TC-1 세포주를 이용한 동물모델에서 As4O6에 의한 혈관폐쇄 효과

목적: As2O3는 다양한 고형암에서 혈관손상을 통하여 항종양 효과를 유도하는 것으로 알려져 있다. 본 연구에서는 자궁경부암동물 모델에서 새로운 비소 화합물인 As4O6와 As2O3를 비교하여 종양 내 혈관폐쇄 현상을 확인하고자 하였다. 연구 방법: HPV 16-E6/E7 유전자를 가지고 있는 TC-1 종양세포를 이용한 자궁경부암 동물모델에서 종양의 부피가 200~250 ㎣에 이르면 PBS, As2O3 and As4O6를 복강 내 투여하였다. 비소 화합물을 주사한 후 종양의 크기는 2~3일 간격으로 측정하고, 종양내혈관 폐쇄현상은 Evans blue 추출법과 Hoechst 33342 염색법 등 2가지 방법을 이용하여 측정하였다. 또한 헤마토실린-에오신 염색을 통하여 조직학적 변화를 관찰하였다. 결과: As2O3와 As4O6 주사 후, 종양 성장은 대조군에 비해 효과적으로 지연되었으며, As4O6 처리군이 가장 우수한 항종양 효과를 나타내었다. 또한 비소화합물의 투여는 종양내의 거대한 세포괴사와 혈관폐쇄 현상을 유도하였다. 결론: TC-1 세포주를 이식한 고형암 동물모델에서 As4O6의 처치는 혈관폐쇄 효과에 의한 항종양 효과를 나타내었다. Objective: Arsenic trioxide (As2O3) is known to have potent anti-vascular activity and significantly suppress solid tumor growth. The present study was conducted to investigate the vascular shutdown effects of a novel arsenic compound, tetraasrsenic oxide (As4O6), in comparison with As2O3 using cervical cancer animal model. Methods: Mice tumor challenge model was used C57BL/6 mice transplanted with TC-1 cells. After the growth of tumors was reached up 200~250 ㎣, mice were divided into 3 groups randomly for control and treatment of either As2O3 or As4O6. As2O3 and As4O6 was treated by i.p. injection. The tumor size was caliperated in twice for weeks and anti-vascular effect were assessed by Evans blue extraction assay and Hoechst 33342 staining. In tumor tissue, histopathological feaure was obserevd by hematoxylin and eosin (H&E) staining. Results: In mice treated with either As2O3 and As4O6 (i.p.), both of As2O3 and As4O6 was significantly suppressed the tumor growth compared with control group. Moreover, effect of As4O6 is more pronounced. These tumor growth inhibition is led to the massive necrosis and vacular shutdown in tumor tissue. Conclusion: This study suggests that As4O6 may have potential anticancer activity via vascular shutdown in C57BL/6 mice transplanted with TC-1 cells.

As the use of preservatives in foods has been increased, the misuse and overuse of preservatives are occasionally occurred. As the variety of food has risen, it is necessary to establish the screening method to determine preservatives in various foods. In this study, benzoic acid, sorbic acid, methyl p-hydroxybenzoate, ethyl p-hydroxybenzoate, i-propyl p-hydroxybenzoate, n-butyl p-hydroxybenzoate were analyzed by TLC and HPLC. In order to determine the recoveries from food, these preservatives were spiked into foods. As a result, solvent systems of 8:2:0.45:0.05 and 12:2:0.45:0.05 of pentane:acetic acid:chloroform:ethanol were appropriate for TLC analysis of preservatives. All preservatives were well resolved with solvent system of 2% ammonium acetate in 2.5% acetic acid:methanol (90/10~10/90) by HPLC. Direct extraction of preservatives from food proved to be more proper than distillation and extraction method. The recoveries by direct extraction method showed 93.7~104.6% of preservatives studied.

The clear cell adenocarcinoma of uterine cervix is very rare tumor, and only 4-9% of entire adenocarcinoma appears to be diagnosed as clear cell adenocarcinoma. Its risk factor and pathogenesis are not exactly known, but intrauterine exposure to DES (diethyl stilbestrol) and associated non-steroidal estrogen during pregnancy before 18weeks is one known risk factor, and also hormonal change or genetic cause are suggested as the risk factors. The peak age of its occurrence has bimodal pattern, which are groups before 24 years-old and after 45 years-old, and clear cell adenocarcinoma arising in latter group is not associated with intrauterine DES exposure. It is also reported that 25% of young women who has clear cell adenocarcinoma had no history of hormonal exposure. The treatment and prognosis is similar to other kinds of uterine cervical adenocarcinoma. With a short literature review, we are reporting a case of 6-year-old girl who visited our clinic because of vaginal mass naturally fallen off with a minor bleeding which was finally diagnosed as clear cell adenocarcinoma, and had no maternal history of DES exposure during pregnancy.

