        Histone 에 관한 연구 ( Ⅶ ) 밤나무 ( Castania crenata S . et Z . ) 꽃가루의 Histone

        It is now generally accepted that there are five main histones in the somatic cells of most species examined. However, some changes in the molecular species of animals and plants during the evolution have been reported. Recently, DeLange et al. (1971) discovered that there is a striking conservation and divergence of sequence during evolution for some of the histones between calf and pea bud. In this respect, it will be worth studying the histones of plant pollens which are unique in some species or class. We have studied on histones of some plant pollens from chestnut, pumpkin and pine tree which are distinctly related families each other. In the present studies, we report the results obtained from chestnut pollens. 1. The yield of whole histone recovered was 8.59 ㎎ per 1 g of pollens. This is very large amount as compared to those of mammary tissues. 2. The yield of DNA was 8.13㎎ per 1 g of pollens. Consequently the DNA to histone ratio was 1:1.06. 3. The relative amounts of five fractions, i.e., H1, H2a, H2b, H3 and H4 were 17.23, 19.32%, 26.89%, 23.06 and 13.50, respectively. 4. The electrophoretic mobility of individual histone fractions gave almost similar patterns to those of corresponding fractions of calf thymus. 5. We found that histone H2b fraction of chestnut pollens contained detectable amounts of ε-N-monomethyllysine. No evidence for the presence of methylated lysine or other side-chain derivatives was reported on this histone fraction. 6. Comparison of amino acid compositions reveals marked quantitative differences between corresponding chestnut pollens and pea bud histone fractions.

        요추부 협착증 치료를 위한 극돌기 삽입술의 생체역학적 효과 분석

        Many types of interspinous distraction devices (IDDs) have been recently developed as an alternative surgical treatment to laminectomy and fusion with pedicle screws for the treatment of the lumbar spinal stenosis (LSS). They are intended to keep the lumbar spine in a slightly flexed posture to relieve pain caused by narrowing of the spinal canal and vertebral foramen. However, their biomechanical efficacies are not well known. In this study, we evaluated the kinematic behaviors and changes in intradiscal pressure (IDP) of the porcine lumbar spine implanted with IDD. For kinematics analysis, five porcine lumbar spines (L2-L6) were used and the IDD was inserted at L4-L5. Three markers (${\phi}{\le}0.8mm$) were attached on each vertebra to define a rigid body motion for stereophotogrammetric assessment of the spinal motion in 3-D. A moment of 7.5Nm in flexion-extension, lateral bending, and axial rotation were imparted with a compressive force of 700N. Then, IDD was implanted at L3-L4. IDPs were measured using pressure transducer under compression (700N) and additional extension moment (700N+7.5Nm). In kinematic behaviors, insertion of IDD resulted in statistically significant decrease 42.8% at the implanted level in extension. There were considerable changes in ROM at the adjacent levels, but statistically insignificant. In other motions, there were no significant changes in ROM as well regardless of levels. IDPs at the surgical level (L3-L4) under compression and extension moment decreased by 12.9% and 18.8% respectively after surgery (p<0.05). At the superiorly adjacent levels, IDPs increased by 19.4% and 12.9% under compression and extension, respectively (p<0.05). Corresponding changes at the inferiorly adjacent levels were 29.4% and 6.9%, but they were statistically insignificant (p>0.05). The magnitude of pressure changes due to IDD, both at the operated and adjacent levels, were far less than the previously reported values with conventional fusion techniques. Our experimental results demonstrated the IDDs can be very effective in limiting the extension motion that may cause narrowing of the spinal canal and vertebral foramens while maintaining kinematic behaviors and disc pressures at the adjacent levels.

