        PRPF8 increases the aggressiveness of hepatocellular carcinoma by regulating FAK/AKT pathway via fibronectin 1 splicing

        Hepatocellular carcinoma (HCC) pathogenesis is associated with alterations in splicing machinery components (spliceosome and splicing factors) and aberrant expression of oncogenic splice variants. We aimed to analyze the expression and potential role of the spliceosome component PRPF8 (pre-mRNA processing factor 8) in HCC. PRPF8 expression (mRNA/protein) was analyzed in a retrospective cohort of HCC patients (n = 172 HCC and nontumor tissues) and validated in two in silico cohorts (TCGA and CPTAC). PRPF8 expression was silenced in liver cancer cell lines and in xenograft tumors to understand the functional and mechanistic consequences. In silico RNAseq and CLIPseq data were also analyzed. Our results indicate that PRPF8 is overexpressed in HCC and associated with increased tumor aggressiveness (patient survival, etc.), expression of HCC-related splice variants, and modulation of critical genes implicated in cancer-related pathways. PRPF8 silencing ameliorated aggressiveness in vitro and decreased tumor growth in vivo. Analysis of in silico CLIPseq data in HepG2 cells demonstrated that PRPF8 binds preferentially to exons of protein-coding genes, and RNAseq analysis showed that PRPF8 silencing alters splicing events in multiple genes. Integrated and in vitro analyses revealed that PRPF8 silencing modulates fibronectin (FN1) splicing, promoting the exclusion of exon 40.2, which is paramount for binding to integrins. Consistent with this finding, PRPF8 silencing reduced FAK/AKT phosphorylation and blunted stress fiber formation. Indeed, HepG2 and Hep3B cells exhibited a lower invasive capacity in membranes treated with conditioned medium from PRPF8-silenced cells compared to medium from scramble-treated cells. This study demonstrates that PRPF8 is overexpressed and associated with aggressiveness in HCC and plays important roles in hepatocarcinogenesis by altering FN1 splicing, FAK/AKT activation and stress fiber formation.

        Simultaneous pulsatile flow and oscillating wall of a non-Newtonian liquid

        In this work, analytical predictions of the rectilinear flow of a non-Newtonian liquid are given. The fluid is subjected to a combined flow: A pulsatile time-dependent pressure gradient and a random longitudinal vibration at the wall acting simultaneously. The fluctuating component of the combined pressure gradient and oscillating flow is assumed to be of small amplitude and can be adequately represented by a weakly stochastic process, for which a quasi-static perturbation solution scheme is suggested, in terms of a small parameter. This flow is analyzed with the Tanner constitutive equation model with the viscosity function represented by the Ellis model. According to the coupled Tanner-Ellis model, the flow enhancement can be separated in two contributions (pulsatile and oscillating mechanisms) and the power requirement is always positive and can be interpreted as the sum of a pulsatile, oscillating, and the coupled systems respectively. Both expressions depend on the amplitude of the oscillations, the perturbation parameter, the exponent of the Ellis model (associated to the shear thinning or thickening mechanisms), and the Reynolds and Deborah numbers. At small wall stress values, the flow enhancement is dominated by the axial wall oscillations whereas at high wall stress values, the system is governed by the pulsating noise perturbation. The flow transition is obtained for a critical shear stress which is a function of the Reynolds number, dimensionless frequency and the ratio of the two amplitudes associated with the pulsating and oscillating perturbations. In addition, the flow enhancement is compared with analytical and numerical predictions of the Reiner-Phillipoff and Carreau models. Finally, the flow enhancement and power requirement are predicted using biological rheometric data of blood with low cholesterol content.

        Specific contact resistance of IGZO thin film transistors with metallic and transparent conductive oxides electrodes and XPS study of the contact/semiconductor interfaces

        In this work, the specific contact resistance (ρc) between amorphous indium-gallium-zinc-oxide (IGZO) semiconductor and different contact electrodes was obtained from thin film transistors (TFTs). Ti/Au (10/100 nm), aluminum doped zinc oxide (AZO, 100 nm) and indium tin oxide (ITO, 100 nm) were used as source/drain electrodes to fabricate IGZO TFTs. Chemical states of the contacts/semiconductor interfaces were examined by depth profile X-ray photoelectron spectroscopy (XPS) analysis to explain the origin of the differences on specific contact resistance. The lowest ρc achieved using Ti/Au was related to the formation of a TiOx interlayer due to oxygen atoms diffusing out from the semiconductor under layer, increasing the carrier concentration of IGZO at the interface and lowering the ρc. On the contrary, no interfacial reactions were observed between IGZO and AZO or ITO source/drain. However, IGZO resistivity increased with ITO contacts likely due to oxygen vacancies filling during ITO deposition. This fact seems to be the origin of the high contact resistance between IGZO and ITO, compared to IGZO-AZO and IGZO-Ti/Au interfaces.

