        연구논문 : Lactobacillus acidophilus을 이용한 홍경천과 홍삼 혼합 발효물의 이화학적 특성 및 항산화 활성

        The study was conducted to investigate the condition for mixed fermentation of Rhodilola sachalinensis with red ginseng using Lactobacillus acidophillus 128 and the changes of physicochemical properties and antioxidant activities before and after the lactic acid fermentation was examined. In the single fermentation of Rhodiola sachalinensis extract, the pH and titratable acidity rarely changed, and the number of lactic acid bacteria decreased greatly. On the other hand, in the lactic acid fermentation of Rhodiola sachalinensis-red ginseng mixed extract of 50% red ginseng content, the pH decreased, whereas the titratable acidity and the number of lactic acid bacteria increased. The solid content of optimal mixed extract for lactic acid fermentation was 0.5%. Sugar content decreased during fermentation, but total phenolic compounds tended to increase during fermentation. The salidroside and p-tyrosol content of the initial Rhodiola sachalinensis-red ginseng mixed extract was 419.5 ㎎% and 60.1 ㎎%, respectively; after fermentation, the salidroside content after lactic acid fermentation decreased greatly to 81.8 ㎎%, and the amount of p-tyrosol increased greatly to 324.9 ㎎%. The DPPH scavenging activity of Rhodiola sachalinensis-red ginseng mixed fermentate was 78.1% at 0.1% concentration, showing a tendency to increase as compared to 50.3% of Rhodiola sachalinensis-red ginseng mixed extract before the fermentation (p<0.05); it was a higher antioxidant activity as compared to the single fermentation of Rhodiola sachalinensis or red ginseng.

        추출 공정 다변화를 통한 홍경천의 항암활성 증진효과

        This study was performed to compare effect of anticancer activities on Rhodiola sachalinensis by ultrasonifi-cation process and solvent. Compared the yield to the water extracts (WE), wafer extracts with ultrasonification (WEU) at60, loot and ethyl alcohol extracts (EE), ethyl alcohol extracts with ultrasonification (EEU) at 60 ˚C from Rhodiola sachalinensis. Experimental studies were progressed through the anticancer activities such as cell cytotoxicity, inhibition activities of cell growth. The cell cytotoxicity using human embryonic kidney cell (HEK293) was showed cytotoxicity of below 26.26%by extracts of Rhodiola sachalinensis in 1.0 mg/ml concentration, The anticancer activities were increased in over 70% by extracts of Rhodiola sachalinensis in A549, AGS, Hep3B and MCF-7 cells. From the results, the extract Rhodiola sachalinensis were showed useful anticancer activities.

        흡착 공정을 활용한 홍경천(Rhodiola sachalinensis) 유산균 발효물의 이화학적 특성 및 항산화 활성

        Rhodiola sachalinensis fermentates by lactic acid bacteria were prepared using the adsorption process, and were investigated for changes of the main compounds and anti-oxidative activities during the adsorption and fermentation process. While the R. sachalinensis extract (RSE), which did not go through the adsorption process, showed little change in pH during fermentation and a significant reduction in the number of lactic acid bacteria, the pre-preparatory adsorption process was found to be helpful for promoting fermentation and for maintenance of bacterial numbers. The contents of total phenolic compounds mostly decreased during the adsorption process, but showed an increasing tendency to rebound during the fermentation process. The contents of salidroside and p-tyrosol in the RSE were 1153.3 mg% and 185.0 mg% respectively, and they did not significantly change after treatment with acid clay or bentonite as adsorbents, which were 1093.0 and 190.5 mg% by acid clay, and 882.2 and 157.3 mg% by bentonite. When the extract was fermented after treatment with acid clay or bentonite, the salidroside contents were decreased by 282.7 and 505.0 mg% respectively, but the p-tyrosol contents were increased by 714.0 and 522.4 mg% respectively. Compared to the DPPH radical scavenging activity of the RSE (66.8%) at the conc. of 0.1%, that of the fermented RSE, which went through adsorption process with acid clay or bentonite, was significantly increased to 79.4 and 72.7% respectively at the same concentration (p<0.05). Though fermentation by lactic acid bacteria was suppressed in the RSE, the results suggested that the adsorption process may promote fermentation without any change in the content of major active compounds. It is expected that fermentation by lactic acid bacteria could improve the antioxidant activity and various associated functionalities of R. sachalinensis.

        참돌꽃 근경의 항산화작용

        Ihe purpose of this study was to determine the antioxidative effects of Rhodiola sachalinensis. Rhodiola methanol extract was fractionated sequentially with dichloromethane and butanol. Each Rhodiola fraction (water, MeOH, BuOH and CH2Cl2 fractions) showed the potent superoxide dismutase (SOD) activity, and had inhibitory effects on peroxide value of linoleic acid (40-57%) and lipid peroxidation (47-70%). In Fe2+/ascorbate system-induced rat liver microsome. Rhodiola methanol extract also recovered carbon tetrachloride-induced decrease in SOD by 42% and catalase activities by 50%, and had inhibitory effects (54%) on carbon tetrachloride-induced lipid peroxidation in rat liver microsome. These results suggest that Rhodiola sachalinensis has the antioxidative effects.

