Identification of HDAC9 as a viable therapeutic target for the treatment of gastric cancer

Histone deacetylase inhibitors (HDACis) are a new class of anticancer drugs confirmed to have good therapeutic effects against gastric cancer (GC) in preclinical experiments, but most HDACis are non-selective (pan-HDACis), with highly toxic side effects. Therefore, it is necessary to screen HDAC family members that play key roles in GC as therapeutic targets to reduce toxic side effects. In this study, we evaluated the targeting specificity of the HDACi suberoylanilide hydroxamic acid (SAHA) for GC via fluorescence molecular imaging (FMI). In vitro FMI results showed that SAHA had higher binding affinity for GC cells than for normal gastric cells. In vivo FMI of gastric tumor-bearing mice confirmed that SAHA can be enriched in GC tissues. However, there was also a high-concentration distribution in normal organs such as the stomach and lungs, suggesting potential side effects. In addition, we found that among the HDAC family members, HDAC9 was the most significantly upregulated in GC cells, and we verified this upregulation in GC tissues. Further experiments confirmed that knockdown of HDAC9 inhibits cell growth, reduces colony formation, and induces apoptosis and cell cycle arrest. These results suggest that HDAC9 has an oncogenic role in GC. Moreover, HDAC9 siRNA suppressed GC tumor growth and enhanced the antitumor efficacy of cisplatin in GC treatment by inhibiting the proliferation and inducing the apoptosis of GC cells in vitro and in vivo. Our findings suggest that the development of HDAC9-selective HDACis is a potential approach to improve the efficacy of chemotherapy and reduce systemic toxicity.
Geft is dispensable for the development of the second heart field

Geft is a guanine nucleotide exchange factor, which can specifically activate Rho family of small GTPase by catalyzing the exchange of bound GDP for GTP. Geft is highly expressed in the excitable tissue as heart and skeletal muscle and plays important roles in many cellular processes, such as cell proliferation, migration, and cell fate decision. However, the in vivo role of Geft remains unknown. Here, we generated a Geft conditional knockout mouse by flanking exons 5-17 of Geft with loxP sites. Cre-mediated deletion of the Geft gene in heart using Mef2c-Cre transgenic mice resulted in a dramatic decrease of Geft expression. Geft knockout mice develop normally and exhibit no discernable phenotype, suggesting Geft is dispensable for the development of the second heart field in mouse. The Geft conditional knockout mouse will be a valuable genetic tool for uncovering the in vivo roles of Geft during development and in adult homeostasis. (BMB reports 2012; 45(3): 153-158)

Reinforcement agglomeration always leads to severe stress concentration and porosity, which is detrimental to the deformation ability and mechanical properties of particulate-reinforced metal matrix composites. In this study, uniform distribution of 32vol%B<SUB>4</SUB>C has been achieved in B<SUB>4</SUB>C/Al composite by means of flake powder metallurgy (Flake PM), in which flake Al powder is used as the starting material. The flake Al powder exhibits higher apparent volume than spherical powders of the same mass, and thus can provide more space to accommodate the B<SUB>4</SUB>C particles. Therefore, compared with conventional PM, Flake PM can lead to more uniform distribution of B<SUB>4</SUB>C particles in the composite powder as well as in the consolidated composite. Meanwhile, the flake Al powder has a nano skin of Al<SUB>2</SUB>O<SUB>3</SUB>, which could be fractured and dispersed inside the fine matrix grains during consolidation, and were found to induce a higher normalized strain hardening rate for the composite during deformation. As a result, the Flake PM 32vol%B<SUB>4</SUB>C/Al composite exhibits an ultimate tensile strength of 305MPa and a uniform elongation of 6.6%, 63% stronger and 13% more ductile than its counterpart fabricated by conventional PM.
Evaluation of the gastroprotective effects of 20 (S)-ginsenoside Rg3 on gastric ulcer models in mice

