        The significance of intertumor and intratumor heterogeneity in liver cancer

        Genomic analyses of primary liver cancer samples reveal a complex mutational landscape with vast intertumor and intratumor heterogeneity. Different primary liver tumors and subclones within each tumor display striking molecular and biological variations. Consequently, tumor molecular heterogeneity contributes to drug resistance and tumor relapse following therapy, which poses a substantial obstruction to improving outcomes of patients with liver cancer. There is an urgent need to the compositional and functional understanding of tumor heterogeneity. In this review, we summarize genomic and non-genomic diversities, which include stemness and microenvironmental causes of the functional heterogeneity of the primary liver cancer ecosystem. We discuss the importance and intricacy of intratumor heterogeneity in the context of cancer cell evolution. We also discuss methodologies applicable to determine intratumor heterogeneity and highlight the best-fit patient-derived in vivo and in vitro models to recapture the functional heterogeneity of primary liver cancer with the aim to improve future therapeutic strategies.

        Design of a type of broadband metamaterial absorber based on metal and graphene

        In this paper, we demonstrate a kind of broadband metamaterial perfect absorber using both graphene and metal resonator elements. Through step by step design and simulation, wider absorption band from about 4.22 THz to 7.48 THz with average absorption rate up to 98.21% is achieved in the absorption spectrum. In addition, the absorber has characteristics of polarization insensitivity and wide incident angle due to its inherent rotational symmetry. Moreover, the absorption band can be adjusted by changing the chemical potential of the graphene. The superiorities of broadband, high absorption rate, polarization independent and wide-angle characteristics make it have potential application prospects in electromagnetic wave absorbing, signal sensing and detection, and other optoelectronic devices.

        Molecular Characterization and Expression Analysis of GhWRI1 in Upland Cotton

        Numerous genes were involved in oil metabolism,transcription factors are the key genes, such as WRINKLED(AtWRI1, At3g54320) in Arabidopsis. A homolog of AtWRI1,GhWRI1 (accession number: JF430801), was cloned in presentstudy. Promoter activity analyses revealed that activityelements were existed in the upstream of GhWRI1 initiationcodon between -912 ~ -720 bp and -372 ~ -177 bp regions. The putative transcriptional activity domain was localized inthe C-terminal of GhWRI1 protein. GhWRI1 was moreabundant in high oil content materials and elevated expressionwith cottonseed development, suggesting the crucial role inseed oil accumulation. Meanwhile, expression level of GhWRI1was induced under multiple abiotic stresses, suggesting thatGhWRI1 might participate in cotton responding to stresses. Overexpression of GhWRI1 increased seed oil content andseed weight in transgenic Arabidopsis thaliana. In transgenicplants, genes downstream of AtWRI1, such as enoyl-ACPreductase (AtEAR, At2g05990), pyruvate kinases (AtPKp2,At5g52920), and pyruvate dehydrogenase subunit (AtPDHE1α,At1g01090) were up-regulated expression two to five timescompared with the wild-type. The results suggested that GhWRI1could be used in regulating oil accumulation in cottonseeds.

        Experimental study on chemical activation of recycled powder as a cementitious material in mine paste backfilling

        To improve the utilization rate of construction waste as mine backfilling materials, this paper investigated the feasibility of using recycled powder as mine paste backfilling cementitious material, and studied the pozzolanic activity of recycled construction waste powder. In this study, alkali-calcium-sulfur served as the activation principle and an orthogonal test plan was performed to analyze the impact of the early strength agent, quick lime, and gypsum on the pozzolanic activity of the recycled powder. Our results indicated that in descending order, early strength agent > quick lime > gypsum affected the strength of the backfilling paste with recycled powder as a cementitious material during early phases. The strength during late phases was affected by, in descending order, quick lime > gypsum > early strength agent. Using setting time and early compressive strength as an analysis index as well as an extreme difference analysis, it was found that the optimal ratio of recycled powder cementitious material for mine paste backfilling was recycled powder:quick lime:gypsum:early strength agent at 78%:10%:8%:4%. X-ray diffraction analysis and scanning electron microscope were used to show that the hydration products of recycled powder cementitious material at the initial stages were mainly CH and ettringite. As hydration time increased, more and more recycled powder was activated. It mainly became calcium silicate hydrate, calcium aluminate hydrate, etc. In summary, recycled powder exhibited potential pozzolanic activities. When activated, it could replace cementitious materials to be used in mine backfill.

