        Indoor feeding combined with restricted grazing time improves body health, slaughter performance, and meat quality in Huang-huai sheep

        Objective: The aim of this study was to evaluate the effects of three feeding systems, i.e., indoor feeding (CON), indoor feeding with 4-h daily access to grazing artificial pasture (ITGP), and indoor feeding with 8-h daily access to grazing artificial pasture (IEGP), on the plasma antioxidant and immunological capacity, slaughter characteristics, meat quality and economic efficiency of Huang-huai lambs. Methods: Thirty-three healthy Huang-huai rams with similar body weight (approximately 5 mo of age, 28.96±1.01 kg) were assigned equally to three experimental groups. When finished fattening, six lambs from each group were collect blood samples for plasma analyses and then slaughtered to determine slaughter characteristics and obtain biceps brachii muscle for further analysis of meat quality and fatty acid profile. Results: Compared to CON group, animals submitted to ITGP and IEGP groups resulted in greater contents of serum glutathione peroxidase, immunoglobulins (IgA, IgG, and IgM), polyunsaturated fatty acids (PUFA), n-6 PUFA, and PUFA/saturated fatty acid (FA) ratio and lower palmitic /oleic acid ratio (p<0.05). Moreover, animals in ITGP group exhibited a higher (p<0.05) loin eye area, content of meat crude protein (CP), and eicosetrienoic acid compared to CON group, while slaughter performance was superior (p<0.05) to that of the IEGP group. The economic efficiency of ITGP group was 70.12% higher than that of CON group, while the IEGP group exhibited a decrease of 92.54% in economic efficiency compared to the CON group. Conclusion: Restricted grazing time combined with indoor feeding was more effective in conferring superior body health, carcass traits and economic efficiency in Huang-huai lambs, as well as higher CP content and healthier FA composition in the resulting meat.

        Biochemical Characterization of a GDSL-Motif Esterase from Bacillus sp. K91 with a New Putative Catalytic Mechanism

        The esterase gene Est8 from the thermophilic bacterium Bacillus sp. K91 was cloned and expressed in Escherichia coli. The monomeric enzyme exhibited a theoretical molecular mass of 24.5 kDa and an optimal activity around 50°C at pH 9.0. A model of Est8 was constructed using a hypothetical YxiM precursor structure (2O14_A) from Bacillus subtilis as template. The structure showed an α/β-hydrolase fold and indicated the presence of a typical catalytic triad consisting of Ser-11, Asp-182, and His-185, which were investigated by site directed replacements coupled with kinetic characterization. Asp-182 and His-185 residues were more critical than the Ser-11 residue in the catalytic activity of Est8. A comparison of the amino acid sequence showed that Est8 could be grouped into the GDSL family and further classified as an SGNH hydrolase. Est8 is a new member of the SGNH hydrolase subfamily and may employ a different catalytic mechanism.

      • A study on the Influencing Factors to China-Pakistan Trade under BRI Initiative

        The Belt and Road Initiative is a grand plan proposed by China to promote the common development and prosperity of China and its partner countries. With the construction of the China-Pakistan Economic Corridor, the flagship of the Belt and Road Initiative, China-Pakistan relations are getting closer, known as the “iron brother”. Economic cooperation is a key priority for both sides. Under the context of the Belt and Road Initiative, this study uses trade data of China and Pakistan to find out the bilateral trade problems and the influencing factors behind them. This study will discuss the above questions and propose some suggestions for policymakers. Specifically, the discussion of the problems was conducted in terms of GDP, population, comparative advantages, international environment, domestic environment, and other factors respectively. In the conclusion part, the author suggests that the two countries should try to improve their domestic/international environment and reduce policy barriers to promote the trade. Modern industrial parks are encouraged to solve the problems. This study on the influencing factors and economic trade relations between China and Pakistan hopes to provide some academic references to the study of the CPEC and provide some guidance to the policy makers.

        Production and Characterization of Ethanol- and Protease-Tolerant and Xylooligosaccharides-Producing Endoxylanase from Humicola sp Ly01

        This paper reports the production and characterization of crude xylanase from the newly isolated Humicola sp. Ly01. The highest (41.8 U/ml) production of the crude xylanase was obtained under the optimized conditions (w/v): 0.5% wheat bran, 0.2% KH2PO4, and 0.5% peptone; initial pH 7.0; incubation time 72 h; 30℃; and 150 rpm. A considerable amount of the crude xylanase was induced using hulless barley bran or soybean meal as the carbon source, but a small amount of the enzyme was produced when supplementary urea was used as the nitrogen source to wheat bran. The crude xylanase showed apparent optimal cellulase-free xylanase activity at 60℃ and pH 6.0, more than 71.8% of the maximum xylanase activity in 3.0-30.0% (v/v) ethanol and more than 82.3% of the initial xylanase activity after incubation in 3.0-30.0% (v/v) ethanol at 30℃ for 2 h. The crude xylanase was moderately resistant to both acid and neutral protease digestion, and released 7.9 and 10.9 μmol/ml reducing sugar from xylan in the simulated gastric and intestinal fluids, respectively. The xylooligosaccharides were the main products of the hydrolysis of xylan by the crude xylanase. These properties suggested the potential of the crude enzyme for being applied in the animal feed industry, xylooligosaccharides production, and high-alcohol conditions such as ethanol production and brewing.

