        Preparation of Thermosensitive Hydrogels by means of Tandem Physical and Chemical Crosslinking

        The tandem physical and chemical crosslinking was used to prepare injectable hydrogels. First, the terpolymers with diene groups were synthesized by the free radical copolymerization of N-isopropylacrylamide (NIPA), N,N-dimethylacrylamide (DMA) and furfuryl methacrylate (FM). Second, dienophile-functionalized terpolymers were obtained from the terpolymers of NIPA, DMA, and 2-hydroxyethyl methacrylate (HEMA) and Nmaleoyl alanine (AMI) by a coupling reaction under N,N'-dicyclohexylcarbodiimide (DCC). When the screened diene-functionalized terpolymer and dienophile-functionalized terpolymer were dissolved separately in water and mixed, physically cross-linked hydrogels formed rapidly within 10 s at 37 ℃. Subsequently, the physically crosslinked hydrogels could change gradually into chemically cross-linked hydrogels by a Diels-Alder reaction and the mechanical strength of the hydrogels could be improved. The swelling behavior study suggests that the hydrogels possess thermosensitivity and good reproducibility at different temperatures. The strategy described, i.e. incorporating the advantages of physically crosslinked hydrogels and chemically crosslinked hydrogels, has potential applications in the preparation of injectable hydrogels.

        Loss of MicroRNA-137 Impairs the Homeostasis of Potassium in Neurons via KCC2

        Neuropsychiatric disorders are the leading cause of mental and intellectual disabilities worldwide. Current therapies against neuropsychiatric disorders are very limited, and very little is known about the onset and development of these diseases, and their most effective treatments. MIR137 has been previously identified as a risk gene for the etiology of schizophrenia, bipolar disorder, and autism spectrum disorder. Here we generated a forebrain-specific MIR137 knockout mouse model, and provided evidence that loss of miR-137 resulted in impaired homeostasis of potassium in mouse hippocampal neurons. KCC2, a potassium-chloride co-transporter, was a direct downstream target of miR-137. The KCC2 specific antagonist VU0240551 could balance the current of potassium in miR-137 knockout neurons, and knockdown of KCC2 could ameliorate anxiety-like behavior in MIR137 cKO mice. These data suggest that KCC2 antagonists or knockdown might be beneficial to neuropsychiatric disorders due to the deficiency of miR-137.

        中國老齡化問題應對策略研究:集體社會資本視角下的老年人力資源開發

        인구의 고령화는 경제발전과 기술진보의 산물이자 삶의 질을 추구하는 인류의 필연적인 결과이다. 이 세계적인 난제를 해결하기 위해서는 인류 공통의 지혜가 필요하다. 중국은 2000년에 인구 고령화 국가 대열에 합류하여 현재는 거의 중등 고령화 수준에 도달했고, “부유해 지기 전의 고령화”, 급속한 고령화, 대규모 고령화의 특징을 보이고 있다. 인구 고령화는 미래 세계에서 영구적인 모습이 될 가능성이 높기 때문에 경제사회의 발전을 위해서는 노인인적자원 활용과 노인 수요 충족에 중점을 두어야 한다. 따라서 노인인적자원개발 사업이 중국의 현 인구 고령화 문제를 해결하는데 기여할 수 있을 것이라 판단된다. 본 연구는 Coleman의 집단의 사회적 자본 개념과 거시적 차원의 사회적 자본 이론을 기반으로 하였으며, 노인인적자원개발의 다자이익 집단은 사회개발, 조직개발, 자기개발의 세 가지 유형으로 구분하였다. 또한 중국의 고령화 현황과 노인인적자원개발의 전제 조건, 미래에 직면할 도전에 대하여 체계적으로 분석하고, 이를 바탕으로 최종적으로 중국의 실정에 맞는 인구 고령화 문제에 대한 해결책을 알아보았다. 본 연구의 목적은 다자이익집단이 함께 참여하여 사회적 자본을 모으고, 시너지 효과를 형성하여 인구 고령화 문제가 가져올 부정적인 결과를 완화하는데 있다. Population aging is the product of economic development and technological progress, as well as the inevitable result of human"s pursuit of high quality of life,which requires the common wisdom of human beings to solve this worldwide problem. China entered the ranks of population aging countries in 2000, and has now reached the aged society, and is characterized by “getting old before getting rich” problem, rapid population aging, and a large-scale elderly population. Since population aging has become an irreversible trend and is likely to become a permanent norm in the future all over the world, economic and social development needs to be closely promoted around the utilization of the elderly human resources and meeting their needs. Therefore, this paper advocates the development of the elderly human resources business as a means to solve the current problem of population aging in China. Based on Coleman"s concept of collective social capital and macro-level social capital theory, this paper classifies the multi-stakeholders in developing elderly human resources into three types: social development, organizational development and self-development. Through this, the paper makes a systematic analysis on the current situation of population aging in China, as well as the preconditions and future challenges of developing elderly human resources. Finally, the paper has constructed a set of solutions to the population aging problem in line with China"s national conditions. The purpose of this paper is to realize the use of multistakeholder groups to participate together, gather social capital and form a synergy to alleviate the adverse consequences of population aging. Findings are as follows: (1) in terms of social development, China should make full preparations under the national strategic framework by taking advantage of the “14th five-year plan” time window period, improve the policy system of elderly human resources development, improve the differences in the degree of aging in different regions according to local conditions, innovate ideas, and encourage high-quality elderly people to return to the labor market. (2) In terms of organizational development, it is necessary to improve the specialization and professionalism of the intermediary services of the elderly labor, break the organizational boundaries of communities. The paper suggests that educational organizations establish lifelong education and training systems, and that enterprises implement flexible management of the elderly human resources. (3) In terms of self-development, both the awareness of selfdevelopment and behavioral change of the elderly should be promoted. We should not only improve the awareness of re-employment and self-learning, but also making full use of the leading role of high-quality elderly human resources. So as to improve more elderly people to integrate into the elderly life with higher social value.

