      • A novel design method for improving collapse resistances of multi-story steel frames with unequal spans using steel braces

        The bearing capacities resisted by the two-bay beams of multi-story planar frames with unequal spans under column removal scenarios differ considerably owing to the asymmetric stress on the left and right beams connected to the failed column and cause the potential for beams with larger span-to-depth ratios to be unable to exert effectively, which is disadvantageous for resisting the vertical load in unequal-span frame structures. To address this problem, the structural measure of adding braces to the weak bays of multi-story unequal-span frames was proposed, with the objective of achieving a coordinated stress state in two-bay beams with unequal spans, thereby improving the collapse resistance of unequal-span frame structures. Before conducting the numerical simulation, the modeling methods were verified by previous experimental results of two multi-story planar frames with and without steel braces. Thereafter, the effects of the tensile and compressive braces on the collapse behavior of the frame structures were elucidated. Then, based on the mechanical action laws of the braces throughout the collapse process, a detailed design method for improving the collapse resistance of unequal-span frame structures was proposed. Finally, the proposed design method was verified by using sufficient example models, and the results demonstrated that the design method has good application prospects and high practical value.

      • Townscape in a High-rise: Imageability and Accessibility of Vertical Malls in Hong Kong

        The increasing integration of public space and consumerism in Hong Kong has yielded new urban forms. The emergent vertical malls in Hong Kong and other East Asian metropolises have overturned the existing vertical order of the city. This vertical order is determined by the level of accessibility, but is being challenged by widely adopted vertical circulation technology. Inspired by Fredric Jameson's and Rem Koolhaas' reflections on the cultural significance of vertical transportation, this article examines the conflict between market logic and urban design requirements in the vertical interior spaces. "Departmentalization," as the current programming formula for vertical malls, can be further optimized by critically applying urban design doctrines such as Kevin Lynch's five elements of city image. It concludes with a statement that the knowledge base of vertical urbanism should be open to a set of new terminology informed by a new technological environment.

        TET2 Regulates 5-Hydroxymethylcytosine Signature and CD4+ T-Cell Balance in Allergic Rhinitis

        Purpose: Previous studies have shown the role of ten-eleven translocation 2 (TET2) in CD4+ T cells. However, its function in CD4+ T cells under allergic inflammation is unclear. We aimed to investigate the epigenomic distribution of DNA 5-hydroxymethylcytosine (5hmC) and the role of TET2 in CD4+ T cells of allergic rhinitis (AR). Methods: The hMeDIP-seq was performed to identify sequences with 5hmC deposition in CD4+ T cells of AR patients. Tet2-deficient or wild type mice were stimulated with ovalbumin (OVA) to develop an AR mouse model. The histopathology in nasal mucosae, Th1/Th2/Treg/Th17 cell percentage, concentrations of Th-related cytokines, expression of Tet and differential hydroxymethylated genes (DhMG), and the global deposition of 5hmC in sorted CD4+ T cells were detected. Results: Epigenome-wide 5hmC landscape and DhMG in the CD4+ T cells of AR patients were identified. Tet2 depletion did not led to spontaneous inflammation. However, under the stimulation of allergen, OVA, loss of Tet2 resulted in the exacerbation of allergic inflammation, which was characterized by severer allergic symptoms, more inflammatory cells infiltrating the nasal lamina propria, sharper imbalances between Th1/Th2 and Treg/Th17 cells, and excessive secretion of OVA-specific IgE and Th2-related cytokines. Moreover, altered mRNA production of several DhMG and sharp decrease in 5hmC deposition were also observed in Tet2-deficient OVA-exposed mice. Conclusions: TET2 may regulate DNA 5hmC, DhMG expressions, and CD4+ T cell balance in AR.

        Cloud computing task scheduling is a multi-objective decision on how to perform a variety of tasks simultaneously reasonable sort is essential. Therefore, this article constructs a population of convergence non-dominated sorting method. This method is based on non-dominated sorting method; the use of distributed estimation method is improved by four steps to complete the task order scheduling. The experimental results show that this method has good convergence and distribution.

      • Stable and Active Oxidation Catalysis by Cooperative Lattice Oxygen Redox on SmMn<sub>2</sub>O<sub>5</sub> Mullite Surface

        <P>The correlation between lattice oxygen (O) binding energy and O oxidation activity imposes a fundamental limit in developing oxide catalysts, simultaneously meeting the stringent thermal stability and catalytic activity standards for complete oxidation reactions under harsh conditions. Typically, strong O binding indicates a stable surface structure, but low O oxidation activity, and <I>vice</I><I>versa</I>. Using nitric oxide (NO) catalytic oxidation as a model reaction, we demonstrate that this conflicting correlation can be avoided by cooperative lattice oxygen redox on SmMn<SUB>2</SUB>O<SUB>5</SUB> mullite oxides, leading to stable and active oxide surface structures. The strongly bound neighboring lattice oxygen pair cooperates in NO oxidation to form bridging nitrate (NO<SUB>3</SUB><SUP>-</SUP>) intermediates, which can facilely transform into monodentate NO<SUB>3</SUB><SUP>-</SUP> by a concerted rotation with simultaneous O<SUB>2</SUB> adsorption onto the resulting oxygen vacancy. Subsequently, monodentate NO<SUB>3</SUB><SUP>-</SUP> species decompose to NO<SUB>2</SUB> to restore one of the lattice oxygen atoms that act as a reversible redox center, and the vacancy can easily activate O<SUB>2</SUB> to replenish the consumed one. This discovery not only provides insights into the cooperative reaction mechanism but also aids the design of oxidation catalysts with the strong O binding region, offering strong activation of O<SUB>2</SUB>, high O activity, and high thermal stability in harsh conditions.</P> [FIG OMISSION]</BR>

