present day, new performances are demanded for automobile body steel sheet for example, light weight, durability, resistance corrosion and environmental protection. For an improvement of resistance corrosion, European automobile companies developed pre-sealed steel in 1990's. Pre-sealed steel have organic coated layer upper zinc coated layer. Evaluating a resistance spot weldability of pre-sealed steel plate is important because resistance spot welding is used mainly in automobile body joining process. In this study, evaluating a weldability of pre-sealed steel plate was carried out using lobe diagram, dynamic resistance and high-speed image. Usually, Pre-sealed steel has poor weldability than GA steel's. To improve a weldability of pre-sealed steel sheet, pre-current was applied to welding process. Pre-current is applied prior to base-current. In conclusion, the weldability of pre-sealed steel has been improved than GA steel's by applying pre-current.

        Cochlodinium polykrikoides 적조출현주의보 발령에 분자탐침기법의 활용

        Real-time PCR을 적조출현주의보 발령에 활용하였다. Cochlodinium polykrikoides 적조예보를 위해 $10cells\;mL^{-1}$ 이상에서 적조출현주의보를 발령하였는데 현미경 검경은 저밀도에서 정확도가 낮기 때문에 이를 보완하기 위해 C. polykrikoides 특이적 real-time PCR을 사용하였다. 6월 말경에 C. polykrikoides이 통영, 남해 해역에 저밀도로 광범위하게 출현하였고 2015년 8월 2일 적조출현주의보를 발령하였다. Real-time PCR was applied to early warning of red tide. For early warning of red tide at $10cells\;mL^{-1}$, Cochlodinium polykrikoides specific real-time PCR was used as a complement of microscopy that has a lower detection sensitivity. C. polykrikoides appeared extensively in Tongyeong, Namhae waters at low densities in the end of June, and early warning of C. polyrkrikoides blooms was issued on 2 August 2015.

        Cochlodinium polykrikoides (Dinophyceae)의 동아시아와 필리핀 유전형의 남해안 분포

        어류폐사 와편모조류인 Cochlodinium polykrikoides는 large-subunit(LSU) ribosomal RNA gene을 기반으로 전 세계적으로 4가지 유전타입이 알려져 있으며, 남해안에는 2가지 유전타입이 출현한다고 보고되었다. 본 연구에서는 C. polykrikoides의 동아시아 타입과 필리핀 타입의 남해안 출현양상을 quantitative real-time PCR(qPCR)을 이용하여 3년간(2014~2016년) 조사하였다. 동아시아 타입의 경우 2014~2016년에 40~100% 비율로 남해안 전 정점(통영~완도)에서 검출이 된 반면, 필리핀타입은 대마난류 유입이 강했던 2016년에만 통영~고흥 일부 해역에서 1~2% 비율로 극미량 검출되었다. 위 결과는 동아시아타입이 남해안의 우점 C. polykrikoides 개체군임을 보여주고 있으며, 일부 유영세포는 대마난류를 따라 외해역으로 부터 유입될 수 있음을 시사한다. Fish killing dinoflagellate Cochlodinium polykrikoides has been separated into four genetically differentiated subpopulations globally based on large-subunit (LSU) ribosomal RNA gene, and two subpopulations have been found in the South Sea, Korea. In this study, distributions of the East Asia and Philippines ribotypes were surveyed in the South Sea for 3 years (2014~2016) using quantitative real-time PCR (qPCR). The East Asia ribotype was detected in all sampling stations of the South Sea (Tongyeong~Wando) by 40~100% positives for 2014~2016, whereas the Philippines ribotype was detected in some areas of Tongyeong~Goheung by 1~2% positives for only 2016 when the Tsushima Warm Current (TWC) was particularly strengthened. These results indicate that the East Asia ribotype is the dominant subpopulation in the South Sea, also some of C. polykrikoides swimming cells might be transported from offshore to the South Sea via TWC.

        In recent years, several significant progresses of the analytical technology for reinforced concrete structures have been achieved and as the result, nonlinear behavior of concrete is of interest in analysis and design. An efficient design concept for earthquake loads, is called performance based design, has been standard design in U.S.A, Europe and Japan since those countries experienced sever earthquake damage at end of 90's. To evaluate the performance of the concrete structures, the sophisticated as well as simple analysis tools are necessary. As usually, strut-tie model well predicts the strength of the disturbed region, however, it does not provide ductility information at the failure. Therefore, simple tools which able to predict both the strength and the ductility of RC structures are in demand. 2D lattice model is introduced on this study as the analysis tool for the RC structures subject to earthquake. Experimental correlation studies indicate the 2D lattice model quite well predict the strength as well as the ductility of RC structures.

      • The Fabrication Method of Tip Array by Glass Deep Dry Etching Process

        The process of making the etch mask for the glass dry etching is complicated and its cost is high. To reduce these disadvantages, we suggest the glass dry etching method using a single photoresist (PR) mask. In this works, we figured out the optimized condition for the dry etching using the photoresist without any metal or silicon mask. We observed the characteristics of quartz wafer, such as the etch profile, etch rate, the selectivity and the critical dimension (CD) loss rate.
