      • miR-340 Reverses Cisplatin Resistance of Hepatocellular Carcinoma Cell Lines by Targeting Nrf2-dependent Antioxidant Pathway

        Many chemotherapeutic agents have been successfully used to treat hepatocellular carcinoma (HCC); however, the development of chemoresistance in liver cancer cells usually results in a relapse and worsening of prognosis. It has been demonstrated that DNA methylation and histone modification play crucial roles in chemotherapy resistance. Currently, extensive research has shown that there is another potential mechanism of gene expression control, which is mediated through the function of short noncoding RNAs, especially for microRNAs (miRNAs), but little is known about their roles in cancer cell drug resistance. In present study, by taking advantage of miRNA effects on the resistance of human hepatocellular carcinoma cells line to cisplatin, it has been demonstrated that miR-340 were significantly downregulated whereas Nrf2 was upregulated in HepG2/CDDP (cisplatin) cells, compared with parental HepG2 cells. Bioinformatics analysis and luciferase assays of Nrf2-3'-untranslated region-based reporter constructor indicated that Nrf2 was the direct target gene of miR-340, miR-340 mimics suppressing Nrf2-dependent antioxidant pathway and enhancing the sensitivity of HepG2/CDDP cells to cisplatin. Interestingly, transfection with miR-340 mimics combined with miR-340 inhibitors reactivated the Nrf2 related pathway and restored the resistance of HepG2/CDDP cells to CDDP. Collectively, the results first suggested that lower expression of miR-340 is involved in the development of CDDP resistance in hepatocellular carcinoma cell line, at least partly due to regulating Nrf2-dependent antioxidant pathway.

        Empirical decomposition method for modeless component and its application to VIV analysis

        Aiming at accurately distinguishing modeless component and natural vibration mode terms from data series of nonlinear and non-stationary processes, such as Vortex-Induced Vibration (VIV), a new empirical mode decomposition method has been developed in this paper. The key innovation related to this technique concerns the method to decompose modeless component from non-stationary process, characterized by a predetermined 'maximum intrinsic time window' and cubic spline. The introduction of conceptual modeless component eliminates the requirement of using spurious harmonics to represent nonlinear and non-stationary signals and then makes subsequent modal identification more accurate and meaningful. It neither slacks the vibration power of natural modes nor aggrandizes spurious energy of modeless component. The scale of the maximum intrinsic time window has been well designed, avoiding energy aliasing in data processing. Finally, it has been applied to analyze data series of vortex-induced vibration processes. Taking advantage of this newly introduced empirical decomposition method and mode identification technique, the vibration analysis about vortex-induced vibration becomes more meaningful.

        Primary Aldosteronism and Cerebrovascular Diseases

        As diagnostic techniques have advanced, primary aldosteronism (PA) has emerged as the most common cause of secondary hypertension. The excess of aldosterone caused by PA resulted in not only cardiovascular complications, including coronary artery disease,myocardial infarction, arrhythmia, and heart failure, but also cerebrovascular complications, such as stroke and transient ischemicattack. Moreover, PA is associated more closely with these conditions than is essential hypertension. In this review, we present up-todate findings on the association between PA and cerebrovascular diseases.

        Rate-based model for predicting and evaluating H2S absorption in the haloalkaliphilic biological desulfurization process

        The highly efficient performance of H2S absorption is the crucial indicator for haloalkaliphilic biologicaldesulfurization (HBDS) because it immediately concerns the H2S removal efficiency and pH change ofalkaline solutions. Therefore, we investigated the effect of operating parameters on the H2S absorption’sperformance under haloalkaline conditions. The gas–liquid ratio and packing height significantly improveH2S removal efficiency, from 80% to 90% and 66% to 99%, respectively. The absorption temperature had atrivial impact on the H2S removal efficiency, and the maximum value appeared at 45 C. Additionally, alloperating parameters caused pH changes that varied in the acceptable range (0.1 to 0.5) during theabsorption process. A rate-based model was successfully developed to predict the haloalkaliphilic H2Sabsorption process accurately. Moreover, this model could be implemented to effectively evaluate theHBDS system’s stability and provide reliable theoretical guidance for the industrial HBDS process toensure good process stability.

