      • The enhanced IL-l8 production by UVB irradiation requires ROI and AP-1 signaling in human keratinocyte cell line (HaCaT)

        Based on our recent observation that enhanced IL-18 expression positively correlates with malignant skin tumors, such as SCC and melanoma, we examined the possible role of UVB, known to be associated with skin cancer development, in the enhancement of IL-18 production using primary human epidermal keratinocytes and human cell line HaCaT. After cells were exposed to UVB irradiation in vitro, IL-18 production was examined by Northern blot analysis and ELISA, and it was found that IL-18 production is enhanced by UVB irradiation in a dose- and time-dependent manner. In addition, we confirmed that it is functionally active form of IL-18 using the inhibitor of caspase-1. The effect of UVB irradiation was blocked by antioxidant, N-acetyl-ι-cysteine (NAC), which suggested the involvement of reactive oxygen intermediates (ROI) in the signal transduction of UVB irradiation-enhanced IL-18 synthesis. We also found that UVB irradiation increased AP-1 binding activity by using EMSA with AP-1-specific oligonucleotide. Furthermore, inhibitors of UVB-induced AP-1 activity, such as PD98059, blocked enhanced IL-18 production, indicating that AP-1 activation is required for UVB-induced IL-18 production. Taken together, our results suggest that UVB irradiation-enhanced IL-18 production is selectively mediated through the generation of ROI and the activation of AP-1.

        열공형과 비열공형 피질하 혈관성 치매에서 위험인자의 차이에 관한 비교 연구

        Backgrounds and Objectives: Vascular dementia is a group of dementing disoders arising from various stroke syndrome. Among these. subcortical ischemic vascular dementia (SIVD) is regarded as a relatively distinct clinical entity. However, MRI patterns of SIVD are not homogenous. In some patients, lacunes are dominant, and in others, subcortical white matter changes are. This study was designed to compare risk factor profiles between SIVD with and without multiple lacunes. Methods: We divided 47 subjects (22 males, mean age. 68 years) recruited from VADAPET (Multicenter Trial For Evaluation Of The Changes In the PET Images Of Subcortical Vascular Dementia Patient) study into two groups one with more than 5 lacunes in deep gray matter (lacune group) and the other with 5 or less(non-lacune group) Clinical characteristics and laboratory findings of two groups were compared. Results: Nineteen of 47 patients (40%) belonged to the lacune group. The lacune and non-lacune groups d d not differ in the following variables: age, hypertension, diabetes mellitus, hyperlipidemia heart disease, history of stroke or TIA, history of trauma or major surgery, family history of hypertension stroke, or dementia, age at diagnosis of dementia, body mass index, white blood cell count, ESR, CRP, fibrinogen, hemoglobin A1C, total cholesterol. LDL cholesterol creatinine, proteinuria, glucosuria, and microhematuria. However, male sex, smoking alcohol. hemoglobin, and HDL cholesterol were possibly associated more with lacune group SIVD than with non-lacune group (p<0 1) Multivariate analyses revealed that smoking, hemoglobin, and HDL cholesterol were independent predictors of SIVD with multiple lacunes Conclusion: Our study suggests that SIVD with multiple lacunes may be significantly different in smoking habits hemoglobin, and HDL cholesterol from SIVD without multiple lacunes.

        감자軟腐病(Erwinia carotovora var. atroseptica)에 依한 감자塊茎腐敗와 water potential에 關한 硏究

        감자 괴경에 타박상을 입혔거나 세균현탁액을 주입했을 때 낮은 water potential(-6.46 bar) 보다 높은 water potential (-6.06 bar)에서 더 쉽게 부패를 초래하였으며, 높은 water potential을 가진 괴경과 낮은 water potential을 가진 괴경 사이에 있어서의 부패의 진전에는 큰 차이를 발견할 수 있었으며, 현탁액의 농도의 종류에 따른 괴경의 연부병의 이병정도는 높은 water potential에서 높았으며, 세균 현탁액 농도의 은 높은 water potential에서 이며 낮은 water potential에 서는 이었으며, 높은 water potential과 낮은 water potential 에서는 작은 차이를 인정할 수 있었다. 이 시험에서 감자는 상처나 타박상이 나지 않도록 다루어야 하며 수확후 충분히 건조하여야 오랜 운반과 저장중에 연부병 발생을 크게 줄일 수 있다는 것을 나타낸다. Results obtained from the experiment conducted to find out e relationships between tuber decay, and water potential and bruising in or on the tubers, are summerized as follows , 1) When potato tubers were bruised or injected with bacterial inoculum, the tubers with high water potential rotted more easily than the tubers with low potential. A big difference in the development of decay between high and low water potential tubers was found. 2) In tubers injected with different levels of inoculum. high water potential tubers were more susceptible to soft rot than low water potential tubers. 3) of inoculum concentration was 8.5(log) at high water potential tubers and 9.8(log) at low water potential. A small difference between low and high water potential was detected. The results of this experiment show that potatoes should be handled carefully and must be dried after harvest to reduce decay development in shipment and storage.