        Evaluation of the gastroprotective effects of 20 (S)-ginsenoside Rg3 on gastric ulcer models in mice

        Background: Gastric ulcer (GU) is a common gastrointestinal disease that can be induced by many factors. Finding an effective treatment method that contains fewer side effects is important. 20 (S)-ginsenoside Rg3 is a kind of protopanaxadiol and has shown superior antiinflammatory and antioxidant effects in many studies, especially cancer studies. In this study, we examined the treatment efficacy of 20 (S)-ginsenoside Rg3 on GU. Methods: Three kinds of GU models, including an alcohol GU model, a pylorus-ligated GU model, and an acetic acid GU model, were used. Mouse endothelin-1 (ET-1) and nitric oxide (NO) levels in blood and epidermal growth factor (EGF), superoxide dismutase, and NO levels in gastric mucosa were evaluated. Hematoxylin and eosin staining of gastric mucosa and immunohistochemical staining of ET-1, inducible nitric oxide synthase (NOS2), and epidermal growth factor receptors were studied. Ulcer index (UI) scores and UI ratios were also analyzed to demonstrate the GU conditions in different groups. Furthermore, Glide XP from $Schr{\ddot{o}}dinger$ was used for molecular docking to clarify the interactions between 20 (S)-ginsenoside Rg3 and EGF and NOS2. Results: 20 (S)-ginsenoside Rg3 significantly decreased the UI scores and UI ratios in all the three GU models, and it demonstrated antiulcer effects by decreasing the ET-1 and NOS2 levels and increasing the NO, superoxide dismutase, EGF, and epidermal growth factor receptor levels. In addition, high-dose 20 (S)-ginsenoside Rg3 showed satisfactory gastric mucosa protection effects. Conclusion: 20 (S)-ginsenoside Rg3 can inhibit the formation of GU and may be a potential therapeutic agent for GU.

        Evaluation of 20(S)-ginsenoside Rg3 loaded hydrogel for the treatment of perianal ulcer in a rat model

        Background: As a kind of common complication of the surgery of perianal diseases, perianal ulcer is known as a nuisance. This study aims to develop a kind of 20(S)-ginsenoside Rg3 (Rg3)-loaded hydrogel to treat perianal ulcers in a rat model. Methods: The copolymers PLGA<sub>1600</sub>-PEG<sub>1000</sub>-PLGA<sub>1600</sub> were synthesized by ring-opening polymerization process and Rg3-loaded hydrogel was then developed. The perianal ulcer rat model was established to analyze the treatment efficacy of Rg3-loaded hydrogel for ulceration healing for 15 days. The animals were divided into control group, hydrogel group, free Rg3 group, Rg3-loaded hydrogel group, and Lidocaine Gel® group. The residual wound area rate was calculated and the blood concentrations of interleukin-1 (IL-1), interleukin-6 (IL-6), and vascular endothelial growth factor (VEGF) were recorded. Hematoxylin and eosin (H&E) staining, Masson's Trichrome (MT) staining, and tumor necrosis factor α (TNF-α), Ki-67, CD31, ERK1/2, and NF-κB immunohistochemical staining were performed. Results: The biodegradable and biocompatible hydrogel carries a homogenous interactive porous structure with 10 ㎛ pore size and five weeks in vivo degradation time. The loaded Rg3 can be released sustainably. The in vitro cytotoxicity study showed that the hydrogel had no effect on survival rate of murine skin fibroblasts L929. The Rg3-loaded hydrogel can facilitate perianal ulcer healing by inhibiting local and systematic inflammatory responses, swelling the proliferation of nuclear cells, collagen deposition, and vascularization, and activating ERK signal pathway. Conclusion: The Rg3-loaded hydrogel shows the best treatment efficacy of perianal ulcer and may be a candidate for perianal ulcer treatment.

