      • On the association analysis of CNV data: a fast and robust family-based association method

        <P><B>Background</B></P><P>Copy number variation (CNV) is known to play an important role in the genetics of complex diseases and several methods have been proposed to detect association of CNV with phenotypes of interest. Statistical methods for CNV association analysis can be categorized into two different strategies. First, the copy number is estimated by maximum likelihood and association of the expected copy number with the phenotype is tested. Second, the observed probe intensity measurements can be directly used to detect association of CNV with the phenotypes of interest.</P><P><B>Results</B></P><P>For each strategy we provide a statistic that can be applied to extended families. The computational efficiency of the proposed methods enables genome-wide association analysis and we show with simulation studies that the proposed methods outperform other existing approaches. In particular, we found that the first strategy is always more efficient than the second strategy no matter whether copy numbers for each individual are well identified or not. With the proposed methods, we performed genome-wide CNV association analyses of hematological trait, hematocrit, on 521 Korean family samples.</P><P><B>Conclusions</B></P><P>We found that statistical analysis with the expected copy number is more powerful than the statistic with the probe intensity measurements regardless of the accuracy of the estimation of copy numbers.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1186/s12859-017-1622-z) contains supplementary material, which is available to authorized users.</P>

        Silage preparation and fermentation quality of natural grasses treated with lactic acid bacteria and cellulase in meadow steppe and typical steppe

        Objective: In order to improve fermentation quality of natural grasses, their silage preparation and fermentation quality in meadow steppe (MS) and typical steppe (TS) were studied. Methods: The small-scale silages and round bale silages of mixed natural grasses in both steppes were prepared using the commercial lactic acid bacteria (LAB) inoculants Chikuso-1 (CH, Lactobacillus plantarum) and cellulase enzyme (AC, Acremonium cellulase) as additives. Results: MS and TS contained 33 and 9 species of natural grasses, respectively. Stipa baicalensis in MS and Stipa grandi in TS were the dominant grasses with the highest dry matter (DM) yield. The crude protein (CP), neutral detergent fiber and water-soluble carbohydrate of the mixed natural grasses in both steppes were 8.02% to 9.03%, 66.75% to 69.47%, and 2.02% to 2.20% on a DM basis, respectively. All silages treated with LAB and cellulase were well preserved with lower pH, butyric acid and ammonia-N content, and higher lactic acid and CP content than those of control in four kinds of silages. Compared with CH- or AC-treated silages, the CH+ AC-treated silages had higher lactic acid content. Conclusion: The results confirmed that combination with LAB and cellulase may result in beneficial effects by improving the natural grass silage fermentation in both grasslands.

        The Optimal Design of Junctionless Transistors with Double-Gate Structure for reducing the Effect of Band-to-Band Tunneling

        The effect of band-to-band tunneling (BTBT) leads to an obvious increase of the leakage current of junctionless (JL) transistors in the OFF state. In this paper, we propose an effective method to decline the influence of BTBT with the example of ntype double gate (DG) JL metal-oxide-semiconductor field-effect transistors (MOSFETs). The leakage current is restrained by changing the geometrical shape and the physical dimension of the gate of the device. The optimal design of the JL MOSFET is indicated for reducing the effect of BTBT through simulation and analysis.

      • Research on Smart Home Interaction in Sleep Scenarios Based on User Needs

        This study explores the interaction methods of smart homes in sleep scenarios based on user needs, aiming to enhance user interaction experience with a user-centric interaction philosophy, and shift the development direction of smart homes from technologydriven to consumer needs-driven. The results indicate that in the sleep aid stage, Focused Interaction is the primary method, supported by Peripheral and Implicit Interactions. During the sleep stage, Implicit Interaction is the main method, supplemented by Peripheral and Focused Interactions. In the awakening stage, a foundation of Peripheral and Implicit Interactions with minimal Focused Interaction aligns more closely with consumer habits. This model can prevent the blind development of new interaction methods in smart homes within sleep scenarios, reducing the occurrence of unnecessary or unreasonable human-machine interactions.

