        Effects of Xylanase on Performance, Blood Parameters, Intestinal Morphology, Microflora and Digestive Enzyme Activities of Broilers Fed Wheat-based Diets

        The study was conducted to investigate the effects of different levels of xylanase on performance, blood parameters, intestinal morphology, microflora and digestive enzyme activities of broilers. The wheat-based diets were supplemented with 0, 500, 1,000, 5,000 U/kg xylanase. Xylanase supplementation significantly (p<0.05) improved the feed:gain ratio of broilers from 1 to 21 d and 1 to 42 d. Supplementing 500 U/kg and 1,000 U/kg xylanase improved (p<0.05) the villus height and the ratio of villus height to crypt depth in the small intestine. Excess supplementation of xylanase (5,000 U/kg) increased the villus height in the ileum (p<0.01) and the ratio of villus height to crypt depth in the duodenum and ileum (p<0.05). The microflora in the ileum and caecum, digestive enzyme activities in the small intestine and the concentrations of serum glucose, uric acid, insulin and IGF-I were not affected by the supplementation of xylanase. Excess level of xylanase (5,000 U/kg) had a tendency to induce the multiplication of E. coli and total aerobes. The results suggested that supplementing 500 U/kg and 1,000 U/kg xylanase was beneficial for broilers and excess xylanase supplementation resulted in no further improvement or negative effects.

        Data Access Control Scheme Based on Blockchain and Outsourced Verifiable Attribute-Based Encryption in Edge Computing

        The arrival of the Internet of Things and 5G technology enables users to rely on edge computing platforms to process massive data. Data sharing based on edge computing refines the efficiency of data collection and analysis, saves the communication cost of data transmission back and forth, but also causes the privacy leakage of a lot of user data. Based on attribute-based encryption and blockchain technology, we design a fine-grained access control scheme for data in edge computing, which has the characteristics of verifiability, support for outsourcing decryption and user attribute revocation. User attributes are authorized by multi-attribute authorization, and the calculation of outsourcing decryption in attribute encryption is completed by edge server, which reduces the computing cost of end users. Meanwhile, We implemented the user's attribute revocation process through the dual encryption process of attribute authority and blockchain. Compared with other schemes, our scheme can manage users' attributes more flexibly. Blockchain technology also ensures the verifiability in the process of outsourcing decryption, which reduces the space occupied by ciphertext compared with other schemes. Meanwhile, the user attribute revocation scheme realizes the dynamic management of user attribute and protects the privacy of user attribute.

        Dexmedetomidine promotes the progression of hepatocellular carcinoma through hepatic stellate cell activation

        Dexmedetomidine (DEX) is an anesthetic that is widely used in the clinic, and it has been reported to exhibit paradoxical effects in the progression of multiple solid tumors. In this study, we sought to explore the mechanism by which DEX regulates hepatocellular carcinoma (HCC) progression underlying liver fibrosis. We determined the effects of DEX on tumor progression in an orthotopic HCC mouse model of fibrotic liver. A coculture system and a subcutaneous xenograft model involving coimplantation of mouse hepatoma cells (H22) and primary activated hepatic stellate cells (aHSCs) were used to study the effects of DEX on HCC progression. We found that in the preclinical mouse model of liver fibrosis, DEX treatment significantly shortened median survival time and promoted tumor growth, intrahepatic metastasis and pulmonary metastasis. The DEX receptor (ADRA2A) was mainly expressed in aHSCs but was barely detected in HCC cells. DEX dramatically reinforced HCC malignant behaviors in the presence of aHSCs in both the coculture system and the coimplantation mouse model, but DEX alone exerted no significant effects on the malignancy of HCC. Mechanistically, DEX induced IL-6 secretion from aHSCs and promoted HCC progression via STAT3 activation. Our findings provide evidence that the clinical application of DEX may cause undesirable side effects in HCC patients with liver fibrosis.

        Investigation on Structure and Properties of a Novel Designed Peptide with Half-Sequence Ionic Complement

        Although the existing design principle of full-sequence ionic complement is convenient for the development of peptides, it greatly constrains the exploration of peptides with other possible assembly mechanisms and different yet essential functions. Herein, a novel designed half-sequence ionic complementary peptide (referred to as P9), AC-Pro-Ser-Phe-Asn-Phe-Lys-Phe-Glu-Pro-$NH_2$, is reported. When transferred from pure water to sodium chloride solution, P9 underwent a dramatic morphological transformation from globular aggregations to nanofibers. Moreover, the rheological experiment showed that the P9 could form a hydrogel with a storage modulus of about 30 Pa even at very low peptide concentration (0.5% (wt/vol)). The P9 hydrogel formed in salt solution could recover in a period of about 1,800 sec, which is faster than that in the pure water. The data suggestcd that the half-sequence, ionic complementary peptide might be worthy of further research for its special properties.

