IL-24 Contributes to Neutrophilic Asthma in an IL-17A-Dependent Manner and Is Suppressed by IL-37

Purpose: Neutrophilic asthma is associated with asthma exacerbation, steroid insensitivity, and severe asthma. Interleukin (IL)-24 is overexpressed in asthma and is involved in the pathogenesis of several allergic inflammatory diseases. However, the role and specific mechanism of IL-24 in neutrophilic asthma are unclear. We aimed to elucidate the roles of IL-24 and IL-37 in neutrophilic asthma, the relationships with IL-17A and the mechanisms regulating neutrophilic asthma progression. Methods: Purified human neutrophils were isolated from healthy volunteers, and a cell coculture system was used to evaluate the function of IL-24 in epithelium-derived IL-17A-dependent neutrophil migration. IL-37 or a small interfering RNA (siRNA) targeting IL-24 was delivered intranasally to verify the effect in a murine model of house dust mite (HDM)/lipopolysaccharide (LPS)-induced neutrophilic asthma. Results: IL-24 enhanced IL-17A production in bronchial epithelial cells via the STAT3 and ERK1/2 signaling pathways; this effect was reversed by exogenous IL-37. Anti-IL-17A monoclonal antibodies reduced neutrophil chemotaxis induced by IL-24-treated epithelial cells in vitro. Increased IL-24 and IL-17A expression in the airway epithelium was observed in HDM/LPS-induced neutrophilic asthma. IL-37 administration or IL-24 silencing attenuated neutrophilic asthma, reducing IL-17A levels and decreasing neutrophil airway infiltration, airway hyperresponsiveness, and goblet cell metaplasia. Silencing IL-24 inhibited T-helper 17 (Th17) immune responses, but not Th1 or Th2 immune responses, in the lungs of a neutrophilic asthma model. Conclusions: IL-24 aggravated neutrophilic airway inflammation by increasing epithelium-derived IL-17A production, which could be suppressed by IL-37. Targeting the IL-24/IL-17A signaling axis is a potential strategy, and IL-37 is a potential candidate agent for alleviating neutrophilic airway inflammation in asthma.

Thin-walled structures with varying thickness (single variable attribute) had been proven to be able improve energy absorption characteristics. In order to further study the energy absorption characteristics of thin-walled structures whose material properties changed as the thickness changing, a novel type of mixed thin-walled tube with axially-varying thickness and material were proposed in this paper. The finite element (FE) model was validated under axial quasi-static loading firstly, and seven groups simulation models of thin-walled tubular structure were established. The simulation results showed that the dual-variable attribute tubes can ensure high energy absorption and reduce the initial peak crushing force effectively in the crushing process. Furthermore, the maximum crushing force efficiency of steel-aluminum mixed tube can reach 2.77, while the aluminum mixed tube increased the crushing force efficiency to 1.24. Subsequently, a analytical model for dual-variable attribute tube was established and the theoretical mean crushing force calculation formula can be derived. The simulation and theoretical results were wirelessly close, except that the error of SUT5 was about 11 %. Therefore, the theoretical prediction results were in good agreement with the simulation results, and the theoretical prediction method can provide some reference for the design of double variable tube.

For detection of single taste intensities and binary interactions using an electronic tongue (E-tongue) and the human tongue (H-tongue), the 4 basic flavor substances of sucrose, caffeine, citric acid, and sodium chloride were used. A linear fit was performed using response values of the E-tongue and concentrations of the basic tastes. The taste intensity of the E-tongue was positively correlated with sour and salty substances, but negatively correlated with sweet and bitter substances. All taste intensities were positively correlated with the 4 flavor substances using the H-tongue. A mathematical model of the 4 taste substances on both the E-tongue and H-tongue exhibited a semi-logarithmic form. There were synergistic, dampening, or offset effects on the taste intensity of each flavor substance in binary systems. H-tongue and E-tongue results were consistent for sweet/bitter and salt/sour interactions. H-tongue and E-tongue results were contradictory for salt/sweet and sweet/sour interactions.
On-line Drift Reduction for Portable Electronic Nose Instrument in Monitoring Indoor Formaldehyde

In this paper, a new on-line drift reduction method based on component correction method is proposed for using in a portable electronic nose (Enose) instrument which is used for real-time monitoring indoor formaldehyde. For a portable Enose instrument which is used indoors, the external conditions are changed from time to time, so the drift direction cannot be easily obtained by using component correction method. Support vector machines (SVM) is used for predicting baseline of each sensor in portable Enose instrument. According baseline prediction and correction, the drift direction is obtained, and reduced by using component correction. And then, another SVM is used for predicting the concentration of formaldehyde. The results show that the method works efficiently for a portable Enose instrument which is used for real-time monitoring indoor formaldehyde.