        LINC01272 Suppressed Cell Multiplication and Induced Apoptosis Via Regulating MiR-7-5p/CRLS1 Axis in Lung Cancer

        LINC01272 is a long non-coding RNA (lncRNA) that has been considered as a biomarker for many diseases including lung squamous cell carcinoma. Here, we investigated the function and mechanism of LINC01272 on lung cancer (LC). The differential expression of LINC01272 in LC and normal samples was analyzed by GEPIA based on the data from TCGA-LUAD database, as survival prognosis was analyzed through Kaplan-Meier Plotter. LINC01272 overexpression plasmid and miR-7-5p mimic were transfected into A549 and PC-9 cells. LINC01272, miR-7-5p and cardiolipin synthase 1 (CRLS1) mRNA expression was measured by quantitative reverse transcription-polymerase chain reaction. Cell viability was detected through MTT assay. Cell multiplication was evaluated by cell formation assay. Cell apoptosis was assessed through flow cytometry assay. Through bioinformatics, the target miRNA of LINC01272 and downstream genes of miR-7-5p were predicted. The targeting relationship was tested by dual luciferase reporter analysis. CRLS1, B-cell lymphoma-2 (Bcl-2), BCL2-associated X (Bax) and cleaved caspase-3 protein levels were detected through western blot. LINC01272 was downregulated in LC and low LINC01272 expression had poor prognosis. In A549 and PC-9 cells, LINC01272 inhibited cell viability and multiplication and induced apoptosis. LINC01272 negatively regulated miR-7-5p and CRLS1 was a target of miR-7-5p. MiR-7-5p reversed the effect of LINC01272 on viability, multiplication, apoptosis and expression of miR-7-5p and CRLS1 as well as apoptosis-related factors (Bcl-2, Bax and cleaved caspase-3). LINC01272 suppressed cell multiplication and induced apoptosis via regulating the miR-7-5p/CRLS1 axis in LC.

        Ion-Imprinted Polymers Modified Sensor for Electrochemical Detection of Cu2+

        An electrochemical sensor (Cu2+-IIPs/GCE) was developed for detection of Cu2+ in water. Cu2+-IIPs/GCE was prepared by dispersing Cu2+ imprinted polymers (Cu2+-IIPs) on a preprocessed glassy carbon electrode. Cu2+-IIPs were synthesized on the surface of modified carbon spheres by ion imprinting technology. The electrochemical performance of Cu2+-IIPs/GCE was evaluated by differential pulse voltammetry method. The response of Cu2+-IIPs/GCE to Cu2+ was linear in 1.0 x 10 -5 mol/L to 1.0 x 10 -3 mol/L. The detection limit was 5.99 x 10 -6 mol/L (S/N = 3). The current response value of Cu2+-IIPs/GCE was 2.14 times that of the nonimprinted electrode. These results suggest that Cu2+-IIPs/GCE can detect the concentration of Cu2+ in water, providing a new way for heavy metal ions adsorption and testing.

        Characteristics of La_2O_3―Y_2O_3―Mo cermet cathode with RE_2O_3 nano particles

        The secondary electron emission property and microstructure of La_2O_3―Y_2O_3―Mo cermet cathode have been studied. It shows that the cathode prepared by Sol―Gel doping method has fine microstructure containing rare earth oxide (RE_2O_3) nanoparticles and uniform distribution of different substances. Auger Electron Spectroscopy (AES) depth profile result shows that an RE_2O_3 layer about 28 nm in thickness could be formed on the cathode surface, which plays an important role in the emission. Pre-activation of the cathode is favorable for the improvement of emission property. La_2O_3―Y_2O_3―Mo cathode preactivated exhibits good secondary emission property. The secondary emission yield could reach 5.24,about 1.8 times higher than that of original cathode. The enhancement on the conductivity of materials resulting from the pre-activation is favorable for the replenishment and transport of electrons, thus the secondary emission property of the cathode could be improved.

        A study on tensile strength mechanism of hypoeutectic gray cast iron reinforced by composite silicon carbide powder

        This study aims to reveal the changes in the microstructure of hypoeutectic gray cast iron under the addition of compositesilicon carbide powder of different mass fractions, and to explore how modified silicon carbide (SiC) particles of different sizesand contents improve the mechanical properties of hypoeutectic gray cast iron. The results showed that the modified SiCparticles increased the nucleation, the undercooling and microstructure of hypoeutectic gray cast iron, and the tensile strengthof the iron at room temperature was increased by 18,28%. The present work solves the problem that traditional SiC ceramicparticles are prone to agglomeration in molten metal, analyzes its action mechanism, and can expand the application of SiCceramic particles to strengthening metal properties of casting materials.
