        참당귀의 흰쥐 4-Vessel Occlusion 전뇌허혈에 대한 신경방어효과

        Objectives : Angelicae Radix, the root of Angelica gigas N_AKAI (Umbelliferae) , which has been used to nourish the blood(補血), regulate menstruation(調經), relieve pain and promote bowel movements(崩漏) and activate blood circulation for the treatment of fatigue, headache, constipation and blood troubles in traditional Korean medicine. It's flavour and channel tropism is sweet(甘), pungent(辛), warm(溫), acting on the lung, heart and spleen(肺, 心, 脾). The purpose of study we report here was to determine the neuroprotective effect of Angelica gigas on global ischemia induced by 4-vessel occlusion in Wistar rats. Methods : Angelica gigas extracts was lyphilized after extraction with 85% methanol. We induced 4-VO for 10 minutes and reperfused again. The number of CA1 pyramidal neurons were counted after 7 days of reperfusion under the cresyl violet staining. Results : The results showed that Angelica gigas had significantly neuroprotective effects (100, 250 and 500mg/kg of AR extracts, p<0.001) compared with control group. Each neuroprotective ratio was about 66.1, 79.3 and 78.6% respectively. Immunohistochemical data partially explained the protective effects of Angelica gigas via attenuation of COX-2 induction in hippocampus. Conclusion : Angelica gigas gas neuroprotective effects on the global ischemia induced by 4-vessel occlusion in Wistar rats and the attenuation of COX-2 induction is thought to be its possible mechanism.

      • 흰쥐 정소 간질세포의 발생에 관한 형태학적 연구

        It is well known that interstitial cell of Leydig is developed from the mesenchymal cell in the intestitial tissue of the testis during fetal period, but it gradually regressed until puberty when mature Leydig cell is derived from the mesenchyme of interstitial tissue of fetal Leydig cell. The function of the Leydig cell is synthesis and secretion of testosterone which influences on differentiation of male geritalia at fetal period, and revealation of male secondary sexual characteristies and differentiation of sperm at puberty. To investigate the differentiation of interstitial cell in interstitium and ultrastructural changes of interstitial cell in rat testis and ultrastructural changes of interstitial cell in rat testis and relation to testosterone secretion, the author has studied this experiment. Fetal rats on the 16th, 18th and 20th gestational days and rats on the 1st, 3rd, 7th day, 2nd, 3rd and 8th week after birth were used as experimental animals. Some of obtained testis were stained with hematoxylin and eosin for light microscopic study, and other specimens were double stained with uranyl acetate and lead citrate for electron microscopic study. The results were as follows: 1. On the 16th day of pregnancy, a few of round eosinophilic Leydig cells are firstly observed in the interstitium of testis in fetal rat. On the 18th and 20th day of pregnance, 1st and 3rd day of neonatal life, a number of Leydig cells forming cellular cord were observed. On the 1st and 2nd week after birth, number and size of Leydig cell are markedly reduced but on teh 3rd week after birth Leydig cells are reappeared. On the 8th week after birth distribution of Leydig cells is similar to that in adult rat testis. 2. On the 18th and 20th day pregnancy, and 1st and 3rd day of neonatal life, circular or oval shaped mitochondria matrix were observed in the Leydig cell. On the 1st and 2nd week after birth, giant mitochondria and mitochondria with disorganized mitochondrial crista were revealed. On the 3rd and 8th week after birth, appearances of mitochondria were similar to that in Leydig cell of the rat testis on the 20th day of pregnancy. 3. Numerous and round cisternae of smooth endoplasmic reticulum were observed in Leydig cell of rat testis on the 18th and 20th day of pregnancy. 1st and 3rd day of neonatal life, and 1st week after birth . Horseshoe-shaped deformity and reduction in number of cisternae of smooth endopalmic reticulum were revealed in Leydig cells on the 2nd week after birth. Numerous and round cisternae of smooth endoplasmic reticulum were seen on the 3rd and 8th week after birth. 4. A few lipid droplets were revealed in Leydig cell of rat testis on the 18th day of pregnancy, but many lipid droplets were observed in Leydig cells of rat testis on the 20th day of gestation, 1st and 3rd day of neonatal life, and 1st week after birth. Numbers of lipid droplets were gradually decreased after 2nd week of postnatal life. 5. Many autophagic vacuoles were observed in Leydig cells would be firstly differentiated from the mesenchymeal cells in interstitium of fetal rat testis on the 16th day of pregnancy, and proliferated from the 18th day of pregnancy to the 1st day of neonatal life, and then gradually regressed before the puberty, but at the begining of puberty adult type of Leydig cell be proliferated.