        Comparison of the efficacy and safety of transurethral laser versus open prostatectomy for patients with large-sized benign prostatic hyperplasia: A meta-analysis of comparative trials

        Purpose: The selection of open prostatectomy (OP) over transurethral laser surgery is controversial for large volume prostates. Thus, we aim to compare the efficacy and safety of transurethral laser versus OP, and provide the latest evidence of clinical practice for large-sized benign prostatic hyperplasia (BPH). Materials and Methods: This meta-analysis used Review Manager V5.3 software and the systematic literature search of Cochrane Library, Embase, PubMed, and Web of Science datasets was performed for citations published from 2000 to 2020 that compared transurethral laser with OP for the treatment of large BPH. Variables of interest assessing the two techniques included clinical characteristics, and the perioperation-, effectiveness-, and complication-related outcomes. Results: The meta-analysis included twelve studies containing 1,514 patients, with 792 laser and 722 OP. The transurethral laser group was associated with shorter hospital stay and catheterization duration, and less hemoglobin decreased in the perioperative variables. There was no significant difference in the international prostate symptom score, post-void residual urine volume, maximum flow rate, and quality of life score. Transurethral laser group had a significantly lower incidence of blood transfusion than OP group (odds ratio, 0.10; 95% confidence interval, 0.05 to 0.19; p<0.001; I2=8%), and no statistical differences were found with respect to the other complications. Conclusions: Both OP and transurethral laser prostatectomy are effective and safe treatments for large prostate adenomas. With these advantages of less blood loss and transfusion, and shorter catheterization time and hospital stay, laser may be a better choice for large BPH.

      • Efficacy of Multiple Low-dose Photodynamic TMPYP4 Therapy on Cervical Cancer Tumour Growth in Nude Mice

