        Gene therapy of intracranial glioma using interleukin 12-secreting human umbilical cord blood-derived mesenchymal stem cells.

        <P>Clinical trials of gene therapy using a viral delivery system for glioma have been limited. Recently, gene therapy using stem cells as the vehicles for delivery of therapeutic agents has emerged as a new treatment strategy for malignant brain tumors. In this study, we used human umbilical cord blood-derived mesenchymal stem cells (UCB-MSCs) as delivery vehicles with glioma-targeting capabilities, and modified interleukin-12 (IL-12p40N220Q; IL-12M) as a novel therapeutic gene. We also engineered UCB-MSCs to secret IL-12M (UCB-MSC-IL12M) via tetrameric cell-permeable peptide (4HP4)-mediated adenoviral transduction. We confirmed the migratory capacity of UCB-MSC-IL12M toward GL26 mouse glioma cells by an in vitro migration assay and in vivo injection of UCB-MSC-IL12M into the ipsilateral hemisphere of implanted gliomas in C57BL/6 mice. In vivo efficacy experiments showed that intratumoral injection of UCB-MSC-IL12M significantly inhibited tumor growth and prolonged the survival of glioma-bearing mice compared with control mice. Antitumor effects were associated with increased local IL-12M levels, followed by interferon-γ secretion and T-cell infiltration in intracranial gliomas, as well as antiangiogenesis. Interestingly, tumor-free mice after UCB-MSC-IL12M treatment were resistant to ipsilateral and contralateral tumor rechallenge, which was closely associated with tumor-specific long-term T-cell immunity. Thus, our results provide the rationale for designing novel experimental protocols to induce long-term antitumor immunity against intracranial gliomas using UCB-MSCs as an effective delivery vehicle for therapeutic cytokines including IL-12M.</P>

        LPS로 유도한 RAW 264.7 세포의 염증반응에서 자초(紫草)의 항염증 효과

        Objective: Lithospermum Erythrorhizon (LE) has been used as an anti-bacterial and anti-inflammatory agent. However, it is unclear that LE aqueous extract could show the anti-inflammatory effects in RAW 264.7cells. The purpose of this study was to investigate the anti-inflammatory effect of aqueous extract from LE on lipopolysaccharide (LPS) - induced inflammatory response. Methods: To measure out the cytotoxicity of LE, we performed the MTT assay. To evaluate the anti-inflammatory effects of LE, we examined the inflammatory mediators such as nitric oxide (NO), prostaglandin E2 (PGE2) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-a, interleukin, (IL)-1β and (IL)-6) on RAW 264.7 cells. We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-κB) activation by western blot. Results : Aqueous Extract from LE itself did not have any cytotoxic effect in RAW 264.7 cells. Aqueous extract from LE inhibited LPS-induced productions of inflammatory mediators such as NO, PGE2, and pro-inflammatory cytokines including TNF-a, IL-1β and IL-6 in RAW 264.7cells. In addition, LE inhibited the phosphorylation of p38 kinases (p38), c-Jun NH2-terminal kinase (JNK), and NF-κB activation in RAW 264.7 cells. Conclusion : LE down-regulated LPS-induced production of inflammatory mediators through the inhibition of p38, JNK and NF-κB activation. Taken together, these results could provide the evidence for the anti-inflammatory effects of LE. Therefore, LE may be a novel target in the management of inflammation and help to support a potential strategy for prevention and therapy of inflammatory diseases.

      • 체육교과 수업에 따른 스트레스 요인에 관한 연구 : 대전시 고등학생을 중심으로 focus on high students in Daejeon city