Onions (Allium cepa L.) are widely used in the food industry for its nutritional and aromatic properties. Our studies showed that ethyl acetate extract of onion (EEO) had potent inhibitory effects on animal fatty acid synthase (FAS), and could induce apoptosis in FAS over-expressing human breast cancer MDA-MB-231 cells. Furthermore, this apoptosis was accompanied by reduction of intracellular FAS activity and could be rescued by 25 mM or 50 mM exogenous palmitic acids, the final product of FAS catalyzed synthesis. These results suggest that the apoptosis induced by EEO occurs via inhibition of FAS. We also found that EEO could suppress lipid accumulation during the differentiation of 3T3-L1 adipocytes, which was also related to its inhibition of intracellular FAS activity. Since obesity is closely related to breast cancer and obese patients are at elevated risk of developing various cancers, these findings suggested that onion might be useful for preventing obesity-related malignancy.

Glucocorticosteroid-induced osteoporosis is the most frequent of all secondary types of osteoporosis, and can increase the risk of vertebral compression fractures (VCFs). There are promising additions to current medical treatment for appropriately selected osteoporotic patients. Few studies have reported on the efficiency of percutaneous vertebroplasty (PVP) or kyphoplasty for whole thoracic and lumbar glucocorticosteroid-induced osteoporotic vertebral compression fractures. We report a case of a 67-year-old man with intractable pain caused by successional VCFs treated by PVP.
Experimental Study of Composite Steel Plate Shear Wall with Flush End-Plate Connection

This paper presents an investigation on the structural performance of steel plate shear wall (SPSW) with fl ush end-plate beam–column connections and infi ll precast reinforced concrete (precast RC) panels. Two single-span two-story SPSW specimens, including unstiff ened SPSW (NBRP) and precast RC panel restrained SPSW (Con-BRP), are fi rst tested, followed with a parametric study by fi nite element method. Precast RC cover panels are installed on both sides of the infi ll steel plate and are disconnected from steel frame. Test results indicate that the use of precast RC cover panels increases the load carrying and energy dissipation capacities of the SPSW structure, but decreases its ductility. It is also eff ective in reducing the inward fl exural deformation of columns. Moreover, the stiff ening eff ect of the infi ll steel plate on the beam–column connections remains, which is a result of the precast RC cover panel’s resistance to the local buckling and the tears of the infi ll steel plates. The infl uence of the gap size between the precast RC cover plate and frame members on the failure mode of the specimen Con-BRP is also investigated, based on which a maximum gap size is recommended. Conclusions are drawn that SPSW structure with fl ush end-plate beam–column connections and precast RC cover panels fully exploits the strength of infi ll steel plates and exhibits excellent structural performance.

A new analytical perturbation method is developed in this study to investigate the general reflectioncoefficients in the frequency-wavenumber domain of the acoustic field in a fluid-filled boreholesurrounded by a transversely isotropic medium (TIM). The transversely isotropic medium with asymmetric axis parallel to the borehole axis, which is usually called a VTI medium, was adoptedbecause its exact solutions exists, and a corresponding isotropic medium was adopted as a referencestate of perturbation solution. The general reflection coefficients were originally calculated byusing the perturbation method and were compared with the analytical solutions. The zero-, firstandsecond-order perturbation solutions for the general reflection coefficients excited by monopole,dipole and quadrupole sources were investigated for a transversely isotropic elastic solid. The resultsshowed that the general reflection coefficients obtained by using the perturbation solutionsand the analytical solutions were similar for all three sources. In summary, our study demonstratedthat the perturbation method is valid and effective in acoustical logging. This work provided atheoretical foundation for extending perturbation analyses to complicated anisotropic acousticallogging applications.
