Diatomaceous earth를 이용한 경제적이고 효율높은 homemade PCR purification column의 제조 및 평가

In recent years the amount of sequencing has increased tremendously, especially due to the introduction of automated DNA sequencing. PCR is principal method for the synthesis of templates for DNA sequencing. Consequently, the need for PCR cleanup for template preparation has increased. Already, many PCR purification kits are marketed. This DNA purification kits uses most glass membrane or silica gel, So, can do rapidly efficient purification by simple miniprep. But, efficiency of purification kits is different according to manufacturer and is very expensive. Therefore, we compare the homemade Diatomaceous earth column with major manufacturer's spin column methods for DNA purification. As a result, we found that the homemade Diatomaceous earth column method resulted in equal or better yields relative to the commercial purification kits.

This study proposes a technique utilizing virtual information instead of real information in a teleoperated wiping operation. A teleoperation system composed of a master manipulator, a slave manipulator, and a real-time control system is used for this study. Both the master and slave manipulator have a kinematic structure effectively conveying and following the operator’s command respectively. The master manipulator uses DC servo motors to do a major role for counter balancing. A force sensor attached at the end of the slave manipulator’s end effector is used to measure a contact force. MATLAB/xPC Target is used for real-time control of the teleoperation system in Windows OS. This study proposes algorithms for generating the virtual force and the virtual velocity. They are generated as the master manipulator’s grip moves up and down, and back and forth. A force-force control technique proposing in this paper uses the virtual force as a reference input for the contact force control, and uses the virtual velocity as a reference input for control the slave manipulator’s horizontal movement. A PID controller is used as a local controller for the contact force control. In order to show the effectiveness of the proposing force-force control technique, experiments are performed on a plastic plate with two bumps.

In this study for the analysis of ultimate bearing capacity of drilled shaft socketed into different rock types, the results of load transfer data of Kimhae bridge, Namhang and Inchon grand bridge were used to compare the measured and theoretical formular values. The most of bearing capacities by the theoretical formular were lower than the results of static load test, which means that the theoretical formular punderestimate the actual bearing capacity of piles. For Kimhae bridge, the amount of maximum settlement was 17.79mm under 1650 ton applied and remaining settlement was 4.71mm. 76.5 percent of side resistance was transfered in sedementary layer, while 23.6 percent in weathered rock layer. To establish the reasonable design method in domestic site, the data of static load test including axial load transfer analysis should be accumulated and analyzed. In recent, new load test like as both direction load test would be used in sites, the ultimate bearing capacity of drilled shaft could be evaluated more precisely.

This paper introduces a new zinc coated copper wire electrode with coiled cone shape which has low surface resistance and tolerance to the motion artifact for promontory stimulation electrically auditory brainstem responses (PSEABR). Auditory brainstem responses (ABR) can be used to predict hearing threshold level with a great deal of accuracy particularly for a young child who cannot cooperate mechanically and some hearing impaired who are exaggerating a hearing loss for economic compensation. While severe profound sensorineural hearing losses may not be implemented by auditory potentials, PSEABR is proven as a useful tool even for some sensorineural related hearing impaired. It was shown that PSEABR gives the electrical stimuli to promontory of the cochlear instead of giving acoustic stimuli. For this reason, PSEABR can be used as an alternative for cochlear implantation, and can also be used as an optimal device selection and neural information for MAP. It was found that the role of electrode is very important in PSEABR. Even though this cone-shaped electrode was applied in animal experiments, waveforms are well produced by PSEABR. Thus, it was concluded that cone-shaped electrode turned out to be a useful preoperative audiological evaluation tool in deciding time for cochlear implantation surgery.

This paper introduces a development of an electrical stimulator for auditory stimulation. The electrical stimulator is useful in neurotological diagnosis, audiological evaluation, candidate selection for cochlear implantation, optimal device selection and decision making of MAP strategy for severe-to-profound hearing impaired persons. The development was based on sound parameters of auditory brainstem responses and auditory electrophysiological characteristic such as effective firing of auditory nerve and recording evoked potentials during refractory period of neuron. Besides pulse parameter could adjustable by programming for more varied electrical stimulation evoked response audiometry. Using the electrical stimulator, electrical square pulse was applied to promontory, and electrically evoked auditory brainstem response and electrically middle latency response were successfully recorded in cats.