        Breviscapine Ameliorates Fear Extinction and Anxiety in BALB/cJ Mice

        Objective In this research, the influence of breviscapine on anxiety, fear elimination, and aggression and the potential mechanism was investigated. Methods Anxiety and locomotion were analyzed by elevated plus maze and open field test in mice. Bussey-Saksida Mouse Touch Screen Chambers were used to perform fear conditioning. Territorial aggression was assessed by resident intruder test. Protein levels were evaluated by Western blot. Breviscapine improved fear-extinction learning in BALB/cJ mice. Results Breviscapine at 20-100 mg/kg increased center cross number, total distance traveled, and velocity in a dose-dependent manner. On the other hand, breviscapine at 20-100 mg/kg decreased the immobility time in open field test. In addition, breviscapine at 20-100 mg/kg increased the ratio of time on the open arm, time on the distal parts of the open arm, and total distance traveled in elevated plus maze. Breviscapine at 100 mg/kg increased the average attack latency and decreased the number of attacks over the last 3 days of resident intruder test. In hippocampus, protein levels of postsynaptic density protein-95 and synaptophysin were elevated by breviscapine at these three doses. Conclusion The administration of breviscapine alleviates fear extinction, anxiety, and aggression, while increases locomotor in a dose-dependent manner, which might be associated with its influence on synaptic function.

        Atomic layer deposited ZrxAl1-xOy film as high κ gate insulator for high performance ZnSnO thin film transistor

        In this work, the high κ Zr x Al 1−x O y fi lms with a diff erent Zr concentration have been deposited by atomic layer deposition,and the eff ect of Zr concentrations on the structure, chemical composition, surface morphology and dielectric propertiesof Zr x Al 1−x O y fi lms is analyzed by Atomic force microscopy, X-ray diff raction, X-ray photoelectron spectroscopy andcapacitance-frequency measurement. The eff ect of Zr concentrations of Zr x Al 1-x O y gate insulator on the electrical propertyand stability under negative bias illumination stress (NBIS) or temperature stress (TS) of ZnSnO (ZTO) TFTs is fi rstlyinvestigated. Under NBIS and TS, the much better stability of ZTO TFTs with Zr x Al 1−x O y fi lm as a gate insulator is due tothe suppression of oxygen vacancy in ZTO channel layer and the decreased trap states originating from the Zr atom permeationat the ZTO/Zr x Al 1−x O y interface. It provides a new strategy to fabricate the low consumption and high stability ZTOTFTs for application.

        Kernel Fisher Discriminant Analysis for Natural Gait Cycle Based Gait Recognition

        This paper studies a novel approach to natural gait cycles based gait recognition via kernel Fisher discriminant analysis (KFDA), which can effectively calculate the features from gait sequences and accelerate the recognition process. The proposed approach firstly extracts the gait silhouettes through moving object detection and segmentation from each gait videos. Secondly, gait energy images (GEIs) are calculated for each gait videos, and used as gait features. Thirdly, KFDA method is used to refine the extracted gait features, and low-dimensional feature vectors for each gait videos can be got. The last is the nearest neighbor classifier is applied to classify. The proposed method is evaluated on the CASIA and USF gait databases, and the results show that our proposed algorithm can get better recognition effect than other existing algorithms.

      • Development of Automatic Measuring System for Fish Length on Board

        In order to meet requirement from International Organizations, each far sea vessel needs report fish length of its catch. Currently, the length is manually measured and recorded which is laborious, time consuming, and tends to manual errors. Therefore, argues occur occasionally between vessel managers and government agents. It needs new approach to improve. This study developed an automatic measuring and recording system for fish length which consists of object detecting unit, length measuring unit, guiding slider (chain type or pneumatic type), PLC (programmable logic controller), and PC (personal computer). The system detected and measured fish length, once the carried fish enter the detecting zoon on deck. Fish length was measured by sensing the signal of object existence by a sensor on the slider. The lasting time of detected object (fish) and the slider speed were multiplied to obtain fish length. This operation was processed by a computer program in Visual Basic. The length data can also be saved in a file along with date, time, and its catch position (longitude and latitude) when worked with GPS (global positioning system). The system was tested in room with four boxes of length of 37, 63, 90, and 126 cm. The accuracy in length was about 95 to 97%, according to test results. Factors influencing on the accuracy included response time of the sensor, moving speed of the slider, and the distance between sensor and object. Real fish (yellow fin tuna) samples were also tested in room and in a CT7 vessel. The length accuracy was about 90 to 95% when a modified coefficient was adopted in the computer program. Detection part of fish (such as fish back or fish abdomen), the height of sensor position, luminance of detection environment were the factors affecting the accuracy. Test result also showed the automatic system completed one reading in 3 second which reduced the time in more than 50% compared to the manual operation. Meanwhile, the system also can avoid the manual record error and work with electronic monitoring system (EMS) by sending catch data thru satellite in real time from far sea to control center on land so as to offer better quality of length data for fishery and nature resource management.

        Understanding nanorheology and surface forces of confined thin films

        Understanding the nanorheology and associated intermolecular/surface forces of fluids in confined geometries or porous media is of both fundamental and practical importance, providing significant insights into various applications such as lubrication and micro/nanoelectromechanical systems. In this work, we briefly reviewed the fundamentals of nanoreheolgy, advances in experimental techniques and theoretical simulation methods, as well as important progress in the nanorheology of confined thin films. The advent of advanced experimental techniques such as surface forces apparatus (SFA), X-ray surface forces apparatus (XSFA) and atomic force microscope (AFM) and computational methods such as molecular dynamics simulations provides powerful tools to study a wide range of rheological phenomena at molecular level and nano scale. One of the most challenging issues unresolved is to elucidate the relationship between the rheological properties and structural evolution of the confined fluid films and particles suspensions. Some of the emerging research areas in the nanorheology field include, but are not limited to, the development of more advanced characterization techniques, design of multifunctional rheological fluids, bio-related nanorheology, and polymer brushes.