        백두산 자생 참돌꽃의 생육과 이용 - 1. 배양세포 유도와 유리아미노산 조성 -

        본 실험은 백두산 자생 참돌꽃의 발아조건과 종자, 유식물 및 뿌리유래 캘러스의 유리 아미노산 조성과 함량을 조사하여 다음과 같은 결과를 얻었다. 뿌리로부터 캘러스 유도는 2,4-D 및 NAA가 첨가된 MS배지에서 유도되었다. 캘러스 형성율은 2,4-D 2.0 mg L^(-1)에 kinetin 1.0mg L^(-1) 혼용 처리구가 NAA 2.0 mg L^(-1)에 kinetin 1.0 mg L^(-1) 혼용 처리구보다 캘러스 형성율이 높았다. 유리 아미노산은 종자, 줄기, 뿌리 그리고 캘러스에서 aspartic acid, glutamic acid, serine 및 g1ycine 등의 25~26종이 검출되었으며 종자, 줄기 그리고 뿌리부위에서는 lysine이 가장 많은 양을 나타냈으며 캘러스에서는 histidine이 높은 양을 나타냈다. 전체적인 유리아미노산 분포는 lysine, histidine 및 arginine의 함량이 높아 참돌꽃 유식물 및 캘러스의 경우 염기성 아미노산의 분포가 높은 식물임을 알 수 있었다. 또한 총 유리 아미노산 함량은 종자와 뿌리, 줄기에 비해 캘러스에서 신선중 g당 500~1,000μmole 정도 높아 우수한 약리작용을 지니고 있는 참돌꽃의 성분을 배양세포를 통하여 대량생산이 가능함을 시사하고 있다. This study was carried out to investigate the callus formation and to deter-mine the composition and contents of the free amino acids in seedlings and callus of Rhodiola sachalinensis. The callus formation from the part of seedling-roots was the most effective on MS (Murashige and Skoog) media supplemented with 2.0 mg L^(-1)2, 4-D included in 1.0 mg L^(-1) kinetin than the other experimental plots which prepared with the different concentration of the various growth regulators. Free amino acids extracted from the explants and the callus were a total of 25~26 kinds. Especially, the basic amino acids such as arginine, lysine and histidine released in callus were found relatively in a large quantity. These results suggest that the cultured callus of Rhodiola sachalinensis could induce for the mass production of the other useful ingredients.

        고산홍경천 에탄올 추출물의 항산화작용과 쥐의 혈압 및 골격근 수축성에 미치는 영향

        To study the effects of ethanol extract of the Rhodiola Sachalinensis on the anti-oxidation and blood pressure and skeletal muscles contractility in rats. The ethanol extracts and fractions of Rhodiola Sachalinensis were determinated the anti-oxidation effects by DPPH (1,1-dipenyl- 2-picrylhydrazyl) method and comparing with the BHA (Butylated hydroxyanisole) and AA (Ascorbic acid). The gastrocnemius and soleus muscle contractility were observed by stmulating the sciatic nerve with electricity after affusing stomach with ethanol extract of the 200 mg/kg dosage of Rhodiola Sachalinensis for 4 weeks. The ethanol extract of the Rhodiola Sachalinensis could shorten latent period, increase maximal contractive extent and time and relaxative time at the twitch and complete tetanus of gastrocnemius and soleus muscles. The ethanol extract of the Rhodiola Sachalinensis shows high anti-oxidation effect and can enhance skeletal muscles contractility in rats.

        홍경천의 생육특성검정 및 재배지역 탐색

        중국의 약용작물로 널리 알려진 홍경천의 충북도내 재배 가능성을 알아보기 위하여 수행 한 시험 결과를 요약하면 다음과 같다. 지상부 생육상황은 최고조에 달했던 7월의 경우 초장은 8.1­15.8cm, 줄기수는 3.1­3.6개였으며, 여름철엔 하고현상이 심하여 지상부의 고사율이 76­94%로 매우 높았다. 지하부 생육상황의 경우 근장은 단양지역이 6.8cm로 가장 컸으나 굵기는 청원이 10.0mm로 가장 굵었으며 , 생체중 또한 3.5g으로 제천, 단양 대비 3.5배 정도 높았다. 한편 뿌리 부패율이 62.3­86.5%로 매우 높아 당분간 농가재배는 어려울 것으로 생각된다. 개화기는 6월 중·하순으로 개화기간은 12­13일 정도 되었고, 개화율은 60­77%정도 되었으며, 복산형화서로 꽃색은 자색, 황녹색, 회자색, 자홍색 등 다양하였다. 종자는 미세종자로 성숙기는 8월 중순­9월 하순이었고, 색깔은 황갈색이었으며 종자의 길이는 1.5­2.0mm, 너 비 는 0 1­0.5mm, 천립중은 0.13­0.15g으로 조사되었다. This study was conducted to search for possible cultivation area of Rhodiola sachalinensis as well as to obtain the basic information of growth characteristics. One­year old nursery plants of Rhodiola sachalinensis were collected from Yonbyeon Hongkyungcheon development society in China This expriment was evaluted in three areas according to sea level such as plain region in Cheongwon(50m at sea level), mid­mountainous region in Jecheon(350m at sea level) mountainous region in Banyans(650m at sea level). The growth status Rhodiola sachalinensis showed wide range of variation in there areas. Plant height showed from 8.1cm to 15.8cm, number of branch showed from 3.1 eachs to 4.6 eachs and rate of withering showed from 76% to 94%, respectively. It was increased rapidly at the early August caused by summer depression. Flowering period of Rhodiola sachalinensis was shown at around middle and late June, total duration of flowering showed from 12 to 13 days and flower colors were various as brown, dark purple, yellow green and brown red color.