      • Fundamental Issues Related to the Origin of Melatonin and Melatonin Isomers during Evolution: Relation to Their Biological Functions

        <P>Melatonin and melatonin isomers exist and/or coexist in living organisms including yeasts, bacteria and plants. The levels of melatonin isomers are significantly higher than that of melatonin in some plants and in several fermented products such as in wine and bread. Currently, there are no reports documenting the presence of melatonin isomers in vertebrates. From an evolutionary point of view, it is unlikely that melatonin isomers do not exist in vertebrates. On the other hand, large quantities of the microbial flora exist in the gut of the vertebrates. These microorganisms frequently exchange materials with the host. Melatonin isomers, which are produced by these organisms inevitably enter the host’s system. The origins of melatonin and its isomers can be traced back to photosynthetic bacteria and other primitive unicellular organisms. Since some of these bacteria are believed to be the precursors of mitochondria and chloroplasts these cellular organelles may be the primary sites of melatonin production in animals or in plants, respectively. Phylogenic analysis based on its rate-limiting synthetic enzyme, serotonin <I>N</I>-acetyltransferase (SNAT), indicates its multiple origins during evolution. Therefore, it is likely that melatonin and its isomer are also present in the domain of archaea, which perhaps require these molecules to protect them against hostile environments including extremely high or low temperature. Evidence indicates that the initial and primary function of melatonin and its isomers was to serve as the first-line of defence against oxidative stress and all other functions were acquired during evolution either by the process of adoption or by the extension of its antioxidative capacity.</P>

      • Two-step enhanced cancer immunotherapy with engineered <i>Salmonella typhimurium</i> secreting heterologous flagellin

        <P>We report a method of cancer immunotherapy using an attenuated Salmonella typhimurium strain engineered to secrete Vibrio vulnificus flagellin B (FlaB) in tumor tissues. Engineered FlaB-secreting bacteria effectively suppressed tumor growth and metastasis inmousemodels and prolonged survival. By using Toll-like receptor 5 (TLR5)-negative colon cancer cell lines, we provided evidence that the FlaB-mediated tumor suppression upon bacterial colonization is associated with TLR5-mediated host reactions in the tumor microenvironment. These therapeutic effects were completely abrogated in TLR4 and MyD88 knockout mice, and partly in TLR5 knockout mice, indicating that TLR4 signaling is a requisite for tumor suppression mediated by FlaB-secreting bacteria, whereas TLR5 signaling augmented tumor-suppressive host reactions. Tumor microenvironment colonization by engineered Salmonella appeared to induce the infiltration of abundant immune cells such as monocytes/macrophages and neutrophils via TLR4 signaling. Subsequent secretion of FlaB from colonizing Salmonella resulted in phenotypic and functional activation of intratumoral macrophages with M1 phenotypes and a reciprocal reduction in M2-like suppressive activities. Together, these findings provide evidence that nonvirulent tumor-targeting bacteria releasing multiple TLR ligands can be used as cancer immunotherapeutics.</P>

      • Rational design of common transition metal-nitrogen-carbon catalysts for oxygen reduction reaction in fuel cells

        <P><B>Abstract</B></P> <P>Bio-inspired non-precious-metal catalysts based on iron and cobalt porphyrins are promising alternatives to replace costly platinum-based catalysts for oxygen reduction reaction (ORR) in fuel cells. However, the exact nature of the active sites is still not clearly understood, and further optimization design is needed for practical applications. Here, we report a rational catalyst design process by combining density functional theory (DFT) calculations and experimental validations. Two sets of square-planar (MN<SUB>x</SUB>C<SUB>4−x</SUB>) and square-pyramid (MN<SUB>x</SUB>C<SUB>5−x</SUB>) active centers (M=Mn, Fe, Co, Ni) incorporated in graphene were examined using DFT. Fe-N<SUB>5</SUB> and Co-N<SUB>4</SUB> sites were identified theoretically to have the best performance in fuel cells, while Ni-N<SUB>x</SUB>C<SUB>4−x</SUB> sites catalyze the most H<SUB>2</SUB>O<SUB>2</SUB> byproduct.         A Fusion Protein of Derp2 Allergen and Flagellin Suppresses Experimental Allergic Asthma

        Purpose: The house dust mite (HDM) is one of the most important sources of indoor allergens and a significant cause of allergic rhinitis and allergic asthma. Our previous studies demonstrated that Vibrio vulnificus flagellin B (FlaB) plus allergen as a co-treatment mixture improved lung function and inhibited eosinophilic airway inflammation through the Toll-like receptor 5 signaling pathway in an ovalbumin (OVA)- or HDM-induced mouse asthma model. In the present study, we fused the major mite allergen Derp2 to FlaB and compared the therapeutic effects of the Derp2-FlaB fusion protein with those of a mixture of Derp2 and FlaB in a Derp2-induced mouse asthma model. Methods: BALB/c mice sensitized with Derp2 + HDM were treated with Derp2, a Derp2 plus FlaB (Derp2 + FlaB) mixture, or the Derp2-FlaB fusion protein 3 times at 1-week intervals. Seven days after the final treatment, the mice were challenged intranasally with Derp2, and airway responses and Derp2-specific immune responses were evaluated. Results: The Derp2-FlaB fusion protein was significantly more efficacious in reducing airway hyperresponsiveness, lung eosinophil infiltration, and Derp2-specific IgE than the Derp2 + FlaB mixture. Conclusions: The Derp2-FlaB fusion protein showed a strong anti-asthma immunomodulatory capacity, leading to the prevention of airway inflammatory responses in a murine disease model through the inhibition of Th2 responses. These findings suggest that the Derp2-FlaB fusion protein would be a promising vaccine candidate for HDM-mediated allergic asthma therapy.
