        High-j Proton h11/2 and g7/2 Intruder Bands in 113In

        Excited states of 113In have been populated through the heavy-ion fusion evaporation reaction 110Pd(7Li, 4n)113In. A new band with the configuration of a proton d5/2 orbital is identified. Two ΔI = 2 intruder bands, built on the πh11/2 and the πg7/2 orbitals, have been extended to spins (63/2-)ħ and (55/2+)ħ, respectively. The negative-parity πh11/2 intruder band shows a smooth increase in aligned spin, which is attributed to a strong proton-neutron interaction. The properties of the positive-parity πg7/2 band are discussed based on tilted axis cranking model calculations, and the features of the antimagnetic rotation for this band are shown after backbend. Furthermore, the contributions of the two-shears-like mechanism, the neutron (gd)ν shell and the core rotation are investigated for the positive-parity πg7/2 band.

        Microstructure and Biocompatibility of Porous Ti-Nb-Zr-CPP Composite Fabricated by Spark Plasma Sintering and Leaching Process

        Metallic biomaterials such as titanium and its alloys have been widely used as permanent implant materials due to their excellent corrosion resistance, fatigue property, and strength. Calcium pyrophosphate (CPP) is an attractive bioceramic for human tissue implantation due to its good biocompatibility and similar composition to human bone. High energy mechanical milling and spark plasma sintering (SPS) can be used to fabricate a sound composite of the Ti-Nb-Zr-CPP. In a biomaterial such as the Ti-6Al-4V ELI alloy, the coating property between the bioceramic and Ti-based alloy is very important. By addition of CPP to the Ti-Nb-Zr alloy, the Ti-Nb-Zr alloy can improve the wetability between the Ti alloy and bioceramic as a coating material. Also the porous surface of the biomaterials can improve the symphyseal ability with osteoblast. In this study, a porous surface can be made by using several different leaching solutions such as aqueous diluted H3PO4. Ti-Nb-Zr-CPP composite was fabricated by spark plasma sintering (SPS) at 1000 ℃ under 70 MPa using high energy mechanical milled powder. A porous surface was successfully fabricated by using a 40 vol%H3PO4+ 60 vol.%H2O leaching solution at room temperature for 24 h.

      • Dynamics and Liver Disease Specific Aspects of Quality of Life Among Patients with Chronic Liver Disease in Yunnan, China

        Background: Patients with chronic liver diseases (CLD) may have compromised health related quality of life (HRQoL). Hepatitis B virus (HBV) infection has long been the leading cause of CLD including liver cancer and cirrhosis. Knowledge on different symptom profiles of CLD should help in development of comprehensive treatment and patient care plans. Objective: To access the facets of HRQoL in chronic liver diseases throughout their spectrum of severity. Materials and Methods: A cross-sectional study was conducted in the First Affiliated Hospital of Kunming Medical University in Yunnan Province of China. Both out- and inpatients undergoing treatment protocols for different HBV related liver disease states were consecutively collected from December 2012 to June 2013. ANOVA was used to compare the mean scores of EQ-5D and chronic liver disease questionnaire (CLDQ) among 5 disease groups. The relationship between demographic variables predicting global CLDQ scores and the domains of CLDQ was analysed. Results: A total of 1040 patients including 520 without complications, 91 with compensated cirrhosis, 198 with decompensated cirrhosis, 131 with HCC and 100 with liver failure were recruited. All domains of CLDQ, the means of EQ-5D value and EQ VAS exhibited significant decline with worsening of disease severity from uncomplicated HBV to liver failure. The multivariate regression demonstrated the reduction of mean scores of CLDQ domain at advanced stage. Patients with liver failure and HCC had more HRQoL impairment than other disease states. No effect of patient gender was found. Patient age was associated with 'fatigue' and 'worry' domains (p=0.006; p=0.004) but not with other domains and global scores of CLDQ and ED-5D. Conclusions: The HRQoL in chronic hepatitis B patients is greatly affected by disease states. Care for HBV-related diseases should consider not only the outcomes of treatment strategies but also improvement in patient wellbeing.