        Influence on SERS enhancement factor of the components of an artificial opal loaded with metal NPs: A systematic study

        We have inquired about the influence of composite artificial opal components on its SERS enhancement factor (EF). Particularly, we considered metal (100 nm) and dielectric spheres (290 nm), and an excitation wavelength of 632 nm. We show that the electric field of a SiO2 sphere is weaker and more uniform as its porosity increases. Additionally, a porous sphere promotes a lower EF compared to that of a non-porous sphere. The optical response of the composite opal is insensitive to the polarization state of the incident field. A SERS EF of 104 is reachable with an opal loaded with Au or Ag NPs. In general, the dielectric spheres affect the SERS EF intensity of the metal NPs. From the optical spectra, we observed that with a 632 nm wavelength, the composite opal is out of resonance.

        Comparative Study of Oswald Ripening and Trans-Interface Diffusion-Controlled Theory Models: Coarsening of γ ' Precipitates Affected by Elastic Strain Along a Concentration Gradient

        According to Lifshitz, Slyozov, and Wagner (LSW) and Trans-Interface Diffusion-Controlled (TICD) theoreticalmodels, this paper reports the microstructure and its coarsening behavior of γ' metastable-coherentprecipitates in concentration gradient of Ni-13.75Ti (at%)/Ni generated by diffusion couple. The coarseningof precipitates was evaluated in two different Ti contents (R1-11.4Ti (at%) and R2-13Ti (at%)) generated alongthe concentration gradient and includes average size, size distributions and growth rate. The solvus andmetastable-coherent bimodal lines as determined at 850 °C of 9.16 (at%) and 9.92Ti (at%) respectively byscanning electron microscopy. This paper suggests that elastic strains produced by the matrix/precipitate latticemismatch caused significant deviations between the experimental results and those predicted by the LSW or TIDCtheories. Activation energies for TIDC (Qi) and LSW (Qr) are Qr: 219.69 and 172.61 kJ mol-1for R1 and R2regions, respectively, and Qi: 218.46 and 164.56 kJmol-1for R1 and R2 regions, respectively. A concentration gradientallows the study of various alloys with different concentration and volume-fraction in a single sample.

        In Vitro and In Vivo Nematicide Effect of Extract Fractions of Pleurotus djamor Against Haemonchus contortus

        Sheep haemonchosis is a disease that causes anemia, diarrhea, weight loss, low production, and even death of young animals. This represents a negative economic impact on the livestock sector. For decades, chemicals have been used to control this parasitic disease; however, cases of anthelmintic resistance have increased around the world. For this reason, the search for control alternatives is necessary. Several studies have shown that edible mushrooms of Pleurotus genus have different medicinal properties, including nematicidal activity. In this study, the chromatographic fractionation of the hydroalcoholic extract of fruiting bodies Pleurotus djamor was carried out to identify and isolate the metabolites responsible for nematicidal activity. From the fractionation of the extract, the fraction PdR2 and the subfraction PdB were obtained, which were evaluated against Haemonchus contortus under in vitro conditions. The nematicidal effect of the fraction PdB in gerbils infected artificially with H. contortus was evaluated. Finally, the analysis of the PdB fraction by gas chromatography-mass spectrometry and nuclear magnetic resonance was carried out. The results showed that the PdB had 100% of egg hatching inhibition from 5 mg/mL. Regarding larvicidal activity, it presented >97.2% from 24 h to 20 mg/mL. The in vivo evaluation of the PdB fraction showed a reduction of H. contortus larvae of 92.56%. The compounds present in this fraction were the mixture of allitol and an unidentified terpene in a ratio of 9:1. The PdB fraction is a potential alternative for the control of H. contortus, where allitol and a terpene could be responsible for nematicidal activity.

