Effect of Light with Different Wavelengths on Nostoc flagelliforme Cells in Liquid Culture

( Yu Jie Dai ),( Jing Li ),( Shu Mei Wei ),( Nan Chen ),( Yu Peng Xiao ),( Zhi Lei Tan ),( Shi Ru Jia ),( Nan Nan Yuan ),( Ning Tan ),( Yi Jie Song ) 한국미생물 · 생명공학회 2013 Journal of microbiology and biotechnology Vol.23 No.4

The effects of lights with different wavelengths on the growth and the yield of extracellular polysaccharides of Nostoc flagelliforme cells were investigated in a liquid cultivation. N. flagelliforme cells were cultured for 16 days in 500 ml conical flasks containing BG11 culture medium under 27 μmol·m-2·s-1 of light intensity and 25oC on a rotary shaker (140 rpm). The chlorophyll a, phycocyanin, allophycocyanin, and phycoerythrin contents in N. flagelliforme cells under the lights of different wavelengths were also measured. It was found that the cell biomass and the yield of polysaccharide changed with different wavelengths of light. The biomass and the yield of extracellular polysaccharides under the red or violet light were higher than those under other light colors. Chlorophyll a, phycocyanin, and allophycocyanin are the main pigments in N. flagelliforme cells. The results showed that N. flagelliforme, like other cyanobacteria, has the ability of adjusting the contents and relative ratio of its pigments with the light quality. As a conclusion, N. flagelliforme cells favor red and violet lights and perform the complementary chromatic adaptation ability to acclimate to the changes of the light quality in the environment.

Study on the Purification of Polysaccharides from Noscoc flagelliforme with Radial Flow Chromatography

        Yu-Jie Dai,Jing-Wen Wang,Shi-Ru Jia,Si-Jun Yue,Meng-Yao Jia,Peng Xu 한국생물공학회 2009 Biotechnology and Bioprocess Engineering Vol.14 No.3

The isolation and purification of polysaccharide from Noscoc flagelliforme by radial flow chromatography were studied. The column (7.7 cm of bed length and 229.6 cm³ of bed volume) was packed with DEAE-01 anion ion-exchange resin and gradient eluted with NaCl solutions. The content of the polysaccharide was determined with the phenol-sulfuric acid method. The effects of sampling weight, elution velocity, and elution concentration gradient on the separation efficiency were examined and three isolated peaks were obtained. The optimal separation conditions are 10 mg of the sampling weight (sampling volume is 20 mL), 1.0 mL/min of the elution velocity, and 1.00 mol/L² of NaCl gradient elution. The adjacent peak resolutions among the three main components (1, 2, and 3 according to their elution order) are 0.660 (o12) and 0.786 (o23), respectively. It is deduced that 39.8 cm of the bed length is required for the fully separation of the three polysaccharides The isolation and purification of polysaccharide from Noscoc flagelliforme by radial flow chromatography were studied. Facile Self-Assembly of Fe3O4 Nanoparticles@WS2 Nanosheets: A Promising Candidate for Supercapacitor Electrode

        Yu Dai,Xiao Wu,Dawei Sha,Ming Chen,Han Zou,Jie Ren,Jingjing Wang,Xuehua Yan 대한금속·재료학회 2016 ELECTRONIC MATERIALS LETTERS Vol.12 No.6

Graphene-like dichalcogenides with huge surface area and nanostructuredtransition metal oxides with extraordinarily high theoretical capacities could becomposited as promising electrode candidates for supercapacitors. In this work, monolayer and few-layers WS2 nanosheets were exfoliated by combination ofball-milling and sonication. A facile strategy for the hierarchical self-assembly ofFe3O4 nanoparticles (Fe3O4NPs) on WS2 nanosheets was developed to synthesizeFe3O4NPs@WS2 nanocomposites via hydrothermal method. Fe3O4NPs areuniformly dispersed on the WS2 nanosheets without aggregation. The particlesize of Fe3O4NPs is about 3 nm. The nanocomposite shows strong enhancementsof electrochemical behaviors. This self-assembly synthesis strategy may havegreat prospects for other 0D/2D nanocomposites in supercapacitors and otherenergy devices.

