      • Histone에 관한 연구(V) Bakers' Yeast의 Histone

        저자들은 bakers' yeast(Saccharomyces cerevisiae)로 부터 핵을 분리 정제한 후 Johns 및 Johns and Phillips 등의 방법에의 하여 histone을 추출 분리하였다. 각 histone 분획들은 15% polyacrylamide disc gel 전기영동에의 하여 순도 및 mobility를 동물조직의 그것과 비교 검토하였다. 또 amino산을 정량하여 다른 동물에서 추출한 각 histone 분획과 비교 검토하였으며 다른 연구자들이 yeast에서 추출한 총 histone의 amino 산의 조성과도 비교 검토하여 다음과 같은 결과를 얻었다. 1. 총 histone의 수율은 yeast 1 g 당 3.257mg 이다. 2. DNA의 함량은 yeast 1 g 당 2.916 mg 으로 DNA: histone의 비는 1 : 1.117 이다. 3. Yeast로 부터 4개의 histone 분획 즉 f1(KAP), f2a(GRK+LAK), f2b(KAS) 및 f3(ARE) 등을 분리하였으며, 이들의 상대적 백분율은 46.6%. 1.4%. 49.4% 및 2.6%이다. 4. Lysine-rich 분획인 f1(KAP)과 slightly lysine-rich 분획인 f2b(KAS)의 백분율은 46.6% 및 49.4%로 정상 조직의 그 분획의 백분율에 비하여 약 2배가 되며, 따라서 yeast의 histone의 약 96%가 lysine-rich histone으로 되어있다. 5. 각 분획의 polyacrylamide disc gel 전기 영동상의 이동도는 다른 조직의 그것과 거의 같고, f2a 분획은 f2a1 및 f2a2 분획으로 되어있다. 그러므로 yeast의 histone 분획은 정상조직의 histone 분획과 같이 5개의 주분획으로 구성되어 있다. 6. 각 histone 분획의 amino산 조성은 정상 조직의 그것과 약간의 차이가 있으며, 염기성 amino산과 산성 amino산의 비는 큰 차이가 없다. 그러나 lysine과 arginine의 비는 f1 분획에서만 약 2배의 차이가 있다. 7. 저자들이 분리한 총 histone의 amino산 조성은 다른 연구자들이 yeast에서 분리한 그것과 큰 차이가 없었다. In an attempt to investigate histone fractions of yeast, nuclei was isolated from a yeast, Saccharomyces cerevisiae, by the procedure of Rozijn and Tonino, and histone fractions f1(KAP), f2a(GRK+LAK), f2b(KAS) and f3(ARE) were prepared from isolated nuclei by the procedure of Johns. The resulting four fractions have all been put on 15% polyacrylamide disc gel electrophoresis and chromatographed for analysis of amino acid composition. DNA was assayed by the method of Burton. The results of the experiments were summarized as follows: 1. The yield of whole histone was 3.257 mg per 1g of yeast. This amount was very similar to calf thymus and liver. 2. The yield of DNA was 2.916 mg per 1g of yeast. The DNA to histone ratio was 1 : 1.117. 3. The relative amounts of four fractions, i. e., f1(KAP), f2a(GRK+LAK), f2b (KAS), and f3(ARE) were 46.6%, 1.4%, 49.4% and 4.9%, respectively. 4. The amounts of fractions f1, f2a, f2b and f3 from yeast were remarkably different to those of corresponding fractions of other source, marked increase in the amount of f1 and f2b, and marked decrease in the amount of f2a and f3 from yeast were observed. 5. The electrophoretic pattern of histone fraction gave similar pattern to those of calf thymus and mouse liver, and fractions f1, f2b and f3 were electrophoretically homogeneous in 15% polyacrylamide disc gels at pH 4.0. 6. Amino acid analysis of the individual histone fractions showed that the over-all compositions were similar but not identical to those of corresponding fractions from calf thymus and other mammalian cells.

