      • 골수유핵세포의 경내피세포 및 경기질세포 이행 과정에서 류코트리엔 B_(4) 역할과 활성산소종 억제의 영향

        Objectives:Leukotriene B_(4)(LTB4) is lipid mediator derived from membrane phospholipids during the process of inflammation, having many roles(ie;inducer of chemotaxis, the production of nitric oxide, transepithelial migration of neutrophil). The major activities of LTB4 include the recruitment and activation of leukocytes, suggesting that it may involve the process for transendothelial migration of nuclear cells in bone marrow environment. Reactive Oxygen Species (ROS) have a cell signaling roles that are involved in signal transduction cascades of numerous growth factor-, cytokine-, and hormone-mediated pathways, and regulate many biological systems. In this present study, we focused on the role of LTB4 and ROS on transmigration of bone marrow nuclear cells across endothelial or stromal cell monolayer. Methods:MS-5, murine stromal cell line cells, or bEnd.3, murine microvascular cell line cells, were grown to confluence on microporous transwell membrane. Murine marrow cells were placed on top of the prepared transwell membrane. The transwells were then seated in wells containing media and LTB4 with or without pretreatment of N-acetylcysteine(NAC), an oxygen free radical scavenger, or diphenylene iodonium(DPI), an inhibitor of NADPH oxidase-like flavoproteins. Cells that migrated through the stromal or endothelial layer into the wells were assayed for transendothelial migration. Results:The numbers of migrated bone marrow nuclear cells through the bEnd.3 were increased by treatment of LTB4(control, 12.5±0.2%;50nM, 22.7±0.9%;100nM, 44.3±1.4%;200nM, 36.3±0.9%;p<0.05). The numbers of migrated bone marrow nuclear cells through the MS-5 were also increased by treatment of LTB4(control, 11.0±0.9%;50nM, 25.7±0.9%;100nM, 35.8±1.8%;200nM, 32.1±0.9%;p<0.05). However, increasing effect of LTB4 to the transmi-gration of bone marrow nuclear cells through the MS-5 or bEnd.3 were inhibited by pretreatment of NAC or DPI. Conclusion:Through our data, it is suggested that LTB4 could induce the transmigration of bone marrow nuclear cells and ROS might be involved on the transendothelial migration of bone marrow nuclear cells by LTB4. It would be very interesting to test the effects of LTB4 and ROS on stem cell mobilization and homing in the future. 류코트리엔 B_(4)(LTB4)는 세포막 인지질로부터 유래된 염증과정에 관여하는 지방매개체이다1-3). LTB4의 주된 역할은 화학주성의 유도, 활성산소의 생산, 백혈구, 특히, 호중구 및 호산구의 이동에 관여하고 있으며, 내피세포와 백혈구의 부착에 중요한 역할을 하는데, 이러한 LTB4의 역할은 케모카인에 의한 백혈구의 이동과 화학주성과 많은 부분에서 유사한 특성을 가진다4-7). 염증반응과 백혈구 이동과 연관된 LTB4의 역할들은 골수내 세포들의 이동 과정에서 다른 시토카인이나 성장인자 혹은 케모카인의 역할과 유사한 점이 많다. LTB4와 비슷한 작용을 가진 여러 케모카인들이 조혈모세포를 포함한 골수내 세포들의 이동과 연관이 있음이 최근 밝혀짐에 따라, LTB4도 골수내 세포의 이동에 관련이 있을 가능성이 있다8-14). 활성산소종(ROS)은 여러 종류의 성장인자, 시토카인, 혹은 호르몬 등으로 유도된 조혈세포내 신호전달체계에 중요한 역할을 하고, 조혈세포내 여러 생명현상의 조절과 관련이 있다15-18). 특히, 염증세포에서 분비된 시토카인 혹은 케모카인들이 세포내 ROS를 증가시키고, 이때 증가된 ROS는 혈관내피세포 단층의 투과성을 증가시켜 백혈구의 경내피세포 이행을 용이하게 한다19-22). 조혈모세포 이식 후 조혈모세포가 말초혈액에서 골수 내로 자리잡는 귀소나, 골수내 조혈모세포를 혈액으로 나오게 하는 가동화의 과정은 매우 복잡하여, 여러 다양한 기전들이 관련이 있지만, 혈관내피세포 및 기질세포와 골수 조혈세포들과의 유기적인 반응과 골수 조혈세포의 경내피세포 이행이 중요한 역할을 한다23)24). 본 연구에서는 골수유핵세포의 경내피세포 및 경기질세포 이행에 LTB4와 ROS가 어떻게 영향을 미치는지 알아보고자 하였다.

        The Accurate Ranging System for Geodetic Observation - Mobile (ARGO-M) was successfully developed as the first Korean mobile Satellite Laser Ranging (SLR) system in 2012, and has joined in the International Laser Ranging Service (ILRS) tracking network, DAEdeoK (DAEK) station. The DAEK SLR station was approved as a validated station in April 2014, through the ILRS station "data validation" process. The ARGO-M system is designed to enable 2 kHz laser ranging with millimeter-level precision for geodetic, remote sensing, navigation, and experimental satellites equipped with Laser Retro-reflector Arrays (LRAs). In this paper, we present the design and development of a next generation high-repetition-rate SLR system for ARGO-M. The laser ranging rate up to 10 kHz is becoming an important issue in the SLR community to improve ranging precision. To implement high-repetition-rate SLR system, the High-repetition-rate SLR operation system (HSLR-10) was designed and developed using ARGO-M Range Gate Generator (A-RGG), so as to enable laser ranging from 50 Hz to 10 kHz. HSLR-10 includes both hardware controlling software and data post-processing software. This paper shows the design and development of key technologies of high-repetition-rate SLR system. The developed system was tested successfully at DAEK station and then moved to Sejong station, a new Korean SLR station, on July 1, 2015. HSLR-10 will begin normal operations at Sejong station in the near future.