Background: Gastric ulcer (GU) is a common gastrointestinal disease that can be induced by many factors. Finding an effective treatment method that contains fewer side effects is important. 20 (S)-ginsenosideRg3 is a kind of protopanaxadiol and has shown superior antiinflammatory and antioxidanteffects in many studies, especially cancer studies. In this study, we examined the treatment efficacy of 20(S)-ginsenoside Rg3 on GU. Methods: Three kinds of GU models, including an alcohol GU model, a pylorus-ligated GU model, and anacetic acid GU model, were used. Mouse endothelin-1 (ET-1) and nitric oxide (NO) levels in blood andepidermal growth factor (EGF), superoxide dismutase, and NO levels in gastric mucosa were evaluated. Hematoxylin and eosin staining of gastric mucosa and immunohistochemical staining of ET-1, induciblenitric oxide synthase (NOS2), and epidermal growth factor receptors were studied. Ulcer index (UI)scores and UI ratios were also analyzed to demonstrate the GU conditions in different groups. Furthermore,Glide XP from Schrödinger was used for molecular docking to clarify the interactions between 20(S)-ginsenoside Rg3 and EGF and NOS2. Results: 20 (S)-ginsenoside Rg3 significantly decreased the UI scores and UI ratios in all the three GUmodels, and it demonstrated antiulcer effects by decreasing the ET-1 and NOS2 levels and increasing theNO, superoxide dismutase, EGF, and epidermal growth factor receptor levels. In addition, high-dose 20(S)-ginsenoside Rg3 showed satisfactory gastric mucosa protection effects. Conclusion: 20 (S)-ginsenoside Rg3 can inhibit the formation of GU and may be a potential therapeuticagent for GU.

Precipitation characteristics influencing fatigue crack propagation contained matrix precipitate, grain boundary precipitateand precipitate free zone for Al–Zn–Mg–Cu alloys. Over-aging treatment could effectively regulate precipitation and then tobe able to change fatigue crack propagation behavior compared with the peak aging state. In the current work, typical T651and T7651 aging tempers of the alloy were extracted via hardness, electrical conductivity and mechanical properties underone-step and two-step aging treatments. Fatigue crack propagation (FCP) rate under them was tested and correspondingprecipitation characteristics and fracture morphology were observed. The results indicated that fatigue crack propagationresistance for the T7651 temper possessed an obvious improvement on the side of that for the T651 temper, which was alsosupported by fracture appearance, including tearing ridge, tearing dimple and fatigue striation. The precipitation observationshowed that the T651 alloy contained GPI zone, GPII zone and ηʹ phase while the T7651 alloy possessed ηʹ phase and η phase.Compared with the T651 temper, matrix precipitate for the T7651 temper distinctly owed an expanding of size distributionand an enlargement of average size while cuttable phase still remained the dominance for both tempers. Grain boundaryprecipitate and precipitate free zone manifested no obvious difference between the two aging tempers. Cut and bypass mechanismsof dislocation–precipitate interactions were used for explanation and it revealed the reinforced cuttable phase was infavor of enhancing fatigue crack propagation resistance. A theoretical model which directly correlated FCP rate with matrixprecipitate characteristics was employed to calculate FCP rate by substituting quantitative precipitate characteristics and thecalculation results were vaguely consistent with the experimental measurement, which proved its reliability and feasibility.

In the present work, the infl uence of various retrogression treatments on hardness, electrical conductivity and mechanicalproperties of a high Zn-containing Al–Zn–Mg–Cu alloy is investigated and several retrogression regimes subjected to a samestrength level are proposed. The precipitates are qualitatively investigated by means of transmission electron microscopy(TEM) and high-resolution transmission electron microscopy techniques. Based on the matrix precipitate observations, thedistributions of precipitate size and nearest inter-precipitate distance are extracted from bright-fi eld TEM images projectedalong ⟨110⟩ Al orientation with the aid of an imaging analysis and an arithmetic method. The results show that GP zonesand η′ precipitates are the major precipitates and the precipitate size and its distribution range continuously enlarge with theretrogression regime expands to an extent of high temperature. The nearest inter-precipitate distance ranges obtained arequite the same and the average distance of nearest inter-precipitates show a slight increase. The infl uence of precipitates onmechanical properties is discussed through the interaction relationship between precipitates and dislocations.

DC/DC converters with hybrid proton exchange membrane (PEM) fuel cell and battery power sources are designed and implemented using sliding mode control (SMC). The converters are typically comprised by unidirectional boost converter and bidirectional converter which are used for regulating the bus voltage and managing power distribution. SMC is selected to control the converters since it can cope with the high non-linear characteristic of the coupled system as well as ensure the stability. The hybrid power control system is constructed in MATLAB/Simulink platform. A comparative approach based on conventional proportional-integral (PI) control is developed to compare the control performance with SMC. The experimental validation is carried out with National Instruments (NI) LabVIEW-based hybrid PEMFC/battery supplied DC/DC converters system prototype.