        The objective of this study was to manifest the food labelling system that would be of help to consumers’ efficient decision making on food labelling. Out of the contents of the labelling standard of food, etc., recently revised by Notification No. 2000-36 of Korea Food &rug Administration, several mooted points were raised of the type of food, the date of manufacture, the term of distribution, ingredients, raw materials, content, etc. Despite these mooted points of the labelling system of food and so forth, the current regulations of the labelling standard of food and so on plays a substantial role in fulfilling the needs of the most important factors in the labelling regulations from the perspective of the protection of consumers and communication, such as the description of nutritive substances, notabilias, cautions and validity of inferior foods degenerated in the process of distribution by regulating false or exaggerated advertisements, along with intention of prevention of misuse and abuse of foods. As a result, the present study revealed results out of the inquiries into the evaluation of and improvements in the labelling system of food as a communications medium, as follows: First, the nutrients that are stipulated to be included in food labelling are confined to calory, carbohydrate, protein, fat and natrium. However, the description of essential nutritive substances as a communications medium that are closely related to frequently invaded diseases such as hypertension, diabetes, heart disease, hyperlipemia, cerebrovascular diseases, etc. in consideration of their frequency of invasion. Furthermore, as regards the nutritive substances to be included in labelling in relation to the recommended quantity of nutrition per day is limited to vitamin and inorganic substances, which also raise the mooted points that they contain a narrow range of nurients and that the recommended quantity of nutrition should be considered according to the invasion of diseases on consumers. In consequence, it is needed to develop and supply computer programs of the contents of nutrients, expand research projects in compliance with consumers’ diet, provide more public relations of the labelling of nutritive ingredients and supply guiding leaflets and then gradually ameliorate systems and regulations of the labelling of nutritive substances. Second, the labelling of notabilias and cautions as a communications medium awakens consumers a sense of terror or a sense of awakening to provide for dangerous cases on the consumers, serving as one of the most important labelling factors to protect the health and lives of consumers by rationally making choice of food. This regulations may be referred to as a prior or a posterior system that can actively protect harmful elements to consumers by the emergence of the food recall system and the product liability law. Therefore, besides the obligatory labelling of notabilias and cautions of food under the food hygiene law, it is necessitated to revise the related law so that it should include all the factors that may do harm to consumers and get to extend the regulation of labelling notabilias and cautions currently applied to part of foods to all. In addition to it, it is recommended for the government and enterprises to prepare compound and comprehensive measures against it to enhance the reliability of foods for consumers. Third, labelling the length of storage of foods consists of the date of manufacture and that of validity. The former only manifests the date at which a food was made but does not show till what time its quality is maintained, while the latter does not know the date of manufacture and the elapse of time but represents how long its quality can last. Today the government is gradually implementing the autonomous establishment of the term of distribution but the mere description of it has its limits. In consequence, when we take into account the fact that almost all the food enterprises are small in scale and weak in their distribution structure, on a short term basis we are in need of the government support to medium and small food companies, the security of consumers’ reliability on the information of the labelling of the term of distribution, the practice of an awarding system of resident reports of illegal and inferior foods, the operation of a fluctuated price system of validity-overdue foods, extended practice of a food bank for the purpose of efficient management of food materials, and on a long term basis the introduction of a pluralistic validity labelling system which can be harmonized with that of advanced countries.

      • 당귀 물추출물의 면역증강 효과

        천연물로부터 생체의 방어력을 증강시킬 수 있는 면역조절물질을 발굴하기 위한 목적의 일환으로, 한방에서 보혈에 관련하여 널리 사용되고 있는 당귀가 면역계에 미치는 영향을 검토하였다. 당귀추출물은 실험관 내에서 lipopoly-saccharides(LPS), phytohemagglutinin(PHA)와 같은 수준의 lymphcoyte 증식 효과를 보였으며, 그 효과는 LPS보다 오랫동안 지속되었다. LPS가 강력한 B cell mitogen이지만 생체내 독성이 심하여 투여가 어려운 반면, 당귀추출물에 의한 생체내 독성은 거의 없으므로 앞으로 생체내 투여 가능성이 있을 것으로 사료된다. Mouse에 투여하였을 때에도 당귀추출물은 SRBC에 대한 항체를 생성하는 lymphocytes의 수를 증가시켰으며, GVH 반응도 증가시켜 주었다. 따라서 당귀의 면역계에 대한 효과는 B cell의 증식과 그에 따른 항체 생성의 증가 뿐만아니라, T cell 매개 면역반응의 증가에 의한 것으로 생각되며, 앞으로 인체의 면역계에 미치는 효과 및 그 기작에 관한 많은 연구를 통하여 당귀가 면역증강제로 개발될 가능성이 있을 것으로 사료된다. The effects of Angelica gigas Nakai, which has been used as a blood-building decoction for recovery from weakness in the Chinese medicine, were examined for the purpose of screening a new nontoxic immunomodulator. The water extract of Angelica gigas Nakai showed a strong mitogenic effect on splenocytes in vitro to the same level of lipopolysaccharide(LPS) and phytohemagglutinin(PHA). Splenocytes stimulated with the extract in vitro, survived longer than that with lipopolysaccharide(LPS). LPS is a powerful B cell stimulant. However, it is now entirely restricted to only experimental purposes because of the highly toxic nature. Compared with this, the extract of Angelica gigas Nakai did not show any acute toxicity in mice injected intraperitoneally at a dosage of 1g/kg-body weight. It was tested in a hemolytic plaque assay whether in vivo treatment of the extract of Angelica gigas Nakai could influence the secretion of antibodies against sheep red blood cells(SRBC). It turned out that intraperitoneal(i.p.) injection of the extract of Angelica gigas Nakai for 4 days increased the number of antibody-secreting cells remarkably. Splenomegaly, due to graft versus host reaction, was also increased after 7 days i.p. treatment of the extract of Angelica gigas Nakai. These results indicated that the extract of Angelica gigas Nakai might be a useful immunomodulator, especially since it is a relatively nontoxic natural product.