        Congenital Heart Disease: a Pictorial Illustration of Putting Segmental Approach into Practice

        The human heart is a complex organ in which many complicated congenital defects may happen and some of them require surgical intervention. Due to the vast complexity of varied anatomical presentations, establishing an accurate and consistent nomenclature system is utmost important to facilitate effective communication among pediatric cardiologists, cardiothoracic surgeons and radiologists. The Van Praagh segmental approach to the complex congenital heart disease (CHD) was developed in the 1960s and has been used widely as the language for describing complex anatomy of CHD over the decades. It utilizes a systematic and sequential method to describe the cardiac segments and connections which in turn allows accurate, comprehensive and unambiguous description of CHD. It can also be applied to multiple imaging modalities such as echocardiogram, cardiac CT and MRI. The Van Praagh notation demonstrates a group of three letters, with each letter representative for a key embryologic region of cardiac anatomy: the atria, ventricles and great vessels. By using a 3-steps approach, we can evaluate complex CHD precisely and have no difficulties in communicating with other medial colleague. This pictorial essay revisits the logical steps of segmental approach, followed by a pictorial illustration of its application.

        Burkitt's lymphoma is an unique subtype of malignant lymphoma, first reported in 1985 by Denis Burkitt among african children. We experienced a case of Burkitt's lymphoma in 11 years old female patient complaining of abdominal distention and dyspnea. Clinical and histopathologic findings were compatible with Bukitt's lymphoma. We have presented this case with literature reviews.

        멍게껍질 추출색소 및 CLA (Conjugated Linoleic Acid)가 함유된 사료를 섭취한 고등어 (Scomber japonicus)의 항산화 활성

        This study was performed to assess the antioxidative properties of lipid from aquacultured mackerel Scomber japonicus fed with a diet fortified with conjugated linoleic acid (CLA) and ascidian Halocynthia roretzi tunic extracts by radical scavenging assay. The fish were separated into squid oil (Control) and 2.5% CLA (CA25) groups during the 8-week feeding period. The reducing power of each sample showed high levels of activity compared with α-tocopherol and butylated-hydroxyanisol (BHA) at 0.2-1.0 mg/mL of lipid. Inhibition of linoleic acid oxidation in samples from Control and CA25 groups showed similar activity after 2 days of incubation at 40℃. The 2,2-diphenyl-1-picrylhydrazyl (DPPH), superoxide, and hydroxyl radical scavenging activities of CLA and carotenoid-deposited sample (CA25) were higher than those of the Control group. The results indicated that the lipid extracted from the viscera of mackerel showed slightly higher antioxidant activities than that from the muscle.

        CLA(Conjugated Linoleic Acid)가 함유된 사료를 섭취한 고등어 (Scomber japonicus)의 가열조리에 의한 지방산조성의 변화

        CLA가 함유된 사료를 섭취하고 성장한 고등어에 함유된 EPA, DHA 등의 고도불포화지방산을 포함한 여러 종류의 지방산이 여러 가지 방법으로 요리하였을 때의 어떠한 변화를 나타내는가를 조사함으로써 소비자에게 양식 고등어의 적절한 조리방법을 제공하고자 하였다. 조리 후 포화산 및 모노엔산은 생육의 Control 27.5%, CA25 28.8%와 차이가 없었다. 폴리엔산 역시 RO-8GM, RO-8CM이 31.2%, 30.7%, BO-8GM, BO-8CM은 27.1%, 31.5%, CA-8GM, CA-8CM은 25.4%, 28.4%를 나타내어 생육의 Control 29.4%, CA25의 31.9%와 비교하였을 때 된장조림 및 통조림의 경우 CLA 무첨가구에서 함량이 감소하였지만, CLA 첨가구에서는 통조림구에서만 감소하는 경향을 보였다. 조리된 고등어중 n-6/n-3의 비는 구이의 경우 Control 및 CA25구 각각에서 0.29, 0.24, 된장조림의 경우 각각 0.28, 0.27 그리고 통조림의 경우 각각 0.28, 0.31로 나타났다. The effects of frying, steaming with soybean paste, and canning on the fatty acid compositions of farmed mackerel fed with CLA were evaluated. Saturates and monoenes acid content of the cooked mackerel control and CA25 groups at 27.5% and 44.6% and at 28.8% and 41.0%, respectively, were not significantly different from the raw samples at 27.1% and 35.6%, respectively. The polyenes acid content of control and CLA-fed groups were 31.2% in RO-8GM and 30.7% in RO-8CM after roasting, 27.1% in BO-8GM and 31.5% in BO-8CM for boiling, and 25.4% in CA-8GM and 28.4% in CA-8CM after canning which were not significantly different from the raw samples with 29.45% and 31.9%, respectively. Ratio of the n-6/n-3 in roasted group were 0.29 and 0.24, in steaming with soybean paste were 0.28 and 0.27, and in canned mackerel were 0.28 and 0.31 for the control and CA25 groups, respectively.