      • The molecular mechanisms of vitamin C on cell cycle regulation in B16F10 murine melanoma

        <P>Vitamin C has inconsistent effects on malignant tumor cells, which vary from growth stimulation to apoptosis induction. It is well known that melanoma cells are more susceptible to vitamin C than any other tumor cells, but the precise mechanism remains to be elucidated. In the present study, the proliferation of B16F10 melanoma cells was suppressed by vitamin C, which induced growth arrest in a dose-dependent manner without cytotoxic effects. Therefore, we investigated the changes in cell cycle distribution of B16F10 melanoma cells by staining DNAs with propidium iodide (PI). The growth inhibition of B16F10 melanoma by vitamin C was associated with an arrest of cell cycle distribution at G1 stage. In addition, the levels of p53-p21<SUP>Waf1/Cip1</SUP> increased during G1 arrest, which were essential for vitamin C-induced cell cycle arrest. The increased p21<SUP>Waf1/Cip1</SUP> inhibited CDK2. Moreover, the activity of p53-p21<SUP>Waf1/Cip1</SUP> pathway was closely related with the activation of checkpoint kinase 2 (Chk2). Inhibitor of the PI3K-family, LY294002 and the ATM/ATR inhibitor, caffeine, blocked vitamin C-induced growth arrest in B16F10 melanoma cells. These results suggest that vitamin C might be a potent agent to inhibit proliferative activity of melanoma cells via the regulation of Chk2-p53-p21<SUP>Waf1/Cip1</SUP> pathway. J. Cell.       • Developmental change of GABAergic postsynaptic current in rat periaqueductal gray

        The present study was designed to examine developmental changes of GABAergic spontaneous miniature inhibitory postsynaptic currents (mIPSCs) in periaqueductal gray (PAG) neurons mechanically isolated from young (12-to 18-day) and adult (8-to 12-week) rats. While the frequency of mIPSCs was similar, the current amplitude in adult rats was significantly smaller than in young rats. In the study of mIPSC kinetics, all kinetic parameters except for the fast decay time in adult rats were smaller or shorter than in the case of young rats. The present study demonstrates that a decrease in the amplitude of GABAergic mIPSC during development may result from a decrease in the GABA contents of synaptic vesicles and from changes in the kinetics of postsynaptic GABA-activated Cl? channels.

      • Development and Present Status of the Seed Potato Program in the Republic of Korea

        한국의 감자 종서 생산체계는 30년 이상의 역사를 지니고 있으나 1985년까지의 일반적인 발전과정이 Horton등(1988)에 의하여 단 한번 보고되었을 뿐이다. 따라서 본 논문에서는 1986년 이후의 변화를 포함한 한국의 종서 생산체계의 발전과정과 현황을 증식체계의 변화와 바이러스병의 발생을 중심으로 보고하고자 한다. 한국의 종서 생산체계는 1961년에 원원종과 보급종을 처음으로 생산한 이래 증식 및 검정체계가 몇차례 수정되어 현재까지 유지되고 있다. 그러나 1960년대와 1970년대에는 주로 종서의 공급량 증대에 중점을 두었을 뿐 바이러스병의 방제에는 소홀히 하였다. 결과적으로 1970년대 후반에는 종서의 바이러스 이병율이 증가하여 1979년에는 주요 재배작형에 필요한 보급종을 생산하지 못한 바 있다. 그후 조직배양 및 망실재배를 이용한 상위급 종서의 생산으로 1980년대 후반까지는 양질의 종서가 공급되었다. 그러나 1980년대 후반부터 종서의 엽권바이러스 이병율이 증가하여 1988년에는 주요 재배품종의 원종 생산에 실패한 바 있다. 이와같은 문제점을 극복하기 위하여 1989년부터 원원종 생산 단계까지 망실재배를 확대이용하고 있으며 엽권바이러스에 저항성인 신품종의 보급과 기내소피경을 이용한 종서 생산체계의 확립이 추진되고 있다. 더불어 1980년대 후반의 엽권바이러스 이병율 증가의 원인에 대하여 논하였다. The present paper reports the development and present status of the Seed Potato Program in the Republic of Korea with focuses on the change in multiplication system and the occurrence of virus diseases. The Seed Program began by producing the first Foundation and Certified Seed in 1961. During the 1960s and 1970s, the Seed Program primarily emphasized producing adequate amounts of seed potatoes and made relatively simple effort for virus control. As a result, the seed stocks became severely infected with viruses, and the Seed Program failed to produce Certified Seed for major cropping systems in 1979. Since the failure, tissue culture techniques and nethouse cultivation systems were introduced for the production of upper-grade seed stocks, effecting the production of Certified Seed in acceptable qualities during the 1980s. During the second half of the 1980s, however, virus infection rates of the seed stocks increased again, and the Seed Program failed to produce the Registered Seed of the major variety due to a high rate of potato leafroll virus (RLRV) occurrence in 1988. To overcome the PLRV problem, the Seed Program extended the use of nethouses even for the production of Foundation Seed from 1989. In addition, the Seed Program is going to practice utilization of PLRV-resistant varieties and establishment of seed potato production systems using microtubers produced in vitro. Possible reasons for the PLRV increase during the late 1980s are also discussed.

