apoptosis protein (IAP) family, which is known to be over-expressed in various carcinomas and associated with their biologically aggressive characteristics. The aim of this study was to investigate survivin expression in human medullary thyroid carcinoma (MTC) and a MTC cell line TT, correlate suvivin expression with clinicopathologic features of MTC, and test effects of antisurvivin oligonucleotides (ASODNs) on growth and apoptosis of TT cells. Survivin expression was immunohistochemically determined in formalin-fixed and paraffinembedded specimens obtained from 10 cases of normal thyroid (NT) and 10 cases of MTC, and in TT cells. In TT cells, we confirmed survivin expression and its down-regulation by ASODNs using RT-PCR and Western blot analyses, and investigated effects of ASODNs on viability and growth by MTT assay and apoptosis by apoptotic analyses including DNA laddering assay, acridine orange/ethidium bromide staining and flow cytometric cell cycle analysis. Immunohistochemical analysis showed high survivin expression in MTC and TT cells, whereas no immunoreactivity was detectable in NT. Statistical analyses revealed no significant correlation of survivin expression with the clinicopathologic features of MTC. In TT cells, survivin expression at both mRNA and protein levels was confirmed and could be down-regulated by ASODNs concomitant with decrease in viability and growth, and increase in apoptosis. Our results suggest that survivin plays an important role in MTC independent of the conventional clinicopathologic factors, and ASODNs is a promising survivin- targeted gene therapy for MTC.

Sine oculis homeobox homolog 3 (Six3) gene is responsible for normal mammalian pituitary development, and its genetic variations or deficiency will cause hypopituitarism, suggesting that this gene is a potential candidate gene for studying association with growth traits in animals. Herein, genetic variants within bovine Six3 gene was screened in 1031 individuals from four Chinese indigenous cattle breeds. Two novel polymorphisms (NC_007309:g.2515G>A and NC_007309:g.2607T>C) locating at positions nt1707 and nt1799 of intron 1 in bovine Six3gene, were found, and could be genotyped by TaqI ACRS PCR-RFLP and Alw26I PCR-RFLP, respectively. The frequencies of allele “A” of TaqI locus varied from 0.004 to 0.309,as well as the frequencies of allele “C” of Alw26I locus waved from 0.025 to 0.340. Association analysis revealed no significant association of TaqI locus with growth traits in Nanyang breed. However, significant relationships between Alw26I locus and body weight (BW) and average daily gain (ADG) in Nanyang breed was found (p<0.05). The individuals with genotype TC had greater body weight and average daily gain than those with genotype TT at 18 months old. Furthermore, based on combinated genotypes from these two loci, diplotypes was found to be associated with growth traits (p<0.05).The individuals with dihaplotype GG-TC had greater body weight and average daily gain at 18 month-old than those of other dihaplotypes. Therefore, the TaqI and Alw26I genetic variants of bovine Six3 gene were recommended as DNA markers related to growth traits through marker-assisted selection for genetics and breeding in cattle.
A New Generalized Resolvent and Application in Banach Spaces

In this paper, we introduce a new generalized resolvent in a Banach space and discuss its some properties. Using these properties, we obtain an iterative scheme for finding a point which is a fixed point of relatively weak nonexpansive mapping and a zero of monotone mapping. Furthermore, strong convergence of the scheme to a point which is a fixed point of relatively weak nonexpansive mapping and a zero of monotone mapping is proved.

Three new diphenylpropanes, HindsiipropaneA–C (1–3), together with one known arylpropyl quinoneGriffithane D (4), were isolated from Celastrus hindsii. Their structures were established by 1D and 2D nuclearmagnetic resonance spectroscopic analysis, and massspectroscopy. Compound 4 was firstly obtained in thisgenus. All the isolated compounds were evaluated in vitrofor cytotoxicity against four human tumor cell lines (A549,HCT116, MDA-MB-231, BEL7404) by the MTT assay.

Odontoglossum ringspot virus (ORSV) is a member of the genus Tobamovirus. It is one of the most prevalent viruses infecting orchids worldwide. Earlier studies reported the genetic variability of ORSV isolates from Korea and China. However, the evolutionary rate, timescale, and phylogeographical analyses of ORSV were unclear. Twenty-one coat protein (CP) gene sequences of ORSV were determined in this study, and used them together with 145 CP sequences obtained from GenBank to infer the genetic diversities, evolutionary rate, timescale and migration of ORSV populations. Evolutionary rate of ORSV populations was 1.25 × 10−3 nucleotides/site/y. The most recent common ancestors came from 30 year ago (95% confidence intervals, 26- 40). Based on CP gene, ORSV migrated from mainland China and South Korea to Taiwan island, Germany, Australia, Singapore, and Indonesia, and it also circulated within east Asia. Our study is the first attempt to evaluate the evolutionary rates, timescales and migration dynamics of ORSV.
Uniaxial Eccentric Load Behavior of L-CFSST Composite Column

This study presents the mechanical behavior of L-shaped concrete-filled square steel tube (L-CFSST) composite columns under uniaxial eccentric compression. Nine specimens were tested and the effects of eccentricity, steel tube thickness, steel yield strength, and slenderness ratio were investigated regarding failure modes, ultimate load, load-lateral displacement curves, and load-strain curves. Test results showed that, under uniaxial eccentric load, these columns showed favorable mechanical properties and steel tubes showed strong cooperative working performance. There were two types of failure modes for these columns: local buckling of the steel tube or overall bending of the specimen. ABAQUS was used to simulate the loading process and the accuracy of the numerical simulation results was verified by comparing failure characteristics, load-displacement curves, and ultimate bearing capacity with the test results. The N/Nu-M/Mu curves of the columns under uniaxial eccentric load with different parameters were simulated by the finite element method. A simplified formula for predicting the N/Nu-M/Mu relationship was established based on the finite element analysis results, which might be a workable approach for designing L-CFSST composite columns in engineering practice.

Characterization and uncertainty of uplift load-displacement behaviour of belled piers

A total of 99 full-scale field load tests at 22 sites were compiled for this study to elucidate several issues related to the load-displacement behaviour of belled piers under axial uplift loading, including (1) interpretation criteria to define various elastic, inelastic, and "failure" states for each load test from the load-displacement curve; (2) generalized correlations among these states and determinations to the predicted ultimate uplift resistances; (3) uncertainty in the resistance model factor statistics required for reliability-based ultimate limit state (ULS) design; (4) uncertainty associated with the normalized load-displacement curves and the resulting model factor statistics required for reliability-based serviceability limit state (SLS) design; and (5) variations of the combined ULS and SLS model factor statistics for reliability-based limit state designs. The approaches discussed in this study are practical and grounded realistically on the load tests of belled piers with minimal assumptions. The results on the characterization and uncertainty of uplift load-displacement behaviour of belled piers could be served as to extend the early contributions for reliability-based ULS and SLS designs.
