      • RE-09 : High mobility group box-1 and Toll like receptor 4 in Endometriosis: Mediating NF-kB pathway

        목적: The aim of the study was to examine the sterile, chronic inflammatory status in endometriosis by detecting High mobility group box-1 (HMGB-1), and HMGB-1 signaling through toll like receptor (TLR) 4 in endometrial stromal cell, which activates NF-kB pathway that might play a pathogenic role in endometriosis. 방법: Total 69 patients who had undergone hysterectomy from March, 2012 to March, 2014 were included: 39 patients who had endometriosis as case group, 30 participants without endometriosis as the control. Expression of HMGB-1 in endometrium was examined by immunohistochemistry. From endometriosis patient, eutopic endometrium was obtained and used to culture in vitro. To confirm the association of HMGB-1 and TLR4 in NF-kB pathway, the cultured human endometrial stromal cells (HESCs) were treated with recombinant HMGB-1 (15 ng/mL) for 0, 12, 24, 48 hrs. TLR4 antagonist (LPS-RS) was added after rHMGB-1 treatment to HESCs to see the change of proliferation after inhibiting TLR4. CCK8 proliferation assay kit. Real-time PCR and western blotting were used to examine the proliferation, quantification of the mRNA and protein. Specific inhibitor of NF-κB signaling pathway (TPCA-1) was used to explore the role of NF-κB signaling pathway in HESCs proliferation. 결과: HMGB-1 expression was significantly higher in glandular epithelial and stromal cells of endometriosis group compared to the controls. rHMGB-1 significantly enhanced cell proliferation in HESCs and increased TLR4 expression by dose dependent fashion. Inhibiting TLR4 by LPS-RS, cell proliferation in HESCs and expression of TLR4 were significantly decreased comparing to the controls. After blocking NF-kB pathway with TPCA-1, cell proliferation in HESCs was significantly decreased by dose dependent fashion. 결론: This study showed that HMGB-1 may play an important role through TLR4 and NF-kB pathway, in inflammatory cascade of pathogenesis in endometriosis.

      • Ar 이온에 의해 Sputtering된 Pt(111) 표면의 NO 흡착

        Adsorption of nitric oxide on the Pt(111) surface sputtered by Ar-ion has been studied using thermal desorption spectroscopy, Auger electron spectroscopy, and low energy electron diffraction. A thermal desorption spectrum obtained from the perfect Pt(111) saturated with NO at 300K is quite simillar to those reported previously. The main portion desorbs at about 370K(α-state) with a shoulder at about 470K(β-state). The chemisorption of nitric oxide is predominantly molecular on the Pt(111) surface, accompanied by a small amount of dissociation which becomes negligible when the Pt(111) surface is perfect. When the Pt(111) surface is sputtered by Ar-ion with 2KeV, the thermal desorption spectrum becomes quite complex. The shoulder peak, which appears on the perfect surface spectrum, increases with Ar-ion sputtering time. The maximum desorption spectra of N₂ and N₂O are observed simultaneously between 470 and 600K. The desorption mechanisms for N₂O are proposed. The increasing N₂ with the β-state of NO indicates that the β-state is a precursor to the NO dissociation.

      • 초고진공 장치의 설계 및 제작과 LEED Optics에 의한 Auger Electron Spectrometer의 분해능 측정

        A multipurpose Ultra High Vacuum chamber was designed and constructed in this laboratory. This enables us to be equipped with various analytical instruments such as Ar-ion gun LEED optics, Electron analyzer, and Quadrupole mass spectrometer etc. The 4-Grid LEED optics, a product of Omicron company in West Germany, was equipped with the UHV chamber and the LEED power supply was designed by this group. Turbo-C language figured out Auger spectra of the ad-species by means of the computer interface. The resolution of Auger electron spectrometer was measured with a constant primary electron beam energy. The results showed that the resolution was independent of the primary electron beam energy, but depends on the modulation voltage. Also, it was revealed that the resolution of 4-Grid optics was more improved than that of 3-Grid optics.

      • 열탈착 분석법에 의한 Pt(111) 표면위의 NO와 CO의 흡착

        The adsorptions of nitric oxide and carbon monoxide on Pt(111) surfaces have been studied by using LEED, AES, and TDS(Thermal Desorption spectroscopy). The adsorbed species of NO is predominantly molecular on the Pt(111) surface at room temperature. A main desorption peak of TDS is attributed to the molecular adsorption at 360K. With increasing No exposures, a shoulder peak at 480K which may be attributed to the defect site adsorption is appeared. The desorption rate order for No desorption is 1st order and the desorption energy for NO calculated by Redhead equation assuming pre-exponential factor v_1=10^13 sec^-1 is 21.7Kcal/mol. The adsorption of carbon monoxide is also molecular on Pt(111) surface at room temperature. The TDS results showed only a single broad peak at 460K, which is attributed to molecular adsorption. The desorption order for CO desorption is 1st order as well as NO and the desorption energy for CO is 28Kcal/mol.