      • A novel design method for improving collapse resistances of multi-story steel frames with unequal spans using steel braces

        The bearing capacities resisted by the two-bay beams of multi-story planar frames with unequal spans under column removal scenarios differ considerably owing to the asymmetric stress on the left and right beams connected to the failed column and cause the potential for beams with larger span-to-depth ratios to be unable to exert effectively, which is disadvantageous for resisting the vertical load in unequal-span frame structures. To address this problem, the structural measure of adding braces to the weak bays of multi-story unequal-span frames was proposed, with the objective of achieving a coordinated stress state in two-bay beams with unequal spans, thereby improving the collapse resistance of unequal-span frame structures. Before conducting the numerical simulation, the modeling methods were verified by previous experimental results of two multi-story planar frames with and without steel braces. Thereafter, the effects of the tensile and compressive braces on the collapse behavior of the frame structures were elucidated. Then, based on the mechanical action laws of the braces throughout the collapse process, a detailed design method for improving the collapse resistance of unequal-span frame structures was proposed. Finally, the proposed design method was verified by using sufficient example models, and the results demonstrated that the design method has good application prospects and high practical value.

        Centromere protein N promotes lung adenocarcinoma progression by activating PI3K/AKT signaling pathway

        Background: As an important member of centromere family, centromere associated protein N (CENPN) was abnormally expressed in varied malignant tumors. Objective: This paper aimed to analyze the expression and related mechanism of CENPN in lung adenocarcinoma (LUAD). Methods: The expression of CENPN in LUAD was analyzed by Gene Expression Profiling Interactive Analysis (GEPIA) database. The mRNA expression, protein expression, cell viability, cell invasion, cell apoptosis, cell stem like characteristics were detected by RT-PCR, western blot, CCK8 assay, transwell assay, flow cytometry and spheroidization assay, respectively. Finally, the pathological changes of xenograft were estimated by H&E staining, and the expression of proteins was detected by immunohistochemistry. Results: GEPIA analysis showed that the CENPN expression in LUAD was significantly higher than that in normal lung tissue, which was negatively correlated with the prognosis. These results were consistent with our clinical data. Besides, CENPN was highly expressed in LUAD cell lines. In addition, the upregulation of CENPN amplified the cell viability, stemness and invasive ability in PC9 cells. However, the knockdown of CENPN inhibited the cell activity, stemness, invasive ability with increased cell apoptosis in A549. Furthermore, CENPN could positively regulate the phosphorylation of PI3K and AKT. The PI3K inhibitor, 740Y-P, could reverse the effect of CENPN silencing on the expression of Ki-67, cleaved caspase 3, OCT4, and snail 1. Finally, the downregulation of CENPN restrained the growth of xenograft and inactivated the PI3K/AKT pathway. Conclusion: CENPN was abnormally overexpressed in LUAD, and promoted tumor progression of LUAD by affecting PI3K/AKT pathway.

      • Instantaneous multi-mode identification and analysis of vortex-induced vibration via a mode decomposition method

        <P><B>Abstract</B></P> <P>The dynamic characteristics of marine risers/pipes often present serried modes with various frequencies due to high levels of structural flexibility and slenderness, especially when the flow velocity is non-uniformly distributed along the span. Therefore, the vortex-induced vibration (hence VIV) for slender risers/pipes is usually characterized by multi-mode motions. In this paper, by means of a newly developed empirical mode decomposition (EMD) method which contributes to more efficient instantaneous multi-mode identification and analysis, new characteristics of a multi-mode “lock-in” vibration process of a large-scale flexible pipe subject to shear flow were discussed. Because the two-degree vibration along the span can be analyzed simultaneously, the effects of multi-mode VIV were investigated systematically. From the given illustrative examples, it was found that the vibration energy diffusion between the fluid and the structure, and among the participating modes, may be repeatable and reversible, or even irreversible, which causes VIV to be highly intricate. The coexistence of multiple modes, energy transfer, and mode switching/jump is observed when the reduced velocity is relatively high. The multi-dominant mode phenomenon is also found in both cross-flow (CF) and in-line (IL) VIVs.         Effect of B-complex vitamins on the antifatigue activity and bioavailability of ginsenoside Re after oral administration

