        An outbreak of highly pathogenic H5N1 avian influenza in Korea, 2008

        In spite of intensive surveillance programs for the control of HPAI, an outbreak of highly pathogenic avian influenza (HPAI) H5N1 in Korea in April 2008 caused serious damage to poultry farms, as did previous outbreaks in 2003/2004 and 2006/2007. Six viruses were selected from the Korean 2008 isolates for genetic analysis, and all eight gene segments from each of the influenza viruses were sequenced. A phylogenetic analysis showed that all of the viruses were of the same virus type and that the hemagglutinin (HA) gene was clustered with that of clade 2.3.2 viruses. However, the internal and neuraminidase (NA) genes were closely related to those of the clade 2.3.4 viruses (recent human and bird isolates from Southeast Asia).

        <P>SETTING: National Masan Tuberculosis Hospital, Masan, South Korea. OBJECTIVE: To evaluate the pharmacokinetics of prothionamide (PTH) in South Korean patients with multidrug-resistant tuberculosis (MDR-TB) and to investigate whether differences in body mass index (BMI) could explain observed differences in PTH disposition. DESIGN: Seventeen patients participated in the study; all had MDR-TB and had received combination anti-tuberculosis treatment, including PTH, cycloserine, ofloxacin, para-aminosalicylic acid and streptomycin or kanamycin, for at least 2 weeks. The patients were divided into two groups based on BMI: Group A (18.5 < or = BMI<23), and Group B (BMI<18.5). Serum samples were collected over 24 h, and the plasma PTH concentration was determined by a validated high-performance liquid chromatography assay. RESULTS: After steady-state administration of PTH, the mean area under the plasma concentration-time curve from time 0 to 12 h (AUC(0-12h)) was 11.0 +/- 3.7 microg h/ml. The mean T(max) and t(1/2) were respectively 3.6 h and 2.7 h. No significant difference in PTH disposition was observed between groups A and B, except for ke and t(1/2). CONCLUSION: In the pharmacokinetic parameter estimates for PTH in MDR-TB patients during routine treatment, the pharmacokinetics of PTH did not appear to correlate with extent of emaciation in MDR-TB patients.</P>

        Cordyceptin induces apoptosis through repressing hTERT expression and inducing extranuclear export of hTERT

        Cordycepin is an adenosine analog originally extracted from Cordyceps militaris that possesses many pharmacological effects including immune activation and antioxidant and antitumor effects. However, the underlying relationship between apoptosis and telomerase activity in response to cordycepin exposure has not been investigated. In this study, we found that cordycepin-induced apoptosis of human leukemia cells (H937 and THP-1 cells) was associated with inactivation of telomerase and downregulation of human telomerase reverse transcriptase (hTERT) as well as the transcription factors c-Myc and Sp1, which are required for basal transcription from the hTERT gene promoter. Cordycepin also attenuated the activation of phosphoinositide-3-kinase (PI3K)/Akt signaling, thereby reducing phosphorylation and nuclear translocation of hTERT. We further showed that the PI3K inhibitor LY29004 significantly decreased telomerase activity in cordycepin-treated cells and increased cordycepin-induced cell death. These findings demonstrate that cordycepin is cytotoxic to human leukemia cells and suppresses telomerase activity through transcriptional and post-translational suppression of hTERT by inactivating the PI3K/Akt signaling pathway.

        Association of Sequence Variations in DGAT 1 Gene with Economic Traits in Hanwoo (Korea Cattle)

        The effects of diacylglycerol O-acyltransferase (DGAT1) candidate gene polymorphism on the economic traits of Hanwoo were studied. Through sequencing analysis, two polymorphism sites at K232A and T11993C were established and were analyzed by PCR-RFLP. The PCR-RFLP analysis for K232A showed that the frequencies of alleles K and A were 0.75 and 0.25, respectively, and the frequencies of genotypes for K/K, K/A and A/A were estimated as 0.509, 0.491 and 0, respectively. In the PCR-RFLP analysis for T11993C, we found allele frequencies of 0.773 and 0.227 for T and A, respectively, and 0.546, 0.454 and 0 for the T/T, T/C and C/C genotype frequencies, respectively. No significant effects on economic traits in Hanwoo were found in the separate analysis of K232A and T11993C polymorphisms, but the interaction between K232A and T11993C showed a significant effect (p<0.005) on marbling score. The DGAT1 candidate gene was found to have a significant effect not only on milk yield and component traits but also on the metabolism of intramuscular fat.