        Refining Characteristics of Hardwood Pulp Using Straight- and Curved-bar Plates: A Time Series Study

        There are obvious differences in the refining characteristics of plates with straight bars compared to curved bar plates having the same bar parameters during refining. The objective of this research is to explore the difference in the refining characteristics of the straight bar and curved bar plates on experimental and theoretical studies. The bar angle of the curved bar plate is defined, and the two plates are used in the refining of bleached sulphate eucalyptus pulp (BSEP). Samples at different refining times were collected, and the properties of the fibers, pulp, and paper were detected and analyzed. It was found that the curved bar and straight bar plates have a similar influence on the pulp SR freeness. However, the curved bar plate effectively retained the fiber length, which means that its fiber cutting effect is weaker than that of the straight bar plate. Meanwhile, the fiber external fibrillation of the samples refined by the curved bar plate was better than that of the samples refined by the straight bar, which showed a higher tensile and tear index of the formed paper. Through the theoretical analysis of refining intensity via characterization of the parameter bar edge length (BEL), the specific edge load (SEL) of the curved bar plate was lower, and the refining characteristics predicted by the refining intensity were similar to the results from the experiments conducted in this paper.

        Fertility-preserving treatment outcome in endometrial cancer or atypical hyperplasia patients with polycystic ovary syndrome

        Objective: This study aimed to investigate the impact of polycystic ovary syndrome (PCOS) on fertility-sparing treatment in young patients with atypical endometrial hyperplasia (AEH) or endometrioid endometrial cancer (EEC). Methods: A total of 285 patients with EEC (n=76, FIGO stage IA, without myometrium invasion) or AEH (n=209) who received progestin-based fertility-sparing treatment were evaluated retrospectively. Among the 285 patients, 103 (36.1%), including 70 AEH cases and 33 EEC cases, were diagnosed with PCOS. General characteristics, cumulative 16- and 32-week complete response (CR) rate, pregnancy outcome and recurrence were compared between patients with or without PCOS. Results: The cumulative 16-week CR rate was lower in the PCOS group than in the non- PCOS group (18.4% vs. 33.8%, p=0.006). Patients with PCOS took longer treatment duration to achieve CR (7.0 months vs. 5.4 months, p=0.006) and shorter time to relapse after CR (9.6 months vs. 17.6 months, p=0.040) compared with non-PCOS group. After adjusting for patient age, body mass index, PCOS, homeostasis model assessment-insulin resistance index, and serum testosterone levels, we found that body mass index ≥25 kg/m2 (HR=0.583; 95% CI=0.365–0.932; p=0.024) and PCOS (HR=0.545; 95% CI=0.324–0.917; p=0.022) were significantly correlated with lower 16-week CR rate. Conclusion: PCOS was associated with lower 16-week CR rate, longer treatment duration and shorter recurrence interval in patients with AEH or EEC receiving fertility-preserving treatment.

        The prognostic significance of estrogen and progesterone receptors in grade I and II endometrioid endometrial adenocarcinoma: hormone receptors in risk stratification