        Background: Both ginsenoside Re and B-complex vitamins are widely used as nutritional supplements. They are often taken together so as to fully utilize their antifatigue and refreshing effects, respectively. Whether actually a drug-nutrient interaction exists between ginsenoside Re and B-complex vitamins is still unknown. The objective of this study was to simultaneously investigate the effect of B-complex vitamins on the antifatigue activity and bioavailability of ginsenoside Re after their oral administration. The study results will provide valuable theoretical guidance for the combined utilization of ginseng and B-complex vitamins. Methods: Ginsenoside Re with or without B-complex vitamins was orally administered to mice to evaluate its antifatigue effects and to rats to evaluate its bioavailability. The antifatigue activity was evaluated by the weight-loaded swimming test and biochemical parameters, including hepatic glycogen, plasma urea nitrogen, and blood lactic acid. The concentration of ginsenoside Re in plasma was determined by liquid chromatography-tandem mass spectrometry. Results: No antifatigue effect of ginsenoside Re was noted when ginsenoside Re in combination with B-complex vitamins was orally administered to mice. B-complex vitamins caused to a reduction in the bioavailability of ginsenoside Re with the area under the concentration-time curve from zero to infinity markedly decreasing from $11,830.85{\pm}2,366.47h{\cdot}ng/mL$ to $890.55{\pm}372.94h{\cdot}ng/mL$. Conclusion: The results suggested that there were pharmacokinetic and pharmacodynamic drug-nutrient interactions between ginsenoside Re and B-complex vitamins. B-complex vitamins can significantly weaken the antifatigue effect and decrease the bioavailability of ginsenoside Re when simultaneously administered orally.

        Ginsenoside Rb1 ameliorates cisplatin-induced learning and memory impairments

        Background: Ginsenoside Rb1 (Rb1), a dominant component from the extract of Panax ginseng root,exhibits neuroprotective functions in many neurological diseases. This study was intended to investigatewhether Rb1 can attenuate cisplatin-induced memory impairments and explore the potentialmechanisms. Methods: Cisplatin was injected intraperitoneally with a dose of 5 mg/kg/wk, and Rb1 was administeredin drinking water at the dose of 2 mg/kg/d to rats for 5 consecutive wk. The novel objects recognition taskand Morris water maze were used to detect the memory of rats. Nissl staining was used to examine theneuron numbers in the hippocampus. The activities of superoxide dismutase, glutathione peroxidase,cholineacetyltransferase, acetylcholinesterase, and the levels of malondialdehyde, reactive oxygen species,acetylcholine, tumor necrosis factor-a, interleukin-1b, and interleukin-10 were measured by ELISAto assay the oxidative stress, cholinergic function, and neuroinflammation in the hippocampus. Results: Rb1 administration effectively ameliorates the memory impairments caused by cisplatin in bothnovel objects recognition task and Morris water maze task. Rb1 also attenuates the neuronal loss inducedby cisplatin in the different regions (CA1, CA3, and dentate gyrus) of the hippocampus. Meanwhile, Rb1 isable to rescue the cholinergic neuron function, inhibit the oxidative stress and neuroinflammation incisplatin-induced rat brain. Conclusion: Rb1 rescues the cisplatin-induced memory impairment via restoring the neuronal loss byreducing oxidative stress and neuroinflammation and recovering the cholinergic neuron functions.