      • KNN 규칙과 새로운 특징 가중치 알고리즘을 결합한 패턴 인식 시스템

        본 논문에서는 유전자 알고리즘을 이용한 새로운 적응적 특징 가중치 방식과 클래스별로 적용된 KNN(Nearest -Neighbor) 규칙을 이용한 새로운 패턴 인식 시스템을 제안한다. 패턴 인식 시스템의 성능을 향상시키기 위하여, 새로운 연산자를 갖는 유전자 알고리즘으로 가중치의 중간값을 결정함으로써 과잉 맞춤(overfitting)을 피하면서, 데이터의 분포에 따라 적절한 특징의 가중치를 찾는 새로운 특징 가중치 알고리즘을 제안한다. 또한, 제안하는 방법은 각각의 클래스를 가장 잘 표현하는 특징 공간들을 개별적으로 찾는다. KNN분류기는 클래스별로 찾은 특징 공간들을 이용하여 클래스에 따라 특징 공간을 변화시켜 미지 패턴의 클래스를 예측한다. 제안된 알고리즘은 Concordia대학의 handwritten numeral database에 적용시켜 그 성능을 확인하였다. This paper proposes a new pattern recognition system combining the new adaptive feature weighting based on the genetic algorithm and the modified KNN(K Nearest-Neighbor) rules. The new feature weighting proposed herein avoids the overfitting and finds the Proper feature weighting value by determining the middle value of weights using GA. New GA operators are introduced to obtain the high performance of the system. Moreover, a class dependent feature weighting strategy is employed. Whilst the classical methods use the same feature space for all classes, the Proposed method uses a different feature space for each class. The KNN rule is modified to estimate the class of test pattern using adaptive feature space. Experiments were performed with the unconstrained handwritten numeral database of Concordia University in Canada to show the performance of the proposed method.

        산풍단(散風丹)이 생쥐의 항경련(抗痙攣), 진정(鎭靜) 및 진통작용(鎭通作用)에 미치는 영향(影響)

        Sanpoongdan(SPD) has been known effective on infantile convulsive disorders in clinical field of oriental medicine. The purpose of this study was to investigate the anti-convulsive, sedation and analgesic effects of SPD in mouse. The anticonvulsive effect was evaluated In mice treated with pentylenetetrazol, stychnine, and picrotoxin. For the sedative effect, observations were made on the sleeping time induced by thiopental sodium and pentobarbital sodium following oral administration of SPD. Furthermore, reduction of spontaneous movements and ataxia using rota rod method were evaluated. Analgesic effects on the writhing syndrome induced by acetic acid and on hindlimb pain induced by pressure were also observed. The findings were as follows : 1. The solid extracts of SPD revealed no effect on convulsions induced by pentylenetetrazol, strychnine, and picrotoxin. 2. Thiopental sodium-induced sleeping time was prolonged by the administration of the solid extracts of SPD, but this result was devoid of statistical significance. 3. The oral administration of SPD enhanced the sleeping induced by pentobarbital sodium. 4. Spontaneous movements were significantly depressed following the oral administration of the solid extracts of SPD. 5. Ataxia was not shown in rota rod method following the oral administration of the solid extracts of SPD. 6. The solid extracts of SPD showed positive analgesic effects on the acetic acid-induced writhing syndrome. 7. The solid extracts of SPD raised the threshold of the hindlimb pressure pain, but the result was not statistically significant. From the results, it can be concluded that SPD has sedative and analgesic effects.

        지성보명단(至聖保命丹)의 항경련(抗痙攣) 작용에 관한 실험적 연구

        Objectives In order to investigate the anticonvulsive effects of Jisungbomuyngtan, the experiments were done in mice. Methods After solid extracts of Jisungbomuyngtan was orally administered to mice, four types of convulsion were induced in mice by ECT, strychnine, picrotoxin, caffein. Each of the time to convulsion and death was measured. Results 1. The solid extracts of Jisungbomuyngtan showed a remarkable effect in delaying the onset of convulsion induced by ECT 2. In case of convulsion induced by strychnine, the solid extracts of Jisungbomuyngtan revealed a slight, but not statistically significant effect. 3. Considerable prolongation of time to death was observed in convulsion induced by picrotoxin due to the anticonvulsive effect of Jisungbomuyngtan. 4. The anticonvulsive effect of Jisungbomuyngtan was not found in convulsion induced by caffein. Conclusions Jisungbomuyngtan effect remarkably in delaying the onset of convulsion induced by ECT. But slightly effect by strychnine. And Jisungbomuyngtan effect in prolonging time to death in convulsion induced by picrotoxin, but not found in convulsion induced by caffein.

