Zinc finger (ZNF) proteins play a critical role in cell growth, proliferation, apoptosis, and intracellular signal transduction. In this paper, we cloned and characterized a novel human KRAB-related zinc finger gene, ZNF425, which encodes a protein of 752 amino acids. ZNF425 is strongly expressed in the three month old human embryos and then is almost undetectable in six month old embryos and in adult tissues. An EGFP-ZNF425 fusion protein can be found in both the nucleus and the cytoplasm. ZNF425 appears to act as a transcription repressor. Over-expression of ZNF425 inhibits the transcriptional activities of SRE, AP-1, and SRF. Deletion analysis indicates that the C2H2 domain is the main region responsible for the repression. Our results suggest that the ZNF425 gene is a new transcriptional inhibitor that functions in the MAPK signaling pathway. [BMB reports 2011; 44(1): 58-63]
Three-dimensional features of MHD flows turning in a right-angle duct

This study presents a numerical simulation of three-dimensional (3D) Liquid metal (LM) Magnetohydrodynamic (MHD) flows turning in a right-angle duct with a square cross section under a uniform magnetic field applied perpendicular to the plane of the main flow. The 3D features of the flow in a fluid region adjacent to the duct walls of the turning segment are investigated. Cases with different Hartmann numbers and conductance parameters are analyzed using the CFX code. The MHD features of the LM flow are examined in terms of fluid velocity, current density, electric potential, and pressure gradient. The formation of a velocity recirculation is observed in the inner region of the right-angle segment immediately after the turning of the flow because of the inertial force therein, thereby yielding a region of low electric potential with a complicated current distribution. In particular, in the right-angle segment, the axial velocity in the side layer near the outer wall (that is, in the outer side layer) is relatively lower than that in the inflow and outflow channels. In addition, the velocity recirculation region in the right-angle segment decreases and the pressure gradient increases with an increase in conductance parameter.
ZNF424, a novel human KRAB/C2H2 zinc finger protein, suppresses NFAT and p21 pathway

Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK- 293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions. [BMB reports 2010; 43(3): 212-218]

Background: The year 2022 marks the 30th anniversary of heart transplant service in Hong Kong (HK). In this study, we describe prevailing trends and outcomes of advanced heart failure (AHF), including heart transplantations (HTx), in HK over the past 30 years. Methods: Trends in heart failure prevalence in HK from 1993 to 2021 were analyzed based on data from the Hospital Authority Clinical Data and Reporting System. All AHF patients referred for HTx consideration between 1992 and 2021 were reviewed. The bridge-to-transplant (BTT) utilization of short-term mechanical circulatory support (ST-MCS) devices, including venoarterial extracorporeal membrane oxygenation (VA-ECMO) and durable left ventricular assist devices (LVADs), from 2010 to 2021 was reviewed. Results: Overall, 237 heart transplants were performed in HK, with 10-year posttransplant and median survival of 68.1% and 18.7 years, respectively. An increase in AHF clinic referrals was correlated with increasing heart failure prevalence (R2=0.635, P<0.001). In total, 146 referrals were made for ST-MCS, and an observed increase in ST-MCS referrals was correlated with increasing VA-ECMO utilization (R2=0.849, P<0.001). Among 62 patients accepted for AHF therapy, those with durable LVAD implementation had better 1-year survival (71.5%) than those receiving an extracorporeal CentriMag (Levitronix) device as BTT (40%, P=0.008). In total, 143 LVADs were implanted, with 130 as BTT or bridge-to-candidacy (BTC) methods. The survival rate among the 130 BTT/BTC LVAD patients resembled that of HTx recipients (73.8% vs. 69.8% at 9 years, P=0.296). Conclusions: The burden of AHF management has increased and gained complexity over the past 30 years in Hong Kong.
ZnO Nanowire Arrays on 3D Hierachical Graphene Foam: Biomarker Detection of Parkinson’s Disease

<P>We report that vertically aligned ZnO nanowire arrays (ZnO NWAs) were fabricated on 3D graphene foam (GF) and used to selectively detect uric acid (UA), dopamine (DA), and ascorbic acid (AA) by a differential pulse voltammetry method. The optimized ZnO NWA/GF electrode provided a high surface area and high selectivity with a detection limit of 1 nM for UA and DA. The high selectivity in the oxidation potential was explained by the gap difference between the lowest unoccupied and highest occupied molecular orbitals of a biomolecule for a set of given electrodes. This method was further used to detect UA levels in the serum of patients with Parkinson’s disease (PD). The UA level was 25% lower in PD patients than in healthy individuals. 2',4'-Dihydroxy-6'-methoxy-3',5'-dimethylchalcone(DMC) is a chalcone isolated from the buds of Cleistocalyxoperculatus (Roxb.) Merr. et Perry, and thehepatoprotective effects of DMC on Kunming mice havebeen studied in previous study. However, the effects ofDMC on hepatocyte toxicity and corresponding mechanismremain unclear. The aim of this study was to evaluate thehepatoprotective mechanism of DMC in human hepatocytes(L02) treated with H2O2. The results demonstrated thatpretreatment with DMC effectively protected H2O2-inducedcell viability loss, cell membrane damage (lactate dehydrogenase,nitric oxide production and caspase-3 accumulation. Besides, DMC pretreatment increased the amount ofglutathione, decreased malondialdehyde and the percentageof apoptotic L02 cells compared with only H2O2 treatedgroup. Taken together, these results indicated that DMC hadhepatoprotective effects against H2O2-induced liver injuryby alleviating oxidative stress and apoptosis process in L02cells, and DMC might be a potential candidate for theintervention of liver diseases.