        Effects of Triterpenoids and Flavonoids Isolated from Alnus firma on HIV-1 Viral Enzymes

        Triterpenoids and flavonoids isolated from Alnus firma S. Z. were found to inhibit HIV-1 virus replication and controlled its essential enzymes. In this study, the inhibition of HIV-1 viral replication and its essential enzymes, such as reverse transcriptase, protease and ${\alpha}-glucosidase$, were observed using 18 Korean plant extracts. Among the extracts, the methanol extract of Alnus firma leaves showed potent inhibition against the HIV-1 induced cytopathic effect (CPE) in MT-4 cells on microscopic observation (the minimum concentration for complete inhibition of HIV-1 induced CPE, IC=50 ${\mu}g/mL$). Thus, 14 compounds were isolated and identified from the methanol extract of Alnus firma leaves. Of these compounds, the alnustic acid methyl ester exhibited inhibition against HIV-1 pretense, with an $IC_{50}$ of 15.8 ${\mu}M$, and quercetin, quercitrin and myricetin $3-O-{\beta}-D-galactopyranoside$ displayed inhibition against H IV-1 reverse transcriptase, all with $IC_{50}$ values of 60 ${\mu}M$. Based on these results, the viral replication inhibition of the methanol extract of Alnus firma leaves was adjudged to be acutely related to the protease inhibition activation of the alnustic acid methyl ester as well as the reverse transcriptase inhibition activation of flavonoids.

        The Effective Preparation of Flavonoids from Scutellaria baicalensis G<sub>EORGI</sub> by Diaion HP-20 Resin

        Scutellaria baicalensis $G_{EORGI}$ (Scutellariae Radix) has been used to clear heat and to dry dampness in the stomach or intestines, which manifests as diarrhea or dysenteric disorder. In this study, we investigated the effective preparation of active components in Scutellariae Radix using the methods of solvent extraction and absorption fractionation for the development of new functional food or pharmaceuticals. The marker substances, baicalin, baicalein, wogonoside, and wogonin were directly isolated from the Scutellariae Radix. There chemical structures were elucidated by spectroscopic analysis. The Scutellariae Radix was extracted with hot water. To enhance yield of flavonoids in Scutellariae Radix, the hot water extract was dissolved in ethanol with concentration dependent manner. The precipitates were separated using centrifugal techniques at 10,000 rpm. Supernatant liquid was applied to the HPLC for quantification of major compounds. Separately, the hot water extract was absorbed on Diaion HP-20 resin. And then, the absorbed fraction was eluted with methanol for HPLC. The contents of baicalin, baicalein, wogonoside and wogonin in different treatment methods were analyzed by HPLC. Total amount of four major components were 16.9% in 50% ethanol extract, 21.7% in 70% ethanol extract, 20.5% in 90% ethanol extract, and 39.3% in absorbed fraction of Diaion HP-20 resin. In these results, we found that resin absorption method is suitable for the extraction of enriched flavonoids from Scutellariae Radix.

        동결건조방식으로 가공된 녹용의 이화학적 특성 연구

        N/A The non-mature antlers of deer, Cervi Parvum Cornu (CPC), have been used as a tonic in traditional Korean medicine. Objectives: The purpose of this study was to evaluate the quality of Cervi Parvum Cornu which is newly processed by lyophiliztion. We compared lyophilized CPC (LCPC) with parboiled CPC (PCPC) by the yield of water extract, the contents of total free amino acids and the patterns of gangliosides. Methods: We analyzed the quantitiative and qualitative determinations of amino acids in LCPC using HPLC method. The contents and patterns of gangliosides in LCPC were measured by gravimetric analysis and HPTLC. Results: The yields of the water extract from LCPC and PCPC gave 18% and 10%, respectively. The contents of total free amino acids in the PCPC were much higher than the LCPC. Comparing the TLC pattern of gangliosides, the both samples showed similar patterns as the purchased gangliosides. The contents of gangliosides in the LCPC (18.6 mg/g) were higher than those in the PCPC (14.3 mg/g). Conclusions: These results suggest that the lyophilizing process may be an alternative method for improving the quality of CPC.