        Histone 에 관한 연구 ( Ⅷ ) 호박 ( Cucurbita moschata Duch . ) 꽃가루의 Histone

        Although studies on the histones from various kinds of animal sources have been reported, information about pollens is not available yet. The present report describes the properties of histones of pumpkin pollens. The results obtained were as follows: 1. The yield of whole histone was 12.2 ㎎ per g of pollens. 2. The yield of DNA was 9.1㎎ per g of pollens. Consequently, the DNA to histone ratio was 1 : 1.34. 3. The relative amounts of five histone fractions, i.e., Hl, H2a, H26, H3 and H4 were 12. 46%, 15. 16%, 27, 38%, 21. 23% and 23, 77%, respectively. The data presented in this report show that there is considerable variation in the relative amounts of histone fractions as compared to those of calf thymus. 4. Amino acid composition of individual histone fractions showed that there are marked quantitative differences between pumpkin pollens and pea bud. 5. Usually the methylated derivatives of lysine have been reported in various histone fractions of animal and plant sources. However, we found none o the methylated lysine residues in all the histone fractions from pumpkin pollens. 6. The electrophoretic mobility of individual histone fractions gave similar patterns to those of corresponding fractions of chestnut pollens.

      • 신뢰도를 고려한 다목적 저수지의 월별 운영율

        본 연구에서는 방류량 결정에 있어 신뢰도 수준을 고려한 저수지 운영율을 개발할 수 있는 방법론을 제시하고 있다. 이와같은 저수지 운영율은 장기간의 유입량 자료와 모의 발생된 자료를 이요, 최적화 기법에 의한 운영결과를 위험도(신뢰도)분석 및 고나련 이자들과의 상관분석을 실시하여 도출하였다. 이와같은 방법론을 본댐과 조정지댐으로 구성되어 있는 충주 저수지 시스템의 운영에 적용하여 월별 운영율을 개발하였으며, 또한 개발된 운영율을 검증하기 위한 모의 운영을 실시하여 과거 운영 실적과 비교하였다. 비교 결과는 금번 개발된 운영율을 이요할 경우 기존 시스템의 기대 편익을 상당히 증가시킬수 있을뿐만 아니라 운영에 따른 신뢰도도 개선시킬 수 있는 것으로 나타났다. This paper presents a methodology of developing reservoir operating rules which can consider the reliability levels incorporated with the discharge policy. The operating rules were derived based on the regression and risk analysis of the optimally operated results by using the long term historical and generated reservoir inflows. The methodology was applied to the operation of the Chungju reservoir system which is consisted of two reservoirs and powerplants, and monthly operation rules were developed. Simulations were performed by using the developed operating rules can not only significantly improve the output from the existing system but also improve the reliability incorporated with the output.

        Histone 에 관한 연구 ( 3 ) 정상인의 요속의 histone

        The histones have been precipitated from concentrate of pooled urine of healthy male middle school students by adding ammonium sulfate to 60% saturation and further fractionated by ethanol and acetone precipitation. A total of 71.5 ㎎ of total histone was recovered from 24ℓ of urine. The resulting four major fractions have all been characterized by analysis of amino acid composition and by polyacrylamide gel disc electrophoresis. The results established that the striking consistency did exist through all the corresponding fractions between urine sample and thymus and liver of calf. This analytical evidence suggests that the histones from human urine and calf thymus and liver seem to be homogeneous protein. The relative amounts of four major fractions, i.e., f1, f2 a, f2 b, and f3 were 21.8%, 24.5%. 22.5% and 31.2%, respectively. The amounts of fractions f1 and f3 from urine were almost similar to those of corresponding fractions of calf thymus and liver, marked increase in the amount of f2 a and concomitant derease in the amount of f2 b from urine were observed. The mechanism of occurrence of histones in urine and the physiological significance of histone excretion in urine remain to be clarified.