        In vitro fertilization outcome in women with diminished ovarian reserve

        Objective This study aimed to identify factors that affect in vitro fertilization (IVF) outcomes in women with diminished ovarian reserve (DOR). Methods We reviewed 99 IVF cycles in 52 women with DOR between September 2010 and January 2015. DOR was defined as serum anti-Mullerian hormone level of <1.1 ng/dL or serum follicle-stimulating hormone level of ≥20 mIU/mL. Total 96 cycles in 50 patients were evaluated after excluding fertility preservation cases. Results The clinical pregnancy rate was 11.5% per cycle, and the total cancellation rate was 34.4%. Clinical pregnancy rate was significantly associated with the antral follicle count and the cause of the DOR. Age, serum anti-Mullerian hormone and follicle-stimulating hormone levels, antral follicle count, peak estradiol level, and the cause of DOR were significantly associated with cycle cancellation. However, history of previous ovarian surgery remained as a significant factor of clinical pregnancy (model 1: odds ratio [OR] 10.17, 95% confidence interval [CI] 1.46 to 70.84, P =0.019; model 2: OR 10.85, 95% CI 1.05 to 111.71, P =0.045). In cancellation models, idiopathic or previous chemotherapy group showed borderline significance (model 1: OR 3.76, 95% CI 0.83 to 17.04, P =0.086; model 2: OR 3.15, 95% CI 0.84 to 11.84, P =0.09). Conclusion DOR caused by previous ovarian surgery may show better pregnancy outcome, whereas that caused by chemotherapy could significantly increase the cycle cancellation rate. Furthermore, patients with DOR who previously received gonadotoxic agents may show reduced efficacy and increased risk of IVF cycle cancellation.

      • RE-12 : HMGB-1 induced toll-like receptor 4 signaling promotes cell proliferation in endometrial stromal cells

        Toll-like receptors (TLRs) are innate immune receptors that mediate the pattern recognition of, and response toward pathogens and host-derived danger signals. We examined whether endogenous ligands released by damaged cells such as high-mobility group box-1 (HMGB-1) signaling through TLR4 may play a pathogenic role in endometriosis. HMGB-1 and TLR4 expression in endometrium were analyzed using immunohistochemistry with HMGB-1, a specific endogenous ligand of TLR4 to activate TLR4 signaling. The effect of HMGB-1-TLR4 signaling on cell proliferation was examined. Specific inhibitor of NF-κB signaling pathway was used to explore the role of NF-κB signaling pathway in HMGB-1-TLR4 pathway. During the menstrual cycle, TLR4 protein was highly expressed. HMGB-1 significantly enhanced cell proliferation in endometrial stromal cells by TLR4 dependent fashion. HMGB-1-TLR4 signaling was associated with regulation on the activation of NF-κB signaling pathway. Our results suggest that HMGB-1-TLR4 signaling may play an important role in establishment of endometriosis through NF-κB signaling pathway.

        Decreased Renal Function Is a Risk Factor for Subclinical Coronary Atherosclerosis in Korean Postmenopausal Women

        Objectives: Decreased renal function is associated with increased cardiovascular risk. Our study was planned to verify the association of decreased renal function and subclinical coronary atherosclerosis in postmenopausal women. Methods: We performed a retrospective review of 251 Korean postmenopausal women who visited the health promotion center for a routine health checkup. Estimated glomerular filtration rate (eGFR) was used to show renal function, which was estimated by calculated using the Cockcroft-Gault (CG) and the modification of diet in renal disease (MDRD) formulas. Coronary atherosclerosis was assessed by 64-row multidetector computed tomography. Results: Women with reduced eGFR (< 60 mL/minute/1.73 m<sup>2</sup>) had significantly higher brachial-ankle pulse wave velocity (baPWV) than women with normal eGFR (≥ 60 mL/minute/1.73 m<sup>2</sup>). The eGFR was negatively correlated with baPWV (r = -0.352, P < 0.001), significantly. The eGFR was lower in women with coronary atherosclerosis than in normal control women, markedly. Reduced eGFR was significantly associated with the presence of coronary atherosclerosis (odds ratio [OR] = 7.528, 95% confidence interval [CI] = 2.728-20.772, P < 0.001). Conclusions: Decreased eGFR was closely associated with increased arterial stiffness and coronary atherosclerosis in postmenopausal women. Evaluating subclinical atherosclerosis by screening the renal function in postmenopausal women may be helpful screening high risk group and considering starting menopausal hormone therapy before atherosclerosis development.       • High mobility group box-1 in ectopic endometrium: Role in inflammatory pathway of endometriosis

        목적: The aim of the study was to examine the relationship between inflammation by oxidative stress and development of endometriosis by High mobility group box-1 (HMGB-1). 방법: Ectopic endometrial tissue was obtained from patient with endometriosis, and used for in vitro stromal cell culture. Ishikawa cell was used for epithelial cell experiments. Endometrial cell proliferation was examined by CCK-8 proliferation assay, cell viability was confirmed by MTT assay. To examine how different cell invasion changes according to the inflammatory status, cell invasion study was performed treating recombinant HMGB-1 by increasing concentration. 결과: The cultured human endometrial stromal cells (HESCs) from ectopic endometrium showed decreasing cell viability and increasing HMGB-1 release according to H2O2 treatment. On the H2O2 treated HESC, adding estradiol showed increasing cell proliferation in HESCs and ishikawa cells. The invasion study showed increasing cell invasion count according to treated rHMGB-1 concentration. From HESCs from ectopic endometrium, HMGB-1 release was increased by concentration of treated estradiol and H2O2. 결론: This study showed that HMGB-1 may play an important role in inflammatory cascade of pathogenesis in endometriosis.

      • Case Reports : A case of isolated peritoneal metastasis in clinically early stage squamous cell carcinoma of the uterine cervix

        Cervix cancer had been the most common gynecologic cancer for decades. Although early diagnosis and treatment have been developed, cervix cancer is the fourth leading cause of cancer death in women worldwide. Unlike other gynecologic cancers, cervical cancer is defined and classified by a clinical staging system. Because this system is based on the spreading pathway in which cancer cell prefers to spread directly to adjacent organs, it is very rare to see distant metastasis of early stage cervical cancer. Herein, we report a case with clinically early stage squamous cell carcinoma of uterine cervix, which showed isolated peritoneal metastasis, and review the relevant literature.
