        Kraft Lignin Biodegradation by Dysgonomonas sp. WJDL-Y1, a New Anaerobic Bacterial Strain Isolated from Sludge of a Pulp and Paper Mill

        Wastewater containing kraft lignin (KL) discharged from pulp and paper industries could cause serious environmental contamination. Appropriate effluent treatment is required to reduce the pollution. Investigations on anaerobic bacteria capable of degrading KL are beneficial to both lignin removal and biofuel regeneration from the effluent. In this paper, an anaerobic strain capable of degrading KL was isolated from the sludge of a pulp and paper mill and identified as Dysgonomonas sp. WJDL-Y1 by 16S rRNA analysis. Optimum conditions for KL degradation by strain WJDL-Y1 were obtained at initial pH of 6.8, C:N ratio of 6 and temperature of 33°C, based on statistical analyses by response surface methodology. For a 1.2 g/l KL solution, a COD removal rate of 20.7% concomitant with biomass increase of 17.6% was achieved after 4 days of incubation under the optimum conditions. After the treatment by strain WJDL-Y1, KL was modified and degraded.

        Power control of CiADS core with the intensity of the proton beam

        This paper reports the control method for the core power of the China initiative Accelerator DrivenSystem (CiADS) facility. In the CiADS facility, an intense external neutron source provided by a protonaccelerator coupled to a spallation target is used to drive a sub-critical reactor. Without any control rodinside the sub-critical reactor, the core power is controlled by adjusting the proton beam intensity. Inorder to continuously change the beam intensity, an adjustable aperture is considered to be used at theLow Energy Beam Transport (LEBT) line of the accelerator. The aperture size is adjusted based on theProportional Integral Derivative (PID) controllers, by comparing either the setting beam intensity or thesetting core power with the measured value. To evaluate the proposed control method, a CiADS coremodel is built based on the point reactor kinetics model with six delayed neutron groups. The simulations based on the CiADS core model have indicated that the core power can be controlled stably byadjusting the aperture size. The response time in the adjustment of the core power depends mainly onthe adjustment time of the beam intensity

      • What is the Best Explainable Artificial Intelligence for Enhancing Self-Regulation Behavior in Healthcare Management? Evidence from A Randomized Field Experiment

        Despite the widespread use of artificial intelligence (AI) in mobile healthcare apps, the need for more transparency in AI algorithms hinders their effectiveness by preventing users from understanding the reasons behind AI-based information provision. To address this challenge, various types of explainable AI (XAI) are adopted to offer transparent explanations of AI. Despite significant debates surrounding AI intervention, limited research has been devoted to whether and how various XAI types affect user behavior differently. In this study, we conducted a randomized field experiment to investigate the effectiveness of three XAI algorithms: 1) feature importance, 2) feature attribution, and 3) counterfactual explanation in promoting users' health behavior. Drawing on the self-regulated learning theory, we expect that XAI focusing on counterfactual explanation increases strategic planning and outcome expectancy, resulting in better self-regulation behavior. Our findings indicate that counterfactual explanation significantly improves users' action planning behavior, leading to a 16.5% increase in workout duration and a 3.49% increase in health records compared to the control group. Our results are salient for users with a high level of AI susceptibility due to age, goal weight loss, and AI outcome. Our finding sheds light on the potential of algorithmic explanations to improve the effectiveness of AI interventions in the healthcare industry, with practical implications for designing more transparent and user-friendly healthcare apps.

      • Efficacy and Safety of Ombitasvir/Paritaprevir/Ritonavir and Dasabuvir in Non-Cirrhotic Asian Patients with Genotype 1b HCV Infection: ONYX-I SVR24 Results

        Aims: HCV genotype 1b is the most common genotype in Asian patients. ONYX-I is a phase 3, randomised, double-blind, placebo-controlled study of the 3-DAA regimen of OBV/PTV/r and DSV in treatment- naive and treatment-experienced non-cirrhotic patients with HCV GT1b infection in China, South Korea and Taiwan. Methods: In this study, the safety/efficacy of OBV/PTV/r + DSV administered for 12 weeks were evaluated in non-cirrhotic Asian patients. Patients in Arm A received active study drug during a 12-week double-blind (DB) period, while patients in Arm B received placebo during the same period followed by an open-label (OL) period in which they received 12 weeks of active study drug. Efficacy was assessed by SVR12 and SVR24. Efficacy and safety were assessed in all patients who received at least one dose of active study drugs. Results: 650 HCV GT1b patients (54% female, 100% Asian, 44% treatment-experienced) were enrolled from China (n=410) South Korea (n=120) and Taiwan (n=120), and randomised 1:1 to Arms A and B. In Arm A, SVR12 and SVR24 rates were 99.5% (183/184) in treatment-naive patients and 100% (141/141) in treatment- experienced patients. Most treatment-emergent adverse events (TEAEs) in patients receiving the active drug were mild in severity. The most common (≥5%) TEAEs in Arm A were upper respiratory tract infection (10.5%), headache (6.2%) and dizziness (5.2%). Seven patients had serious AEs during active treatment (Arm A) and one patient in Arm A discontinued treatment. Conclusions: In non-cirrhotic Asian adults with HCV GT1b-infection, treated with OBV/PTV/r + DSV for 12 weeks, SVR24 rates equalled previously reported SVR12 rates from this study (99.5% of treatment- naive and 100% of treatment-experienced patients), and are consistent with other clinical trials with this drug combination. The treatment was generally well tolerated with mostly mild TEAEs reported.

        Functional Analysis of Wheat TaPaO1 Gene Conferring Pollen Sterility Under Low Temperature

        Thermosensitive male sterility plays an important role in wheat fertility and production. As a key enzyme for chlorophyll degradation, pheophorbide a oxygenase (PaO) can suppress cell death in plants. We cloned the wheat gene TaPaO1 from the thermosensitive genetic male sterile (TGMS) line BS366; it encodes a typical PaO protein, containing a conserved Rieske [2Fe-2S] iron–sulphur motif, a mononuclear non-heme iron-binding motif, and a C-terminal CxxC motif. TaPaO1 was expressed in all tissues and was upregulated during the meiosis stage of BS366 anthers under low temperature. Subcellular localization of TaPaO1 specifically labelled the surrounding of chloroplasts. TaPaO1 regulated by RD29A promoter which responded to low temperature led to pollen sterility in transgenic tobacco. Expression analysis showed that TaPaO1 exhibited a higher level of expression in the anther than in other tissues in transgenic tobacco plants during low temperature treatment. We propose that the higher senescence-related activity of TaPaO1 may lead to the cell death of anthers, which happens at an early developmental stage under low temperature. These results provide new insights into the function of PaO during the early developmental stage of anthers. PaO is closely related to cell death regardless of whether it exhibits increased activity or inactive.