        This study, based on analyzing the factors of stresses caused by physical education class on the 7th Curriculum was intended to provide the data which needed to raise the rate of participation for physical education class and to propose a method of physical education which met to the students' demands. This study focused on only second-year students of six academic high schools(two boys' high school, two girls' high school and two coeducational high school) in Daejeon city. The confidence level of this data was examined by the modulus of Cronbach's alpha(α) and the characteristics of questionaries were examined through the standard deviation and the average. The results of this study are as follow; First, the students prefer to learn in order of soccer, shooting, tennis, badminton and archery. A soccer was the most popular event among the boy students and a shooting was the most popular event among the girl students. Second, the students felt stress strongly to the examination and the next was in the lack of confidence, and the students were under the least stress in the friction among friends. In the facilities and the condition of environment, the students were under so much stresses in the lack of sports equipments, and in the teachers. They were dissatisfied with the teachers' teaching methodologies. Third, in the factors of the stresses by gender, the girl students were under the more stress to the learning contents than the boy students. Fourth, in the difference classified by the school, the girl students of coeducational high school were under the most stresses and the next was in order of the girl students of girls' high school, the boy students of coeducational high school and the boy students of boys' high school in the learning contents. In the learning contents by Duncan's multi-comparison, there was no difference among the girl students. The girl students were under more stresses than the boy students, and the boy students of coeducational high school were under more stresses than the boy students of boys' high school. Fifth, in the factors of stresses classified by the satisfaction for physical education class, we divided two groups, one is group which satisfied for the class, the other is a group which unsatisfied for the class. In both groups, the students were under the most stress to the examination. In the group which satisfied for the class, the students were under a little stress in the friction among friends, in the learning contents and in the factors of unsatisfaction for the teacher. The students of the group which unsatisfied for the class were under so much stresses to the six questionaries, and the difference of 't' value was the highest in learning contents.

      • Use of Array Detector for Real-time Spectoscopic Ellipsometer

        고속분광 엘립소미터를 위해 photodiode array 검출기를 이용하였다. 이때 각 부품의 시간적 요구사항을 만족시키기 위하여 array 구동용 전자회로를 직접 설계 제작하였는데 512의 채널을 읽어 내는데 필요한 시간인 read-out time이 편광기의 회전 속도를 제한하였다. 하지만 광학 주기당 3-4회의 부분 적분법을 적용함으로써 한 쌍의 엘립소미터 스펙트럼을 측정하는데 20 m초 이내의 측정속도를 실현할 수 있었다. 각 채널로부터 일정한 반응값을 얻을 수 있었으며 연속측정에서도 평균정도에 따라 0.04에서 0.007도 정도의 정밀도를 얻었다. We employed photodiode array as a detection system for fast scanning spectroscopic ellipaometer. Controller for data acquisition was designed to accommodate all timing requirements. Pixel read-out time for 512 channels limited the rotational frequency of polarizer. However, when we employed partial-integration method to take 3 or 4 spectra per optical cycles, we could achieve the fastest data acquisition speed of less than 20 ms for a pair of ellipsometric spectra. We obtained quite uniform response from ll channels and the repetitive measurements showed 0.04-0.007 degree of precision depending on averaging.

        Infra-structure : 도시공간의 연속성을 위한 건축적 방법 제안 : 울산광역시 북구 효문역을 중심으로

        Contemporary urban's expansion is accomplished continuously and quickly. However, high completion urban infra such as road and railroad acts as an obstruction cause upon big space about quick change of contemporary urban's structure. The research is to define threshold's concept in the urban space, to analyze meaning and cause of urban space's interruption by urban infra, to construct plan in order to solve it. Recently the existing cities' developments have been partial except newtown plan, and progressed in several in South Korea. The fact is that an existence of high completion urban infra element is a main cause, so that a continuity of urban flow is absent, and it is hard to have an integrative and sequential progress in point of urban development. In addition, development speed around infra is lowed, ghettoization is given. In order to solve the problem, the urban infra pointed out as a main problem should be accommodated, urban context's continuity should be recovered through passive design method that inserts municipal structure system for macro urban development. Expected pursuit through the method is summarized like following four theme. First, the urban flow's continuity is granted. The linear urban infra passing over the urban center by urban's expansion acts as an obstruction cause, so the continuity of urban flow is recovered through design parameter's insertion on the boundary. Second the design method of transfer system is suggested through complexed transportation infra, and fortified position is acquired in order to connect transportation of inter-city later on. The target place is composed of spaces functioned as station buildings at past, it will be used as a terminal station of SLRT which would be established later. Because a large dwelling site and circulation complex have been progressed around the target place, the place will function as fortified position of transportation connecting cities as well as suggesting architecturalization of transfer system interlocking rail-light rail-bus. Third complexed function related with round program is conducted. The existing target place is used as farmland, but it will be changed on functional view totally by recent public land development and circulation complex plan. Complexed cultural commercial space related with expected program has been planed. Forth, formation of roadside's landscape in urban scale is incited. The target place is a field existing urban infra associated with a industrial road and rail as well as an entrance to Ulsan Bukgu, so visual architecture seen by driving road and railroad would be produced.