        Objective: Photodynamic therapy (PDT) is an emerging therapeutic procedure suitable for the treatment of cervical cancer. However, the side effects of PDT are severe, including skin ulceration, so we designed an experiment to examine the effects of multiple low-dose photodynamic therapy of 5, 10, 15, 20-tetrakis(1-methylpyridinium-4-yl) porphyrin (Tmpyp4) on tumour growth by utilizing a model in nude mice implanted with Hela cervical cancer cells. Materials and Methods: Female BALB/c nude mice (aged 5-6 weeks, weighing 18-20 g) were used. Hela cervical cancer cells were injected subcutaneously ($1{\times}10^7cells/200{\mu}L$). Ten days after injection, the mice were divided into three groups (n=6), the A group of controls without any treatment, the B group receiving a single-treatment with Tmpyp4 (10 mg/kg, intratumor injection) and irradiation (blue laser, $108J/cm^2$), and the C group given three-treatments with Tmpyp4 (10 mg/kg, intratumor injection) and irradiation at intervals of two days. After starting treatment, tumours were measured every two days, to assess growth. At 2 weeks after the last treatment of C group, tumour tissue and organs were collected from each mouse to evaluate tumor histology and organ damage. Results: Tumour growth in C group was significantly inhibited compared with A and B groups (P<0.05), without any injury to the skin and internal organs. Conclusion: Our novel findings demonstrated that multiple low-dose photodynamic therapy of Tmpyp4 could inhibit cervical cancer growth significantly with no apparent side effects.

        Two new ursane-type triterpenoid saponins from Elsholtzia bodinieri

        Two new ursane-type triterpenoid saponins,bodiniosides M (1) and N (2), along with three knownsaponins, oblonganosides I (3), pseudobuxussaponin B (4)and bodinioside A (5), were isolated from the aerial partsof Elsholtzia bodinieri. The structures of compounds 1 and2 were characterized by spectroscopic data as well as acidhydrolysis and GC analysis as 3-O-b-D-xylopyranosyl-19ahydroxy-23-acetoxy-urs-12(13)-en-28-oic acid 28-O-a-Lrhamnopyranosyl-(1-2)-b-D-glucopyranoside and 3-O-b-Dglucopyranosyl-2a,19a-dihydroxy-urs-12(13)-en-28,20blactone. Compounds 1 and 5 exhibited potent anti-HCVactivities in vitro with a selective index of 6.53 and 4.41,respectively.

        A Spraying Path Planning Algorithm Based on Point Cloud Segmentation and Trajectory Sequence Optimization

        Spraying trajectory planning is a key and challenging work for intelligent spraying robot. In order to effectively fulfill spraying on complex surfaces without CAD model, a novel spraying trajectory planning method based on segmentation and trajectory sequence optimization is proposed in this paper, which is mainly composed by three steps: surface segmentation, trajectories generation and trajectories connection. In surface segmentation, a method named regional growth with minimum curvature point (RGMCP) is proposed to segment a 3D entity into different subsurfaces by taking normals and curvatures into consideration simultaneously. In trajectories generation step, an intersection of plane and point cloud (IPPC) algorithm is used to generate the optimal spraying trajectory for each segmented subsurface. Finally, for trajectories connection, a sequence optimization algorithm based on swap-evolution particles (SOSP) is proposed to connect all the subsurface trajectories as a complete spraying one in an optimum manner by regarding it as a sequence optimization problem. The effectiveness of the proposed method is validated by simulation and practical experiment simultaneously. Comparatively, our method can improve the efficiency of a spray task with 367 trajectories and 627 s time-consuming to 215 trajectories and 413 s, while the coating thickness variances are lowered from 51.9 µm2 and 30.4 µm2 to 3.64 µm2 and 7.89 µm2 respectively, which shows that the proposed method is more effective and can keep better coating thickness uniformity.

        Coenzyme Q10 Production by Sphingomonas sp. ZUTE03 with Novel Precursors Isolated from Tobacco Waste in a Two-Phase Conversion System

        Coenzyme Q10 (CoQ10) is a widely used supplement in heart diseases treatment or antioxidative dietary. The microbial production of CoQ10 was enhanced by addition of solanesol and novel precursors recovered from waste tobacco. The novel precursors were separated by silica gel and identified as α-linolenic acid (LNA) and butylated hydroxytoluene (BHT) based on the effect on CoQ10 production and GC-MS. The effects of novel precursors on CoQ10 production by Sphingomonas sp. ZUTE03 were further evaluated in a two-phase conversion system. The precursor`s combination of solanesol (70mg/l) with BHT (30mg/l) showed the best effect on the improvement of CoQ10 yield. A maximal CoQ10 productivity (9.5 mg l-1 h-1) was achieved after 8h conversion, with a molar conversion rate of 92.6% and 92.4% on BHT and solanesol, respectively. The novel precursors, BHT and LNA in crude extracts from waste tobacco leaves, might become potential candidates for application in the industrial production of CoQ10 by microbes.