        CM1 Ligation Induces Apoptosis via Fas-FasL Interaction in Ramos Cells, but via Down-regulation of Bcl-2 and Subsequent Decrease of Mitochondrial Membrane Potential in Raji Cells

        Background: CM1 (Centrocyte/-blast Marker I) defined by a mAb developed against concanavalin-A activated PBMC, is expressed specifically on a subpopulation of centroblasts and centrocytes of human germinal center (GC) B cells. Burkitt lymphoma (BL) is a tumor consisting of tumor cells with the characteristics of GC B cell. Previously we reported that CM1 ligation with anti-CM1 mAb induced apoptosis in Ramos $(IgM^{high})$ and Raji $(IgM^{low})$ cells. Methods & Results: In the present study, we observed that CM1 ligation with anti-CM1 mAb induced Fas ligand and Fas expression in Ramos cells, but not in Raji cells. Furthermore, anti-Fas blocking antibody, ZB4, blocked CM1-mediated apoptosis effectively in Ramos cells, but not in Raji cells. Increased mitochondrial membrane permeabilization, which was measured by $DiOC_6$, was observed only in Raji cells. In contrast to no significant change of Bax known as pro-apoptotic protein, anti-apoptotic protein Bcl-2 was significantly decreased in Raji cells. In addition, we observed that CM1 ligation increased release of mitochondrial cytochrome c and upregulated caspase-9 activity in Raji cells. Conclusion: These results suggest that apoptosis induced by CM1-ligation is mediated by Fas-Fas ligand interaction in Ramos cells, whereas apoptosis is mediated by down-regulation of Bcl-2 and subsequent decrease of mitochondrial membrane potential in Raji cells.

      • Electroacupuncture Enhancement of Natural Killer Cell Activity Suppressed by Anterior Hypothalamic Lesions in Rats

        Background/Objective: Neuroendocrine hormones are derived from the hypothalamus. The central nervous system, particularly the hypothalamus, is capable of modulating the cytolytic activity of adherent natural killer (NK) cells. In addition, electroacupuncture (EA) stimulation of the Zusanli (ST36) acupoint enhances splenic NK cell and cytokine activities in rats. However, it is still unclear whether the anterior hypothalamus affects this immunomodulation. Therefore, the aim of the present study was to examine the effect of EA stimulation at the Zusanli acupoint on the NK cell activity modulated by an anterior hypothalamic area lesion. Methods: Male Sprague-Dawley rats were used. Lesions were placed by means of a direct current through a concentric electrode. The electric acupuncture stimulation was delivered for 30 min per each experiment at the right ST36 acupoint with an electrical stimulator. The NK cell activity of the spleen was measured by a fluorescence assay. Results: The NK cell activity was significantly reduced on the 2nd day after the lesion, but was restored to that of the sham group by the 7th day. However, when EA was applied for 2 days after the operation, the NK cell activity of the lesion group was restored to that of the sham group. After 7 days of EA, the NK cell activity of the lesion group was slightly higher than that of the sham group. Conclusion: From these results, we can suggest that EA enhances or restores the NK cell activity suppressed by an anterior hypothalamic area lesion.