        Histone 에 관한 연구 ( Ⅸ ) Walker 256 carcinosarcoma 의 Histone 에 대하여

        In attempt to investigate histone fractions of tumor cells, nuclei was isolated from Walker 256 carcinosarcoma by the procedure of Pogo et al., Histone fractions Hl, H2a, H2b, H3 and H4 were prepared from isolated nuclei by the procedure of Johns. The five histone fractions found in most tissues were also present in the Walker 256 carcinosarc:oma histones. These histone fractions were characterized by amino acid analysis and by polyacrylamide disc gel electrophoresis. The results obtained were as follows: 1. The yield of whole histone was 3.75me per g of Walker 256 carcinosarcoma. 2. The yield of DNA was 3.35mg per g of Walker 256 carcinosarcorna. Consequently the DNA to histone ratio was 1 : 1.12. 3. The relative amounts of five fractions, i.e., Hl, H2a, H2b, H3 and H4 were 20.00%, 9.33 %, 26.67%, 33.33 % and 10.67%, respectively. 4. The electrophoretic mobility of individual histone fractions gave almost similar patterns to those of corresponding fractions of calf thymus. 5. Amino acid analysis of the individual histone fractions showed that the over-all com positions were similar but not identical to those of the corresponding fraction from calf thymus. 6. We found that histone H2b fraction of Walker 256 carcinosarcoma contained detectable amounts of ε-N-monomethyllysine and ε-N-dimethyllysine. No evidence for the presence of methylated lysine or other side-chain derivatives was reported on this histone fraction. We also found that histcne H1 fraction contained detectable amounts of 3-methylhistidine.

        저자들은 실험암세포인 Walker 256 carcinosarcoma로 부터 핵을 분리한 후 Johns 및 Phillips and Johns 방법에 의하여 histone분획 즉 H1, H2a, H2b, H3 및 H4 등 5개의 주분획을 분리하였다. 각 histone 분획들은 15% polyacrylamide disc gel 전기영동에 의하여 순도 및 mobility를 검토하였다. 또 각 분획의 아미노산을 정량하여 아미노산 및 methyl화된 아미노산을 분리 확인하였으며, 동물의 정상조직의 그것과 비교 검토하여 다음과 같은 결과를 얻었다. 1. 총 histone의 수율은 Walker 256 carcinosarcoma조직 1g당 3.75mg이다. 2.         Histone 에 관한 연구 ( Ⅷ ) 호박 ( Cucurbita moschata Duch . ) 꽃가루의 Histone

        Although studies on the histones from various kinds of animal sources have been reported, information about pollens is not available yet. The present report describes the properties of histones of pumpkin pollens. The results obtained were as follows: 1. The yield of whole histone was 12.2 ㎎ per g of pollens. 2. The yield of DNA was 9.1㎎ per g of pollens. Consequently, the DNA to histone ratio was 1 : 1.34. 3. The relative amounts of five histone fractions, i.e., Hl, H2a, H26, H3 and H4 were 12. 46%, 15. 16%, 27, 38%, 21. 23% and 23, 77%, respectively. The data presented in this report show that there is considerable variation in the relative amounts of histone fractions as compared to those of calf thymus. 4. Amino acid composition of individual histone fractions showed that there are marked quantitative differences between pumpkin pollens and pea bud. 5. Usually the methylated derivatives of lysine have been reported in various histone fractions of animal and plant sources. However, we found none o the methylated lysine residues in all the histone fractions from pumpkin pollens. 6. The electrophoretic mobility of individual histone fractions gave similar patterns to those of corresponding fractions of chestnut pollens.

        Studies on Histones ( Ⅴ ) : Histones from Yeast

        Histones from active dry yeast have been studied. The protein has been precipitated from nuclear fraction of yeast which has been cultured in suitable media, and histone fractions were further prepared according to the method of Johns. The resulting four major fractions have all been put on polyacrylamide disc gel electrophoresis and chromatographed for analysis of amino acid composition. The results obtained were as follows: 1. The yield of whole histone recovered was 651.5 ㎎ per 100 g of active dry yeast. 2. The relative amounts of four major fractions, i.e., KAP (f1). GRK+LAK(f2al+f2a2), KAS (f2b) and ARE (f3) were 46.6%, 1.4%, 49.4% and 2.6% respectively. A marked deceases in the amounts of fraction GRK+LAK and ARE and concomitant increases in the amounts of KAP and KAS were observed as compared with the corresponding histone fractions from mammalian liver and spleen. 3. Electrophoretic behavior on the polyacrylamide gel and amino acid composition were compared with corresponding fractions from calf thymus histones and discussed.