        Overview of KSTAR research progress and future plans toward ITER and K-DEMO

        <P>A decade-long operation of the Korean Superconducting Tokamak Advanced Research (KSTAR) has contributed significantly to the operation of superconducting tokamak devices and the advancement of tokamak physics which will be beneficial for the ITER and K-DEMO programs. Even with limited heating capability, various conventional as well as new operating regimes have been explored and have achieved improved performance. As examples, a long pulse high-confinement mode operation with and without an edge-localized mode (ELM) crash was well over 70 and 30 s, respectively. The unique capabilities of KSTAR allowed it to improve the capability of controlling harmful instabilities, and they have been instrumental in uncovering much new physics. The highlights are that the L/H transition threshold power is sensitive to the resonant magnetic perturbation (RMP) and insensitive to non-resonant magnetic perturbation. Co-<I>I</I> <SUB>p</SUB> offset rotation dominated by an electron channel predicted by general neoclassical toroidal viscosity theory was confirmed. Improved heat dispersal in a divertor system using three rows of rotating RMP was demonstrated and predictive control of the ELM-crash with <I>a priori</I> modeling was successfully tested. In magnetohydrodynamic physics, validation of the full reconnection model (i.e. <I>q</I> <SUB>0</SUB>  >  1 right after the sawtooth crash) and self-consistent validation of the anisotropic distribution of turbulence amplitude and flow in the presence of the 2/1 island with theoretical models were achieved. The turbulence amplitude induced by RMP was linearly increased with the slow RMP coil current ramp-up time (i.e. the magnetic diffusion time scale). The <I>D</I> <SUB> <I>α</I> </SUB> spikes (i.e. ELM-crash amplitude) was linearly decreased with the turbulence amplitude and not correlated with the perpendicular electron flow. In the turbulence area, a non-diffusive ‘avalanche’ transport event and the role of a quiescent coherent mode in confinement were studied. To accommodate the anticipation of a higher performance of the KSTAR plasmas with the increased heating powers, a new divertor/internal interface with a full active cooling system will be implemented after a full test of the new heating (neutral beam injection II and electron cyclotron heating) and current drive (CD) (Helicon and lower hybrid CD) systems. An upgrade plan for the internal hardware, heating systems and efficient CD system may allow for a long pulse operation of higher performance plasmas at <I>β</I> <SUB>N</SUB>  >  3.0 with <I>f</I> <SUB>bs</SUB> ~ 0.5 and <I>T</I> <SUB>i</SUB>  >  10 keV.</P>

      • Telomere length, TERT and shelterin complex proteins in hepatocellular carcinomas expressing ''stemness''-related markers

        Background & Aims: Hepatocellular carcinomas (HCCs) expressing ''stemness''-related markers have been associated with aggressive biological behavior and poor prognosis. We examined the relationship between ''stemness''-related protein expression and telomere length, hTERT and shelterin complex protein expression and chromosomal instability. Methods: Quantitative fluorescent in situ hybridization for telomere length, immunohistochemistry for K19, EpCAM, CD133, c-kit, HepPar1, hTERT, TRF1, TRF2, POT1, RAP1 and TPP1, and TUNEL assay were performed in 137 HCCs, and array comparative genomic hybridization was performed with 24 HCCs. Results: Telomeres were significantly longer in HCCs expressing ''stemness''-related proteins (K19: p<0.001, EpCAM: p=0.002, CD133: p=0.002). On analyzing different tumor cells within EpCAM-expressing HCCs, EpCAM-positive tumor cells showed longer telomeres (1.329+/-0.246) compared to EpCAM-negative tumor cells (0.996+/-0.381) within the same HCCs (p=0.031). Telomeres were significantly longer in HCCs expressing hTERT (p=0.048) and RAP1 proteins (p=0.031). K19-expressing HCCs expressed hTERT (p=0.002), TRF2 (p=0.001) and TPP1 (p=0.013) more frequently compared to K19-negative HCCs. EpCAM-positivity was associated with more frequent hTERT (p=0.028), TPP1 (p=0.017), TRF2 (p=0.027) and POT1 (p=0.004) expression. Copy number alterations were more frequent in K19 and EpCAM-expressing HCCs compared to HCCs without these markers (K19: p=0.038, EpCAM: p=0.009). HCCs with longer telomeres were associated with a shorter overall (p=0.019) and disease-free survivals (p=0.049), and decreased disease-free survivals were seen in TRF2-positive HCCs (p=0.018). Conclusions: HCCs expressing ''stemness''-related proteins are characterized by increased telomere length, increased expression of hTERT and shelterin complex proteins, and increased chromosomal instability compared to conventional HCCs. Longer telomeres and TRF2 expression in HCCs are associated with poor patient outcomes.

