        Preparation of High-Purity Urokinase Using Single-Step Hydrophobic Interaction Chromatography with p-Aminobenzamidine Ligand

        A novel process for urokinase purification was studied using p-aminobenzamidine as the ligand and sepharose 4B as the matrix. The adsorption, washing, and elution conditions were optimized by an unusual method. An adsorption buffer containing 2.5 M NaCl and 1% Tween 80 facilitated the adsorption of urokinase on the affinity media and prevented contaminants from binding to the p-aminobenzamidine affinity gel. It was found that 5% Tween 80 removed most of the contaminants from the affinity column. A 0.2 M glycine elution buffer containing 0.5 M NaCl (pH 3.0) was found to have a strong elution ability with a high recovery and purity of urokinase. A crude urokinase material of 231 IU/mg protein from human urine was purified to 124,300 IU/mg protein with a purification factor of 538 and yield of 86.7%. As a result, a high purity urokinase was obtained with only a single affinity chromatography step. The purification process was successfully scaled-up to a 2-1 chromatography column. The resulting urokinase eluate could be directly lyophilized, thereby complying with Chinese pharmacopoeia (1995 version) standards.

      • 패시브 컨버터를 이용한 단상 SRM의 운전 특성 해석

        본 논문에서는 기존의 비대칭 컨버터와 비교 했을 때, 턴-오프 구간에서 높은 음전압이 인가되어 감자구간을 감소시키는 동시에 이에 따른 부토크의 발생을 억제하며, 정토크의 활용영역을 확대할 수 있는 패시브 컨버터회로를 제안한다. 제안된 패시브 컨버터는 세 개의 다이오드와 추가된 부스터 커패시터를 기존의 비대칭 컨버터에 추가 하는 구조로, 컨버터의 동작 모드에 따라 기존의 직류전압을 평활하기 위한 커패시터와 추가된 커패시터는 직렬 또는 병렬로 연결된다. 여자 모드에서는 병렬로 연결된 커패시터 회로에서 안정적인 직류 링크전압의 공급이 가능해지고, 감자모드에서는 두 개의 커패시터가 직렬로 연결되어 직류링크 전압의 2배에 해당하는 높은 감자전압이 상권선에 인가되어 감자구간을 감소시킬 수 있는 장점이 있다. 이 효과로 감자모드의 턴-오프시점을 기존의 비대칭 컨버터에 비하여 연장할 수 있어, 여자 구간을 좀 더 확장 시킬 수 있는 장점을 가진다. 그리고 패시브 컨버터의 높은 감자전압은 추가적인 제어기의 제어가 필요 없고, 비대칭 컨버터의 스위치를 모두 턴-오프 시키는 동작에 의하여 자동으로 발생하므로 제어가 매우 용이한 장점이 있다. 제안된 패시브 컨버터는 6/6 단상 SRM에 적용하여 컴퓨터 시뮬레이션을 통해 기존의 비대칭 컨버터와 비교하여 그 성능을 검증 하였다.

        Prognostic Accuracy of the SIRS, qSOFA, and NEWS for Early Detection of Clinical Deterioration in SARS-CoV-2 Infected Patients

        Background: The case fatality rate of coronavirus disease 2019 (COVID-19) is estimated to be between 4.3% and 11.0%. Currently there is no effective antiviral treatment for COVID-19. Thus, early recognition of patients at high risk is important. Methods: We performed a retrospective observational study of 110 patients with severe acute respiratory syndrome coronavirus 2 infection. We compared the effectiveness of three scoring systems: the Systemic Inflammatory Response Syndrome (SIRS), quick Sequential Organ Failure Assessment (qSOFA), and National Early Warning Score (NEWS) systems, for predicting the prognosis of COVID-19. The area under the receiver operating characteristic curve (AUROC) was used for these assessments, and Kaplan-Meier survival curves were used to identify the cumulative risk for 28-day mortality according to the NEWS stratification. Results: For predicting 28-day mortality, NEWS was superior to qSOFA (AUROC, 0.867 vs. 0.779, P < 0.001), while there was no significant difference between NEWS and SIRS (AUROC, 0.867 vs. 0.639, P = 0.100). For predicting critical outcomes, NEWS was superior to both SIRS (AUROC, 0.918 vs. 0.744, P = 0.032) and qSOFA (AUROC, 0.918 vs. 0.760, P = 0.012). Survival time was significantly shorter for patients with NEWS ≥ 7 than for patients with NEWS < 7. Conclusion: Calculation of the NEWS at the time of hospital admission can predict critical outcomes in patients with COVID-19. Early intervention for high-risk patients can thereby improve clinical outcomes in COVID-19 patients.

