Streptococcus lactis $KB_{21}$의 lactose constitutive mutant에 관한 연구

From S. lactis $KB_{21}$, stabilized strain by intergration of the lactose plasmid into the host chromosome, several lactose constitutive mutants were obtained by UV irradiation and spontaneous mutation. The rapid growth of the mutants in the medium containing lactobionate confirmed their constitutive nature. The mutants synthesized $phospho-{\beta}-galactosidase$ with an activity of 1.7 to 3.4 times that of the parent. Lactose plasmid로 host chromosome에 도입시킨 S. lactis $KB_{21}$으로부터 spontaneous mutant와 자외선 처리로 lactose constutive mutant를 분리하였다. lactobionate를 탄소원으로 하는 배지에서 parent는 거의 생육하지 못하는 반면 mutant는 빠를 생육을 보임으로써 lactose constitutive mutant임을 확인하였다. Mutant의 $phospho-{\beta}-galactosidase$ activity는 parent의 약 $1.7{\sim}3.4$배를 나타내었다.

According to an analysis on differences in aged women`s hairstyle behaviors and hair satisfaction, the following results have been obtained. First, aged women prefer going to a beauty shop to get beauty services. They liked a short hairstyle. When asked about current hairstyles, ``short cut`` was the most responded, followed by ``perm.`` In terms of use cycle, ``45 days`` was most common. When asked about who was influential in current hairstyles, most respondents said, ``my own decision.`` When asked what hair care products they use, ``just shampoo and hair rinse`` was mostly answered. Second, a significant difference was found in the location of hairstyle setting, preferred hairstyle, use cycle, current hairstyle and hair care depending on socio-demographic characteristics. Third, a significant difference was observed in hair satisfaction depending on how they obtain money to cover the cost of living, economic standards, current pocket money, location of hairstyle setting and hair care method. It would be a good idea for beauty shops to use the results of this study as basic data for customer management and customer satisfaction. In terms of beauty services, the results would be useful in providing systematic beauty services to aged women.
신생 쥐 간의 Progesterone $6{\alpha}-Steroid$ Hydroxylase에 대한 연구

신생 쥐 간의 subcellular fraction의 특징과 $6{\alpha}$-steroid hydroxylase의 subcellular localization및 특성을 살펴본 결과는 다음과 같다. 1000g 30초 침전 fraction은 총 DNA의 95%를 차지하므로써 crude nuclei fraction으로 나타났으며 또한 5'-nucleotidase의 specific activity가 높은 것은 파괴되지 않은 세포에 기인된 것으로 생각이 되며, 1450g 10분 침전 fraction은 5'-nucleotidase의 activity가 가장 높으므로 crude plasma membrane fraction으로 동정하였으나 mitochondria가 같이 침전되어 있었으며, 9000g 20분 침전 fraction은 succinate-cytochrome C reductase의 activity가 가장 높아 mitochondria fraction으로 하였고, 105,000g 60분 상등액은 LDH가 가장 높아 cytosol로 하였으며, 105,000g 60분 침전은 남은 부분으로 microsome fraction으로 추정하였다. 각 fraction에 기질인 $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one을 incubation시킨 후의 생성된 steroid를 보면 crude nuclei fraction에서는 pregnadiol($5{\alpha}$-pregnane-$3{\alpha}/{\beta}$, $20{\alpha}$-diol)의 형성으로 $20{\alpha}$-reduction과 $3{\alpha}$-$3{\beta}$ iso${\beta}$merization을 볼 수 있었으며 crude plasma membrane fraction에서는 $20{\alpha}$-reduction과 $6{\alpha}$-hydroxylation을 볼 수 있었으며 crude mitochondria와 cytosol에서는 $20{\alpha}$-reduction만을 crude microsome에서는 $16{\alpha}$-hydroxylation을 볼 수 있었으므로 $6{\alpha}$-hydroxylase는 crude plasma membrane에 존재함을 확인하였다. 한편 $6{\alpha}$-steroid hydroxylase에 존재함을 확인하였다. 한편 $6{\alpha}$-steroid hydroxylase의 최대 활성도는 pH 7에서 나타났으며 progesterone은 hydroxylation을 시키지 못한 반면 $3{\alpha}$-hydroxy-$5{\alpha}$-pregnan-20-one은 $6{\alpha}$-hydroxylation이 되었다. Five subcellular fractions were obtained by successive centrifugation from the liver of rats within 6 hours of life and characterized by comparing marker compound or marker enzyms. After incubating $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one with the each fraction, the steroids were analyzed by TLC, GLC and GC-MS. A $6{\alpha}$-hydroxylase which hydroxylizes the tetra-hydrogenated compound of progesterone, $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one, was localized in the crude plasma membrane fraction, but not in the microsome fraction. The maximum 6α-hydroxylation was observed at pH 7.0. While this 6α-steroid hydroxylase was not able to hydroxlyze the progesterone, the $3{\alpha}$-isomer was hydroxylized at the $6{\alpha}$-position.
Cross-flow filtration에 의한 Bifidobacterium longum의 고농도 배양