        Neural stem cells injured by oxidative stress can be rejuvenated by GV1001, a novel peptide, through scavenging free radicals and enhancing survival signals

        <P>Oxidative stress is a well-known pathogenic mechanism of a diverse array of neurological diseases, and thus, numerous studies have attempted to identify antioxidants that prevent neuronal cell death. GV1001 is a 16-amino-acid peptide derived from human telomerase reverse transcriptase (hTERT). Considering that hTERT has a strong antioxidant effect, whether GV1001 also has an antioxidant effect is a question of interest. In the present study, we aimed to investigate the effects of GV1001 against oxidative stress in neural stem cells (NSCs). Primary culture NSCs were treated with different concentrations of GV1001 and/or hydrogen peroxide (H2O2) for various time durations. The H2O2 decreased the viability of the NSCs in a concentration-dependent manner, with 200 mu M H2O2 significantly decreasing both proliferation and migration. However, treatment with GV1001 rescued the viability, proliferation and migration of H2O2 injured NSCs. Consistently, free radical levels were increased in rat NSCs treated with H2O2, while co-treatment with GV1001 significantly reduced these levels, especially the intracellular levels. In addition, GV1001 restored the expression of survival-related proteins and reduced the expression of death associated ones in NSCs treated with H2O2. In conclusion, GV1001 has antioxidant and neuroprotective effects in NSCs following treatment with H2O2, which appear to be mediated by scavenging free radicals, increasing survival signals and decreasing death signals.       • 1987년 한국에서 발생한 렙토스피라병의 혈청역학적 조사

        Human leptospirosis was an unfamiliar disease in Korea until 1984 that outbreak of leptospirosis occurred among farmers and soldiers after field works for harvesting rice. During that time, Lee and Jo confirmed the first Korean cases of leptospirosis by serological test, isolation of causative agent and autopy findings. Afterward several outbreaks occurred also during autumn especially after flood in every years and some characterisitcs of leptospirosis in Korea such as clinical manifestations, serotypes and seroepidemiological features has been revealed by many investigators. Because of the major mode of transmission between rodents and human is by direct contact with leptospiral urine of rodents or contaminated soil by the urine, leptospirosis in Korea has been primarily a disease of person in occupations heavily exposed to contaminated soil or infected urine such as farmer, army and etc. Therefore it seems that leptospirosis is one of the main communicable diseases to be controlled urgently in Korea, for an agricultural people account for almost half of total Korean people. For clarifying the seroepidemiological patterns of human leptospirosis in Korea by sex, month region and main reacting serovars of L. interrogans among acute febrile disease occurred in 1987, 1,773 patient's sers with acute febrile episodes were tested by microagglutination test using 19 representative strains of leptospiral serogroup as antigen. All of those sera were collected from 10 collaborative clinics located in Kyunggi, Kangwon, Chungbuk, Chungnam, Chonbuk, Chonnam province and Seoul. The results wee summerized as follows. 1) Among 1,773 sera of patients with acute febrile episodes, 219 (12.4%) were seropositive to L. interrogans, 487(27.5%) to R. tsutsugamushi, 241(13.6%) to R.typhi and 160(90.0%) to Hantaan virus. 2) Among seropositives to L.interrogans, the male outnumbered the female, 65% and 35%. 3) For age distribution, 26.9% of seropositives to L.interrogans were fifties, 19.6% were forties, 9.1% were sixties, 5.9% were thirties and 4.1% were twenties. 4) Eighty three percent of seropositives had occurred between September and October in 1987 with a peak in September. 5) Main leptospiral serovars reactive to patient's sera were Icterohaemorrhagiae(54.3%), Canicola(31.0%), CH-48(13.2%), Tarassovi(0.9%)and Cynopteri(0.5%). 6) For regional distribution, 65.8% of seropositives to L.interrogans were residents from Chonbuk, 12.3% were Chonnam, 7.3% were Chungnam, 5.5% were Kyunggi and 1.4% were Kangwon.