        Objectives: Although patients with grade I and II endometrioid endometrial adenocarcinoma (EEA) are considered with good prognosis, among them 15%–25% died in 5 years. It is still unknown whether integrating estrogen receptor (ER) and progesterone receptor (PR) into clinical risk stratification can help select high-risk patients with grade I–II EEA. This study was to investigate the prognostic value of ER and PR double negativity (ER/ PR loss) in grade I–II EEA, and the association between ER/PR loss and The Cancer Genome Atlas (TCGA) classification. Methods: ER and PR were assessed by immunohistochemistry on hysterectomy specimens of 903 patients with grade I–II EEA. ER and PR negativity were determined when <1% tumor nuclei were stained. Gene expression data were obtained from the TCGA research network. Results: Compared with ER or PR positive patients (n=868), patients with ER/PR loss (n=35) had deeper myometrial infiltration (p=0.012), severer FIGO stage (p=0.004), and higher rate of pelvic lymph node metastasis (p=0.020). In univariate analysis, ER/PR loss correlated with a shorter progression-free survival (PFS; hazard ratio [HR]=5.25; 95% confidence interval [CI]=2.21–12.52) and overall survival (OS; HR=7.59; 95% CI=2.55–22.60). In multivariate analysis, ER/PR loss independently predicted poor PFS (HR=3.77; 95% CI=1.60–10.14) and OS (HR=5.56; 95% CI=1.37–22.55) for all patients, and poor PFS for patients in stage IA (n=695; HR=5.54; 95% CI=1.28–23.89) and stage II–IV (n=129; HR=5.77; 95% CI=1.57–21.27). No association was found between ER/PR loss and TCGA classification. Conclusion: Integrating ER/PR evaluation into clinical risk stratification may improve prognosis for grade I–II EEA patients.

        Instability analysis under part-load conditions in centrifugal pump

        In this study, a centrifugal pump with a specific speed of 39.12 m×min -1 ×m 3 s -1 is treated to analyze the flow instability under part-load conditions by numerical simulation and experimental test. For calculations, the RANS method, coupled with the k-ω SST turbulence model, is adopted. Numerical results at different operation points are compared with available experimental data, such as hydraulic performance and flow field information by particle image velocimetry. The numerical and experiment results agree well. The flow simulation indicates a strong reverse flow at the passage upstream impeller inlet, and the energy loss in the impeller is the largest under partload conditions among all flow components in the pump. In one impeller revolution, one blade-to-blade flow passage is always nearly blocked off by the rotating stall occurring at the impeller inlet for each instant, and the blockage induces a jet flow with large velocity at the next blade-to-blade flow passage along the rotational direction of the impeller. The blockage and the jet flow in the blade-to-blade flow passages will make the flow unstable inside the impeller and cause performance breakdown and pressure vibration under part-load conditions for the pump.

        Insect chemosensory proteins (CSPs) are small, water-soluble proteins involved in detection and recognition of chemical cues and semiochemicals. A better understanding of CSPs and their function could aid in the development of pest control strategies that are based on chemosensory regulation. We screened our unpublished transcriptome data of Bemisia tabaci and identified five CSP genes (BtabCSPs) including two novel BtabCSPs viz. BtabCSP4 and BtabCSP5. Phylogenetic analysis of amino acid sequences of the proteins encoded by these genes revealed that BtabCSPs are distinct fromCSPs of other hemipterans. Quantitative PCR analysis showed differential expression of these genes in different stages. In the adult stage, BtabCSP1, 2 and 4 had significantly higher expression than BtabCSP3 and 5 respectively. On the other hand, BtabCSP2 and 5 were relatively highly expressed in stage of pseudopupae compared to BtabCSP1, 3 and 4. In the third instar BtabCSP3, 4 and 5 exhibited higher expression compared to other CSP genes. Expression of all CSP genes was relatively low in the first and second instars compared to later developmental stages. The identification of novel BtabCSPs and the expression profiling of CSP genes in general may help facilitate development of pest management strategies based on disruption of CSPdriven behaviors.

        Study on Fiber Cutting Performance of Isometric Straight Bar Plate with Different Bar Angle

        The refining plate is a direct-acting component of refiners in pulping, and the bar profile of it have a major impact on fiber cutting, which affects the paper forming and strength of the formed paper. In this paper, the fiber average length, the fiber length distribution, the original fiber cutting rate and adjacent fiber cutting rate of samples collected in different refining time are used to analyze the fiber cutting mechanism of the isometric straight bar refining plates with different bar angles. The results show that the fiber cutting performance of plates gradually decreases and the limit fiber cutting length of the plate increases with the increase of the bar angle which is opposite to the conclusion predicted by specific edge load (SEL). However, the fiber cutting action of plates is gradually weakened until the average fiber length is reduced to the limit cutting length of plates. Meanwhile, the cutting mechanism of the straight bar plates with different bar angles is different. The larger bar angle of plate, the lower fiber cutting rate and the adjacent cutting rate of which is basically kept constant during refining, while the adjacent cutting rate of smaller bar angle plate on fibers gradually decreases with refining and finally remains constant. The study on the cutting performance of different bar angle plates has positive significance for the selection and design of refining plates for different pulping process.