Bifidobacterium longum을 고농도 배양하기 위하여 최적 생육 조건을 조사하고 cross-flow filtration으로 생육 저해 대사산물인 lactic acid와 acetic acid를 제거하면서 균체 순환 배양을 사용하였다. 회분 배양에서는 탄소원으로 glucose를 50 g/l 농도로 사용하고 초기 pH 6.5인 경우 생육이 가장 높았다. B. longum을 cross-flow filtration으로 12시간 배양한 결과, 희석속도 $0.31h^{-1}$에서 최대 균체농도에 도달하여 16.4g/l의 건조균체량을 얻었으며 이로써 회분 배양시 보다 약 4배 높은 균체를 생산하였다. The conditions for production of high cell density of Bifidobacterium longum were investigated and the cross-flow filtration system was used to remove the inhibitory metabolites, lactic acid and acetic acid. The maximum cell growth was observed with glucose as carbon source at the concentration of 50 g/l at $37^{\circ}C$ with the initial pH 6.5. When B. longum was cultured in a cross-flow filtration system, the maximum cell growth was observed at a dilution rate(D) of $0.31\;h^{-1}$ and the dry cell weight was 16.4 g/l($3.5{\times}10^{10}\;cell/ml$), which was about four times higher than that obtained in the batch culture with pH control.

Bacteriocin-producing lactic acid bacteria, Lactococcus sp. HY 449, was isolated from dairy products. Using response surface methodology, the various concentrations of medium compo- nents (tryptone, glucose, yeast extract, tween 80, and initial pH) were tested to find the optimum conditions for maximum bacteriocin production and growth of Lactococcus sp. HY 449. Central composite design was used to control the concentrations of medium components in the experiment. Bacteriocin production and cell growth of Lactococcus sp. HY 449 were most affected by glucose and yeast extract. Estimated optimum growth conditions of Lactococcus sp. HY 449 were as follows; tryptone 1.08%, glucose 1.129%, yeast extract 0.674%, tween 80 0.11%, and initial pH 7.19. Also estimated optimum conditions for bacteriocin production were tryptone 0.937%, glucose 1.108%, yeast extract 0.163%, tween 80 0.09%, and initial pH 6.98.
김치 공장의 절임 폐수 및 배추 쓰레기를 이용한 효모 배양

김치 공장의 배추 철임 폐수 빛 배추 쓰레기를 이용한 효모 균체 생산 가능성을 조사하였다. 철임 폐수는 약 19/L의 환원당과 7-12%의 높은 NaCl을 함유하고 있으며 pH는 약 5에서 6사이로 내염성 효모가 생육 가능한 것으로 밝혀졌다. 4종의 내염성 효모의 생육을 조사한 결과 Candida guillieγmond ii A TCC 6260 이 가장 우수한 생육을 나타내었고, 배양 조건에 따른 절임 폐수에서의 생육 특성을 조사한 결과 C. guilliermondii는 25~35℃의 배양 온도,절임 폐수의 초기 pH가 pH 3~6 그리고, 염농도 9%까지는 생육에 영향받지 않았으며 질산엽이나 인산엽의 첨가는 불필요한 것으로 나타났다. 이 효모를 절임 폐수에서 배양할 경우는 약 85%의 BOD 감소 효과를 보였다. 한편,김치 공장에서 발생하는 배추 쓰레기의 자원화 방법으호 절임 폐수에 첨가한 결과 20% 첨가했을 경우 절임 폐수만 사용한 경우보다 약 5.5 배 가량 균체 생산이 증가하였다. The possibility of using waste brine and cabbage waste from kimchi industry as raw materials for the production of yeast cell mass was investigated. Among four strains of osmotolerant yeast, Candida gz강lliermondii A TCC 6260 showed the best growth in the waste brine containing about 1. 0 g/L of reducing sugar and 7% to 12% of NaCl. The growth of C. guilliermondii in waste brine was affected slightly between the temperature range of 250C to 350C and the initial pH of 3 to 6. The NaCl concentration up to 9% was not inhibitory to the growth of C. guilliermoηdii and the addition of 10mM of ammonium salts or 5 mM of potassium phosphate had no effect on the growth. The growth of yeast reduced BOD of the waste brine by 85% within 24 hours. When C. guilliermondii was cultured in waste brine added with cabbage juice extracted from waste cabbage, the cell mass was increased significantly.