      • 지구성 및 파워선수에서의 안정시 혈청효소 분석 : An object on cyclists and weight lifters 사이클 및 역도선수를 대상으로

        This study was performed to evaluate the changes on serum enzymes in resting state by regular, prolonged aerobic and power exercise. Subjects were consisted of 21 male and 11 female cyclists and 16 male and 13 female weight lifters. They were highly traininged state. (1) Serum glucoses and triglycerides showed values within normal limits but the glucose levels of weight lifters were lower than cyclists. (2) Serum AST and ALT also showed within normal values and male athletes values were larger than female athletes but no statistical significance. (3) Total CPK levels showed apparent difference between two type of exercise, weight lifters had higher CPK levels than cyclists, especially male athletes had statistical significance. The isoenzyme CPK-MM also showed higher values and had statistical significance in male and female athletes. (4) Total LDH was higher in weight lifters than cyclists and isoenzyme LDH-4 and LDH-5 had significant difference. (※) From the results, we could be suggested serum total CPK and isoenzyme LDH-4, and LDH-5 showed changes of physical adaptation by prolonged power exercise in resting state and they would be used indicator of exercise ability.

      • Jump 트레이닝이 쥐(RAT)의 뼈 형태 및 강도에 미치는 영향

        Structural and mechanical adaptations of the femur and tibia to jump and run training were investigated in female Fischer 344 rats. Rats aged 4 week were trained for 8 weeks after 1 week of stabilization. In experiment A, the forced run-trained (speed : 30m/min, duration : lh/day)group was compared with the control group. In experiment B, voluntary run and jump-trained (height : 40㎝, 100 times/day) group were compared with the control group. The limb bones of the jump-trained group had greater cross-sectional areas and greater maximum load in a fracture test than the limb bones of the control group, but there was no significant difference in bone length between the jump-trained group and the controls. The bone adaptations to forced running and voluntary running were similar. The limb bones of both run groups were longer than those of each control group. The cross-sectional areas and the maximum load in the run-trained groups were greater than those in each control group but less than those in the jump-trained group. The present results indicate that bone adaptations to jump training and run training differ and that jump training is more effective for building stronger bones.

      • 이단평행봉 경기의 연기내용에 관한 분석 : 제82회 전국체육대회 여자고등부 결승경기를 중심으로 The 82th National sports high school girls' Final Game

        This thesis is to be helpful in promoting competitive power in two-line parellel bar competition of women's gymnastics through showing the right way of efficiently training players. For this purpose, the refrees' grade list and video-recorded tapes of women's high school players in the 82nd National 체육대회 were used in analysing difficulty, composition, performance, bonus points, penalty count factors and their details. 1. There were, in difficulty-level, A-grade 4.13, B-grade 4.0, C-grade 2.13, D-grade 3.0 and E-grade 0.25. 2. In performance perfection rate, Ms. Oh from Kyeonggi-do has 94.24%, Ms. Park from Seoul 81%, and Ms. Cheon from Jeonbuk 80.5%, which is in accordance with the ranking order. This simply means that perfection rate and the grade, to a great extent, have something to do with each other. In other hand, despite Ms. Jeon from Chungbuk and Ms. Park from Gangwon-do have 9.6 and 9.5 each as the SV grade, they happened to be far away from Top 3 rank due to the failure in decent and landing. This proves an importance of the repetitive training, which results in the maturity in performance. 3. It also proves the necessity of over D-level performance in landing skill that only 2 players out of 8 satisfied the special landing requirments. 4. As shown in the example that Ms. Jeon from Chungbuk earned at most 7.80 with perfection rate of 64% in spite of plus points of 0.7, and Ms. Park 6.925 with perfection rate of 48.5% despite plus points of 0.5, the failure risk in high-level performances is so high that trainers should encourage their players to acquire the proper skills necessary for such high-levels. I understand I can hardly generalize these results, simply because I limited my research materials to the 82nd National sports Nevetheless, The research results above definitely show us that the players should have the environments in which they are able to feel psychologically stable. And the stability, I believe, comes from the accumulated experiences of participating in the game. Furthermore, the scientific program to improve players' skills should be urgently created and adopted to their training.