      • Knockdown of HMGN5 Expression by RNA Interference Induces Cell Cycle Arrest in Human Lung Cancer Cells

        HMGN5 is a typical member of the HMGN (high mobility group nucleosome-binding protein) family which may function as a nucleosomal binding and transcriptional activating protein. Overexpression of HMGN5 has been observed in several human tumors but its role in tumorigenesis has not been fully clarified. To investigate its significance for human lung cancer progression, we successfully constructed a shRNA expression lentiviral vector in which sense and antisense sequences targeting the human HMGN5 were linked with a 9-nucleotide loop. Inhibitory effects of siRNA on endogenous HMGN5 gene expression and protein synthesis were demonstrated via real-time RT-PCR and western blotting. We found HMGN5 silencing to significantly inhibit A549 and H1299 cell proliferation assessed by MTT, BrdU incorporation and colony formation assays. Furthermore, flow cytometry analysis showed that specific knockdown of HMGN5 slowed down the cell cycle at the G0/G1 phase and decreased the populations of A549 and H1299 cells at the S and G2/M phases. Taken together, these results suggest that HMGN5 is directly involved in regulation cell proliferation in A549 and H1299 cells by influencing signaling pathways involved in cell cycle progression. Thus, our finding suggests that targeting HMGN5 may be an effective strategy for human lung cancer treatment.

      • Rs895819 within miR-27a Might be Involved in Development of Non Small Cell Lung Cancer in the Chinese Han Population

        MicroRNA-27a (miR-27a) is deemed to be an oncogene that plays an important role in development of various cancers, and single nucleotide polymorphism (SNP) of miR-27a can influence the maturation or aberrant expression of hsa-miR27a, resulting in increased risk of cancer and poor prognosis for non-small cell lung cancer (NSCLC). This study aimed to assess the effects of rs895819 within miR-27a on susceptibility and prognosis of NSCLC patients in 560 clinical confirmed cases and 568 healthy check-up individuals. Adjusted odds/hazard ratios (ORs/HRs) and 95% confidential intervals (CIs) were calculated to evaluate the association between rs895819 and the risk and prognosis of NSCLC. The results showed that allele A and genotype GG of rs895819 were significantly associated with an increased risk of NSCLC (38.9% vs 30.8%, adjusted OR=1.26, 95%CI=1.23-1.29 for allele G vs A; 18.1% vs 11.7%, adjusted OR=1.67, 95%CI=1.59-1.75 for genotype GG vs AA). Moreover, positive associations were also observed in dominant and recessive models (53.7% vs 49.9%, adjusted OR=1.17, 95%CI=1.13-1.20 for GG/AG vs AA; 18.1% vs 11.7%, adjusted=1.65, 95%CI=1.58-1.73). However, no significant association was found between rs895819 and the prognosis of NSCLC in genotype, dominant and recessive models. These results suggested that miR-27a might be involved in NSCLC carcinogenesis, but not in progression of NSCLC. The allele G, genotype GG and allele G carrier (GG/AG vs AA) of rs895819 might be genetic susceptible factors for NSCLC. Further multi-central, large sample size and well-designed prospective studies as well as functional studies are warranted to verify our findings.

      • Prognostic Role of MicroRNA-21 in Non-small Cell Lung Cancer: a Meta-analysis

        Introduction: Many studies have reported that microRNA-21 (miR-21) mihght predict the survival outcome in non-small cell lung cancers (NSCLCs) but the opposite opinion has also been expressed. The aim of this study was to summarize the evidence for a prognostic role of miR-21. Materials and Methods: All the eligible studies was searched by Medline and EMBASE and patients' clinical characteristics and survival outcome were extracted. Then a meta-analysis was performed to clarify the prognostic role of the miR-21 expression in different subgroups. Results: A total of 8 eligible articles were yielded covering survival outcomes or clinical characteristics. The combined hazard ratio (HR) and 95% confidence interval (95% CI) for overall survival (OS) was 2.19 [0.76, 6.30], while the combined HR (95% CI) of Asian group for OS had a significant result, 5.49 [2.46, 12.27]. The combined HR (95% CI) for recurrence free survival or disease free survival (RFS/DFS) was 2.31 [1.52, 3.49]. Odds ratios (ORs) showed that the miR-21 expression was associated with lymph node status and histological type. Conclusion: miR-21 expression could predict the prognostic outcome of NSCLC in Asians, despite some deficiencies in the study data.