        Detecting Malware in Cyberphysical Systems Using Machine Learning: a Survey

        Among the scientific literature, it has not been possible to find a consensus on the definition of the limits or properties that allow differentiating or grouping the cyber-physical systems (CPS) and the Internet of Things (IoT). Despite this controversy the papers reviewed agree that both have become crucial elements not only for industry but also for society in general. The impact of a malware attack affecting one of these systems may suppose a risk for the industrial processes involved and perhaps also for society in general if the system affected is a critical infrastructure. This article reviews the state of the art of the application of machine learning in the automation of malware detection in cyberphysical systems, evaluating the most representative articles in this field and summarizing the results obtained, the most common malware attacks in this type of systems, the most promising algorithms for malware detection in cyberphysical systems and the future lines of research in this field with the greatest potential for the coming years.

        Analysis of virulence traits of Staphylococcus aureus isolated from bovine mastitis in semi-intensive and family dairy farms

        Background: Staphylococcus aureus is one of the main microorganisms that causes bovine mastitis, and its well-known virulence characteristics and interactions with the environment are used to aid the design of more efficient therapies. Objectives: To determine whether the virulence traits, such as antibiotic resistance and biofilm-forming and internalization abilities, of S. aureus isolated from bovine mastitis are related to dairy production system types. Methods: The study was performed in the Mexican states of Guanajuato and Michoacan. Semi-intensive dairy farms (SIDFs) and family dairy farms (FDFs) (454 and 363 cows, respectively) were included. The 194 milk samples from mastitis affected quarters were collected and 92 strains of S. aureus were isolated and identified by biochemical and molecular tests. Antibiotic resistance, biofilm and internalization assays were performed on 30 randomly selected isolated strains to determine virulence traits, and these strains were equally allocated to the 2 dairy production systems. Results: All 30 selected strains displayed a high degree of resistance (50%–91.7%) to the antibiotics tested, but no significant difference was found between SIDF and FDF isolates. S. aureus strains from SIDFs had an average biofilm forming capacity of up to 36% (18.9%–53.1%), while S. aureus strains from FDFs registered an average of up to 53% (31.5%–77.8%) (p > 0.05). Internalization assays revealed a higher frequency of internalization capacity for strains isolated from FDFs (33.3%) than for those isolated from SIDFs (6.7%) (p > 0.05). fnbpA gen was detected in 46.6% of FDF strains and 33.3% of SIDF strains, and this difference was significant (p < 0.05). Conclusions: Our findings show that the virulence traits of S. aureus isolates analyzed in this study, depend significantly on several factors, such as phenotype, genotype, and environmental conditions, which are significantly related to dairy production system type and daily management practices.

        <B>Abstract</B><P>The response of <I>Grapholita molesta</I> (Busck) males to three-component sex pheromone blends containing a 100% ratio of the major sex pheromone component, (<I>Z</I>)-8-dodecenyl acetate and a 10% ratio of (<I>Z</I>)-8-dodecenol, but with varying ratios of (<I>E</I>)-8-dodecenyl acetate (0.4, 5.4, 10.4, 30.4, and 100.1% <I>E</I>-blends) was tested with populations in eight stone and pome fruit orchards in Europe, Asia, and North and South America. Traps baited with the 5.4% <I>E</I>-blend caught significantly more males than traps with any other blend with all populations. Significantly more males were caught in traps baited with the 10.4% <I>E</I>-blend than in traps with the remaining blends, except with the 0.4% <I>E-</I>blend in Turkey. Significant differences in male moth catches occurred between the other blends with the 0.4>30.4% <I>E-</I>blend, and the 30.4>100.1% <I>E-</I>blend. Male moth catches with the 100.1% <I>E</I>-blend only differed from the hexane control in Chile. No apparent differences were noted to these blends in populations collected from pome or stone fruits. Flight tunnel assays to synthetic blends with a subset of populations were similar to the field results, but the breadth of the most attractive <I>E-</I>blends was wider. Flight tunnel assays also demonstrated a high level of male-female cross-attraction among field-collected populations. Female gland extracts from field-collected populations did not show any significant variation in their three-component blends. The only exceptions in these assays were that long-term laboratory populations were less responsive and attractive, and produced different blend ratios of the two minor components than recently collected field populations.</P>
