Pseudorabies virus (PRV), a member of the Alphaherpesviridae, is the causative agent of Aujeszky’s disease in pigs. Glycoprotein B (gB) of PRV, a major constituent of the viral envelope, consists of 916 amino acids. We continuously combined three gB epitopes, E1 (aa 62-129), E2 (aa 217-282), and E3 (aa 543-737). The DNA fragment containing the PRV gB epitopes was fused with polyhedrin gene in order to generate recombinant baculovirus that expresses the recombinant polyhedra with PRV gB epitopes under the control of the Bombyx mori nucleopolyhedrovirus polyhedrin promoter. Recombinant baculoviruses were injected into fifth-instar B. mori larvae. SDS-PAGE and Western blot analyses revealed that recombinant polyhedra constitute polyhedrin and PRV gB epitopes, and that the recombinant PRV gB epitopes showed cross-reactivity against antiserum of PRV gB produced from pig. To examine the immunogenicity of recombinant PRV gB epitopes, we injected into mice as model animals. ELISA results indicated that antibody production is increased in a similar manner in the injection of recombinant polyhedra with PRV gB epitopes, either injected recombinant polyhedra as a granule form antigen without adjuvant or injected recombinant polyhedrin as a soluble form antigen with adjuvant. Taken together, these data show that PRV gB epitopes were produced as a granule form antigen by fusing recombinant polyhedra in baculovirus-infected silkworm larvae and displayed the immunogenicity in mice, indicating the efficacy of the granule form antigen as a PRV gB vaccine.

There are some places such as bridges in the heavily industrialized area where the pavement should have a strong resistance against heavy axle loading and waterproof function. In those places, many polymer-modified asphalt (PMA) pavements were applied to protect premature cracking, severe rutting and water intrusion without success. Therefore, a much tougher pavement material with waterproofing function was developed for those places. This study evaluated important properties of the special type asphalt mixture which is highly condensed to be almost void-free condition. A high-quality PMA binder with PG82-34 grade was used for preparing the mixture and the optimum binder content was determined to allow near 0% air void in the mix design. The deformation strength(SD) by Kim Test and rut depth by wheel tracking test were measured at 60℃ as high temperature properties. The flexural strength and fracture toughness was measured at -10℃ as low temperature property. The void-free AC showed the higher performance in all four properties than any other asphalt concretes which were prepared for comparison. Therefore, it was shown that the normal concern about limiting air voids within 3-5% was just an apprehension. The void-free AC can be applied for heavy duty pavement on the bridge where the water-proofing function and higher rutting and cracking resistance are required.

Purpose: Recent mechatronics technology is the most appropriate high technology in agricultural applications to save repetitious labor. Method: Cow`s body parameters were measured by several traditional measurers. Image processing technology was used to measure automatically their parameters to reduce labor and time. The parameters were measured form a small model cow which is easily measured, instead to a real cow. The image processing system designed and built for this project was composed of a PC, grabber card, and two cameras, which are located on the side and the top of the model cow. Tests of verification had measured 10 dairy cows. Result: Nine parameters of the model cow`s body were measured, and the difference between the real data and the data by image processing was less than 16.7%. Based on the results of the research, the parameters of a real cow had measured of chest depth, withers height, Pelvic arch height, body length, slope body length, chest width, hip width, thurl width, and pin bone width were compared with image processing data. Conclusions: In the Demonstration test, Result had obtained similar data of cow model experiments, and the most of errors were shown less than 5% relatively good result.
다짐온도에 따른 변형강도 변화를 통한 아스팔트 콘크리트의 철도노반 적용성 평가

아스팔트 콘크리트 철도노반은 여러 가지 장점 때문에 선진국에서는 많이 적용되고 있어 국내에서도 철도에 적용성 연구가 필요하다. 아스팔트 포장은 소성변형 발생의 우려가 커 사전에 강도시험 등을 통해 공용성 확인이 필요하며, 철도용 아스팔트 콘크리트도 같은 맥락에서 소성변형에 대한 평가가 필요하다. 이에 본 연구에서는 변형강도(SD)를 사용하여 가열 아스팔트(HMA) 콘크리트와 준고온 아스팔트(WMA) 콘크리트의 소성변형 저항성을 평가하여 적정다짐온도를 추정하였다. 연구 결과 일반 아스팔트 혼합물은 HMA 135℃, WMA 105℃ 이상, PMA에서는 HMA155℃, WMA 125℃ 이상이 적정 다짐온도로 확인되었다. Since asphalt concrete is widely used for railroad base in advanced countries due to many advantages, the preliminary research for its applicability is necessary in Korea. Asphalt concrete is prone to deform (rut) in hot environment, and therefore, the rut resistance of asphalt concrete for railroad is also need to be evaluate. The rut resistance of hot-mix asphalt (HMA) concrete and warm-mix asphalt (WMA) concrete was evaluated to determine optimum compaction temperature (OCT) using deformation strength (SD). The results showed that OCT values of normal asphalt HMA and WMA were 135℃ and 105℃ , respectively, and those of polymer-modified HMA and WMA were 155℃ and 125℃, respectively.