GM-CSF augmented the photothermal immunotherapeutic outcome of self-driving gold nanoparticles against colorectal cancer

        Jie Dai,Jianmei Li,Yuqin Zhang,Qian Wen,Yun Lu,Yu Fan,Fancai Zeng,Zhiyong Qian,Yan Zhang,Shaozhi Fu 한국생체재료학회 2023 생체재료학회지 Vol.27 No.00

Background Hypoxia is a frequent characteristic observed in solid tumors and is strongly associated with tumor metastasis, angiogenesis, and drug resistance. While the vasculature of hypoxic tumor tissues poses obstacles to the efficient administration of conventional drugs, it may prove advantageous in sustaining hyperthermia. Photothermal therapy (PTT) offers a promising treatment strategy that utilizes the activation of photosensitizers to produce heat, thus facilitating the selective ablation of tumor tissues. Method To enhance the accumulation of photothermal agents in tumor tissue and improve the effectiveness of PTT, we developed a self-propelled hybrid called Bif@PAu-NPs. This hybrid consists of polydopamine (PDA)-coated gold nanoparticles (Au-NPs) loaded onto the anaerobic Bifidobacterium infantis (Bif ). Results The Bif@PAu-NPs actively aggregated at the tumor site because the ability of Bif can target hypoxic regions, and PAu-NPs achieved precise PTT due to their high photothermal conversion efficiency (η = 67.8%). The tumor tissues were ablated by PTT, resulting in the release of antigens through immunogenic cell death (ICD), which stimulates an immune response. The inclusion of GM-CSF enhanced the immune response by recruiting dendritic cells and initiating long-term anti-tumor immunity. Conclusion The Bif@PAu-NPs hybrid effectively suppressed the growth of both primary tumors and re-challenged tumors. The utilization Bif@PAu-NPs in conjunction with GM-SCF exhibits great potential as a photothermal-immunotherapeutic strategy for precisely treating solid tumors.

      • Changes in buccal facial depth of female patients after extraction and nonextraction orthodontic treatments

        Fanfan Dai,Jie Yu,Gui Chen,Tianmin Xu,Ruoping Jiang 대한치과교정학회 2018 대한치과교정학회지 Vol.48 No.3

Objective: This study was performed to investigate buccal facial depth (BFD) changes after extraction and nonextraction orthodontic treatments in postadolescent and adult female patients, and to explore possible influencing factors. Methods: Twelve and nine female patients were enrolled in the extraction and nonextraction groups, respectively. Changes in BFD in the defined buccal region and six transverse and two coronal measuring planes were measured after registering pretreatment and posttreatment three-dimensional facial scans. Changes in posterior dentoalveolar arch widths were also measured. Treatment duration, changes in body mass index (BMI), and cephalometric variables were compared between the groups. Results: BFD in the buccal region decreased by approximately 1.45 mm in the extraction group, but no significant change was observed in the nonextraction group. In the extraction group, the decrease in BFD was identical between the two coronal measuring planes, whereas this differed among the six transverse measuring planes. Posterior dentoalveolar arch widths decreased in the extraction group, whereas these increased at the second premolar level in the nonextraction group. The treatment duration of the extraction group was twice that of the nonextraction group. No differences were found in BMI and Frankfort horizontal-mandibular plane angle changes between the groups. BFD changes in the buccal region moderately correlated with treatment duration and dental arch width change. Conclusions: BFD decreased in adult female patients undergoing extraction, and this may be influenced by the long treatment duration and constriction of dentoalveolar arch width. However, nonextraction treatment did not significantly alter BFD.

Three-dimensional changes in lip vermilion morphology of adult female patients after extraction and non-extraction orthodontic treatment

        Zhi-Yu Liu,Jie Yu,Fan-Fan Dai,Ruo-Ping Jiang,Tian-Min Xu 대한치과교정학회 2019 대한치과교정학회지 Vol.49 No.4

Objective: To investigate the three-dimensional lip vermilion changes after extraction and non-extraction orthodontic treatment in female adult patients and explore the correlation between lip vermilion changes and incisor changes. Methods: Forty-seven young female adult patients were enrolled in this study (skeletal Class III patients were excluded), including 34 lip-protruding patients treated by extraction of four first premolars (18 patients requiring mini-implants for maximum anchorage control and 16 patients without mini-implants) and 13 patients requiring non-extraction treatment. Nine angles, seven distances, and the surface area of the lip vermilion were measured by using pre- and posttreatment three-dimensional facial scans. Linear and angular measurements of incisors were performed on lateral cephalograms. Results: There were no significant changes in the vermilion measurements in the non-extraction group. The vermilion angle, vermilion height, central bow angle, height/width ratio, and vermilion surface area decreased significantly after the orthodontic treatment in the extraction groups, but the upper/lower vermilion proportion remained unchanged. Significant correlations were found between the changes in incisor position and those in vermilion angles, vermilion height, and surface area. Conclusions: Extraction of the four first premolars probably produced an aesthetic improvement in lip vermilion morphology. However, the upper/ lower vermilion proportion remained unchanged. The variations in the vermilion were closely related to incisor changes, especially the upper incisor inclination changes.