게르마늄(Ge)은 거의 대부분의 영양분, 동물, 식물에서 발견되는 미량원소로 많은 연구를 통해 항균성 항바이러스성 항종양성 항돌연변이성 및 면역조절 효과를 지닌 것으로 알려져 있다. 또한, 동충하초는 중국의 전통적인 약용 버섯으로 항종양성, 항산화성 및 면역조절 효과 등 다양한 생물학적 활성을 지니고 있으며 미량원소를 축적시키기 효과적인 것으로 알려져 있다. 이런 특성을 이용하여 본 연구는 게르마늄 농도를 증가시킨 동충하초(CMGe)의 열수 추출액을 얻어내고 그 생물학적 특성을 실험하였다. 네 가지 암세포주 Ramos, A549, HepG2, CEM와 MSC에 대한 CMGe의 세포독성을 MTT assay를 통해 확인한결과, CEM에서 항증식성이 가장 높고 MSC에 대한 세포독성은 낮은 것으로 확인되었다. 마지막으로 누드마우스에 인간 신경아교세포종(U87MG)을 이종이식하여 종양의 부피와 무게를 조사한 결과, CMGe열수 추출액 처리군에서 종양 부피와 무게의 감소를 확인할 수 있었다. 이런 결과는 CMGe가 앞으로 의약 및 제약산업에 다양하게 적용될 수 있을 것으로 기대된다. Cordyceps militaris has been used in traditional Chinese medicine owing to its anticancer and immunomodulatory activities. Germanium compounds have also been shown to be associated with many pharmacological functions, such as antimicrobial, antiviral, antitumor, antimutagenic, and immunomodulating effects. In this study, we examined the biological properties of hot water extract from mycelial liquid culture of germanium-enriched C. militaris (CMGe). CMGe displayed a concentration-dependent antiproliferation activity against four human cancer cell lines. The antiproliferative activity of CMGe was 2-4-fold lower than that of hot water extract from mycelial liquid culture in C. militaris (CM). However, CM had a concentration-dependent cytotoxicity to human bone marrow-derived mesenchymal stem cells (MSCs). Contrastingly, CMGe did not cause any cellular damage to MSCs. MSCs cultured with CMGe displayed an increased proliferative activity with no cytotoxic effect. The oral administration of CMGe inhibited increased tumor volume and weight compared with the control group. CMGe has the potential to be used as an industrial product in medicinal foods as well as in pharmaceutical products.

최근 민간의 교통SOC투자가 활발히 이루어지는 가운데 장래 교통수요예측의 불확실성으로 인한 논란이 큰 이슈로 부각되고 있 다. 그러나 교통SOC투자를 위해 시행되는 투자평가제도에서는 장래 사회·경제적 여건변화에 대한 체계적인 대응이 미흡한 실정이 다. 본 연구는 이러한 문제를 인식하여 타당성 평가에 위험관리를 도입함으로써 사업 완료까지의 지속적인 사업관리방안을 제시하 였다. 국내·외 교통SOC사업에서의 위험관리방안을 조사하여 시사점 및 개선방안을 제시하고, 일반적으로 널리 알려진 위험관리 수 행과정을 교통SOC사업에 적용하고, 사업단계별 대응형식에 따른 대응방안을 마련함으로써 사업이 진행되는 과정에서 발생하는 사회·경제적 변화에 능동적으로 대응할 수 있는 기틀을 마련하였다.

<P><B>Abstract</B></P> <P><B>Background and purpose</B></P> <P>Glycosylation of phenolic compounds has been reported to increase water-solubility, reduce toxicity, and sometimes give improved or novel pharmacological activities. Present study was aimed to evaluate and compare the beneficial effects of quercetin aglycone (Quer) and its glycosylated derivative, quercetin 3-<I>O</I>-xyloside (Quer-Xyl), against acute pancreatitis (AP).</P> <P><B>Methods</B></P> <P>The cellular acute pancreatitis model was established by treating the rat pancreatic acinar cells (AR42J) with lipopolysaccharide (10 µg/ml) and cerulein (10<SUP>−7</SUP> M). The cytotoxicity of Quer or Quer-Xyl on AR42J cells was assessed by MTT assay. Calcium and ROS levels were fluorometrically determined. The ER stress levels (PERK, GRP78), expression levels of amylase and lipase, and apoptotic markers (caspase-3 and -9) were measured by RT-PCR, western blotting, or fluorometric assay.</P> <P><B>Results</B></P> <P>While Quer increased the mRNA expressions of AP marker enzymes, amylase and lipase, Quer-Xyl dose-dependently reversed their expressions. Quer-Xyl suppressed intracellular ROS production and both mRNA and protein levels of GRP78 and PERK, which were significantly elevated in cerulein and LPS-treated AR42J cells. Further, RT-PCR and fluorescence assay revealed that Quer-Xyl dose-dependently augmented the mRNA expressions and activities of caspase-3 and -9.</P> <P><B>Conclusion</B></P> <P>These results showed that Quer-Xyl, but not Quer, has a significant anti-pancreatitis activity through attenuating intracellular ROS production and ER stress response and enhancing apoptotic cell death, suggesting that it might be useful as a potent functional ingredient in health-beneficial foods or as a therapeutic agent to prevent or treat AP.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