      • Poster Session : PS 1469 ; Infectious Diseases : A Fatal Case of Primary Multiseptated Psoas Abscess in Diabetic Patient

        Primary psoas abscess occurs as a result of hematogenous or lymphatic spread from distant site. It is very rare kind of retroperitoneal abscess, but the motality rate is 2.4 percent. Risk factors include diabetes mellitus, intravenous drug use, human immunode ficiency virus infection, renal failure, and other forms of immunosuppression. Here we present a fatal case of primary multiseptated psoas abscess in patient with type 2 diabetes. A 27-year old man with poorly controlled type 2 diabetes presented to our hospital with a 3-week history of progressive lower abdominal pain after falling down on the bed. Initial vital sign was stable. Laboratory studies showed a white cell count 17030/uL, hemoglobin 10.7g/dL, platelet 409000/uL, C-reactive protein 22.599mg/dL, erythrocyte sedimentation rate 53mm/H, procalcitonin 4.64ng/mL, glucose 701mg/ dL, HbA1c 12.2%, and normal renal function. Arterial blood gas analysis showed PH 7.346, Pco2 33.6mmHg, Po2 123mHg, HCO3 17.9mmol/L. A CT scan of the abdomen showed a 12cm-sized multiseptated abscess in the left psoas muscle. (figure. 1) He was promptly treated with percutaneous drainage and empirical antibiotics (ceftriaxone and metronidazole). Afterward methicillin-sensitive Staphylococcus aureus was cultured in drained pus, so we changed antibiotics to combination of nafcillin and gentamicin. Follow-up CT showed improvement of the abscess size (figure. 2b). But a few days later, he advanced to septic shock and acute respiratory distress syndrome. (figure. 2a) So we started the new antibiotics (vancomycin and carbapenem) and applied a mechanical ventilator during extracorporeal membrane oxyenation in intensive care unit. Nevertheless, he expired on hospital day 15. Percutaneous catheter drainage is much less invasive and has been proposed as the draining method of choice. So, most psoas abscess rarely requires surgical intervention. But early surgical drainage may be necessary, considering the patient`s risk factor and the characteristics of abscess.

        중국 벤처기업의 성과요인 - 한국 벤처기업과의 비교연구

          본 연구는 중국 하얼빈시 개발구의 벤처기업들을 대상으로 경영특성과 기업성과에 영향을 미치는 요인들을 조사 하였다. 그리고 이를 한국에서 연구된 벤처기업의 성과요인(창업자, 환경, 전략, 자원능력, 조직특성)과 비교함으로써 성과에 영향을 미치는 요인들의 차이점을 확인하고 그 원인의 규명을 시도하였다. 그 결과, 중국 벤처기업의 경우 환경의 불확실성과 창업자의 위험감수 성향이 기업성과에 부(-)의 영향을 미쳤으며 기술혁신 차별화전략은 한국 벤처기업과 달리 부(-)의 영향을 미쳤다. 반면에 높은 기술능력과 구성원간 친밀감을 바탕으로 원가우위전략을 구사할수록 높은 성과를 창출하는 것으로 나타났다. 또한, 경영특성에 있어서 중국 벤처기업은 상대적으로 국내 벤처기업에 비해 시장세분화 수준은 비교적 높은 수준이나 창업자의 성취욕구가 낮고 원가우위, 기술혁신 등의 전략실행에 있어 한국 벤처기업에 비해 부족한 것으로 나타났다.   This study attempts to analyze the factors affecting the performance of Chinese high-tech ventures. Based on the existing studies in Korea, characteristics of entrepreneur, environmental characteristics, resource capabilities, organizational characteristics, and competitive strategies were considered as the factors affecting the performance of high-tech ventures. We surveyed 93 companies which were located in northern China. To ensure the measurement equivalence when the questionnaire was converted from Korean to Chinese, the questionnaire was pretested in China and was back-translated. Most of the Chinese venture companies were founded by highly educated entrepreneurs. We found significant differences between Chinese and Korean ventures in terms of success factors and managerial characteristics.