Isoindolin-1-ones from the stems of Nicotiana tabacum and their antiviral activities

        Guang-Yu Yang,Jia-Meng Dai,Zhen-Jie Li,Jin Wang,Feng-Xian Yang,Xin Liu,Jing Li,Qian Gao,Xue-Mei Li,Yin-Ke Li,Wei-Guang Wang,Min Zhou,Qiu-Fen Hu 대한약학회 2022 Archives of Pharmacal Research Vol.45 No.8

In previous studies, several isoindolin-1-oneanalogs that exhibited signifi cant anti-tobacco mosaic virus(anti-TMV) activities were isolated from Nicotiana tabacum . Since gene-editing mutants provide a new sample for thediscovery of active metabolites, we focused on the stems ofYN-18–23 (a mutant N. tabacum for gene editing with thealkaloid metabolic pathway cultivated by Yunnan TobaccoCompany), which led to the isolation of four new ( 1–4 )and four known ( 5–8 ) isoindolin-1-ones. To the best of ourknowledge, nicindole C ( 3 ) is the fi rst subclass of isoindolin-1-one bearing a pentacyclic ketone, while nicindole D ( 4 )is the fi rst example of isoindolin-1-one bearing a methylpyridin-2-(1 H )-one moiety. Compounds 1–4 were testedfor their anti-TMV activities, and the results revealed thatcompounds 1 , 3 , and 4 exhibited high anti-TMV activities atconcentrations of 20 μM with inhibition rates of 48.6, 42.8,and 71.5%, respectively. These rates are higher than the inhibitionrate of the positive control (33.2%). The mechanisticstudy of compound 4 , which had the highest anti-TMV activityrevealed that increased potentiation of defense-related enzyme activities and downregulation of expression of theNtHsp70 protein may induce resistance in tobacco againstthe viral pathogen TMV. Molecular docking studies alsorevealed that the isoindolin-1-one substructure is fundamentalfor anti-TMV activity. The methyl-pyridin-2-(1 H )-onemoiety in compound 4 and the 2-oxopropyl groups in compounds1 and 3 at the N -2 position may increase inhibitoryactivities. This study of the structure–activity relationshipis helpful for fi nding new anti-TMV activity inhibitors. Tostudy whether the isoindolin-1-ones have broader antiviralactivities, compounds 1–4 were also tested for their antirotavirusactivities. Compound 4 exhibited high anti-rotavirusactivity with a therapeutic index (TI) value of 20.7. This TI value is close to that of the positive control (20.2).

      • Combining ICA and SVR in Times Series Predication

        Wensheng Dai,Jui-Yu Wu,Chi-Jie Lu 인하대학교 정석물류통상연구원 2009 인하대학교 정석물류통상연구원 학술대회 Vol.2009 No.10

In this paper, a time series prediction approach by combing independent component analysis (ICA) and support vector regression (SVR)is proposed ICA is a novel statistical signal processing technique that was originally proposed to find the latent source signals from observed mixture signal without knowing any prior knowledge of the mixing mechanism. SVR is and artificial intelligence forecasting technique and has been widely applied in time series prediction problems. The proposed approach First uers ICA to the forecasting variables for generating the independent components(ICs). After identifying and removing the ICs containing the noise, the rest of the ICs are then used to reconstruct the forecasting variables which contain less noise. The SVR then uses the denoised forecasting variables to build the forecausting model. in order to evaluaate the performance of the proposed approach the TAIEX(Taiwan Stock Exchange Capitalization weighted Steock index) closing cash index is usde as the illusrtative example. Experimental results show that the proposed model outperforms the SVR model with mon ?filtered forecasting variables and random walk model

Optical Properties of Zinc-oxide Films Determined Using Spectroscopic Ellipsometry with Various Dispersion Models

        Zhong-Hong Dai,Jie Shao,Yi-Ming Chen,Yu-Xiang Zheng,Jia-Da Wu,Liang-Yao Chen,Rong-Jun Zhang 한국물리학회 2009 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.55 No.3

In this work, we have studied the optical properties of wurtizite zinc-oxide films grown on silicon (100) substrates by means of pulsed laser deposition (PLD). Spectroscopic ellipsometry and three dispersion models, namely, the Sellmeier, Cauchy, and Forouhi-Bloomer models, were applied for determining the optical constants of the ZnO thin films. A comparison was made between two samples that were deposited for 30 minutes (sample I) and 60 minutes (sample II), respectively. X-ray diffraction analysis indicates that there are two types of preferred-orientation, i.e., (101) and (100) orientations for sample I and II, respectively. Results show that the Cauchy model gives the best fit for the samples with least root mean square error (RMSE) whereas the Forouhi-Bloomer model is most suitable for the data analysis in both the transparent and the absorption regions. The optical properties extracted from different dispersion models have been compared with the data reported in the literature. The results given in this work show that different dispersion models should be applied to obtain the optical constants In this work, we have studied the optical properties of wurtizite zinc-oxide films grown on silicon (100) substrates by means of pulsed laser deposition (PLD).