        Circ-MAN1A2 Contributes to the Acquired Resistance of Gefitinib by Binding to miR-409-3p to Induce TWIST1 Expression in Non-small-cell Lung Cancer

        Gefitinib has been widely used as a firstgeneration epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor for non-small-cell lung cancer (NSCLC) patients with EGFR mutation. In this study, we explored the key molecules responsible for acquired Gefitinib resistance in NSCLC cells. 3-(4,5-Dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide assay was performed to analyze the Gefitinib resistance and proliferation ability of NSCLC cells. Cell proliferation was also assessed by colony formation assay. Flow cytometry was performed to analyze cell apoptosis. Wound healing assay and transwell invasion assay were performed to the migration and invasion abilities of NSCLC cells, respectively. The target relationship between microRNA-409-3p (miR-409-3p) and circular RNA mannosidase alpha class 1A member 2 (circ- MAN1A2) or twist family bHLH transcription factor 1 (TWIST1) was verified by dual-luciferase reporter assay and RNA immunoprecipitation assay. Circ-MAN1A2 level was markedly elevated in Gefitinib-resistant NSCLC cell lines and tissues. Circ-MAN1A2 interference sensitized Gefitinib-resistant NSCLC cells to Gefitinib. Furthermore, circ-MAN1A2 interference suppressed the proliferation, migration and invasion and promoted the apoptosis of Gefitinib-resistant NSCLC cells. Circ-MAN1A2 overexpression negatively regulated miR-409-3p level by directly binding to it. miR-409-3p silencing partly counteracted circ-MAN1A2 silencing-mediated anti-tumor effects in Gefitinib-resistant NSCLC cells. TWIST1 was a target of miR-409-3p, and miR-409-3p overexpression-induced antitumor effects in Gefitinib-resistant NSCLC cells were partly reversed by TWIST1 overexpression. Circ-MAN1A2 silencing aggravated Gefitinib-mediated inhibition of tumor growth in vivo. In conclusion, circ-MAN1A2 facilitated the acquired resistance of Gefitinib and other malignant behaviors of NSCLC cells through mediating miR-409-3p/ TWIST1 axis.

        Genome-wide single-nucleotide polymorphism data and mitochondrial hypervariable region 1 nucleotide sequence reveal the origin of the Akhal-Teke horse

        Objective: The study investigated the origin of the Akhal-Teke horse using genome-wide single-nucleotide polymorphism (SNP) data and mitochondrial hypervariable region 1 (HVR-1) nucleotide sequences Methods: Genome-wide SNP data from 22 breeds (481 horses) and mitochondrial HVR-1 sequences from 24 breeds (544 sequences) worldwide to examine the origin of the Akhal-Teke horse. The data were analyzed using principal component analysis, linkage disequilibrium analysis, neighbor-joining dendrograms, and ancestry inference to determine the population relationships, ancestral source, genetic structure, and relationships with other varieties. Results: A close genetic relationship between the Akhal-Teke horse and horses from the Middle East was found. Analysis of mitochondrial HVR-1 sequences showed that there were no shared haplotypes between the Akhal-Teke and Tarpan horses, and the mitochondrial data indicated that the Akhal-Teke horse has not historically expanded its group. Ancestral inference suggested that Arabian and Caspian horses were the likely ancestors of the Akhal-Teke horse. Conclusion: The Akhal-Teke horse originated in the Middle East.

        Criteria for evaluating working fluids in loop gravity-assisted heat systems

        A loop gravity-assisted heat pipe (LGHP) is characterized by passive heat dissipation and extensive application prospect. Dissimilar working fluids give rise to noticeable differences in their working performance (Critical heat flux (CHF)). Consequently, it is of great significance to evaluate the performance of LGHP working fluid for its design. In accordance with the basic theories of CHF and hydrodynamics, the pressure drop models of laminar, smooth turbulence, and completely rough turbulence flows were established, and an extensible two-phase composite property parameter was obtained. On this basis, the physical property parameter of transitional rough turbulent flow was also derived. Finally, as persuasively illustrated by the experimental results, there was almost a linear association between CHFs and the derived physical parameter. As a result, the physical parameters derived in this paper can be employed as favorable criteria for the selection of LGHP working fluid.