        Early Hydroxychloroquine Administration for Rapid Severe Acute Respiratory Syndrome Coronavirus 2 Eradication

        There are no proven therapeutics for Coronavirus disease 2019 (COVID-19) pneumonia outbreak. We observed and analyzed the clinical efficacy of the most used hydroxychloroquine (HCQ) for 30 days. In this study, administration of HCQ <5 days from diagnosis (odds ratio: 0.111, 95% confidence interval: 0.034 - 0.367, P = 0.001) was the only protective factor for prolonging of viral shedding in COVID-19 patients. Early administration of HCQ significantly ameliorates inflammatory cytokine secretion by eradicating COVID-19, at discharge. Our findings suggest that patients confirmed of COVID-19 infection should be administrated HCQ as soon as possible.

      • Differential Roles of Glycogen Synthase Kinase 3 Subtypes Alpha and Beta in Cortical Development

        <P>Glycogen synthase kinases 3 (GSK3) α and β are expressed in the nervous system, and disruption of GSK3 signaling has been implicated in a wide range of neurodevelopmental and psychiatric disorders. Although several studies have established a role of GSK3 signaling in the nervous system, much less is known about isoform-specific functions. Here, we have examined the role of GSK3α and GSK3β in the developing neocortex by performing <I>in utero</I> electroporation with specific small interfering RNAs targeting each isoform. We found that depletion of either GSK3α or GSK3β commonly promoted the proliferation of neural progenitor cells in the ventricular zone, but at later stages, knocking down of each isoform resulted in distinct outcomes. In particular, the transformation of radial progenitors to intermediate progenitor cells was promoted in GSK3α-depleted cells, but markedly prevented in GSK3β-depleted cells. Moreover, knocking down of GSK3β but not GSK3α prevented the generation of upper-layer Cux1<SUP>+</SUP> neurons. Consistent with the distinct outcomes, protein levels of c-Myc and β-catenin, well-known substrates of GSK3, were differentially affected by depletion of GSK3α and GSK3β. Together, these results suggest that GSK3α and GSK3β might play distinct roles in the genesis and differentiation of neuronal lineage cells during neocortex development by differential regulation of downstream signaling pathways.</P>

        Yeungnam University type drive-through (YU-Thru) coronavirus disease 2019 (COVID-19) screening system: a rapid and safe screening system

        Active and prompt scale-up screening tests are essential to efficiently control the coronavirus disease 2019 (COVID-19) outbreak. The goal of this work was to identify shortcomings in the conventional screening system (CSS) implemented in the beginning of the outbreak. To overcome these shortcomings, we then introduced a novel, independently developed system called the Yeungnam University type drive-through (YU-Thru), and distributed it nationwide in Korea. This system is similar to the drive-throughs utilized by fast food restaurants. YU-Thru system has shortened the time taken to test a single person to 2–4 minutes, by completely eliminating the time required to clean and ventilate the specimen collection room. This time requirement was a major drawback of the CSS. YU-Thru system also reduced the risk of subjects and medical staff infecting one another by using a separate and closed examination system. On average, 50 to 60 tests were conducted per day when using the CSS, while now up to 350 tests per day are conducted with the YU-Thru system. We believe that the YU-Thru system has made an important contribution to the rapid detection of COVID-19 in Daegu, South Korea. Here, we will describe the YU-Thru system in detail so that other countries experiencing COVID-19 outbreaks can take advantage of this system.