        Molecule-based electrorheological material assembled using β-cyclodextrin as substrate

        Molecule-based electrorheological (ER) materials as a novel type of ER materials, the inclusion compound [H2(β-CD-A)-PTA] between p-toluenesulfonic acid (PTA, guest) and H2(β-CD-A) (host) that is dicarboxylic acid of β-cyclodextrin (β-CD) , and the rare earth (RE) complexes [(β-CD-A)-PTA]3RE2 (RE=La, Gd, Y)of H2(β-CD-A)-PTA, were synthesized. The ER performance and dielectric property of the materials were studied. Our results show that the molecule-based ER materials assembled using β-cyclodextrin as a substrate,especially the inclusion compound and its rare earth (RE) complexes exhibit clear ER effect. The inclusion PTA can markedly enhance the ER performance of H2(β-CD-A) material. The ER activity of the yttrium complex is the highest among these materials. The characteristic of the molecule in molecule-based ER materials is an important factor in influencing ER property.

        Neurotrophic Effects of Schwann Cell Secretions from Wallerian Degenerating Facial Nerves on Neonatal Rats Facial Motoneurons in vitro

        Regeneration and functional recovery of facial nerves after injury are still far from satisfaction. Studies on whether Schwann cells of injured facial nerves could secrete neurotrophic factors to promote nerve regeneration are rare, and that is the pbjective of this study. Schwann cells from Wallerian degenerating segments of rats facial nerves were isolated and cultured in conditioned culture medium, collected, concentrated and fractionated by ultra-filtration. The neurotrophic bioactivity of the soluble secretions from cultured Schwann cells were examined on an established neuron culture model. Dil fluorescence labeling facial motoneurons from neonatal rats, MTT assay, and image analysis for survival of facial motoneurons were used. The results show that facial motoneuron survival activity in Schwann cell culture medium containing a molecular weight larger than 30 kDa component, was significantly higher than in medium with and without serum (p<0.05 and p<0.01 respectively). The effect could be extinguished by either boiling or trypsinizing. At the 2nd, 4th and 7th days after conditioned culture, mean perikaryon area and neurite length of facial motoneurons in a medium containing molecular weight larger than 30kDa component, were significantly larger than in serum containing medium. The results show that Schwann cells from Wallerian degenerating facial nerves in serum-free and neurite re-growth of facial motoneurons in vitro. The results also suggest that the neurotrophic effect might be derived from protein(s) or peptide(s) with molecular weight larger than 30 kDa component of the soluble secretions. It may be potent for functional repair of facial verve and other neural disorders.

        Progranulin Controls Sepsis via C/EBPα-Regulated <i>Il10</i> Transcription and Ubiquitin Ligase/Proteasome-Mediated Protein Degradation

        <P>Progranulin (PGRN) is a widely expressed, pleiotropic protein that is involved in diverse biological processes, including cellular proliferation, neuron development, and wound healing. However, the role of PGRN in the regulation of pathogen-induced systemic inflammation and the mechanisms involved have not been established. In this study, we show that PGRN-deficient mice display heightened mortality in models of polymicrobial sepsis and endotoxinemia, with increased tissue levels of inflammatory cytokines and reduced IL-10 production. Conversely, administration of rPGRN decreases the susceptibility of PGRN-deficient mice to LPS-induced endotoxemic shock and augments IL-10 production by LPS-activated macrophages in a TNFR-dependent manner. Molecular analysis reveals a direct role of the transcription factor C/EBP alpha in PGRN-regulated IL-10 expression. C/EBP alpha-deficient macrophages produce less IL-10 in response to LPS. Furthermore, mice deficient in C/EBP alpha in hematopoietic cells are highly vulnerable to LPS-induced septic shock. Lastly, the defective IL-10 production by PGRN-deficient cells is primarily due to reduced C/EBP alpha protein stability via the E3 ubiquitin-conjugating enzyme E6AP and proteasome-mediated degradation. To our knowledge, this study provides the first evidence that PGRN is a nonredundant regulator of systemic inflammation via modulating the levels and activity of C/EBP alpha, IL-10, and the ubiquitin-proteasome proteolysis pathway. The results bear strong and profound implications for PGRN insufficiency and its mutation-associated systemic and organ-specific inflammatory human diseases.</P>

      • PROGRAMMABLE INDUCTANCE AND CAPACITANCE GENERATOR

        This paper introduces an impedance converter principle of simulated pure inductance and capacitance by using operational amplifiers, in addition, it provides the circuitry configurations according to this principle. The generator can produce equivalent inductance from 0.1H to 6553500H or equivalent capacitance from 10^(-10) F to 65535 × 10^(-7) by programming control. Its circuit is very simple, this generator can be programmed control conveniently and integrated easily. It especially has certain advantages in simulating large inductance or capacitance and programming control.
